[n-Alkane oxidation by propionic acid bacteria].

Propionic acid bacteria were found for the first time to be capable of oxidizing n-alkanes. The value of QO2 is higher on a mixture of n-alkanes (C12--C20) than on individual hydrocarbons whose availability is determined by the length of a hydrocarbon chain: C15 and C16-alkanes are oxidized best of all; C14, C13 and C12-alkanes are oxidized at a lower rate; and alkanes whose chain comprises less than ten carbon atoms are not oxidized at all. Hexadecane is oxidized to CO2, cetyl alcohol and palmitic acid being intermediates; this suggests the existence of the monoterminal pathway for oxidation of n-alkanes in propionic acid bacteria.

[Synthesis of organic acids by immobilized propionic bacteria in the flow system and stabilization of the process].

The capacity of immobilized cells of propionic bacteria to synthesize organic acids was examined. Propionibacterium shermanii cells incorporated into polyacrylamide gel were capable to synthesize propionic, acetic and pyruvic acids in the flow system. As a carbon source glucose, lactate-Na or whey lactose was used. The greatest amount of the acids was synthesized with the use of lactate-Na. The life-time of the biocatalyst (immobilized cells) can be increased by its reactivation with a nutrient medium required for optimal cell proliferation.

[Testosterone and dihydrotestosterone radioimmunoassays in Müllerian ducts of control and testosterone propionate injected quail embryos (author's transl)].

Testosterone (T) and dihydrotestosterone (DHT) have been quantitated by means of radioimmunoassay in Müllerian ducts (CM) from control quail embryos (6 to 8-day male and 6 to 15-day female) and from female embryos injected with 50 nanograms of testosterone propionate (PT) on the 8th day. These hormones are demonstrable in undifferentiated CM from 6-day control embryos. In control males although a highly significant decrease of the CM weight occurs during the CM involution, the detected amounts of androgens remain at a constant level. In control females, the right CM shows a slight increase of the androgen content during the rudimentation, i.e. from day 8 on; in the left CM: the highest steroid levels are found on the 6th day; while the CM differentiate, concentrations decrease and become similar to those found in neutral tissues. Given testosterone propionate on day 8 female embryos: right CM: both testosterone and DHT levels highly increase until day 14; the CM of treated embryos contain 8 times as much steroid as control; left CM: testosterone and DHT increase after injection until day 9,5; they slightly decrease between days 9,5 and 12, and then remain constant on day 14. Differences in concentrations are highly significant between CM of control and of PT - injected embryos. It seems that the binding sites of these androgens are more numerous during the involution and that bound testosterone or DHT could take a part in CM regression in male embryos and right CM rudimentation in female embryos.

[Sensitivity of Sherman's propionic acid bacilli to antibacterial preparations and vitamin B12 synthesis].

Sherman propionic acid bacilli were sensitive to benzylpenicillin, ampicillin, ceporin, tetracyclines, oleandomycin, oletetrin, tetraolean, sigmamycin, levomycetin and furadonine. Methicillin, oxacillin, monomycin, kanamycin, polymyxin and furazolidone had an insignificant effect on the above organism. The subbacteriostatic concentrations of methicillin, oxacillin, streptomycin, monomycin, kanamycin, neomycin, tetraolean, sigmamycin, polymyxin M and ristomycin increased the biosynthesis of vitamin B12 by Sherman propionic acid bacilli, while benzylpenicillin, ampicillin, tetracyclines, oleandomycin, oletetrin, levomycetin and furadonine in the subbacteriostatic concentrations inhibited this process.

The effect on psoriasis of clobetasol propionate used alone or in combination with UVB.

A study has been carried out to compare the effect of a combination of clobetasol propionate and UVB versus placebo/UVB. No significant reduction in time to healing was observed. However, symptoms abated more rapidly in the steroid/UVB group. Recurrences occurred at an earlier stage following the steroid UVB combination for long periods. A third group, treated solely with clobetasol propionate has also been investigated, but results were poorer than those described above. It is concluded that steroids in combination with UVB should be used only during the initial period in order to achieve a more rapid alleviation of symptoms and to avoid the side effects.

[The effect of testosterone propionate, 5 alpha-dihydrotestosterone and 5 alpha-androstan-3 beta, 17 beta-diol on acid phosphatase activity in the prostate of castrated rats].

The acid phosphatase is studied in the prostate gland of rats. It is shown that 10 days after gonadectomy the activity of acid phosphatase lowered considerably. Administration of testosterone propionate or 5 alpha-dihydrotestosterone to castrated animals restored the enzyme activity whereas 5 alpha-androstan-3 beta,17-diol was not effective. Administration of testosterone propionate with one of its metabolites increased the activity of acid phosphatase in the prostate gland, however to a less extent than with the use of the hormone itself. The lowest activity is detected with the simultaneous application of three androgens.

A double-blind comparison of a propionic acid derivative (ibuprofen) and a fenamate (mefenamic acid) in the treatment of dysmenorrhea.

A double-blind three-way crossover design in the treatment of dysmenorrhea comparing a propionic acid derivative (ibuprofen) and a fenamate (mefenamic acid) with a placebo showed that both ibuprofen and mefenamic acid are generally superior to placebo. Statistically significant results were obtained in favor of the study drugs over placebo for the pain relief afforded by the treatments (as graded by patients) and the visual analog pain relief score, which not only ranks but also indicates the degree of pain relief as a percentage of total relief (100%). Pairwise comparisons for the ranks found mefenamic acid significantly superior to placebo (P less than .001) and ibuprofen marginally superior to placebo (P less than .06), while the visual analog pain relief scale demonstrated mefenamic acid and ibuprofen superior to placebo (P less than .001 and P less than .01, respectively). For both the patient ranking of pain relief by treatment and the visual analog pain relief scale, the results showed no significant differences between ibuprofen and mefenamic acid. Side effects occurred in 11 ibuprofen cycles, five mefenamic acid cycles, and ten placebo cycles of the 48 cycles with each agent. These were generally of minor severity or importance and were not statistically different. The need for additional analgesics and the ability to pursue normal daily activity were not different for any treatment group. The findings of this study indicate no clinical difference between a propionic acid derivative such as ibuprofen and a fenamate such as mefenamic acid in the treatment of dysmenorrhea.

Protection by testosterone propionate against the nephrotoxicity of 2-aminoanthraquinone in Fischer rats.

The role of gonadal hormones in the nephrotoxicity of the hepatocarcinogen 2-aminoanthraquinone (2-AAQ) was investigated. Intact and castrated four week old Fischer rats of both sexes and castrated female rats with subcutaneous implants of testosterone propionate pellets were fed 2% 2-AAQ in a Wayne meal diet. After 12 weeks of ad libitum feeding the animals were then given control diet. Based on survival, body, liver and kidney weights, and the histopathological evaluation of the kidneys, castrated female rats given testosterone propionate were afforded the greatest protective effect against the nephrotoxicity of 2-AAQ.

[Superoxide dismutase, catalase and peroxidase of propionic acid bacteria].

The respiration of different species of propionic acid bacteria was found to be resistant to the inhibiting action of cyanide. Propionibacterium globosum, P. globosum (emm), P. vannielii, P. shermanii, P. petersonii, P. pentosaceum and P. raffinosaceum were shown to contain superoxide dismutase, catalase and peroxidase. The highest activity of catalase and superoxide dismutase was found in P. globosum and P. vannielii, the lowest one in P. shermanii and P. raffinosaceum. Peroxidase was detected in propionic bacteria for the first time.

Glutamate receptor (gamma-amino-3-hydroxy-5-methyl-4-isoxazole propionate and kainate subtype) activity enhanced by dizocilpine (MK801) in rat hippocampus. Rapid chemical kinetic measurements of sodium flux and receptor desensitization with native membranes.

L-Glutamate-mediated transmembrane influx of 22Na+ into a native membrane vesicle preparation from rat hippocampus was resolved into components due to glutamate receptor and Na(+)-glutamate symport, by kinetic analysis and pharmacological specificity. Measurements made with a quench-flow technique showed that receptor-mediated cation exchange proceeded in two phases, the faster phase progressively attenuated by a desensitization process with a half time of approximately 90 ms. The influx of 22Na+ continued in the second phase with a half time of 6 s. Receptor-mediated 22Na+ influx was replicated with the glutamate mimetics, alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate and kainate but not with N-methyl-D-aspartate and was inhibited by 6-cyano-7-nitroquinoxaline-2,3-dione but not by 2-amino-5-phosphonovaleric acid. Receptor-mediated 22Na+ influx did not require glycine and was pH dependent; influx was inhibited at pH values less than pH 5.0. Thus, the receptor-mediated activity was of the gamma-amino-3-hydroxy-5-methyl-4-isoxazole propionate-kainate subtype. The N-methyl-D-aspartate receptor inhibitor 5-methyl-10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5,10-imine (dizocilpine or MK801), increased receptor-mediated transmembrane 22Na+ flux in both phases, but did not accelerate receptor desensitization. Receptor-mediated radiotracer exchange was the major contribution to 22Na+ influx in times less than 5 s and the receptor-mediated responses could be measured with a small correction for sodium-glutamate symport or in the presence of a glutamate-uptake inhibitor. A third phase of 22Na+ influx accessed a different internal volume and proceeded with the same first-order rate constant as [3H]glutamate uptake when measured simultaneously. This was shown to be mediated by the glutamate uptake mechanism, with a half time of 20-40 s varying with the preparation. The Na(+)-glutamate symport became the largest cause of 22Na+ influx after at least 5 s, the final magnitude being, on average, twofold larger than the receptor-mediated 22Na+ influx.

Topical glucocorticosteroid (fluticasone propionate) inhibits cells expressing cytokine mRNA for interleukin-4 in the nasal mucosa in allergen-induced rhinitis.

Allergen-induced late nasal responses are associated with recruitment and activation of T lymphocytes and eosinophils and preferential mRNA expression for T-helper type 2 (Th2) cytokines. We tested the hypothesis that topical corticosteroids may inhibit late responses by inhibiting cells expressing mRNA for Th2 cytokines. A randomized double-blind placebo-controlled trial of topical corticosteroid (fluticasone propionate) was performed in 48 adult grass pollen-sensitive patients. Nasal biopsies were taken at baseline and repeated 24 hr after local nasal allergen provocation following 6 weeks treatment with either fluticasone propionate 200 micrograms or placebo nasal spray twice daily. Baseline mRNA expression for interleukin-4 (IL-4) (P = 0.01) and IL-5 (P = 0.002) was higher in the patients than in normal controls. Topical corticosteroid treatment significantly inhibited immediate nasal symptoms, with almost complete inhibition of the late response following allergen challenge. This was associated with a marked decrease in the allergen-induced increases in cells expressing mRNA for IL-4 (P = 0.002) but not for IL-5. Inhibition of the late response was also accompanied by decreases in CD25+ cells, presumed T lymphocytes and eosinophils. A significant correlation was observed between the decreases in IL-4 mRNA+ cells and in eosinophils after treatment (r = 0.46, P < 0.05). These results suggest that prolonged treatment with topical corticosteroid inhibits allergen-induced early and late nasal responses and the associated tissue eosinophilia, and that, at least in part, this may result from inhibition of cells expressing mRNA for IL-4.

Humoral immune responses characteristic of testosterone-propionate-treated chickens.

White Leghorn chickens treated with testosterone-propionate on the 3rd day of embryonation were immunized with a mixture of sheep red blood cells, Brucella abortus and Salmonella pullorum at various ages, and the resulting agglutinins were titrated. The production of IgM antibody against sheep red blood cells was not affected significantly by testosterone-propionate. On the contrary, immune responses against the bacterial antigens were strongly suppressed by the same treatment. Production of IgG antibodies was strongly suppressed by the same treatment. There was little correlation between the production of IgM antibody against sheep red blood cells and the presence of bursal follicles. Immune responses against bacterial antigens correlated with the presence of the follicles. Production of IgG antibodies also correlated with the maintenance of bursal lymphoid structure.

Comparison of fluticasone propionate cream, 0.05%, and hydrocortisone-17-butyrate cream, 0.1%, in the treatment of eczema.

This is a randomized, double-blind, parallel-group, multicenter study involving 120 patients comparing the safety and tolerability of two midpotency topical preparations, fluticasone propionate cream, 0.05%, and hydrocortisone-17-butyrate cream, 0.1%, in the treatment of moderate-to-severe eczema. Safety of the study medications was determined over a twelve-week period using laboratory tests for selected fasting blood chemical levels, hematologic analysis, urinalysis, and morning plasma cortisol levels, and by analyzing both the nature and frequency of reported adverse events. Efficacy was evaluated during the first four weeks of the study. None of the fluticasone-treated patients experienced any severe drug-related adverse events, but one hydrocortisone-17-butyrate-treated patient's eczema was severely exacerbated by drug therapy. Plasma cortisol monitoring revealed minimal hypothalamic-pituitary-adrenal axis suppression. Overall, the nature of drug-related adverse events in patients as young as 12 years old treated with fluticasone propionate cream, 0.05%, indicates this topical application was safe and well tolerated throughout the twelve-week study. Fluticasone cream was also found to be similar in efficacy to hydrocortisone-17-butyrate cream.

Mechanisms of KE298, 2-acetylthiomethyl-3-(4-methylbenzoyl) propionic acid, to suppress abnormal synovial cell functions in patients with rheumatoid arthritis.

OBJECTIVE: 2-Acetylthiomethyl-3-(4-methylbenzoyl) propionic acid, KE298, a derivative or propionic acid developed in Japan has been shown to be effective for suppressing disease activity of rheumatoid arthritis (RA) in clinical trials in Japan. It is thus a candidate as a new disease modifying antirheumatic drug (DMARD). We analyzed effects of KE298 on synovial fibroblast-like cells in patients with RA to obtain insight into the clinical application of this medication. METHODS: RA synovial fibroblast-like cells were co-cultured with KE298 at 10(-4)-10(-5) M in the presence or absence of tumor necrosis factor-alpha 2 ng/ml, and their subsequent proliferative responses and proinflammatory cytokine and matrix metalloproteinase (MMP) production at the mRNA and protein levels were measured. Effects of KE298 on MMP-1 gene transcription and AP-1 transcription factor expression of RA synovial cells were studied by chloramphenicol acetyltransferase assay and gel shift assay, respectively. RESULTS: KE298 inhibited proliferation of RA synovial cells, proinflammatory cytokine production, and MMP-1 production mainly by reducing their transcription via downmodulation of AP-1 transcription factor. CONCLUSION: KE298 inhibits aberrant synovial cell functions of patients with RA by downregulating gene transcription, suggesting clinical application and usefulness of this new DMARD.

Eosinophil markers in seasonal allergic rhinitis. Intranasal fluticasone propionate inhibits local and systemic increases during the pollen season.

BACKGROUND: The purpose was to study activation markers of the eosinophil granulocytes in seasonal allergic rhinitis, and the impact of topical steroid therapy thereupon. METHODS: Sixty-three rhinitis patients with monoallergy to grass were examined before and at peak pollen season. Blood eosinophil count, eosinophil cationic protein (ECP), and eosinophil peroxidase (EPO) in serum and nasal lavage fluid were measured. During the season, patients were randomized to treatment with intranasal fluticasone propionate 0.1 mg o.d. (n=26), 0.2 mg o.d. (n=25), or placebo (n=12). Six healthy persons served as controls. RESULTS: During the season, all parameters, except nasal lavage ECP, increased in the placebo group (P<0.001-P<0.05). Significant differences were seen between the steroid groups and the placebo group for all parameters (P<0.001-P<0.05). Higher eosinophil count (P<0.05), serum EPO (P<0.02), and nasal lavage EPO (P<0.05) were found in patients before season than in controls. The following winter, 44 patients returned for repeated measurement. Lower levels of nasal lavage EPO were observed for patients than levels at the beginning of the season (P<0.0001). CONCLUSIONS: Intranasal fluticasone propionate reduced inflammation of the nasal mucosa, demonstrated locally by nasal lavage ECP and EPO, and systemically by blood eosinophils, serum ECP, and serum EPO. EPO seemed more sensitive than ECP as indicator of allergic inflammation. EPO demonstrated some perennial eosinophil activity in hay fever patients, increasing locally during spring.

Neuroprotective efficacy of YM872, an alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid receptor antagonist, after permanent middle cerebral artery occlusion in rats.

The neuroprotective efficacy of YM872, a novel, highly water-soluble alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid receptor antagonist, was investigated in rats subjected to permanent occlusion of the left middle cerebral artery. The rats were assessed either histologically or neurologically 24 hr or 1 wk after ischemia. YM872 was intravenously infused for either 4 or 24 hr at dose rates of 0 to 20 mg/kg/hr starting 5 min after ischemia to examine the effect of prolonged treatment. YM872 was then infused at 20 mg/kg/hr beginning 0 to 4 hr after ischemia to determine the efficacy time window. Additionally, a 20 mg/kg/hr dose rate of YM872 was infused for 4 hr in single day- or 5-day repetitive-administrations to evaluate long-term benefits of the drug. YM872 significantly reduced infarct volume in both 4- and 24-hr treatment groups measured 24 hr after ischemia. No difference was observed in the degree of protection between length of infusion. Significant neuroprotection was maintained even when drug administration was delayed up to 2 hr after ischemia. A single YM872-administration significantly improved neurological deficit and reduced infarct volume (30%, P <.01) measured 1 wk after ischemia. YM872 treatment did not induce such adverse effects as physiological changes, serious behavioral abnormalities or nephrotoxicity. These data suggest that the alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid receptor plays a crucial role in the progression of neuronal damage in the early phase of ischemia and that YM872 may be useful in treating acute ischemic stroke.

Prenatal diagnosis of propionic acidemia.

In this report we summarize our experience in prenatal diagnosis of propionic acidemia (PA) since 1987. Overall, we have investigated 25 pregnancies at risk from 19 unrelated families. Until genetic structure of the genes involved in PA was elucidated, prenatal diagnosis has been successfully performed by means of metabolite quantitation and/or enzymatic assays in foetal issue. Today, direct propionyl-CoA carboxylase activity assay in combination with molecular analysis in chorion villi can be regarded as a fast and reliable method of choice for prenatal diagnosis of this organic acidemia.
