Salicylic acid disposition in children with rheumatoid arthritis.

The plasma level profile of SA and SUA after a single oral dose of ASA was studied in 8 children with juvenile rheumatoid arthritis, aged 3.5-15.0 years. Pharmacokinetic parameters were on average similar to those reported in the literature for adult subjects, although a somewhat larger intersubject variability was found.

Transvalencin Z, a new antimicrobial compound with salicylic acid residue from Nocardia transvalensis IFM 10065.

Transvalencin Z was isolated from a culture broth of Nocardia transvalensis IFM 10065, a clinical isolate from a Japanese patient with actinomycotic mycetoma. The transvalencin Z structure was determined using NMR and mass spectrometric analyses. The structure is similar to a partial structure of siderophores such as mycobactins and nocobactins, but the compound has no cytotoxic activity. Transvalencin Z shows a strong antimicrobial activity against Gram-positive bacteria, but shows no activity against Gram-negative bacteria, fungi and tumor cells.

Salicylic acid derivatives form stable complexes with scandium(III) ion in aqueous solution.

The stability constants of 5-nitrosalicylic acid (5-NSA) and 5-sulfosalicylic acid (5-SSA) complexes of Sc(III) were determined by potentiomeric pH titration. ML and ML2 type first and second complexes were observed in the solutions of 5-NSA and 5-SSA with Sc(III) at 25 degrees C in I=0.1 M ionic medium. The stability constants of Sc(III)-5NSA and Sc(III)-5SSA systems were also investigated by spectrophotometry to determine the stoichiometries of the complexes formed in the reactions. Our results showed that Sc(III)-5SSA complexes are more stable than the Sc(III)-5NSA complexes in aqueous solutions.

Levels of prostaglandins and arachidonic acid in UV-B irradiated human skin before and after topical application of benzyl-2,5-diacetoxybenzoate, a salicylic acid derivative.

Benzyl-2,5-diacetoxybenzoate (BDAB) was tested for anti-inflammatory activity on experimentally inflamed skin. Human abdominal skin was irradiated with three minimal erythema doses of UV-B (290-320nm) radiation to give a maximum erythema at 24 hours with an associated rise in PGE2 and PGF2 alpha levels measured by gas chromatography-mass spectrometry. Topical application of a 1% w/w preparation of BDAB neither decreased the evoked erythema nor diminished the rise in PGE2 and PGF2 alpha concentrations. BDAB, applied topically, was judged to be ineffectual as an anti-inflammatory agent in UV-B erythema.

[Induction of sesquiterpene cyclase gene expression and antioxidant enzymes regulated by exongenous salicylic acid in leaves of Capsicum annuum].

An analysis of the induction of sesquitèrpene cyclase gene expression and antioxidant enzymes in Capsicum annuum by exongenous salicyclic acid (SA) pointed out that the sesquiterpene cyclase gene expression in detached pepper leaves was induced by 0.5-4 mmol.L-1 of exongenous SA, whereas the enzyme activity and accumulation of cyclase mRNA were much lower, and it took a longer time (36 h) for the gene to be induced to express after SA treatment, comparing to other elicitors such as UV and fungi treatment. The activities of SOD and POD were enhanced, while the CAT activity was inhibited to some degree. Consequently, H2O2 content was increased in SA treated pepper leaves. H2O2 accumulation was related to the integrated influences from antioxidant enzymes.

Smooth muscle relaxant action of benzyl benzoates and salicylic acid derivatives from Brickellia veronicaefolia on isolated guinea-pig ileum.

A dichloromethane-MeOH extract prepared from the aerial parts of Brickellia veronicaefolia inhibited the spontaneous contractions (IC50 = 39.22 microg/mL) of the guinea-pig ileum. Bioassay-guided fractionation of the extract led to the isolation of three new benzoic acid derivatives, 1,2-bis-O-(2-methoxybenzoyl)-beta-d-glucopyranoside (1), 3-(beta-glucopyranosyloxy)benzyl 2,6-dimethoxybenzoate (2) and 3-hydroxybenzyl 2,6-dimethoxybenzoate (3), together with the known compounds taraxasteryl acetate (4), 4-allyl-2-methyloxyphenyl-beta-glucopyranoside (5), 2-hydroxy-6-methoxybenzoic acid (6), 2-methoxybenzoic acid (7), chamazulene (8), 2-methoxybenzyl 2-hydroxybenzoate (9), benzyl 2,6-dimethoxybenzoate (10), 3-methoxybenzyl 2-hydroxy-6-methoxybenzoate (11), benzyl 2-hydroxy-6-methoxybenzoate (12), benzyl 2,3,6-trimethoxybenzoate (13), benzyl 2-hydroxy-3,6-dimethoxybenzoate (14) and 3-methoxybenzyl 2,6-dimethoxybenzoate (15). The isolates were characterized by spectral means. Compounds 2 - 6, 8 - 11, 14 and 15 induced a concentration-dependent inhibition of the spontaneous contractions of the guinea-pig ileum with IC50 values ranging from 1.49 to 4.96 microM. Their activity was comparable to that of papaverine (IC50 = 4.23 microM).

The slow wound-response of gammaVPE is regulated by endogenous salicylic acid in Arabidopsis.

Vacuolar processing enzyme (VPE) is a cysteine protease responsible for the maturation of various vacuolar proteins in higher plants. The Arabidopsis thaliana (L.) Heynh. gammaVPE gene, encoding a VPE homologue, is slowly up-regulated in both local and systemic leaves in response to wounding. To clarify the activation mechanism of gammaVPE, we examined the accumulation of gammaVPE mRNA after hormone treatments or after wounding in wild-type and various mutant plants of Arabidopsis. Both ethylene and jasmonic acid (JA) are known as signal molecules that activate the wound-responsive genes. However, treatment with exogenous JA had little effect on the gammaVPE response, although JA activated the vegetative storage protein ( VSP) gene, a typical wound-responsive gene. Wounding activated gammaVPE even in two ethylene-insensitive plants (etr1-1 and ein2-1). Thus, the wound-induced expression of gammaVPE was independent of ethylene and JA. We found that the wound-induced expression of gammaVPE was reduced in two SA-deficient plants (pad4-1 and NahG), while the wound-induced expression of VSP increased in these mutants. Appreciable accumulation of SA was not observed in either the local or systemic leaves after wounding. These results suggest that endogenous SA enhances the wound-induced expression of gammaVPE and attenuates the wound-induced expression of VSP, although SA is not a wound-signal that directly activates these genes.

Cell cycle arrest and modulation of HO-1 expression induced by acetyl salicylic acid in hepatocarcinogenesis.

BACKGROUND AND AIMS: Control of cell proliferation is important for cancer prevention since cell proliferation has an essential role in carcinogenesis. In rodent carcinogenesis models, antioxidant agents suppress carcinogen-induced cellular hyper proliferation in the target organs. Strict control of cell division is an essential process to ensure that DNA synthesis and mitotic division are accurately and coordinately executed. We studied the interplay between cell cycle and heme oxygenase-1 (HO-1) and the effect of the acetylsalicylic acid (ASA) in hepatic carcinogenesis. METHODS: Male CF1 mice pre-treated with dietary p-dimethylaminoazobenzene (DAB; 0.5%, w/w) were fed with ASA (0.16%, w/w). We investigated the hepatic expression of cyclin D1, cyclin E, Cdk2, Cdk4, p21, p27, p53; the level of bcl-2, an antiapoptotic protein and of heme oxygenase-1 (HO-1), a marker of oxidative stress, by Western blot analysis. RESULTS: The treatment with ASA produced an important attenuation in the induction of cyclin E and cyclin D1 provoked by DAB. p21 and p27 levels were increased when animals received both drugs. The administration of ASA to DAB treated animals induced Cdk2 (29%). HO-1 induction (65%) provoked by DAB was diminished by ASA administration reaching lower induction levels (23%). CONCLUSION: The deregulation of cyclin/CDK expression and the up-regulation of p21 and p27 with the administration of ASA, post-treatment of the carcinogen administration, would block the pass through out to the G0/G1 check point to permit the cells to repair their DNA and HO-1 protected the liver from reactive oxygen species produced from DAB.

Hydrophilic polymer drug from a derivative of salicylic acid: synthesis, controlled release studies and biological behavior.

Hydrophilic polymeric drugs bearing "Triflusal" (4-trifluoromethylsalicylic acid), a drug widely used as antithrombogenic agent (Disgren), have been prepared by free radical copolymerization of methacryloyloxyethyl [2-(acetyloxy)-4-(trifluoromethyl)] benzoate (HTRF) and N,N'-dimethylacrylamide (DMA). The reactivity ratios of both monomers have been determined by 1H NMR spectra by applying non-linear least square treatments to the copolymerization equation (terminal model), and the kinetic parameters obtained indicated that the microstructure of copolymer chains is homogeneous, with a random distribution of the active HTRF units along the copolymer chains. That means that for the copolymer system THDMA22 used in this work, HTRF units are mainly isolated in relatively long DMA sequences. Therefore, in this structure the intramolecular interactions between adjacent HTRF units are negligible. Release of Triflusal from THDMA22 has been studied in vitro using buffered solutions at pH = 2, 7.4 and 10 and 37 degrees C. The system showed an interesting pseudo-zero order release profile at pH = 7.4 during several months. It has been also evaluated the pharmacological activity and the behavior of the system in contact with biological media. In this sense, we have carried out some in vitro studies about the antiaggregant properties and biocompatibility of THDMA22. Results demonstrate that this copolymer inhibits platelet aggregation in its macromolecular form and presents a good biocompatibility with Human Osteoblastic Cells (HOS).

Myocardial microdialysis of salicylic acid to detect hydroxyl radical generation during ischemia.

We examined in vivo monitoring of hydroxyl radical (.OH) generation during myocardial ischemia for 30 min by occluding the left anterior descending (LAD) in dog heart using a microdialysis technique. The hydroxyl radical reacts with salicylate and generates 2,3- and 2,5-dihydroxybenzoic acids (DHBA) which can be measured electrochemically in picomole quantity by a high pressure liquid chromatography-electrochemical (HPLC-EC) procedure. When the premature ventricular contraction (PVC) occurred at almost 25 sec intervals, marked elevation of the levels of 2,3- and 2,5-DHBA was observed in the heart dialysate of 30-min ischemia. This study demonstrated the generation of .OH in the canine heart subjected to 30-min ischemic insult.
