Combination and cleavage of HBV DNA fragments by triple helix-forming oligonucleotides modified with manganese porphyrin in vitro.

OBJECTIVE: To observe the ability of triple helix-forming oligonucleotides (TFOs) modified with manganese porphyrin to combine with and cleave HBV DNA fractions. METHODS: TFO were modified with manganese porphyrin and acridines, and then reacted with the (32)P labeled HBV DNA fragments at 37 degrees C in vitro (pH 7.4). Electrophoretic mobility shift assays and DNase I footprinting tests were used to show the affinity and specificity of TFO to bind to target sequences. The ability of TFO to cleave HBV DNA fragments was tested by cleavage experiments. RESULTS: TFO modified with manganese porphyrin and acridine could bind to the target sequence in a sequence-dependent manner, with a Kd value of 3.5 x 10(-7) mol/L and a relative affinity of 0.008. In the presence of potassium monopersulfate (KHSO(5)), TFO modified with manganese porphyrin and acridine could cleave the target sequence where the triplex DNA was formed. CONCLUSION: In the presence of KHSO(5), TFO modified with manganese porphyrin and acridine could bind and cleave the target HBV-DNA in a sequence-dependent manner.

[Voltammetric behaviors of diethylstilbestrol and its determination at multi-wall carbon nanotubes modified glassy carbon electrode].

AIM: To fabricate multi-wall carbon nanotube (MWNT) modified electrode and study the electrochemical behaviors of diethylstilbestrol at the MWNT-modified electrode. METHODS: Cyclic voltammetry and linear sweep voltammetry. RESULTS: The oxidation peak current of diethylstilbestrol increased remarkably and the peak potential shifted negatively at the MWNT-dihexadecyl hydrogen phosphate (DHP) modified glassy carbon electrode (GCE), in contrast to that at the bare GC electrode and DHP-modified GC electrode. The oxidation peak current is linear with the concentration of diethylstilbestrol over the range from 1 x 10(-8) to 2 x 10(-6) mol.L-1. The detection limit was 2.5 x 10(-9) mol.L-1. The relative standard deviation (n = 10) was 2.9% for 1 x 10(-6) mol.L-1 diethylstilbestrol. CONCLUSION: The MWNT-DHP modified GCE exhibits catalytic activity to the oxidation of diethylstilbestrol.

[Modified FLAG regimen in the management of refractory acute myeloid leukemia].

OBJECTIVE: To evaluate the therapeutic effect of modified FLAG regimen in the management of refractory acute myeloid leukemia (AML). METHODS: Sixteen patients with refractory AML were divided into two groups. In modified FLAG regimen group (n=10), the patients received fludarabine (Flu, 50 mg/d, VDx5 d) and Ara-c (200 mg/d, VDx5 or 7 d). The regimen for classic FLAG group (n=6) consisted of Flu (50 mg/d, VDx5d), Ara-C (500 or 1,000 mg/d, VDx5d) and G-CSF (300 microg/d, x5 d, subcutaneously injected 4-6 hours before chemotherapy). Each patient received subcutaneous G-CSF (300 microg/d) when the white blood cell count was lower than 1.0x10(9)/L till the condition was corrected. RESULTS: The total complete remission(CR) rate of the 16 patients was 50% (8/16). Seven patients in modified group achieved CR (70%) and only one of the classic group did (17%, P<0.05). Episodes of infections were lower in modified group than in the classic group (50% vs 83%). CONCLUSION: Modified FLAG regimen is more likely than classic FLAG regimen to achieve CR and reduce infections in patients with refractory AML.

[Effect of Tpo and/or IL-11 gene modified stromal cells on the expansion of CD34+ CD38- hematopoietic primitive progenitor cells].

OBJECTIVE: To investigate the effects of Tpo and/or IL-11 gene modified stromal cells on the expansion of CD(34)(+) hematopoietic stem/progenitor cells in cord blood. METHODS: Retroviral vectors containing Tpo or IL-11 gene were constructed and used to transfect the stromal cell line HFCL. Tpo and/or IL-11 mRNA was assayed by Northern blot. Non-modified stromal cells were used, CD(34)(+) hematopoietic stem/progenitor cells from cord blood were expanded on gene-modified stromal cells for 7 days. The phenotype of CD(34)(+)CD(38)(-) primitive progenitors was detected by flow cytometry. RESULTS: HFCL expressed Tpo and/or IL-11 mRNA after transfected by the retroviral vectors. The percentages of CD(34)(+)CD(38)(-) primitive progenitors in the cultures of Tpo, IL-11 and Tpo + IL-11 modified HFCL were (1.8 +/- 0.24)%, (1.62 +/- 0.23)%, and (2.45 +/- 0.28)%, respectively, which were higher than that in the control [(0.8 +/- 0.23)%]. CONCLUSION: The stromal cells modified by Tpo and/or IL-11 gene were able to enhance ex vivo expansion of CD(34)(+) and CD(34)(+)CD(38)(-) hematopoietic stem/progenitor cells from cord blood.

[Experimental study for repair of cranial defects with bone marrow stromal cells and modified alginate].

OBJECTIVE: The primary aim of this investigation was to determine whether expanded BMSCs in vitro mixed with modified alginate gelatin could repair critical defects in rats without the addition of exogenous growth or bone morphogenetic factors. METHODS: Bone marrow stem cells from syngeneic rats cultured in vitro and mixed with modified alginate gel to paint the cranial critical size defect. A full-thickness cranial plate defect was created without damage of dura mater. Modified alginate gelatin with or without BMSCs were painted over the cranial defects. Animals being made cranial defect but received no implant served as sham-operated controls. Craniotomy defects were divided into three groups, which included defects left unpainted (group I, n=6), defects painted with modified alginate gelatin alone (group II, n=6), and defects painted with a modified alginate mixed with BMSCs (group III, n=6). A total of 18 implant experiments were carried out, with postsurgical radiographic and histological analysis completed at 12 week. RESULTS: None of the implants exhibited extrusion or infection. Radiographs showed a likely increased calcification in group III, without finally new calcification in group I and in group II. Histology showed that group I and group II were featured by thinning of the bone at the edges of the defect margins with minimal bone growth inward and dense fibrous tissue with rudimentary alginate material spanning the intervenient gap. The results demonstrated that a great amount of new bone in growth took place in BMSCs-alginate group, stemming from cranial defect edges and proceeding inward. CONCLUSION: Transplantation of syngeneic BMSCs with alginate gel can serve as an example of a cell-based treatment for skeletal reformation and would be especially useful for augmenting or regenerating bone in skeletal defects. So syngeneic BMSCs with alginate gel demonstrate a potential technique to regenerate a variety of skeletal defects that occur in different clinical scenaries.

[The enhancing effect of tomato lectin modified liposomes of insulin on oral absorption in mice].

AIM: To investigate the enhancing effect on insulin absorption through GI tract in mice by using tomato lectin (TL) modified liposomes as the carrier. METHODS: TL-phosphatidylethanolamine (PE) conjugate (TL-PE) was synthesized by using carbodiimide cross-linking method, then the compound was incorporated into the conventional liposomes of insulin. The agglutination test was performed to examine TL biological activities after synthesis and incorporation. When TL modified liposomes were administrated orally to the normal mice or diabetic mice at insulin dose of 350 u x kg(-1), the hypoglycemic effect was determinated according to the blood glucose level. RESULTS: The blood glucose levels of the normal mice were reduced by modified liposomes. The glucose levels were (85 +/- 5)% at 4 h, (54 +/- 11)% at 8 h, (57 +/- 6)% at 12 h postdose compared with the glucose levels prior to oral administration respectively. However, the conventional liposomes and saline have no hypoglycemic effect. The blood glucose levels of the diabetic mice were obviously reduced by TL modified liposomes, the glucose levels were (38 +/- 13)% at 4 h, (50 +/- 15)% at 8 h, (50 +/- 16)% at 12 h respectively. CONCLUSION: TL modified liposomes promote the oral absorption of insulin due to the specific-site combination on GI cell membrane.

Standardization of mPEG-modified allergens.

Standardization of mPEG-modified allergens can be achieved by: standardization of allergens according to established principles; production of mPEG-modified allergens by a reproducible method; control of the batch-to-batch consistency by estimation of the allergenicity and the protein content in relation to the corresponding IHR and by determination of the degree of modification by estimation of the amount of PEG per unit allergen in the modified product. Standardization of mPEG-modified allergens should be looked upon only as a model for standardization of modified allergens. I hope that the authorities in Europe will set up strict and relevant criteria for the registration of similar products and also criteria for prolonging the registration of the older products without relevant standardization and documentation of safety and efficacy.

The Systemic Lupus Activity Measure-revised, the Mexican Systemic Lupus Erythematosus Disease Activity Index (SLEDAI), and a modified SLEDAI-2K are adequate instruments to measure disease activity in systemic lupus erythematosus.

OBJECTIVE: To assess the validity, reliability, and feasibility of the Systemic Lupus Activity Measure-Revised (SLAM-R), the Mexican version of the Systemic Lupus Erythematosus Disease Activity Index (MEX-SLEDAI), and a Modified SLEDAI-2000 (SLEDAI-2K) compared with the SLEDAI-2K in a multiethnic population of patients with SLE. METHODS: We studied 92 SLE patients from 3 US geographic areas (Alabama, Texas, and Puerto Rico). Assessment occurred during regular outpatient, inpatient, or study encounters. A trained physician scored the 4 instruments and also assessed disease activity globally [physician global assessment (PGA)]. Convergent (with SLEDAI-2K) and construct validity (with PGA) were determined by Spearman rank (rs) correlation test. Level of agreement between the instruments was assessed using Bland-Altman plots. Discriminant validity (distinguishing clearly active vs mildly/nonactive disease) was assessed considering the SLEDAI-2K (and the PGA) as the gold standard. Feasibility was explored by cost analyses. RESULTS: The SLAM-R, the MEX-SLEDAI, and the Modified SLEDAI-2K were highly correlated with the SLEDAI-2K (rs = 0.566, 0.755, 0.924, respectively) and with the PGA (rs = 0.650, 0.540, 0.634, respectively). The 3 instruments showed good agreement with the SLEDAI-2K (Bland-Altman plots). The Modified SLEDAI-2K had better discriminant validity than the SLAM-R and the MEX-SLEDAI. The Modified SLEDAI-2K was the least expensive instrument. CONCLUSION: The SLAM-R, the MEX-SLEDAI, and the Modified SLEDAI-2K are adequate options for assessment of SLE disease activity; they are also less costly than the SLEDAI-2K.

Modified Middlebrook 7H9 broth for the rapid detection of mycobacteria.

OBJECTIVE: To compare the growth of acid-fast bacilli on classically formulated Lowenstein-Jensen media and Middlebrook 7H11 agar to modified Middlebrook 7H9 broth. DESIGN: Microbiology culture results were evaluated to determine the number of days required to detect growth of acid-fast bacilli from clinical specimens inoculated on different media. DATA: Three thousand one hundred fifty-seven acid fast bacilli cultures were performed from May 1996 through May 1999. There were 140 positive cultures. Initial growth was detected only on Lowenstein-Jensen medium and/or Middlebrook 7H11 agar for 108 cultures, Lowenstein-Jensen medium and/or Middlebrook 7H11 agar and 7H9 for 12 cultures, and only modified Middlebrook 7H9 broth for 20 cultures. Of the 20 cultures that were initially detected only in 7H9, 11 grew in a mean time of 21.1 days compared with 32.5 days for the later growing solid media. The remaining nine grew in a mean time of 26.0 days but did not grow on solid media within the eight-week incubation period. Mycobacterium avium grew in a higher proportion of broth than solid medium cultures, suggesting a preference for the broth. CONCLUSION: The modified Middlebrook 7H9 broth was inexpensive to prepare, allowed for quick and easy growth interpretation, required no special appliances, e.g., ultraviolet light, and allowed for faster growth of some cultures. Although Lowenstein-Jensen medium and Middlebrook 7H11 agar remained the main-stays of our routine acid fast bacilli culture protocol, modified Middlebrook 7H9 broth provided earlier or additional positive results in 14% of the positive cultures. We recommend the use of modified Middlebrook 7H9 broth as described in this paper for improving early detection of acid fast bacilli by culture.

[Detection of severe acute respiratory syndrome probable patients' virus RNA in Hangzhou by using a two loci and a modified nested real-time reverse transcription polymerase chain reaction].

OBJECTIVE: To detect the RNA of severe acute respiratory syndrome virus (SARS-CoV) by using reverse transcription polymerase chain reaction (RT-PCR) targeted for a two loci and a modified nested real-time RT-PCR as to improving the reliability and sensitivity of tests. METHODS: A nested RT-PCR was used for detecting one fragment of SARS-CoV RNA in oropharyngeal swabs from 3 SARS probable patients, 4 SARS suspect patients and other 27 patients with fever in Hangzhou, and the nested RT-PCR product from one SARS probable patient was sequenced. Meanwhile in these 3 SARS probable patients, other three RT-PCR methods, including a hemi-nested RT-PCR targeted for another fragment of SARS-CoV RNA, a real-time RT-PCR and a modified nested real-time RT-PCR, were employed to detect SARS-CoV RNA. RESULTS: Two positives were found in the 3 SARS probable patients, and none positive in 4 SARS suspect patients and other 27 patients with fever, using the nested RT-PCR. The sequence of the nested RT-PCR product from one SARS probable patient was identified with the counterpart of SARS-CoV genomes published in public database. The results of the hemi-nested RT-PCR, the real-time RT-PCR and the modified nested real-time RT-PCR in the 3 SARS patients were consistent with the one of the nested RT-PCR. During detecting specimen with low copies of RNA, a weak positive signal was produced after about 35 cycles in the real-time RT-PCR, but a strong positive signal was found only after 10 cycles in the modified nested real-time RT-PCR. CONCLUSION: It might improve the reliability of test by employing RT-PCR targeted for two or more fragments in SARS-CoV genome. The modified nested real-time RT-PCR might have higher sensitivity than the routine real-time RT-PCR.

Modified magnetic resonance angiography of the liver using sensitivity encoding in comparison with digital subtraction angiography and CT arterial portography.

BACKGROUND: Over 355 patients have received orthotopic liver transplantation (OLT) at this hospital since 1993. Preoperative imaging studies of both hepatic vessels and parenchyma in these recipients bettered surgical planning or even precluded the necessity of surgery. Here we report our preliminary results of modified magnetic resonance angiography (MRA) using sensitivity encoding (SENSE) through comparative study with conventional digital subtraction angiography (DSA) and CT arterial portography (CTAP). METHODS: Sixteen patients with suspected liver diseases were included in the study. All of them received both dynamic MRI of the liver using SENSE and digital DSA with CTAP within a two-week interval. The four-phase MRA was reconstructed from source images of the coronal dynamic study. The arterial phase of the modified MRA was compared with DSA in the evaluation of hepatic arteries and the portal phase compared with CT portography reconstructed from source images of CTAP. In dynamic study of the liver, a fixed dose (20 ml) of contrast medium and scan timing were used. RESULTS: The main branches and variations of the hepatic arterial system were well shown on the modified MRA, although the marginal branches of hepatic arteries were of poor quality. The figures of portal veins on MRA were as clear as or superior to those of CTAP. In addition, the suprarenal inferior vena cava (IVC) was well demonstrated on MRA and/or non contrast-enhanced coronal balanced fast-field echo (b-FFE) scan sequence in most cases. MRI detected most parenchymal lesions of the liver and hemodynamics of these lesions could be evaluated on source images of the modified MRA. MRI/MRA also serendipitously revealed several extrahepatic disease entities or variations that were not found on DSA/CTAP. CONCLUSIONS: The modified MRA using SENSE is a cost-effective modality of examination for the demonstration of the whole hepatic vascular system. Combined with MRI, it has the potential as a one-stop imaging modality in the preoperative evaluation in fields such as OLT.

[Surgical technique of modified free vascularized fibular grafting for treatment of osteonecrosis of the femoral head].

OBJECTIVE: To evaluate the surgical technique, clinical results, and the complications of modified free vascularized fibular grafting for the treatment of osteonecrosis of the femoral head. METHODS: From October 2000 to August 2004, 124 patients (139 hips) with osteonecrosis of the femoral head were treated with modified free vascularized fibular grafting. There were 83 males(93 hips) and 41 females (46 hips), with a mean age of 36.4 years (16-57). The disease was caused by trauma in 49 cases(54 hips), use of steroids in 29 cases (32 hips), consumption of alcohol in 19 cases (21 hips) and idiopathic condition in 27 cases (32 hips). Of 139 hips, 50 were classified as stage II ; 71 as stage II, 18 as stage IV according to Steinberg system; the Harris hip scores were 79.3, 69.3 and 58.4, respectively. At the operation, modified technique of the fibular osteotomy was adopted. A front-hip operative approach was designed and a modified technique of removing the necrotic bone in femoral head was applied. During operation, the duration of operation, the bleeding volume, and the length of incisions were recorded. The follow-up items included the results of X-ray examination, the Harris score of the hip, and the evaluation of the complications. RESULTS: The duration of the fibular osteotomy was 10 to 30 min (15 min on average). The duration of the total operation was 80 to 120 min (90 min on average). The length of incision at the hip was 6 to 12 cm (8 cm on average). The bleeding volume was 100 to 300 ml (200 ml on average). The average hospitalization days was 7 days. After operation, Harris hip scores in most cases were improved. According to postoperative X-ray, 62 hips (79.5%) were improved to different extents and 14 hips (17.9%) had no significant changes. Deterioration occurred in 2 hips (2.6%). CONCLUSION: The modified free vascularized fibular grafting has lots of virtues, such as less bleeding volume, more clear anatomic structure, more convenience for operation, less damage, less complications, and better results of function recovery. It is an effective method for treating osteonecrosis of the femoral head.

[A study on cytocompatibility of poly (lactic acid) membrane modified by polymer microspheres with different surface charges].

Poly(lactic acid) microspheres with different surface charges have been prepared by using cationic, anionic or nonionic surfactants as the microspheres' surface stabilizers. Embedded with these microspheres, the modified PLA membranes with different surface charges have been obtained. The test of stability by CLSM and the morphological test by SEM confirmed that we obtained the microspheres modified PLA membranes with different surface charges successfully. The chondrocyte compatibility test of these modified PLA membranes showed that the attachment, proliferation and activity of chondrocytes on the positive surface of the modified PLA were better than those of other modified PLA membranes. The positive charge on the surface of PLA membrane could improve the cell-compatibility of PLA well.

[Modified guanidine hydrochloride method for DNA extraction from cord blood used in HLA genotyping].

To compare two different methods for extracting genomic DNA from cord blood and to evaluate their applications for HLA genotyping, the genomic DNA from 72 samples was extracted by guanidine hydrochloride (Gu * HCl) and modified guanidine hydrochloride, the DNA yield and purity were evaluated by spectrophotometry and detected by PCR with sequence-specific primers. The result showed that the genomic DNA was successfully isolated from whole blood by both methods. The modified Gu * HCl method used was better than Gu * HCl method as the modified method produces better quality of DNA and less ambiguous bands in PCR. It is concluded that modified Gu * HCl method has the advantages of low-cost, simple operation, high quality output and clear positive bands in HLA-genotyping, the modified method is optimal for extracting DNA from multiple samples of cord blood bank.

[Preparation of amylopectin modified dipyridamole liposome and its tissue distribution in mice].

AIM: To prepare amylopectin anchored dipyridamole (DIP) liposome and to study its tissue distribution in mice. METHODS: The regular DIP liposomes were prepared by film-scatter method. The amphiphilic O-palmitoyl amylopectin was synthesized and added to modify the surface of liposome. The entrapping efficiency, zeta potential, mean diameter, span of modified and regular liposomes were assayed. The RP-HPLC was used for the determination of DIP concentration in mice tissue. RESULTS: After modification, the entrapping efficiency depressed, zeta potential was raised, mean diameter and span had no obvious change. The level of DIP in lung, liver and spleen for regular liposomes were higher than that of injections. Compared with regular liposomes, the modified liposomes increased the DIP level in lung, and decreased the DIP level in liver, spleen, moreover, lengthened the retention time of DIP in lung. CONCLUSION: The distribution of modified liposome in mice was markedly changed as compared with regular liposomes and injections. The modified liposomes had obvious lung targeting property.

[Therapeutic effects of treatment of pancreatic carcinoma by interleukin-23 and apoptotic cell antigen-modified dendritic cell vaccine: experiment with mice].

OBJECTIVE: To investigate the therapeutic effects on pancreatic carcinoma by dendritic cells (DCs) transfected with interleukin (IL)-23 and acquiring apoptotic cell antigen. METHODS: Murine IL-23 cDNA was subcloned into the vector so as to construct the eukaryotic dual-gene repression vector pcDNA3-IL-23. Bone-derived DCs were obtained from the long bones of extremities of normal C(57)BL/6 mice and were transfected with pcDNA3-IL-23. DCs cultured routinely and IL-23-transfected DCs were co-cultured with apoptotic tumor cells for 24 h so as to collect sensitized DCs acquiring the antigen of the apoptotic cells. Primary pancreatic carcinoma models were created in C(57)BL/6 mice with dimethyl-benzanthracene so as to obtain suspension of tumor cells. Thirty C(57)BL/6 mice were randomly divided into 5 groups: Group I (IL-23-transfected DC vaccine group, to be injected subcutaneously with IL-23 and apoptotic cells-modified DC vaccine twice with an interval of 7 d), Group II (apoptotic cells-sensitized DC vaccine group, to be injected subcutaneously with apoptotic cells-modified DC vaccine), Group III (IL-23 transfected DC group, to be injected with IL-23 transfected DCs), Group IV (un-modified DC group, to be injected with un-modified DCs), and Group V (control group, injected with normal saline). Seven days after the second injection suspension of tumor cells was injected subcutaneously, and then tumorigenesis was observed every other day. Another 30 C(57)BL/6 mice were divided into 5 groups as above-mentioned (Groups I - V(1)) and were killed 1 week after the inoculation of tumor cell suspension. Their spleens were taken out. The cytotoxic lymphocytes (CTLs) were isolated, co-cultured with IL-25 and apoptotic cells, and then tested for the secretion of interferon (IFN)-gamma and IL-4 by ELISA. Thirty C(57)BL/6 mice loading pancreatic carcinoma were randomly divided into 5 groups as above-mentioned (Groups I - V(2)) so as to observe the survival time. RESULTS: ELISA showed that the IFN-gamma expression in the supernatant of culture fluid of IL-23 transfected DCs in Groups I(1) was significantly up-regulated (P < 0.01), and the IL-4 secretion of Group I(1) and Groups III(1) were both significantly lower than that of Group II(1) (both P < 0.05). The CTL civilities of Group I(1) and II(1) were both significantly higher than those of the other 3 groups (all P < 0.05). Four weeks after the inoculation tumorigenesis was not seen in Group I and was seen in Groups III and IV with an incidence of 33% - 50%. Tumorigenesis occurred only 1 week later in Group V. The tumor sizes of Groups I(2) and II(2) were both significantly smaller than those of the other 3 groups and the survival times of Groups I(2) and II(2) were both significantly lower than those of the other 3 groups (all P < 0.01). CONCLUSION: IL-23 helps enhance the presenting ability of DC antigen. IL-23 modified vaccine strengthens the immune response of CTLs against specific tumor, thus inducing defensive immune response in the host and strengthening their active immune ability.

[First experience with the modified program NHL-BFM-90 application in adult patients with primary diffuse large B-cell gastric lymphosarcoma with unfavourable prognosis].

AIM: To assess efficacy of a modified program NHL-BFM-90 in adult patients with primary diffuse large B-cell gastric lymphosarcoms (PDLBGL) with unfavourable prognosis. MATERIAL AND METHODS: Modified courses of NHL-BFM-90 were conducted in 5 patients aged 27-67 years from January 2004 to September 2005. Four patients received chemotherapy of the first line, in one patient block therapy followed monotherapy with chlorambucil and a CHOEP course. All the patients were in a severe clinical condition and had several initial factors of unfavourable prognosis: size of the tumor more than 10 cm; stage IE and more advanced; B-symptoms; proliferative activity above 70%. The program NHL-BFM-90 was modified because of the patients' age. Chemotherapy was conducted according to the middle arm of the original program NHL-BFM-90, but methotrexate was introduced in a dose 1 g/m2 for 12 hours, while leukovorin was given 18 hours after the start of methotrexate injection. In two cases the blocks were enhanced with rituximab, 2 patients had doxorubicin in block A, in one case block C was enhanced with methotrexate. A total of 23 modified blocks NHL-BFM-90 were performed: one patient was given 6 blocks, two patients--5, one patient--4 blocks and one patient--3 blocks. RESULTS: Four patients after block 2 and one patient after block 3 of polychemotherapy NHL-BFM-90 achieved remission of the disease of 6 to 22 months duration which still continues. Infectious complications related to hematological toxicity arose more frequently at the latest courses of chemotherapy. CONCLUSION: Treatment according to the modified program NHL-BFM-90 in adult patients with PDLBGL and unfavourable prognosis is highly effective. For a mean follow-up of 10.2 months no recurrences occurred. The number of courses can be reduced to decrease accumulated hematological toxicity and in case of rapid achievement of remission.

[Sleeve circumcision and preputioplasty with modified incision].

OBJECTIVE: To evaluate the sleeve circumcision and preputioplasty with modified incision for the treatment of patients with phimosis or redundant prepuce. METHODS: Five hundred and seventy-six patients with phimosis or redundant prepuce underwent operations of sleeve circumcision or preputioplasty with modified incision. The conventional incision was modified and changed into two opposite tortuous incisions. RESULTS: The operation with modified incision had the following advantages: less blood loss, slight postoperative edema, no secondary bleeding or infection, quick recovery and good appearance of the penis. CONCLUSION: The sleeve circumcision and preputioplasty with modified incision is an excellent therapeutic option for phimosis and redundant prepuce.
