Metals and arsenic in soils and corresponding vegetation at Central Elbe river floodplains (Germany).

Floodplain soils at the Elbe river are frequently polluted with metals and arsenic. High contents of these pollutants were detected down to subsoil layers. NH4NO3-extractable (phytoavailable) Cd, Ni, and Zn were elevated in horizons with high acidity. Among five common floodplain plant species, Artemisia vulgaris showed highest concentrations of Cd, Cu, and Hg, Alopecurus pratensis of As and Phalaris arundinacea of Ni, Pb, and Zn. Relationships were weak between metal concentrations in plants and phytoavailable stocks in soil. As and Hg uptake seems to be enhanced on long submerged soils. Enrichments of Cd and Hg are linked to a special plant community composition. Grassland herbage sampled in July/August revealed higher concentrations of As (+122%), Hg (+124%), and Pb (+3723%) than in May. To limit harmful transfers into the food chain, low-lying terraces and flood channels revealing highest contaminations or phytoavailabilities should be excluded from mowing and grazing.

Screening and applying wheat microsatellite markers to trace individual Haynaldia villosa chromosomes.

Wheat (Triticum aestivum L.) microsatellite markers were screened for detecting Haynaldia villosa L. chromosomes introduced into wheat background. Two hundred and seventy six primer pairs mapped on 7 homeologous groups of wheat were used to amplify the gDNA of T. aestivum and H. villosa. The results showed that 148 of 276 microsatellite primers amplified polymorphic bands between common wheat cv. Chinese Spring and H. villosa. Primers wmc49 (1BS), wmc25 (2BS), gdm36 (3DS), gdm145 (4AL), wmc233 (5DS), wmc256 (6AL) and gwm344 (7BL) produced a specific polymorphic DNA fragment on chromosome 1V to 7V of H. villosa, respectively. In addition, gwm469 (6DS) detected a specific band on 2V; gdm107 (2DS) amplified a specific band on 6V. These microsatellite markers were effective in identifying individual H. villosa chromosomes in other T. aestivum-H. villosa chromosome addition, substitution and translocation lines involved in different H. villosa accessions and wheat backgrounds. Therefore, these chromosome-specific microsatellite markers could be used as molecular markers for detection of chromosomes of H. villosa in common wheat.

Fructans in crested wheatgrass leaves.

Crested wheatgrass is an important cool-season grass that has become naturalized in many semiarid regions of the western U.S. It provides ground cover and reduces soil erosion caused by water and wind. Additionally, crested wheatgrass produces important forage for livestock and wildlife on 6 to 8 million hectars of western rangeland. It is well adapted to semiarid cold desert regions because of its cool temperature growth and drought tolerance. Understanding the biosynthesis of fructans in crested wheatgrass is important because of their likely role in both cool temperature growth and drought tolerance. Recent research described a major gene (6-SFT) in crested wheatgrass that is involved in fructan biosynthesis. 1-kestotriose, the major DP3 fructan in crested wheatgrass, serves as the substrate for the two major DP4 fructans, 1&6-kestotetraose and 1,1-kestotetraose. The three major DP5 fructans are 1&6,1-kestopentaose, 1,1&6-kestopentaose and 1,1,1-kestopentaose. The major DP6 fructan is 1&6, 1&6-kestohexaose. We postulate that 1&6,1&6-kestohexaose is synthesized from the addition of a fructose to 1&6, 1-kestopentaose. This paper provides structures of the various DP 3, 4, 5 and 6 fructan types produced by crested wheatgrass and provides suggested biosynthetic pathways for all major fructan linkage types present.

Fine root distribution and persistence under field conditions of three co-occurring Great Basin species of different life form.

Fine roots of an annual grass, a perennial grass and a perennial shrub were examined. Based on life histories and tissue composition, we expected the greatest root persistence for the shrub and shortest for the annual grass. Roots were observed with minirhizotrons over 2 yr for number, length and diameter changes. A Cox proportional hazard regression correlated root persistence with soil water, depth, diameter and date of production. In 2001, grass roots had similar persistence times, but shrub roots had the shortest. In 2002, the annual had the longest median root persistence, the perennial grass intermediate and the perennial shrub had the shortest. All species responded similarly to the magnitude of seasonal precipitation; root numbers increased with favorable soil moisture and disappeared with drying; fewer, thinner roots at greater soil depths were found in the drier year (2001). Root persistence increased with soil moisture, diameter and earlier appearance in the spring. Plasticity in root morphology and placement was influenced by water availability, yet persistence was surprisingly contrary to expectations.

Molecular cytogenetic discrimination and reaction to wheat streak mosaic virus and the wheat curl mite in Zhong series of wheat--Thinopyrum intermedium partial amphiploids.

Thinopyrum intermedium (2n = 6x = 42, JJJsJsSS) is potentially a useful source of resistance to wheat streak mosaic virus (WSMV) and its vector, the wheat curl mite (WCM). Five partial amphiploids, namely Zhong 1, Zhong 2, Zhong 3, Zhong 4, and Zhong 5, derived from Triticum aestivum x Thinopyrum intermedium crosses produced in China, were screened for WSMV and WCM resistance. Zhong 1 and Zhong 2 had high levels of resistance to WSMV and WCM. The other three partial amphiploids, Zhong 3, 4, and 5, were resistant to WSMV, but were susceptible to WCM. Genomic in situ hybridization (GISH) using a genomic DNA probe from Pseudoroegneria strigosa (SS, 2n = 14) demonstrated that two partial amphiploids, Zhong 1 and Zhong 2, have almost the identical 10 Th. intermedium chromosomes, including four Js, four J, and two S genome chromosomes. Both of them carry two pairs of J and a pair of Js genome chromosomes and two different translocations that were not observed in the other three Zhong lines. The partial amphiploids Zhong 3, 4, and 5 have another type of basic genomic composition, which is similar to a reconstituted alien genome consisting of four S and four Js genome chromosomes of Th. intermedium (Zhong 5 has two Js chromosomes plus two Js-W translocations) with six translocated chromosomes between S and Js or J genomes. All three lines carry a specific S-S-Js translocated chromosome, which might confer resistance to barley yellow dwarf virus (BYDV-PAV). The present study identified a specific Js2 chromosome present in all five of the Zhong lines, confirming that a Js chromosome carries WSMV resistance. Resistance to WCM may be linked with J or Js chromosomes. The discovery of high levels of resistance to both WSMV and WCM in Zhong 1 and Zhong 2 offers a useful source of resistance to both the virus and its vector for wheat breeding programs.

A new species of Epichloë symbiotic with Chinese grasses.

Epichloë species are fungal symbionts (endophytes) of grasses, six European and four North American biological species in genus Epichloë have been described in previous researches. In this study we describe a new Epichloë species, Epichloë yangzii Li et Wang, found in natural symbioses with Roegneria kamoji native to China. We investigated the host specificity, morphology, interfertility tests and molecular phylogenetic evidences of this new species. The results indicated that E. yangzii is host specific and seedborne. Most morphological characteristics of this new species are typical in the genus. However differences are evident in several features including size of perithecia, asci and ascospores. In mating tests E. yangzii was not interfertile with E. elymi isolates from related hosts in genera Elymus. Phylogenetic relationships based on sequences of beta-tubulin gene (tub2) introns and translation elongation factor 1-alpha gene (tef1) introns showed that members of the new species grouped into exclusive clades with high bootstrap value.

[Prometaphase aberrations leading to the nuclei restitution].

The mechanism of restitution nuclei formation in male meoiosis of Triticum-Agropyrum F1 hybrid and monosomic wheat line Milturum 553 has been studied. Visualization of division spindle with Navashin's routine fixationle distribution method accompanied with acetocarmine-coloured squashed preparations revealed that a block in the kinetochore spindle thread formation, i.e. the absence of chromosome association with the division spindle, is the basic reason of restitution in these genotypes. On this background, the prometaphase spindle in abnormal meiosis of these lines demonstrates a number of vivid and inordinate aberrations revealing some regularities of spindle structure in the normal plant cell.
