Facile preparation of biodegradable chitosan derivative having poly(butylene glycol adipate) side chains.

Various modes are being explored for the construction of functional materials from nanoparticles. Despite these efforts, the assembly of nanoparticles remains challenging with respect to the requirement of multiple component organization on varying dimensions and length scales. The graft copolymers of chitosan with poly(butylene glycol adipate) (PBGA) were prepared due to the esterification reaction between PBGA and 6-O-succinate-N-phthaloyl-chitosan (PHCSSA) in the presence of toluene as a swelling agent. The graft copolymers are nanoparticles with the size of few hundred nanometers as observed from TEM. It is a potential method to combine chitosan with the hydrophobic synthetic polymers. The grafting reactions were conducted with various PBGA/PHCSSA feed ratios to obtain chitosan-g-PBGA copolymers with various PBGA contents. FT-IR, NMR, XRD, spectrofluorophotometer, and TEM were detected to characterize the copolymers.

Effects of DL-alpha-amino adipic acid on Müller cells in frog and chicken retinae in vivo: relation to ERG b wave, ganglion cell discharge and tectal evoked potentials.

In both frog and chicken, an intravitreal injection of DL-alpha-amino-adipic acid, (DL-alpha aaa) provoked a progressive depression and eventually the disappearance of the ERG b wave that was concomitant with severe damage to the Müller cells without any apparent damage to retinal neurons. Ganglion cell discharges as well as tectal evoked potentials were still recorded, i.e. a visual message was still generated in the retina and transmitted to the optic tectum, when the Müller cells had been damaged to as to provoke an abolition of the ERG b-wave. The whole of the drug-induced effects proved to be reversible.

Uptake and metabolism of novel biodegradable poly (glycerol-adipate) nanoparticles in DAOY monolayer.

A useful route for the development of antitumour therapies is by creating improved methods for delivering therapeutic agents to tumour cells or subcellular compartments and increasing retention of drugs within target cells. In this study, we have characterized nanoparticle (NP) uptake and metabolism by DAOY cells, a human medulloblastoma cell line. NPs were formed from a novel polymer, poly (glycerol-adipate) (PGA), containing Rhodamine B Isothiocyanate (RBITC) as a fluorescent marker. It was observed that the cellular uptake of NPs depends on the incubation time and the concentration of NPs in the culture medium. The studies of retention and metabolism of NPs within cells indicated that 1) faster degradation of NPs within cells compared with that in cell culture medium in vitro; 2) a small fraction of NPs were recycled back to the outside of cell, whereas most NPs entered endosomes and lysosomes; and 3) recycled NPs were re-taken up in the following 2 h incubation time. These studies thus suggested that PGA NPs could be used for localising therapeutic agents into cells, and could provide prolonged drug effects because of their long sustained release in physiological conditions and their rapid release when taken up into cells.

Di(2-ethylhexyl) adipate (DEHA) induced developmental toxicity but not antiandrogenic effects in pre- and postnatally exposed Wistar rats.

Di(2-ethylhexyl) adipate (DEHA) has replaced the phthalates in thin plasticized polyvinyl chloride films used for food packaging, mainly because some phthalates induce testis toxicity and antiandrogenic effects. A dose-range finding study followed by a dose-response/effect study in Wistar rats investigated whether pre- and postnatal DEHA doses of 0, 800, or 1200mg/kg/day body weight and doses of 0, 200, 400, or 800mg/kg/day (main study) elicited developmental toxicity including antiandrogenic effects. In the main study, DEHA induced a prolonged gestation period (800mg/kg/day) and a dose-related increase in postnatal death (400 and 800mg/kg/day). DEHA also induced a permanent decrease in offspring body weight (800mg/kg/day). No antiandrogenic endpoints were affected. We conclude that DEHA induced developmental toxicity and the NOAEL is 200mg/kg. DEHA did not induce antiandrogenic effects similar to those of di(2-ethylhexyl) phthalate even though the chemical structures have similarities and the two chemicals have a common metabolite.

Isolation of a thermophile degrading poly(butylene succinate-co-butylene adipate).

A thermophile, strain 73, which degrades poly(butylene succinate-co-butylene adipate) (PBSA) film was isolated from 95 soil samples obtained from different locations by cultivation using an enrichment culture technique at 60 degrees C. At this temperature, the strain grew on PBSA and the dissolved total organic carbon (TOC) concentration in the medium changed according to the growth stage, i.e., after the TOC concentration rapidly reached the minimum, it increased rapidly until it reached a peak and then decreased thereafter. During cultivation, PBSA was almost eliminated and the solution viscosity of the residual PBSA decreased markedly. Gel permeation chromatograms of the residual PBSA showed a significant decrease in the main peak and the appearance of a new peak at the low molecular weight region. The strain was identified as Bacillus stearothermophilus, which has an optimum growth temperature of approximately 63 degrees C.

Metastability and transformation of polymorphic crystals in biodegradable poly(butylene adipate).

Polymorphism phenomenon of melt-crystallized poly(butylene adipate) (PBA) has been studied by wide-angle X-ray diffraction (WAXD), small-angle X-ray scattering (SAXS), and differential scanning calorimetry (DSC). It has been found that the isothermal crystallization leads to the formation of PBA polymorphic crystals, simply by changing the crystallization temperature. The PBA alpha crystal, beta crystal, and the mixture of two crystal forms grow at the crystallization temperatures above 32 degrees C, below 27 degrees C, and between these two temperatures, respectively. The relationship between PBA polymorphism and melting behaviors has been analyzed by the assignments of multiple melting peaks. Accordingly, the equilibrium melting temperatures Tm degrees of both alpha and beta crystals were determined by Hoffman-Weeks and Gibbs-Thomson equations for the purpose of understanding the structural metastability. The Tm degrees of the PBA alpha crystal was found to be higher than that of the beta crystal, indicating that the PBA alpha crystal form is a structurally stable phase and that the beta crystal form is a metastable phase. The analysis of growth kinetics of PBA polymorphic crystals indicates that the metastable PBA beta crystal is indeed the kinetically preferential result. Based on the thermal and kinetic results, the phenomenon of stability inversion with crystal size in melt-crystallized PBA was recognized, in terms of the growth mechanisms of PBA alpha and beta crystals and the transformation of beta to alpha crystals. The PBA beta --> alpha crystal transformation takes place at a sufficiently high annealing temperature, and the transformation has been evident to be a solid-solid-phase transition process accompanied by the thickening of lamellar crystals. The molecular motion of polymer chains in both crystalline and amorphous phases has been discussed to understand the thickening and phase transformation behaviors.

Production of starch foams by twin-screw extrusion: effect of maleated poly(butylene adipate-co-terephthalate) as a compatibilizer.

Free-radical-initiated grafting of maleic anhydride (MA) onto poly(butylene adipate-co-terephthalate) (PBAT), a biodegradable aliphatic-aromatic copolyester, was performed by reactive extrusion. 2,5-Dimethyl-2,5-di(tert-butylperoxy)hexane was used as the free-radical initiator. The peroxide concentration was varied between 0.0 and 0.5 wt % at 3.0 wt % MA concentration; the MA concentration was varied between 1.0 and 5.0 wt % at 0.5 wt % peroxide concentration. The reaction temperature was maintained at 185 degrees C for all experiments. Under these conditions, between 0.194% and 0.691% MA was grafted onto the polyester backbone. Size-exclusion chromatography, melt flow index, intrinsic viscosity measurements, thermal gravimetric analysis, and differential scanning calorimetry were used to characterize the maleated copolyester. Increasing the initiator concentration at a constant MA concentration of 3% resulted in an increase in the grafting of MA while decreasing the molecular weight of the resulting polymer. Increasing the feed MA concentration also increased the grafting percentage. The maleation of the polyester proved to be very efficient in promoting strong interfacial adhesion with high amylose cornstarch in starch foams as prepared by melt blending. Thus, the use of maleated copolyester as a compatibilizer between starch and PBAT allowed the reduction of the density of resulting starch foams to approximately 21 kg/m3 and improved the resilience from 84% to as high as 95%. Also, the resulting starch foams exhibited improved hydrophobic properties in terms of lower weight gain and higher dimensional stability on moisture sorption.

Study of biodegradable polylactide/poly(butylene adipate-co-terephthalate) blends.

Both polylactide (PLA) and poly(butylene adipate-co-terephthalate) (PBAT) are biodegradable polymers. They are thermoplastics which can be processed using most conventional polymer processing methods. PLA is high in strength and modulus (63 MPa and 3.4 GPa, respectively) but brittle (strain at break 3.8%) while PBAT is flexible and tough (strain at break approximately 710%). In view of their complementary properties, blending PLA with PBAT becomes a natural choice to improve PLA properties without compromising its biodegradability. In this study, PLA and PBAT were melt blended using a twin screw extruder. Melt elasticity and viscosity of the blends increased with the concentration of PBAT. Crystallization of the PLA component, phase morphology of the blend, mechanical properties, and toughening mechanism were investigated. The blend comprised an immiscible, two-phase system with the PBAT evenly dispersed in the form of approximately 300 nm domains within the PLA matrix. The PBAT component accelerated the crystallization rate of PLA but had little effect on its final degree of crystallinity. With the increase in PBAT content (5-20 wt %), the blend showed decreased tensile strength and modulus; however, elongation and toughness were dramatically increased. With the addition of PBAT, the failure mode changed from brittle fracture of the neat PLA to ductile fracture of the blend as demonstrated by tensile test and scanning electron microcopy (SEM) micrographs. Debonding between the PLA and PBAT domains induced large plastic deformation in PLA matrix ligaments.

Templated metal-organic frameworks: synthesis, structures, thermal properties and solid-state transformation of two novel calcium-adipate frameworks.

Two novel calcium-adipate framework materials have been synthesized hydrothermally. GWMOF-7 ([Ca(C6H8O4)(H2O)2]*(C10H8N2)) and GWMOF-8 ([Ca(C6H8O4)(H2O)2]*(C12H12N2)) both formed three-dimensional structures and were characterized with single-crystal X-ray diffraction, powder X-ray diffraction, IR spectroscopy and elemental analysis. Thermal properties were also studied with thermogravimetric analysis, and show that these structures undergo a solid-state transformation into a denser three-dimensional framework.

Highly efficient one-step conversion of cyclohexane to adipic acid using single-site heterogeneous catalysts.

A solid source of 'active' oxygen (acetylperoxyborate, APB), when dissolved in aqueous solution in the presence of a single-site microporous catalyst containing redox centres (Fe(III)AlPO-31, Mn(III)AlPO-5, Fe(III)AlPO-5), converts cyclohexane with high efficiency (ca. 88%) and exceptionally high selectivity (ca. 81%) to adipic acid at 383 K; this procedure is also effective in converting styrene to styrene oxide and -pinene and (+)-limonene to their corresponding epoxides.

Theoretical interpretation of the line shape of crystalline adipic acid.

A general quantum theoretical approach of the upsilon(X-H) IR line shape of cyclic dimers of weakly H-bonded species in the crystal state is proposed. In this model, the adiabatic approximation (allowing to separate the high-frequency motion from the slow one of the H-bond bridge) is performed for each separate H-bond bridge of the dimer and a strong nonadiabatic correction is introduced into the model via the resonant exchange between the fast-mode excited states of the two moieties. Quantum indirect damping and Fermi resonances are taken into account. The present model reduces satisfactorily to many models in the literature dealing with more special situations. It has been applied to the cyclic dimers of adipic acid in the crystal phase. It correctly fits the experimental line shape of the hydrogenated compound and predicts satisfactorily the evolution in the line shapes with temperature and the change in the line shape with isotopic substitution.

Identification and quantification of alpha-amino adipic acid in bovine dentine phosphoprotein.

A unique phosphoprotein, which contains an uncommon amino acid, alpha-amino adipic acid (alpha-AAA), was isolated from unerupted bovine teeth by extensive EDTA demineralization. The alpha-AAA in the protein hydrolysates was identified based on the elution time on an amino acid analyzer using the sodium and lithium citrate buffer in a dual column system, and its identity was confirmed by comparisons of its DNS derivative with that of authentic alpha-AAA. This result suggests that the lysine residues in the phosphoprotein may be deaminated by an amine oxidase into allysine and further oxidized to alpha-AAA.

Immunohistochemical profile of galectin-8 expression in benign and malignant tumors of epithelial, mesenchymatous and adipous origins, and of the nervous system.

This study aims to investigate whether the immunohistochemical expression of galectin-8 could be used as a diagnostic marker in tumor tissues of various histogenetic origins including specimens from epithelial (n=145), mesenchymatous (n=16), adipous (n=10) and central and peripheral nervous system (n=25) tissue, and 4 mesotheliomas. Immunohistochemical reactions were carried out with a polyclonal anti-galectin-8 antibody and histological slides from tissues derived from the files of the Laboratory of Anatomopathology of University Erasmus Hospital, Brussels. Formalin-fixed paraffin-embedded tissues of 45 normal cases as well as 41 benign and 114 malignant tumors were studied. Marked decreases in immunohistochemical galectin-8 expression were observed in colon (p=0.001), pancreas (p=0.007), liver (p=0.0008), skin (p=0.002) and larynx (p=0.02) tissue when comparing malignant tissue to normal tissue and/or benign tumors. The reverse relationship was observed for breast tissue (p=0.007). No statistically significant differences (p>0.05) were detected when comparing normal tissue and/or benign to malignant tumors in lung, bladder, kidney, prostate and stomach tissue. Significant galectin-8 expression was also measured in non-epithelial tissue including tumors of the central and peripheral nervous system as well as in skeletal muscle and mesotheliomas. Immunohistochemical monitoring of galectin-8 thus reveals an organ-type-dependent regulation of expression upon malignant transformation of various tissue types of epithelial origin. This observation will prompt further studies to delineate any relationship with prognosis.

Synthesis, absolute configuration and activity at N-methyl-D-aspartic acid (NMDA) receptor of the four D-2-amino-4,5-methano-adipate diastereoisomers.

The four D-2-amino-4,5-methano-adipates 26, 27, 32, 33 were synthesized and their biological activity at the N-methyl-D-aspartate (NMDA) receptor was assessed. The synthesis involved as a key step a rhodium acetate dimer catalyzed addition of ethyl diazoacetate to the protected D-allylglycine (17). In vitro receptor binding using L-[3H]glutamate as the radioligand provided affinity data, while modulation of [3H]TCP binding was used as a functional assay. The analogues were also evaluated in [3H]kainate and [3H]AMPA binding to assess selectivity over non-NMDA glutamate receptors. Three of the four diastereoisomer, D-CAA B (27), C (32) and D (33) were shown to have agonist properties at the NMDA-site, while the fourth, (2R,4R,5R) D-CAA A (26) was characterized as an NMDA-site atypic antagonist.
