BoLA antigens are associated with increased frequency of persistent lymphocytosis in bovine leukaemia virus infected cattle and with increased incidence of antibodies to bovine leukaemia virus.

The association between bovine major histocompatibility system (BoLA) type and persistent lymphocytosis in cattle with antibodies to bovine leukaemia virus was examined by comparing antigen frequencies in cattle with persistent lymphocytosis to controls matched for age, sex, breed and presence of antibodies to BLV. The cattle came from nine dairy herds in south-east Queensland, Australia; six herds were Australian Illawarra Shorthorn (AIS), two herds were Jersey and one herd was Friesian. Antigen W6 and Eu28R were more common in cattle with persistent lymphocytosis than in controls. Antigen W8 was less common in AIS cattle with persistent lymphocytosis. A study of 24 offspring from one sire, heterozygous for W10 and Eu28R, showed that offspring inheriting Eu28R from the sire were significantly more likely to have antibodies to BLV than offspring inheriting the opposing W10 haplotype.

Partial nucleotide sequence of a novel bovine major histocompatibility complex class II beta-chain gene, BoLA-DIB.

A bovine genomic clone that hybridized to HLA-DQ beta cDNA was isolated and fragments containing the beta 1, beta 2 and transmembrane (TM) exons subcloned. The nucleotide sequences of the exons and flanking intron regions were determined. Comparisons of these exon nucleotide sequences and derived amino acid sequences to human class II beta-chain sequences showed that this gene is only 77% identical to HLA-DQ beta and about 75% identical to bovine DQ beta-like genes. The exon sequences were more divergent from other class II beta-chain genes. However, structural features such as conserved cysteines and regions of amino acids strongly suggest this to be a class II beta-chain gene. When exon-containing fragments were used as hybridization probes on Southern blots of bovine genomic DNA digested with Eco RI or Pvu II, each exon hybridized to a single band. Based on these results we have referred to this gene as a novel bovine class II beta-chain gene, BoLA-DIB.

A bovine monoclonal antibody detecting a class I BoLA antigen.

The bovine IgG1 monoclonal antibody (mAb) ILA70 was made by immunizing a calf with peripheral blood mononuclear cells (PBM) from a BoLA-w10 homozygous heifer and subsequently fusing lymphocytes from the local lymph-node with the heterohybridoma 53B3. Family and population studies, antibody binding inhibition and immunoprecipitation of the target antigen all indicate that ILA70 detects a polymorphic epitope on a bovine class I major histocompatibility complex (MHC) molecule. The antibody is complement fixing and so may be used in a standard cytotoxicity assay. Ascitic fluid with antibody activity many times greater than that of the tissue-culture supernatant has been prepared in nude mice. The antibody-producing heterohybridoma has been subcloned three times and appears to be stable. Such heterohybridomas may prove to be a valuable source of particularly discriminating and informative mAbs for the serological analysis of the products of the bovine MHC.

Absorption analysis of boLA sera.

BoLA specificities w1 w2 w5 w8 w9 w10 and w11 have been re-examined by absorption. All of the clusters except w9 showed evidence of sub-group specificities, but only in some cases (w1, w5, w8 and w11) is it suggested that the definition should be revised. The results show a very high degree of reproducibility between sera from different laboratories with clusters of narrow and wide sera for most specificities. These behave in all the cases reported here as linear sub groups or inclusion groups.

[The major histocompatibility complex of West African cattle. Typing of lymphocyte antigens (BoLA) of Baoulé cattle (Bos taurus) and Sudanese zebus (Bos indicus) of Burkina Faso (West Africa)].

Lymphocyte antigen (BoLA) typing on 247 Baoule cattle (Bos taurus) and 106 Sudanese zebus (Bos indicus), allowed us to determine gene frequencies of 43 class 1 specificities, as international "W" and European "EU", as African local from Kenya "KN" and Burkina Faso "BF". In comparing these frequencies, it appears that some specificities could be considered as significant breed markers for Baoule cattle and zebus.

[Frequent occurrence of BoLA w16 in cows susceptible to mastitis].

The association between BoLA class I antigens and mastitis was studied in Boheman Pied breed and its crosses. The mastitis status was followed during two years according to three diagnostic criteria: California Mastitis Test, bacteriological findings and the altered taste of milk. The antigen w16 was found to be significantly associated with susceptibility to mastitis in all diagnostic tests. On the other hand there were some indications that the wll animals were rather resistant to mastitis.
