ScienceLabDatabase: a computer program to organize a molecular biology laboratory.

A description of a novel laboratory management software is provided. ScienceLabDatabase (SLD) offers a useful platform to organize a molecular biology research laboratory. The program manages stocks of biological samples, including plasmids, antibodies and cell lines, laboratory protocols and addresses, and it includes an easy ordering and funds managing system. Preformed data sheet templates help to store and maintain valuable information on samples or reagents and to facilitate the transfer of accurate information between researchers. Password protection regulates access, and simple button functions allow the use of this system without prior database knowledge. SLD is based on FileMaker Pro Version 4.0, which allows easy customization and import of preexisting data from other applications. The SLD program was successfully tested in several independent research groups and proved a useful tool to efficiently organize a molecular biology laboratory.

PREDICTOL: a computer program to determine the thermophysiological duration limited exposures in various climatic conditions.

PREDICTOL is a PC program used to determine the thermophysiological duration limited exposures (DLE) in humans, nude or clothed, submitted to various climatic conditions (hot and cold climates) at rest or during a physical exercise. DLE are determined following different standards of the International Standardization Organization (ISO), especially ISO 7933 for hot environment and ISO-TR 11079 for cold environment. The original aspect of this program is that it can be used whatever the climatic conditions. The program presents two modes: an educational interactive mode and a scenario mode. The educational interactive mode demonstrates the thermophysiological effects, expressed as DLE, of different parameter changes (temperature, humidity, wind speed, metabolic heat production by physical exercise, clothing insulation and water vapor permeability). The scenario mode determines DLE for given various linked sequences as encountered in occupational, military or even recreational activities, each sequence being characterized by its climatic conditions, physical activities performed and by physical clothing properties. DLE given by PREDICTOL are correlated to those obtained in various controlled climatic laboratory conditions (r = 0.86; P < 0.001). PREDICTOL is written in Visual Basic 6.0. A "help menu" is provided to explain the use of the program and give information concerning the equations used to calculate both the thermal balance and DLE.

Increasing reading and communication skills in children with autism through an interactive multimedia computer program.

This paper reports on the effect of using an interactive and child-initiated microcomputer program (Alpha) when teaching three groups of children (N = 30) reading and communications skills: (a) 11 children with autism (M chronological age, CA = 9:4 years), (b) 9 children with mixed handicaps (M CA = 13:1), and (c) 10 normal preschool children (M CA = 6:4 years). Their mental age varied from 5:8 years to 6:9 years and all children received computer instruction supplementary to their regular reading and writing activities. Tests of reading and phonological development were carried out at the onset of the training (Start), at the end (Post 1), and at a follow-up evaluation (Post 2). In addition, video observations of the childrens' verbal and nonverbal communication were added at Start and Post 1. The children with autism increased both their word reading and their phonological awareness through the use of the Alpha program. Clearly significant gains were observed during the intervention, but none during the follow-up period. A similar but weaker pattern is observed for the children with mixed handicaps. In contrast, the normal preschool children increased their scores regardless of the program. Analyses of the children's classroom behavior indicate that the intervention succeeded in stimulating verbal expressions among the children with autism and mixed handicap. A significant increase in enjoyment was also noted for the children with autism. It is concluded that the intervention with a motivating multimedia program might stimulate reading and communication in children with various developmental disabilities, but that such interventions must be individually based and include both detailed planning and monitoring from teachers, and parents, as well as from clinicians in charge.

ESTRIP, a BASIC computer program for obtaining initial polyexponential parameter estimates.

A new BASIC exponential stripping program, ESTRIP, allows the relatively rapid calculation of initial polyexponential parameter estimates, as does the previously published FORTRAN IV program, CSTRIP. The potential advantages of the new program are that it can be run on microcomputers and minicomputers with BASIC capability and a relatively small core and that it can be easily modified by the user.

A computer program for the analysis of crossover designs.

We describe a program, CROSS, which we have written to obtain potency estimates and other parameters for bioassay data from assays of crossover design. The program permits testing of all assays for statistical validity and calculates the complete analysis of variance for assays of balanced design. The form of data input and the complete documentation of assay results make this program particularly useful for anyone carrying out crossover assays on a routine basis. The analysis of variance presented is also useful for more general biological or medical situations.

MINMOD Millennium: a computer program to calculate glucose effectiveness and insulin sensitivity from the frequently sampled intravenous glucose tolerance test.

The Bergman Minimal Model enables estimation of two key indices of glucose/insulin dynamics: glucose effectiveness and insulin sensitivity. In this paper we describe MINMOD Millennium, the latest Windows-based version of minimal model software. Extensive beta testing of MINMOD Millennium has shown that it is user-friendly, fully automatic, fast, accurate, reproducible, repeatable, and highly concordant with past versions of MINMOD. It has a simple interface, a comprehensive help system, an input file editor, a file converter, an intelligent processing kernel, and a file exporter. It provides publication-quality charts of glucose and insulin and a table of all minimal model parameters and their error estimates. In contrast to earlier versions of MINMOD and some other minimal model programs, Millennium provides identified estimates of insulin sensitivity and glucose effectiveness for almost every subject.

Computer program for determining fluorescence resonance energy transfer efficiency from flow cytometric data on a cell-by-cell basis.

The determination of fluorescence resonance energy transfer (FRET) with flow cytometry (FCET) is one of the most efficient tools to study the proximity relationships of cell membrane components in cell populations on a cell-by-cell basis. Because of the high amount of data and the relatively tedious calculations, this procedure should be assisted by powerful data processing software. The currently available programs are not able to fulfill this requirement. We developed a Windows-based program to calculate fluorescence resonance energy transfer efficiency values from list mode flow cytometry standard (FCS) files. This program displays the measured data in standard plots by generating one- and two-parameter histograms on linear or logarithmic scales. A graphical gating tool allows the user to select the desired cell population according to any combination of the parameter values. The program performs several statistical calculations, including mean, S.D., percent of the gated data. We have implemented two types of data sheet for FRET calculations to aid and guide the user during the analysis: one with population-mean-based autofluorescence correction and the other with spectrum-based cell-by-cell autofluorescence correction. In this paper, we describe the gating algorithms, the file opening procedure and the rules of gating. The structure of the program and a short description of the graphical user-interface (GUI) are also presented in this article.

An interactive computer program for randomization analysis of response curves with facilities for multiple comparisons.

An interactive Fortran program, MUCRA, is presented. The program can perform randomization analysis of a completely randomized or randomized-blocks design extended to growth and response curves. A single-step Scheffé-type procedure as well as the Peritz's closed step-down procedure have been implemented which control the familywise type I error-rate. In general, MUCRA is suitable as a computer tool for a distribution-free analysis of variance with repeated measures. The use of MUCRA is demonstrated by analyzing the effects oxprenolol and atenolol have on exercise heart rate. Oxprenolol is a non-selective beta-blocker with moderate intrinsic sympathomimetic activity (ISA), given by the Oros delivery system. Atenolol is a beta 1-selective blocker without ISA. A randomized placebo-controlled crossover design was used to compare the effects of the beta 1-blockers on heart rate during a progressive maximal exercise test on a bicycle ergometer. Application of the Scheffé-type procedure showed that the two drugs significantly (alpha = .05) reduce the heart rate during the exercise test at the three prechosen times (2, 5, and 24 hr) after intake. The reduction from atenolol is more pronounced than from oxprenolol Oros at 2 and 5 hr.

HYDRO: a computer program for the prediction of hydrodynamic properties of macromolecules.

HYDRO is a program for the calculation of sedimentation and diffusion coefficients, rotational relaxation times, and intrinsic viscosities of rigid macromolecules of arbitrary shape that are represented by bead models. Actually, HYDRO contains various FORTRAN callable subroutines that can be linked to the user's own programs to account for variability of shape or flexibility. Some hints are given for the use of HYDRO in various situations.

Development and evaluation of a CD-ROM computer program to teach residents telephone management.

OBJECTIVE: Under managed care, telephone management is crucial to pediatric practice, but an effective method is needed to teach residents telephone skills. Our objective was to design an interactive CD-ROM program to teach residents an organized, consistent approach to telephone complaints and to determine whether use of the program was associated with better subsequent telephone management than reading the same information. SETTING: The general pediatric ambulatory center of a tertiary care children's hospital. PARTICIPANTS: A total of 24 PL-2 and PL-3 pediatric residents. DESIGN: A randomized, prospective, controlled comparison was conducted of resident management of two telephone calls: a 5-year-old with cough and trouble breathing, and a 7-year-old with fever. Thirteen residents were randomized to the computer group and 11 to the reading control group. Intervention. Scripts, scoring, and feedback for 10 CD-ROM-simulated calls were developed from texts and pediatrician survey using a modified Delphi technique. Volunteers acted out the caller's role in scenario scripts and were recorded onto a CD-ROM. The computer simulated calls by recognizing questions typed in a free-form format and answering with a voice response. Feedback was provided for omissions in history-taking and errors in assessment, triage, and home management. The computer group worked through the CD-ROM calls while the control group had equal time to read the same information. Evaluation Measures. A trained, standardized patient acted as the mother in pretest calls placed at the beginning of the month and posttest calls at the end. Calls were recorded and scored in a blinded manner using scoring templates and on interpersonal skills using the Patient Perception Questionnaire. RESULTS: Pretest scores for the two calls were similar in the computer versus the control group (cough, 70.33% +/- 8.36 vs 68.46% +/- 6.73; fever, 75.64% +/- 9.82 vs 73.59% +/- 9.06). Posttest scores were significantly higher in the computer group than in the control group on both calls (cough, 79.08% +/- 8.17 vs 69 +/- 13.3; fever: 83.33% +/- 9.96 vs 70.35% +/- 9.66). Interpersonal skills also were similar pretest (19 +/- 3.4 vs 20 +/- 2.7). There was modest improvement in both groups without a statistically significant difference in posttest scores (24.2 +/- 2.9 vs 22.5 +/- 3.1). CONCLUSIONS: Use of this CD-ROM telephone management program was associated with better postintervention telephone management. The program augments faculty instruction by teaching a consistent, general approach to telephone management.

A computer program for predicting protein antigenic determinants.

A computerized method for predicting the locations of protein antigenic determinants is presented, which requires only the amino acid sequence of a protein, and no other information. This procedure has been used to predict the major antigenic determinant of the hepatitis B surface antigen, as well as antigenic sites on a series of test proteins of known antigenic structure [Hopp & Woods (1981) Proc. natn. Acad. Sci. U.S.A. 78, 3824-3828.] The method is suitable for use in smaller personal computers, and is written in the BASIC language, in order to make it available to investigators with limited computer experience and/or resources. A means of locating multiple antigenic sites on a homologous series of proteins is demonstrated using the influenza hemagglutinin as an example.

A computer program for the management of small cosmid banks.

A Fortran program has been written to permit comparative analysis of Grunstein-Hogness hybridisation data from small plasmid or cosmid banks. This program is particularly useful for restriction mapping using genome walking techniques and will facilitate the analysis of genomes in the 200 kb to 5000 kb size range.

A computer program for the analysis of serial electrocardiograms from patients who suffered a myocardial infarction.

A program is described, which compares serial ECG recordings of patients, who suffered a myocardial infarction. The comparison of the ECGs is based on the measurements, delivered by the Modular ECG/VCG Interpretation System. The two most recent ECGs are compared and a trend analysis based on all ECG recordings of the patient is performed. The criteria in the comparison program are based, among others, on data obtained from a combined cardiological and respiratory survey in The Netherlands in 1973 and 1976.

A computer program for the interpretation of exercise tolerance tests.

A software package has been developed to interpret exercise test data recorded by a metabolic measurement cart (MMC). This package reduces and interprets the exercise test data relative to population and patient specific criteria. The data flow for the data reduction and report printing is described. The data structures used in this processing are illustrated. The application of structured programming, a real-time executive and an easy-to-read language--such as Pascal--is illustrated for the implementation of a medical instrument. Issues of prediction and interpretation relative to exercise testing are discussed.

Detection of negative allosteric effects between monoclonal antibodies by using an antigenic model-builder computer program.

The ability of monoclonal antibodies (MAb) to bind or not simultaneously to the antigen (Ag) is used to establish antigenic maps considering that two different MAb do not bind to the Ag when the corresponding epitopes are overlapped (steric effect). Nevertheless, MAb inducing negative allosteric effect on the Ag could prevent the binding of the second MAb even if it is directed to a separate epitope. We report here that a knowledge-based expert module included in our previously described antigenic model-builder program (MAPAG) was able to differentiate between steric and negative allosteric effects between some MAb.
