Simultaneous identification and quantitation of diphenhydramine and methaqualone.

Methods for the simulataneous identification and quantitation of diphenhydramine and methaqualone in urine have been investigated. Suitable thin-layer and gas chromatographic techniques have been developed. The extraction techniques were evaluated by extracting human urine after the ingestion of therapeutic dosages of the two drugs. A charcoal cartridge technique proved to be superior to direct solvent extraction. Although enzyme hydrolysis gave slightly better recoveries than the charcoal cartridge technique, it is a time-consuming procedure.

Methaqualone discrimination in gerbils: interactions with bemegride and imidazoline (DH-524).

Gerbils were trained in a T-maze to discriminate methaqualone (MTQ), either 16 mg/kg, 12 mg/kg, 8 mg/kg, or 4 mg/kg, from the no drug condition. Discrimination was proportional to dose, MTQ 16 mg/kg vs. no drug being the most rapidly discriminable condition. Challenge of MTQ discrimination with bemegride and imidazoline showed that bemegride, but not imidazoline reversed the MTQ discrimination. The median effective dose of bemegride was related to the amount of MTQ used (16-8 mg/kg).

Metabolite sensitivity in the methaqualone radioimmunoassay.

Recent literature reflects the increased use of RIA as a qualitative and quantitative tool. This study exemplifies the problems that may arise when this technique is used for quantitation. Caution must be exercised, and a physiological and toxicological understanding of each particular drug is essential to provide credible results. It is evident that the sensitivity and ease of the RIA method for methaqualone makes it invaluable as a screening test, but confirmation and quantitation of this drug must remain with those methods that can quantitate each metabolite.

[In vitro and in vivo photochemical reactivity of methaqualone-1-oxide].

Methaqualone-1-oxide (1) exhibits photochemical reactivity. By irradiation of 1 with solar light the oxaziridin 3 is formed at first, which reacts in vitro (human proteins) and in vivo (rats) with macromolecules. As result of the photochemical in vitro and in vivo reactions of 1 the photoproduct 2-acetamidobenzoic acid-2'-methylanilide (6), involved in oxidation of protic compounds, was detected and after hydrolysis of proteins it appears that the short-lived 3 was adding to proteins.

The isolation and identification of precursors and reaction products in the clandestine manufacture of methaqualone and mecloqualone.

Abuse of the hypnotic quinazolinone is well recognized and increasing. Clandestine laboratories producing methaqualone (2-methyl-3-ortho-tolyl-4(3H) quinazolinone) and mecloqualone (2-methyl-3-ortho-chlorophenyl-4(3H) quinazolinone) have been discovered throughout the United States. These laboratories utilize one of many synthesis routes to produce the illicit quinazolinone. Frequently, the clandestine chemist has little, if any, formal education in chemistry; does not keep notes; and does not label flasks and beakers containing solutions. The forensic chemist may be asked to analyze unmarked reaction mixtures that were seized in a clandestine laboratory raid. As a result, a rapid method of isolation and identification of the precursors and products of such a mixture is presented.

[Studies on the kinetics of methaqualone during the late stage of pregnancy (author's transl)].

After oral administration of 0.4 g of methaqualone to pregnant and non-pregnant women, the authors stated that the half-life of this drug was significantly reduced during the last trimester of pregnancy. The two experimental groups showed no significant differences in the volume of distribution and the fictitious initial concentration. The commonly used therapeutical dosages are not likely to produce increased blood levels during normal pregnancy.

[Studies on the kinetics of methaqualone in the course of labour (author's transl)].

A 0,4 g (1598,06 mumol) dose of methaqualone was administered orally to 19 parturient at the beginning of the first stage of delivery. In comparison with former results, got from a control-group of nonpregnant and pregnant women in the last third of pregnancy, it was found a significantly prolonged biological half-life and a decrease of the fictitious initial concentration.

Regional localization of methaqualone in rat brain.

Distribution of orally administered methaqualone-14C (25 mg/kg) in several rat brain areas was examined. Peak concentration attained at 1.5 hr. Among brain areas, highest concentration was measured in pineal gland (8.2 microgram/g). MTQ levels in the cortex, pituitary, hypothalamus and hippocampus ranged from 3 to 3.5 microgram/g, those in the mid-brain, medulla-pons, striatum and cerebellum from 2 to 2.5 microgram/g. Significant amounts of MTQ in all brain regions were still present at 3 hr. These results indicate that MTQ was somewhat uniformly distributed in various brain regions as a result of its lipophilic nature.

Age-dependent variations in the distribution of methaqualone in rat.

Rats at 1, 4, 8, 12, 20 and 60 postnatal days received orally 5 mg/kg 14C-methaqualone (MTQ) and were sacrificed at 0.5, 1.5 and 6 h. Several tissues were analyzed for unchanged MTQ. In most cases, concentrations reached peak levels at 1.5 h. Highest concentrations were found in the liver followed by the lung, spleen, heart, kidney, eye, and brain. Levels of MTQ were higher in all organs of 1- to 12-day-old rats at all time intervals than the corresponding organs of 20- and 60-day-old rats, apparently resulting from initial higher accumulation and slower rate of disappearance. The 1.5-hour brain:plasma concentration ratio in 8-day-old rats was 0.82 and progressively declined to 0.27 in 60-day-old rats; a similar trend with respect to changes in concentration ratio also occurred for the eye, spleen, kidney, and heart. The fact that the age-related reduction in tissue:plasma concentration ratios occurred in several tissues including brain suggests generalized decreases in tissue accumulation of MTQ. The reduction in brain:plasma concentration ratios, therefore, cannot be ascribed to the blood-brain barrier which develops progressively with the advancing age.

[The problem of polyneuropathy in methaqualone medication].

Thirteen years after the introduction of methaqualone into therapy, investigators (namely, Finke, Spiegelberg, and Suchenwirth), by reference to individual cases, pointed out the possible development of polyneuropathies. This has not so far been confirmed despite worldwide use of the drug. Experiments on rats, which were conducted by the present authors, show a statistically significant decrease in maximum motor conduction speed, which is believed to be functional disorder. Clinical symptoms or nervous structural variations in the sense of polyneuropathy could not be observed.

The analysis of illicit methaqualone containing preparations by gas chromatography-mass spectrometry for forensic purposes.

A validated gas chromatographic-mass spectrometric method for quantitative analysis of methaqualone (MTQ) in illicit preparations is reported. The method proved to have a coefficient of variation of below 5%. Four batches of seized tablets, two pairs with similar imprints, were analyzed. It was found that the average MTQ concentration in all four batches of tablets differed significantly (p = 0.01) rendering it impossible to conclude that, on the basis of MTQ concentration alone, the batches with a similar logo originated from the same manufacturer or manufacturing batch. Conversely, it can be said that in this case, the four batches originated from either different clandestine laboratories or manufacturing batches.
