A large number of 1-aryl-3,3-dialkyl triazenes and 1,2-diphenyl triazenes variously substituted in the 3 position with OH and alkyl groups were prepared and studied for acaricide activity against Panonychus ulmi and chemosterilizing and insecticidal activity against Musca domestica. The results highlighted the marked chemosterilizing activity of 1-(2-methoxyphenyl)-3,3-di-n-propyltriazene and the interesting acaricidal activity of 1-(4-benzoylphenyl)-3,3-dimethyltriazene. The examination of such a large number of compounds for insecticidal activity allowed interesting conclusions to be drawn regarding structure/activity relationships.

The novel tricyclic ring systems 14 can be prepared by reaction of thienopyrazoles 5 with 1,3-dielectrophiles, such as acetylacetone, dibenzoylmethane or 1,2,4-dithiazole 8. An other synthetic route for the preparation of the tricyclic systems, based on the pyrazoloazines 13, was investigated. The compounds 5 and 14 were tested for antiulcer activity.

The results of experimental studies of three drugs from aziridinyltriazine group, dioxadet, trisadet, and furizil, are presented. These drugs exhibit a pronounced therapeutic effect against a broaden spectrum of tumors grafted in mice and rats. Toxic effect of the drugs is exhibited mainly in suppression of blood formation. Other side-effects are slightly pronounced. One of the drugs, dioxadet passed the I and II stages of clinical trials and is recommended for application in chemotherapy of malignant tumors.
The hypnotic effect of hexobarbital, measured in male NMRI-mice, was reduced during the first 24 hours after a unique or fractionated whole-body X-ray irradiation with 700 and 1400 R resp. 70 to 280 R and twelve hours after a unique intravenous injection of 16,0 mg tretamine per kilogram of body weight. Immediately after a unique injection of a high dose of tretamine and 24 hours after the last of four applications of 0,25 to 2,0 mg/kg which were repeated each after one day, the sleeping time after hexobarbital was prolonged. 24 hours after fractionated injection of tretamine (0,5 mg/kg each time), the concentration of hexobarbital in the brain showed a significant increase compared with the control animals. These results show that the decreased effect of hexobarbital in the early post-irradiation phase is based on pharmacodynamic processes while the increase of the effect produced by continued administration of tretamine can be explained by pharmaco-cinetic reasons.
An investigation has been made of the possibility of using the retention times t obtained by reversed phase high pressure liquid-liquid chromatography in the study of quantitative structure-activity relatiionships. Taking as an example a group of herbicides which inhibit the Hill reaction, it was found that log t is additive and can be used as the hydrophobic parameter in regression analyses. The characteristics, advantages and possible developments of the method are discussed and the analytical interest is highlighted.

We recently found that a triazinyl dye, cibacron blue F3GA (CB), has a sensitizing effect on the in-vitro susceptibility of methicillin-resistant Staphylococcus aureus (MRSA) to oxacillin (C. Shirai, M. Sugai, H. Komatsuzawa, K. Ohta, M. Yamakido, H. Suginaka, Antimicrob. Agents Chemother. 42: 1278-1280, 1998). Among nine triazinyl dyes tested, CB the strongest sensitizing effect. Population analysis demonstrated that CB reduced the resistance level of MRSA. In the presence of oxacillin at subinhibitory concentrations, CB inhibited the growth of MRSA, but its effect on the cells appeared to be bacteriostatic. Under experimental conditions, CB did not affect the amount of PBP2', binding of [14C]benzylpenicillin to PBP2', or peptidoglycan susceptibilities to bacteriolytic enzymes. Autolytic enzyme-deficient MRSA mutants were equally as sensitive as the parent strain to the effect of CB on the susceptibility to oxacillin. CB affected the resistance level of MRSA irrespective of the status of the mecI/mecR1 element and/or penicillinase plasmids. The sensitivities of several bacteriolytic enzymes to heat-inactivated MRSA cells were not affected when the cells grown in the presence of CB. CB did not stimulate the release of lipoteichoic acid from the cells. These results suggest that the sensitization effect of CB cannot be simply explained by its effect on mecA related products, autolysins, femAB products or the release of lipoteichoic acid.
1,3,5-Triazabicyclo[3.1.0]hexane-2,4-diones proved to be potent antineoplastic and cytotoxic agents in murine and human cancer cells. In L1210 lymphoid leukemia cells DNA synthesis was significantly suppressed over 60 min by the agents from 25 to 100 microM. DNA synthesis was blocked at multiple sites including DNA polymerase alpha, ribonucleoside reductase, dihydrofolate reductase, PRPP-amido transferase, and nucleoside kinases which would be additive overall in suppressing DNA synthesis. The DNA molecule itself did not appear to be at target of the agents since no alkylation of nucleotide bases, intercalation between base-pairs or cross-linking of strands occurred after 24 h incubation at 100 microM. Nevertheless, L1210 DNA fragmentation did occur after 24 h incubation at 100 microM which is usually associated with tumor cell apoptosis.
