cDNA macroarray analysis of gene expression in ineffective nodules induced on the Lotus japonicus sen1 mutant.

The Lotus japonicus sen1 mutant forms ineffective nodules in which development is arrested at the stage of bacterial differentiation into nitrogen-fixing bacteroids. Here, we used cDNA macroarray systems to compare gene expression in ineffective nodules induced on the sen1 mutant with gene expression in wild-type nodules, in order to identify the host plant genes that are involved in nitrogen fixation. Macroarray analysis coupled with Northern blot analysis revealed that the expression of 18 genes was significantly enhanced in ineffective sen1 nodules, whereas the expression of 30 genes was repressed. Many of the enhanced genes encoded hydrolase enzymes, such as cysteine proteinase and asparaginase, that might function in the early senescence of sen1 nodules. By contrast, the repressed genes encoded nodulins, enzymes that are involved in carbon and nitrogen metabolism, membrane transporters, enzymes involved in phytohormone metabolism and secondary metabolism, and regulatory proteins. These proteins might have a role in the establishment of nitrogen fixation. In addition, we discovered two novel genes that encoded glutamate-rich proteins and were localized in the vascular bundles of the nodules. The expression of these genes was repressed in the ineffective nodules, which had lower levels of nitrogenase activity.

Intensity of nitrogen fixation in yellow lupine in presence of different doses of mineral nitrogen.

The effect of different doses of mineral nitrogen on the nitrogen-fixing activity of lupine nodules during vegetation was studied. It was shown that 0.5 of the dose of mineral nitrogen had an inhibitory effect only in the initial period of development of the nodules, whereas full and double doses of mineral nitrogen inhibited nitrogen fixation throughout the period of vegetation. Supplements of mineral nitrogen switch the lupine nutrition from the symbiotic to the autotrophic type, which is accompanied by a decrease in the nitrogen-fixing activity in the nodules and by an increase in the nitrate-reducing capacity of the roots and leaves.

[Relationship between ultrasound-findings of low-echoic nodule of hepatic parenchyma in liver cirrhosis and development of hepatocellular carcinoma].

We investigated development of hepatocellular carcinoma (HCC) in 90 patients with liver cirrhosis by follow up study for a period from 6 months to three and half years, making special reference to ultrasound-findings of the parenchyma of the liver. Liver cirrhosis was classified into four types and two, non-nodular and nodular, groups according to low-echoic nodules distributed in the parenchyma. The development of HCC was higher at the rate of incidence in patients of the nodular group than those of the non-nodular. It was more closely related with the male in patients of the non-nodular group, and both the old age and the advanced hepatic injury in those of the nodular group. A rise of the level of serum AFP during a period of the follow up was not observed in relation to the development of HCC and seemed to be ineffective for detection of carcinoma small sized. The echo-types of the parenchyma in liver cirrhosis were most significantly correlated with the development of HCC in comparison with the other etiologic factors.

Beta-galactosidase and alpha-mannosidase inhibit formation of multicellular nodules in breast cancer cell cultures.

In response to an estrogen, confluent monolayers of MCF-7 cell cultures develop multi-cellular nodules, termed foci. Post-confluent development of foci occurs with physiologic levels of 17beta-estradiol and are inhibited by various anti-estrogens acting through either the estrogen or aryl hydrocarbon receptors. In the present paper we report that disruption of the terminal sugars on membrane receptors results in inhibition of foci. Treatment with 0.013-0.05 units/ml of beta-galactosidase completely inhibited the development of foci while leaving the monolayer of cells intact. Trials with alpha-mannosidase resulted in a similar but less potent inhibition of foci. Lectin-fluorescent conjugates, RCA (Ricinus communis agglutinin), and ConA (Canavalia ensiformis agglutinin) were used to identify membrane surface carbohydrates on MCF-7 cells. Binding of the RCA-fluorescent conjugate was inhibited by co-treatment with galactose or lactose. Binding of ConA-fluorescent conjugate was significantly inhibited by mannose and n-acetyl-glucosamine. This is the first report of inhibition of foci development in MCF-7 cell cultures by disruption of surface carbohydrates on membrane receptors.

Idiopathic hyperplasia of the adrenal gland behaving like an aldosterone producing adenoma.

Primary hyperaldosteronism (adrenal adenoma and idiopathic hyperplasia) is a disorder with hypertension, hypokalemia, elevated serum aldosterone and suppressed plasma renin activity. Hyperplasia is managed medically whereas adenomas are treated surgically. Selective adrenal venous catheterization and aldosterone measurement is a useful tool in making the distinction in 95% of cases. We report a case of bilateral idiopathic hyperplasia of the adrenal glands adequately treated with medications for 6 years followed by worsening. Selective catheterization was consistent with a right sided adenoma. Surgical removal of the right adrenal gland alleviated her symptoms. Pathological examination showed focal nodular hyperplasia. We propose that in the course of the disease the focal hyperplastic nodule became autonomous and behaved like an adenoma. Monitoring of patients with adrenal hyperplasia for recurrence of symptoms is prudent as surgery is beneficial in patients who develop an autonomous nodule.

[First report of demodicosis in goats in the Czech Republic].

The first case of demodectic (Acari: Demodecidae) mange in the Czech Republic was diagnosed in a domestic White Shorthaired goat hospitalized in our clinic. A total of 128 solid protruding nodules, similar in size to lens or pea, were found on lateral parts of the neck and trunk and on proximal part sof the limbs. A pasty, creamy mass containing immature and adult mites could be pressed out of the nodules. Parasitological examinations identified the mites as Demodex caprae Railliet, 1895 (Fig. 1). Antiparasitic treatment was started with the pyrethroid deltamethrine (Butox 7.5 Pour On, Roussel Uclaf) although demodectic mange is not given in the spectrum of its applications. Ten ml of the drug were poured onto the back of the animal twice within an interval of 7 days. The next drug tested was another pyrethroid, Neostomosan sol. ad us. vet. (Sanofi Santé Animale) containing tetramethrine and transmix as the effective ingredients and recommended by the manufacturer for the treatment of canine and feline demodicosis. The animal was bathed twice within an interval of 14 days in a 1 : 400 dilution as recommended by the manufacturer. The mites survived the treatments with both drugs and their motility was not affected. The size of nine of the nodules (4.5 to 6.8 mm), measured before and after the treatments, remained unchanged. The third drug tested was amitraz (Taktic, Hoechst) known to be effective against canine demodicosis (Ectodex, Hoechst). Seven baths with 10-day intervals in the drug diluted 1 : 400 resulted in the decrease of the number of nodules to 83 (by 35.2%). The diameters of 7 of the measured nodules diminished by approx. 1.6 mm. No movements of the mites were observed on Day 56 after the treatment, but the immature stages probably survived because new nodules and allergy developed after the patient had been released from the clinic. The unsatisfactory effect of amitraz can be explained by the limited penetration of the drug into the nodules surrounded by fibrous tissue. Demodectic mange was subsequently diagnosed in one highly pregnant animal housed together with the patient. Further cases of demodicosis were detected by intentional examinations in the districts of Nový Jicín and Olomouc (three cases in the latter). It is apparent that the infection may remain undetected at current health checks, although it is world-wide spread and causes considerable losses to the hide-and-skin industry.

[Lymphoid structures and their cellular content in the cecum in 1-3 years old children (early childhood)].

In 1-3 Ys old children (early childhood) coecal lymphoid nodules are well developed with lymphatic capillaries and high endothelium venules in their base. The highest concentration of lymphoid cells is also noted there. In the apical zone, central part and nodule base cellular content has certain features. Lamina propria of the coecal mucosa contains great number of plasmic cells.

The regulation of serine dehydratase and glucose-6-phosphatase in hyperplastic nodules of rat liver during diethylnitrosamine and N-2-fluorenylacetamide feeding.

Changes in the levels of serine dehydratase and glucose-6-phosphatase induced by dietary stimuli or starvation in hyperplastic nodules of rat liver during diethylnitrosamine or N-2-fluorenylacetamide feeding were studied by immuno- and enzyme histochemical methods. The study was performed during carcinogenesis through a combined method of enzyme histochemistry and radioautography. Serine dehydratase was observed diffusely in the cytoplasm of the original hepatocytes in the periportal zone and was induced markedly during diethynitrosamine feeding but only slightly during N-2-fluorenylacetamide feeding. The enzyme was deficient and not inducible in hyperplastic nodules during their developing phase. Later during the feeding period, however, there was an elevation of the level of serine dehydratase and its inducibility with time in the majority of the nodules. A good correlation was observed between serine dehydratase and glucose-6-phosphatase in their elevated levels and response to enviornmental stimuli. There was a minor group of hyperplastic nodules in which the deficiencies of these enzymes persisted and enzyme induction was not observed. A greater number of hyperplastic nodules with persistent enzyme deficiency was seen during diethylnitrosamine carcinogenesis. These results provide further information about the changing biological nature of hyperplastic nodules with respect to their metabolic adaptability and enzyme levels during hepatocarcinogenesis.

Calcified cutaneous nodules on the heels of children: a complication of heel sticks as a neonate.

We report the presence of calcified nodules on the heels secondary to heel sticks in healthy monozygotic twins in childhood. The subjects were born preterm at 34 weeks. The heels were normal at birth. While in the hospital, they underwent multiple heel sticks as a means of venesection. The wounds failed to heal normally and warty lesions developed. Calcified cutaneous nodules secondary to heel sticks have been reported primarily in high-risk, preterm infants. There have also been two reports of their development in normal healthy infants. To date, all reported lesions have been asymptomatic, with spontaneous resolution by 18-30 months. These cases indicate that such lesions can in fact be chronic, extending into childhood, when they may become symptomatic. Dermatologists should therefore consider this diagnosis as a cause of warty, calcified nodules on the heel in childhood as well as in infants and in neonates.

Primary pleural epithelioid haemangioendothelioma with metastases to the skin. A case report and literature review.

Epithelioid haemangioendothelioma (EHE) is a rare vascular tumour of intermediate behaviour. It can arise from various sites including the liver, spleen, pleura, or lung. Cutaneous EHE can be primary or secondary. This report describes the case of a 51 year old man who presented with a history of dry cough, shortness of breath, and pleural effusion, and who developed two cutaneous nodules in the anterior abdominal wall a few weeks later. He had a previous history of asbestos exposure. Computed tomography scan showed a left sided pleural effusion and nodular pleural mass. Histology of both the pleural and cutaneous lesions was compatible with EHE. Electron microscopic examination demonstrated the presence of Weibel-Palade bodies. The patient underwent elliptical excision of the metastatic cutaneous nodules after decortication of the primary pleural tumour and adjuvant treatment. A few reports have described metastasis of intrathoracic EHE to the skin. Despite treatment with interferon, the patient developed more cutaneous lesions two years after the initial diagnosis. Even though the tumour has the classic light histological and ultrastructural features of EHE, it behaved in an aggressive manner.

Hepatocarcinogenesis due to chronic liver cell injury in hepatitis B virus transgenic mice.

Fifty-nine transgenic mice from a lineage that overproduces the hepatitis B virus large envelope polypeptide and accumulates high intrahepatic concentrations of hepatitis B surface antigen were followed for evidence of liver disease throughout their 24-month life span. By 4 months of age all mice displayed biochemical and histological evidence of moderately severe chronic hepatitis which was followed sequentially by the development of regenerative nodules and oval cell hyperplasia (by 6 months), liver cell adenomas (by 8 months), and hepatocellular carcinomas (by 12 months of age). One hundred % of mice in this lineage developed hepatocellular carcinoma by 20 months of age, whereas no histopathological changes were observed in age- and sex-matched nontransgenic littermate controls over the same period of observation. These results indicate that overproduction of the hepatitis B virus large envelope polypeptide initiates a process characterized by liver cell injury, inflammation, and regenerative hyperplasia, which places large numbers of hepatocytes at risk for the development of transforming mutations, and inexorably progresses to hepatocellular carcinoma. We suggest that this is a general mechanism of hepatocarcinogenesis that may be operative in human hepatitis B virus infection and other necroinflammatory liver diseases as well.

Transformation from SAA2-fibrils to AA-fibrils in amyloid fibrillogenesis: in vivo observations in murine spleen using anti-SAA and anti-AA antibodies.

Early amyloid fibrillogenesis from serum amyloid A protein (SAA) has been observed in the murine spleen after an injection of casein-Freund's complete adjuvant in the presence of amyloid enhancing factor, using anti-SAA C-terminal (anti-SAA) and anti-amyloid A (AA) antibodies. In Western immunoblotting of sera, both SAA1 and SAA2 reached a maximum after 24 h and began to decrease after 48 h. In spleen extracts, SAA2, but not SAA1 or AA, was found from 48 h, when amyloid was first deposited in the marginal zone. Electron microscopic immunohistochemistry of this stage showed reaction products from SAA in the marginal zone as fine granules along the cell membrane of mononuclear cells and focal intercellular aggregates, which contained fine fibrils originating from the cell membrane. Amyloid nodules, surrounded by mononuclear cells, developed from this stage. In the nodules, fibrils were positive for anti-SAA only in the vicinity of the cell membrane, while anti-AA stained fibrils throughout. Our hypothesis for fibrillogenesis is thus as follows: Serum SAA2 is specifically deposited on mononuclear cells in the marginal zone and polymerized extracellularly into fibrils, retaining its antigenicity (SAA2 amyloid fibrils); these fibrils are then processed to AA amyloid fibrils in situ by cleavage of the C-terminal portion of SAA2.

Follicular lymphomatoid papulosis.

In two patients who episodically developed eruptions of papules and nodules for many years, the lesions developed at the sites of hair follicles or around ruptured follicular cysts. They were pseudomalignant lymphomas in histologic appearance and benign in biologic behavior. From its limitation largely to follicular adnexa, the condition appears to be a distinct type of lymphomatoid papulosis.

Multiple malignant eccrine poroma and a linear epidermal nevus.

A 68-year-old woman developed three reddish nodules on a linear epidermal nevus on the right arm. The nodules and linear lesion were resected. The histology revealed the two larger nodules to be eccrine porocarcinoma and the other to be eccrine poroepithelioma. The linear lesion was actually epidermal nevus. To our knowledge, this is the first description of adnexal tumors developing on epidermal nevus.

A new species of Devosia that forms a unique nitrogen-fixing root-nodule symbiosis with the aquatic legume Neptunia natans (L.f.) druce.

Rhizobia are the common bacterial symbionts that form nitrogen-fixing root nodules in legumes. However, recently other bacteria have been shown to nodulate and fix nitrogen symbiotically with these plants. Neptunia natans is an aquatic legume indigenous to tropical and subtropical regions and in African soils is nodulated by Allorhizobium undicola. This legume develops an unusual root-nodule symbiosis on floating stems in aquatic environments through a unique infection process. Here, we analyzed the low-molecular-weight RNA and 16S ribosomal DNA (rDNA) sequence of the same fast-growing isolates from India that were previously used to define the developmental morphology of the unique infection process in this symbiosis with N. natans and found that they are phylogenetically located in the genus Devosia, not Allorhizobium or RHIZOBIUM: The 16S rDNA sequences of these two Neptunia-nodulating Devosia strains differ from the only species currently described in that genus, Devosia riboflavina. From the same isolated colonies, we also located their nodD and nifH genes involved in nodulation and nitrogen fixation on a plasmid of approximately 170 kb. Sequence analysis showed that their nodD and nifH genes are most closely related to nodD and nifH of Rhizobium tropici, suggesting that this newly described Neptunia-nodulating Devosia species may have acquired these symbiotic genes by horizontal transfer.

Progressive hypereosinophilia with lung nodules due to thyroid carcinoma.

An elderly woman had dyspnea, a thyroid mass and multiple lung nodules on chest radiographs. She developed hypereosinophilia over a three-month period in which multiple diagnostic procedures were performed, without definition of the disease process. At post-mortem examination, the primary lesion was found to be a poorly differentiated carcinoma of the thyroid, metastatic to lung. This is the second case of hypereosinophilia related to metastatic thyroid carcinoma. The intensity of the hypereosinophilia in this case was increased by the administration of thyroid-stimulating hormone and by the development of necrosis of the lung nodules.

Prospective study of laparoscopic findings with regard to the development of hepatocellular carcinoma in patients with hepatitis C virus-associated cirrhosis.

BACKGROUND: The laparoscopic findings of hepatocarcinogenesis are not well defined. The purpose of this prospective study was to evaluate the predictive value of laparoscopic findings with regard to the development of hepatocellular carcinoma (HCC) in patients with hepatitis C virus (HCV)-associated cirrhosis. METHODS: One hundred nineteen patients with HCV-associated cirrhosis who underwent laparoscopy were enrolled in this study. Ten laparoscopic variables, including degree of development and size of regenerating nodules, irregularity of regenerating nodules, and size of both hepatic lobes, were measured. The predictive value of each finding for the development of HCC was investigated by using univariate and multivariate analyses. RESULTS: HCC developed in a total of 42 of the 119 patients (35.3%) during a mean follow-up period of 62.9 months. The degree of regenerating nodules, the presence of irregular regenerative nodules, and atrophic right lobe were significant predictive factors for HCC with univariate analysis. In particular, 67.1% of patients with irregular regenerative nodules had HCC develop within 5 years. Multivariate analysis revealed that irregular regenerative nodules (relative risk 6.32, p = 0.012), the degree of regenerative nodules (relative risk 4.78, p = 0.029), and atrophic right lobe (relative risk 3.87, p = 0.048) were independent predictive factors. CONCLUSIONS: Morphologic observation of the liver surface by laparoscopy is important and provides information on factors that are statistically significant early predictors of the development of HCC.

Localization of Bacteria and Hemoglobin in Root Nodules of Parasponia andersonii Containing Both Bradyrhizobium Strains and Rhizobium leguminosarum biovar trifolii.

Dual occupancy of Parasponia andersonii nodules with different Bradyrhizobium strains and Rhizobium leguminosarum biovar trifolii was frequently obtained when two strains were inoculated into plants grown aseptically in tubes. Since reisolates of Bradyrhizobium strains from dually occupied nodules acquired the ability to nodulate Trifolium repens, the spatial relationship of the two species of bacteria during nodule initiation and development was investigated and their proximity was demonstrated. By using light microscopy and electron microscopy and immunogold labeling, R. leguminosarum biovar trifolii NGR66 inoculated alone onto P. andersonii produced small ineffective nodules, with bacteria embedded in matrix material in intercellular spaces and in a few nonliving host cells rather than in infection threads (CP299). In dual infections, the two bacterial species were shown to be adjacent to one another in the matrix of nodule intercellular spaces and in some host nodule cells. However, when two different Bradyrhizobium strains occupied a single nodule, they were located in different lobes of the same nodule. Immunogold labeling showed that Parasponia hemoglobin was localized in the cytoplasm of young infected nodule cells. This suggests that the nitrogen-fixing phase of Parasponia nodule cells is short-lived and correlates with previous acetylene reduction data from nodule slices. Hemoglobin was associated only with areas of nodule tissue infected with the effective nitrogen-fixing strain CP299 and absent from areas infected with R. leguminosarum biovar trifolii.
