Outgrowth of interleukin-2-dependent T cell lines with cytotoxic activity against mesothelial cells from cultures of peritoneal effluent cells of children on CPD.

Previous studies on the peritoneal immune system described the presence of activated T lymphocytes in peritoneal effluents (PE) from patients on chronic peritoneal dialysis (CPD), and showed that mesothelial cells (MC) can present antigens to T cells. In order to better define phenotypic and functional characteristics of T cells and their interactions with MC, we isolated PE cells from 15 children. At the immunophenotypic analysis, high percentages of activated T cells were identified (mean value: 15% double staining for CD3/DR; 12% CD25+). T cells with gamma/delta T cell receptor (mean 5%) and natural killer cells (mean 17%) were also present in elevated numbers. MC lines (n = 7) and interleukin-2-dependent T cell lines (9 CD4+; 1 CD8+) were also obtained by incubating PE cells under different conditions. Two cell lines showed a major histocompatibility complex (MHC) restricted cytotoxic activity against autologous MC; two lines killed allogeneic MC; one line killed both autologous and allogeneic MC. Although the hypothesis that activated T cells could kill MC after recognition of surface structures modified by dialysis fluid, or during antigen presentation, needs to be further investigated, our data suggest that the subsets of lymphocytes we identified could play an important role in the mechanisms of peritoneal membrane defense.

Chorioretinopathy and pituitary dysfunction. The CPD syndrome.

Four reports of a rare syndrome characterized by severe, early-onset chorioretinopathy, trichosis, and evidence of pituitary dysfunction are reviewed. Recently obtained follow-up information about these four patients is presented. A new example of this disease, the fifth case to be reported, to our knowledge, is described.

Autosomal recessive Oliver-McFarlane syndrome: retinitis pigmentosa, short stature (GH deficiency), trichomegaly, and hair anomalies or CPD syndrome (chorioretinopathy-pituitary dysfunction).

We describe a brother and sister with retinitis pigmentosa (RP), growth failure, long eyelashes, and sparse hair. They were born to young healthy consanguineous parents and presented at birth with IUGR. Evolving pigmentary retinopathy was diagnosed at the age of 5 years. A similar condition (Oliver-McFarlane) syndrome was reported previously. Our two sibs confirm the existence of this autosomal recessive syndrome.

Histamine-releasing effect of a corticotrophin derivative. II. Mechanism of action of histamine release by C 44 680-Ba, compared with that of Cpd. 48/80, dextran and triton.

The mechanism of the histamine-liberating action of the synthetic polypeptide C 44 680-Ba, an alkyl-prolyl derivative of beta 1-19 corticotrophin, was investigated and compared with those of Compound 48/80, dextran, Melittin and Triton X-100. It was found that the release of histamine from rat peritoneal cells induced by the polypeptide is dependent on temperature, pH, calcium ions and energy-providing processes. In regard to these criteria, the mode of action of this histamine liberator resembles that of Compound 48/80 but is quite distinct from that of the unspecific substance Triton X-100.

Shall red cell units stand upright, lie flat or be mixed during storage? In vitro studies of red cells collected in 0.5 CPD and stored in RAS2 (Erythrosol).

Red cells were prepared using a new anticoagulant with half the normal amount of citrate and a new additive solution (RAS2, Erythrosol) previously shown to give improved storage conditions, and stored in a highly gas permeable plastic container (PL 2209). Mixing daily and weekly resulted in lower PCO2, higher PO2 and more rapid oxygen saturation of the haemoglobin than storage unmixed in an upright position. Storage horizontally in a lying position unmixed resulted in similar blood gas values as with mixing. The haemolysis was lowest in units mixed once weekly: 0.21 +/- 0.09% after 28 days and 0.26 +/- 0.06% after 56 days. The morphology was better maintained in mixed than in unmixed units. Horizontal storage and mixing once per week seem to be optimal as judged from these in vitro studies.

PAH Growth from the pyrolysis of CPD, indene and naphthalene mixture.

In this study, the addition of cyclopentadienyl (CPDyl) moieties to aromatic rings has been investigated experimenNaphthalene was selected as the representative aromatic compound and its pyrolytic reactivity was studied first to obtain background information for the pyrolysis of cyclopentadiene-indene-naphthalene mixture. The experiments were conducted in a 5 s laminar flow reactor over the temperature range of 700-850 degrees C with 50 degrees C increments. PAH growth from naphthalene pyrolysis is mainly attributed to aryl-aryl addition of naphthyl radicals and naphthalene fragmentation, with lower product formation rates comparing with hydrocarbons with CPDyl moieties. The results indicate that naphthalene is less reactive than the CPDyl containing hydrocarbon radical-molecule addition of CPDyl radicals to naphthalene results in phenanthrene, which can also be formed substantially from other growth pathways. This addition occurs mainly at low temperatures and is less favored due to the competition from more reactive indenyl and CPDyl radicals. CPDyl radical addition to naphthalene exhibits limited aromatic growth due to the aromaticity restrictions of naphthalene. The studies of hydrocarbons with and without CPDyl moieties suggest that the reaction pathways of CPDyl bearing hydrocarbons are different from those without these moieties and cannot be adequately accounted for by the existing acetylene addition and aryl-aryl addition mechanisms.

Combined PI-DAPI staining (CPD) reveals NOR asymmetry and facilitates karyotyping of plant chromosomes.

This paper presents a preparative and staining procedure for plant mitotic chromosomes that uses a combination of PI (propidium iodide) and DAPI (4',6-diamidino-2-phenylindol) and which reveals a pattern of high-affinity regions for these fluorochromes. Nucleolar organiser regions (NORs), telomeres and centromeric regions exhibit high PI affinity (red), whereas other chromosomal regions exhibit high affinity for either PI (red) or DAPI (blue). NOR-bearing and other chromosomes are readily distinguished, facilitating karyotyping. The dual staining pattern was observed in all the plants tested. Aspects of NOR size, number and occurrence are discussed. A karyotype of rice metaphase chromosomes is presented, based on their fluorescent banding patterns.

Verifiable CPD paper: introduction, history of lasers and laser light production.

The word laser conjures in the mind's eye many aspects of what might be described as 'modern' life. The words 'powerful', 'precise' and 'innovative' complement our conception of the world in terms of technology, whereas patients often associate the words 'magical' and 'lightening quick' with the use of lasers in medical practice. The purpose of this series of articles is to explore the history and development of lasers, the integration of lasers into clinical dentistry and the safeguards as to their regulated use.

Verifiable CPD paper: factors that influence the dental attendance pattern and maintenance of oral health for people with multiple sclerosis.

OBJECTIVE: To determine the impact of multiple sclerosis (MS) on patient attendance at dental practices and maintenance of oral health. DESIGN: A cross-sectional postal questionnaire-based study. SETTING: Leicestershire, United Kingdom. SUBJECTS AND METHODS: People with MS in Leicestershire identified from local health authority records (n = 476). MAIN OUTCOME MEASURES: Number registered at dental practice, frequency of attendance, issues and perspectives relating to attendance and maintenance of oral health. RESULTS: A response rate of 61% (n = 289) was obtained. When compared to the general population, a higher number of people with MS were registered with a dentist (49%:88%) and displayed more frequent practice attendance (71%:81%) in the past year. People with MS reported difficulties in attending a dentist and maintaining oral health, which were exacerbated by deterioration in general health. Problems relating to reduced personal mobility had the greatest impact on attendance. CONCLUSIONS: MS has a negative impact on perceived patient attendance and maintenance of oral health. Patients with a progressive disability could benefit greatly from the provision of preventive oral health care. The importance of seeking care earlier rather than later needs to be emphasised to both professionals and patients alike. Further efforts are required to increase awareness of the importance of oral health to the quality of life of people with MS and ensure that individuals with physical disabilities receive the same access to dental services as the able-bodied.
