[Characteristics of acute and chronic intoxication induced by rocket propellant nitrogen tetroxide].

OBJECTIVE: To investigate the characteristics of acute and chronic injuries of the nitrogen tetroxide, a kind of propellant of rocket. METHOD: 128 male rats were divided randomly into 4 groups: acute control group (56), acute nitrogen tetroxide intoxication group (56), long-term response group (8). The animals were killed sequentially at 3, 6, 12, 24, 48 and 72 h. Nitrogen tetroxide was administrated through inhalation at the concentration of 81 mg/m3 for 15 min. Chronic injuries and pathologic changes were also observed one year after the intoxication. RESULT: Pulmonary edema was the main pathological changes after intoxication, complicated with partial haemorrhage. Data acquired from long-term observations showed 75% pulmonary fibrosis and one case of adenocarcinoma of lung. CONCLUSION: The first 2 to 6 h after intoxication is the most severe stage of the injury. During the long-term observation, we find that intoxication with nitrogen tetroxide can induce pulmonary fibrosis and adenocarcinoma.

[Determination of alpha-fetoprotein in the serum of pregnant women by rocket-linear immunoelectrophoresis].

Rocket-linear immunoelectrophoresis enabled to estimate the alpha-fetoprotein content in blood serum of pregnant women. Sensitivity of the procedure constituted as low as 13-22 ng of the protein per I ml of blood serum. Concentration of the protein in blood serum depended on the period of pregnancy; middle values of normal content of alpha-fetoprotein in blood serum were estimated within various periods of pregnancy in women of the district studied. The procedure might be used simultaneously with immunoenzymatic and radioimmune methods in prenatal diagnosis of impairments in development of the fetus neural tube.

Transection of the first portion of jejunum from blast injury in accidental discharge of a (2.75-inch aircraft) rocket from an F-15.

Injury to the small intestine and its mesentery, while common with penetrating abdominal wounds, is relatively uncommon in patients who have received nonpenetrating abdominal trauma. Review of larger series indicates that small bowel injuries occur in only 5-10% of trauma patients. The focus of this paper is: 1) to present a case of transected jejunum and retroperitoneal hematoma incurred through a blast injury from accidental discharge of a 2.75-inch folding fin aircraft rocket from an F-15; and 2) to discuss pathogenesis, diagnosis, and treatment that apply specifically to injuries of the small intestine exclusive of the duodenum.

[Quantitation of human serum apolipoprotein E by Rocket immuno-electrophoresis].

Apo E was purified from the very low density lipoprotein (VLDL) of normal and hypertriglyceridemic subjects by Sephadex G-75, heparin-Sepharose 4B affinity chromatography, and preparative SDS polyacrylamide gel electrophoresis. The antisera were produced by injecting apo E into goats. With apo E antisera, Rocket immuno-electrophoretic method was developed to quantitate apo E levels in normal and hypertriglyceridemic human serum. The detection limit was 5.5 ng apo E. The coefficients of variation of intra- and interassay were 2.6% and 5.2%, respectively. The serum apo E level of 50 normal lipodemic subjects in Chengdu area was 4.7 +/- 1.2 mg/dl.

Experiences of rocket seat ejections in the Swedish Air Force: 1967-1987.

From 1967 to 1987 there were 83 successful and 9 fatal ejections with Saab rocket seats in the Swedish Air Force. Medical consequences and injury factors are reviewed. Thirty-nine survivors had nontrivial injuries; four were seriously injured. The risk for injury has been approximately 50% throughout the period. Two-thirds of the survivors resumed flying within a week, the remainder after up to 1 year, except for three pilots who terminated their flying status.

(Radio) rocket immunoelectrophoresis a useful screening method for house dust extracts.

House dust (HD) extracts prepared from HD collected in households from West Germany, USA, and Spain were investigated by (radio) rocket immunoelectrophoresis. By pouring agarose gels containing different antisera side by side in strips onto a glass plate, the antigen/allergen components of HD extracts could be detected simultaneously in one electrophoretic separation. In addition to mite and animal dander, antigens/allergens of pollens, mold and food (ovalbumin and cow serum) could be detected in most of the extracts.

[Intra-orbital explosion of a rocket].

Orbito-palpebral ballistic wounds are relatively common, most often due to firearms, and raise the problem of their surgical management. Excision of the intra-orbital foreign bodies remains difficult, above all when there is impaction of the anterior portion. The authors report a model case of orbital trauma by rocket splinter, excision of which was possible via the entry orifice.

Simultaneous quantification by double rocket immunoelectrophoresis of apolipoproteins A-I and B in blood spotted on filter paper.

We describe double rocket immunoelectrophoresis for simultaneous quantification of apolipoprotein A-I (apo A-I) and B (apo B) in blood on filter paper. The apolipoproteins from blood spots on filter paper were eluted with detergents (sodium dodecyl sulfate and Triton X-100). The eluates were subjected to electrophoresis on agarose gel containing antisera against both apolipoproteins. Within- and between-assay CVs for apo B/A-I ratios were less than 5.5% and 7.2%, respectively. The apo B/A-I ratio was influenced by length and temperature of storage. In results for 121 venous blood samples, the apo B/A-I ratios in dried blood spots correlated well with those in serum (r = 0.92) and correlated somewhat with the ratios for low-density lipoprotein/high-density lipoprotein cholesterol in serum (r = 0.87). Of these specimens, 68 were from patients with known familial hypercholesterolemia, all of whom had an apo B/A-I ratio greater than 0.90. We think this method will be of value for detecting familial hypercholesterolemia and possibly familial hyperapobeta- and hypoalphalipoproteinemia.

[Determination of the monospecificity of antibodies to different forms of cytochrome p-450 using rocket immunoelectrophoresis].

Antibodies against different forms of cytochrome P-450 were analyzed, using rocket immunoelectrophoresis technique. The technique was found capable of revealing antibody monospecificity. The antibodies against a certain form of cytochrome P-450 may be regarded as monospecific when they form a single precipitation line during the analysis of microsomal preparations which contain a mixture of different forms of cytochrome P-450.

The assay of influenza virus structural antigens in vaccines by rocket immunoelectrophoresis.

The internally situated influenza virus structural antigens (matrix and nucleoprotein) and the glycoprotein hemagglutinin were assayed using rocket immunoelectrophoresis or single radial diffusion following disruption of purified virus with 1% w/v sodium sarcosyl detergent. Electrophoresis was carried out for 1-4 h in 0.05 M barbitone buffer at pH 8.6. The potency estimations of HA and M activity were similar for the two methods for bivalent beta-propiolactone inactivated whole virus vaccines.

[Isolation and quantitative determination of apoprotein C-11 in the blood using rocket immunoelectrophoresis].

Apoprotein C-II was isolated from human blood plasma by means of column chromatography-gel filtration and DEAE ion exchange chromatography by modified method of Brown et al (1969). Production of antiserum to apoprotein C-II as well as a procedure for rocket immunoelectrophoresis are described for estimation of the apoprotein in blood plasma of men and women of various age, exhibiting different lipid spectra. Content of the apoprotein C-II in blood plasma, estimated by the immunoelectrophoretic procedure, was higher in men as compared with women and correlated with concentration of triglycerides in blood plasma.

Low-density-lipoprotein apoprotein B in plasma as measured by radial immunodiffusion and rocket immunoelectrophoresis.

Radial immunodiffusion (RID) and rocket immunoelectrophoresis (RIE) are compared with respect to determination of LDL-bound apo B in plasma. Isolated VLDL could not enter a 15 g/L agarose gel when either technique was used. However, in the presence of plasma proteins, migration of VLDL into agarose was enhanced. Only when plasma samples were kept frozen before the assay was plasma VLDL unable to enter the agarose gel when RID was used. With RIE the contribution of plasma VLDL to the apo B determination under these conditions was not always negligible. Besides enhancing the entry of VLDL into the agarose, the presence of proteins also influences apo B immunoreactivity of LDL and VLDL. For measuring LDL-bound apo B directly in unfractionated plasma we recommend: (a) RID in 15 g/L agarose gel; (b) freezing the plasma samples before assay; (c) diluting the plasma samples in saline supplemented with protein in the same concentration as is present in plasma (70 g/L); and (d) using plasma as the assay standard.

[The background of the serological cross-reaction between Yersinia and Brucella and the possibility of differential diagnostics by the ELISA technique and the electroimmuno assay (rocket electrophoresis) (author's transl)].

A strong serological cross-reaction appears between different species of genus Brucella and Yersinia enterocolitica (Y.e.) serotype 0--9, which seriously complicates the diagnostic works of brucellosis and yersiniosis both in humans and animals. This cross-reaction makes it impossible to perform a differential serological diagnosis using common routine procedures such as the agglutination test and the complement fixation test. By the use of immunological, immunochemical and structural chemical methods it has been shown that the common antigenic structures of Brucella and Yersinia are situated in the lipopolysaccharides (LPS) of the cellwalls. In order to do structure analysis gaschromatography in combination with mass spectrometry of LPS from Brucella and Yersinia bacteria has been achieved. The results show that two monosaccharides (glucose and galactose) constitute common components of the LPS-molecules. A discovered cross-reaction between Y.e. 0--9 and Salmonella urbana (0--30) confirms the hypothesis that glucose and galactose are responsible for the serological cross-reaction between Brucella and Y.e. 0--9. An enzyme-linked immunosorbent assay (ELISA) has been developed in order to be used as a differential routine test. The results show that by using ELISA, a differentiation between antibodies against Y.e. 0--9 and B. abortus can be done with high sensitivity and accuracy. Another differential diagnostic system has been developed by the use of electroimmuno assay (rocket electrophoresis). This method raises the possibility of a rapid and a simple qualitative differentiation of antibodies against Brucella and Yersinia.

Quantitation of C2 by rocket immunoelectrophoresis in 120 pathological sera.

A potent monospecific anti-C2 was used for quantitation of serum C2 by rocket immunoelectrophoresis (RIE). This ready procedure was compared to the one-step hemolytic titration utilizing C2-deficient human serum. RIE detected 4 nanograms C2 in 5 microliters samples. Using highly purified C2 as a reference, C2 concentration in pooled normal human sera was estimated around 50 micrograms/ml by both test systems. The reliability of RIE was assessed when immunochemical and hemolytic determinations were comparatively performed in 120 pathological sera. The correlation coefficient was significant (r = 0.69) and only 8 sera were found outside the expected range, exhibiting an abnormally high C2 protein. They were obtained from 6 patients with cryoglobulinemia associated connective tissue disease and 2 with acute glomerulonephritis. These occasional findings suggested the possibility of an activation of C2 hemolytic activity without simultaneous loss of C2 protein. In 16 sera of individuals with familial C2 deficiency (r = 0.83) and 29 sera of patients with angioneurotic edema (r = 0.72), RIE provided an extremely simple and reliable alternative to the time consuming hemolytic titration of C2.

[The use of rocket immunoelectrophoresis for determining glycoprotein in concentrated rabies vaccines].

Rocket immunoelectrophoresis (RIE) was shown to be useful for the evaluation of glycoprotein (GP) content in concentrated rabies vaccines, and disintegron B., a zwitterionic detergent made in this country, for treatment of the vaccines for these evaluations. The values of GP content obtained by RIE and single radial immunodiffusion test were similar. The highest values were obtained for all the vaccines tested with anti-PM GP serum compared with anti-ERA and Vnukovo-32 GP serum. When anti-PM and anti-Vnukovo-32 GP sera were used for RIE two components were revealed in the vaccines. All the preparations under study contained soluble GP.
