Autocorrelation of molecular electrostatic potential surface properties combined with partial least squares analysis as alternative attractive tool to generate ligand-based 3D-QSARs.

A database of 106 human A3 adenosine receptor antagonists was used to derive two alternative PLS models: one starting from CoMFA descriptors and the other starting from the autocorrelation descriptors. The peculiarity of this work is the introduction of autocorrelation vectors as molecular descriptors for the PLS analysis. The autocorrelation allows comparing molecules (and their properties) with different structures and with different spatial orientation without any previous alignment. In particular, Molecular Electrostatic Potential (MEP) was the property computed and its information encoded in autocorrelation vectors. The 3D spatial distribution and the values of the electrostatic potential is in fact largely responsible for the binding of a substrate to its receptor binding site. Validation was done with an external test set and the results of the two models were compared. Interestingly, our preliminary results seem to indicate that this new alternative approach could robustly compete with the already well consolidated CoMFA approach. In particular, we have suggested that it could be a very interesting tool to filter large structural database in several virtual screening applications.

Electrostatic interaction of microcapsules with guinea-pig polymorphonuclear leucocytes.

Electrostatic interaction of microcapsules with guinea-pig polymorphonuclear leucocytes was investigated at different ionic strengths of the medium through measurements of the degree of phagocytosis by the cells of microcapsules. At any ionic strength, the interaction exhibited a maximum when the surface potential of microcapsules was equal to that of the leucocytes to make phagocytosis of microcapsules by the cells minimum. The experimental findings were analyzed by a novel model for the electrostatic interaction between ion-penetrable membranes. In this analysis, it was assumed that the leucocytes have an area (or areas) which permits cations to partition much more into the membrane than anions on their surface to reduce the electric potential of the area.

Electrostatic free energy and spontaneous curvature of spherical charged layered membrane.

A biophysical model for the equilibrium curvature of a composite membrane element is derived taking into account the mechanical bilayer properties and the adjacent charged protein layers. The minimum of the total free energy density with respect to the curvature of such a membrane curved was estimated from the sum of the electrostatic free energy density of the charges of the membrane and the elastic surface energy density due to bending the lipid bilayer membrane. It was shown that the equilibrium curvature, i.e. the spontaneous curvature, of such a charged composite sandwich-like membrane depends inversely on the bending stiffness of the lipid membrane itself and directly on the charge amount inside and outside the membrane to the second power. Furthermore the geometric and electrostatic structure of the protein layers and the physico-chemical environment conditions are involved. Corresponding to the model developed a "standard RBC" membrane element has a negative spontaneous curvature, accounting for a discocyte RBC shape. The shape change from a discocyte to a more stomatocytic shape (increase in the negative spontaneous curvature) after reducing the charges in the glycocalyx is also explained within this model.

Comparison of methods used for measuring the electrostatic field of video display terminals.

The electrostatic field of video display terminal (VDT) operators was measured with different methods. Using the Wilson plate method the field strength at different locations at the face of three human subjects working with a VDT was determined. The strongest fields were found at protruding facial features. Measurements were also made with a measuring instrument known as a field mill. Comparative measurements were made with the field mill as such and combined with a metal plate or a hemisphere. The measured field strength was highly dependent on the method of measurement. A conducting hemisphere (diameter 0.20 m), used in combination with the field mill, was found to be a useful model of the human head in electrostatic measurements.

Modulation of phospholipase A2 by electrostatic fields and dipole potential of glycosphingolipids in monolayers.

Phospholipase A2 activity against mixed monolayers of dilauroylphosphatidic acid or dilauroylphosphatidylcholine with glycosphingolipids can be reversibly modulated by external constant electrostatic fields. The changes of enzymatic activity are correlated to the depolarization or hyperpolarization of the film caused by specific dipolar properties of glycosphingolipids. Hyperpolarizing fields enhance the enzymatic activity against pure dilauroylphosphatidic acid while depolarizing fields induce a decrease of activity. Compared to the pure substrate, the interface of mixed films containing neutral glycosphingolipids or gangliosides is already partially depolarized and the magnitude of activation induced by an external hyperpolarizing field is decreased; conversely, depolarizing fields cause an increased inhibition of activity. Differing from gangliosides, sulfatides bring about a hyperpolarization of the mixed lipid monolayer and external hyperpolarizing or depolarizing fields cause enhanced activation and reduced inhibition, respectively. The effects of glycosphingolipids depend on their relative proportion in the monolayer. Results were similar with dilauroylphosphosphatidylcholine but the field effects were less than half of those found with dilauroylphosphatidic acid. Our work shows that the activity of phospholipase A2 in addition to responding reversibly to external electrostatic fields, is directly modulated by the polarity and magnitude of the lipid polar head group dipole moments.

[Effect of electrostatic field on bacteria E. coli].

On the basis of the quantum mechanical theory of phase transitions and chemical reactions developed by the authors it is shown that the mentioned processes are controlled by the definite active centers. The electrostatic fields (without electric current included) are capable of changing the degree of activity and structure of these centers and so are capable of controlling the results of processes under study. The reported experimental studies performed on the living objects validate this hypothesis. It was concluded that electrostatic fields (natural and technogenic) exert some action on life activity of the organisms and therefore it is necessary to provide the proper optimal conditions for the life activity in the inhabited spacecraft. For this purpose the special equipment is proposed by the authors.

A geometry optimization and molecular electrostatic potential mapping study of structure-activity relationship for some anti-Alzheimer agents.

Molecular geometries of some substituted (pyrroloamino)pyridines which possess anti-Alzheimer activity were optimized and potential-derived CHelpG point charges were computed using ab initio SCF molecular orbital approach employing the 3-21G basis set. AM1 molecular orbital calculations were performed using these optimized geometries and thus optimized Hybridization. Displacement Charges (HDC) combined with Löwdin charges continuously distributed in three dimension were obtained. Molecular electrostatic potential (MEP) maps of the molecules were obtained in two ways: (i) using the HDC-based model with the help of which MEP minima near the molecules were located, and (ii) using the CHelpG point charges, MEP values on the van der Waals surfaces of the molecules were computed. The MEP maps computed using both the methods have negative MEP regions near the pyridine nitrogen atom which appears to be the main binding site of the molecules with the appropriate receptor. Both electrostatic interaction and lipophilic association between these molecules and the receptor appear to contribute to biological activity.

Theoretical study of the electrostatically driven step of receptor-G protein recognition.

This study proposes a theoretical model describing the electrostatically driven step of the alpha 1 b-adrenergic receptor (AR)-G protein recognition. The comparative analysis of the structural-dynamics features of functionally different receptor forms, i.e., the wild type (ground state) and its constitutively active mutants D142A and A293E, was instrumental to gain insight on the receptor-G protein electrostatic and steric complementarity. Rigid body docking simulations between the different forms of the alpha 1 b-AR and the heterotrimeric G alpha q, G alpha s, G alpha i1, and G alpha t suggest that the cytosolic crevice shared by the active receptor and including the second and the third intracellular loops as well as the cytosolic extension of helices 5 and 6, represents the receptor surface with docking complementarity with the G protein. On the other hand, the G protein solvent-exposed portions that recognize the intracellular loops of the activated receptors are the N-terminal portion of alpha 3, alpha G, the alpha G/alpha 4 loop, alpha 4, the alpha 4/beta 6 loop, alpha 5, and the C-terminus. Docking simulations suggest that the two constitutively active mutants D142A and A293E recognize different G proteins with similar selectivity orders, i.e., G alpha q approximately equal to G alpha s > G alpha i >> G alpha t. The theoretical models herein proposed might provide useful suggestions for new experiments aiming at exploring the receptor-G protein interface.

[Electrostatic air filters for dental practice].

The filtrering effect of an electrostatic filtrattion equipment (Vortronic mod. 75) for air filtration was examined by sampling the air in a general dental practice by means of a Casella Slit Sampler for Airborne Bacteria (C. F. Casella & Co. Ltd., London). A reception, a sterilization, two treatment chairs and a resting cabinet were seperated by 180 cm high walls in a rectangular room of 33 m2 and with an air volume of approximately 80 m3. The staff including one dentist and one or two assistents worked from 8 a.m. till 4 p.m. with a 30 min. lunch break. A mean of 17 patients visited the practice every day. The electrostatic filter was placed in front of one of the treatment chairs and the slit sampler was, for one period, placed at a distance of 5 m from the filter. In the next period both filter and slit sampler were placed within 140 cm from the treatment chair. The air was sampled at noon and at 4 p.m. Micro-organisms from the air samples were cultured on blood agar by incubation at 37 degrees C for 48 hours. After incubation all colonies visible to the eye were counted. A total of 36 air samples were examined. Without filtration of the air the bacterial contamination amounted to 14 cultivable organisms per ft3 air at noon and 54 organisms per ft3 at 4 p.m. The filtration equipment proved to reduce the air contamination to 6 cultivable organisms per ft3.

[Characteristics of electrostatic interaction of Escherichia coli RNA polymerase with promoters of T4 phage DNA].

A comparative analysis of electrostatic potential distribution for "early" T4 phage promoters was undertaken. The data obtained indicate that there are some particular elements in the patterns of electrostatic potential distribution of promoter DNA specific for promoter groups differing by their functional response to ADP-ribosylation of the alpha-subunit as well as to rpoB403- or rpoB409 mutationals of the beta-subunit of RNA-polymerase.

[The influence of electrostatic fields on osmolality and concentrations of neutral fat and cholesterol (author's transl)].

Rats were exposed to electrostatic fields of different intensities and exposure times, after which the osmolality of the serum and the concentration of neutral fat and cholesterol in the serum were measured. The triglyceride and cholesterol concentrations of the sera were independent of the strength of the electrostatic field and independent of the length of exposure of the animals in the constant field or Faraday cages. They always lay within normal limits, but moved according to the conditions of the constant field: the neurtal fat concentrations to the upper, and the cholesterol concentrations to the lower limit. The differences compared to the controls could be confirmed statistically.

[Simultaneous separation of organic acid and organic salts by electrostatic ion chromatography].

The electrostatic ion chromatographic column was prepared by coating conjugated acid salt micelles on the surface of octadecyl silica stationary phase. Pure water was used as mobile phase, and the conductance detector was connected on-line to electrostatic ion chromatograph. The conditions under which organic acid and organic salts were detected were studied. The mechanism for the above separation is discussed. Sodium benzoate and citric acid in Lichee drink were separated and determined. This method is rapid, simple with little interference and good reproducibility without any pollution since the mobile phase is water. This is an environmental friendly analytical method.

[Spectroscopic studies on electrostatically self-assembled gold nanoparticulate thin films].

Colloidal gold was prepared by citrate-induced reduction of hydrogen tetrachloroaurate. Gold nanoparticulate thin films were built up using the electrostatic self-assembly technique and characterized by different spectroscopes. The UV-Vis absorbance spectrum indicated that the gold colloid was a monodisperse suspension. The calculation using the Scherrer's equation on base of the X-ray diffraction spectrum indicated that the average size of the gold particles assembled on the substrates was about 21 nm. The X-ray photoelectron spectra showed that the reduction of hydrogen tetrachloroaurate was almost complete and that the gold was present in the thin films largely as Au zero. The X-ray photoelectron spectra also showed that the gold particles were attracted and fixed onto the PDDA-modified substrates by electrostatic forces. The UV-Vis absorbance spectra and the surface enhanced Raman scattering spectra indicated that, arising from the interparticle coupling, the gold nanoparticulate thin films displayed strong collective plasma resonance absorption and sur faced enhanced Raman scattering effect.

Electrostatic interactions lead to the formation of asymmetric collagen-phosphophoryn aggregates.

In bone and dentin the formation and mineralization of the extra cellular matrix structure is a complex process highly dependent on intermolecular interactions. In dentin, the phosphophoryns (PP) and type I collagen (COL1) are the major constituents implicated in mineralization. Thus, as a first step in understanding the tissue organization, we have initiated a study of their interaction as a function of pH, ionic strength, and relative concentrations or mixing ratios. Complex formation has been analyzed by dynamic light scattering to detect aggregate formation and by rotary shadowing electron microscopy (EM) to determine aggregate shape. The EM data showed that at the pH values studied, the PP-COL1 interaction leads to the formation of large fibrillar aggregates in which the PP are present along the fibril surfaces. The quantitative phase distribution data showed a 1/1 molar equivalence at the maximum aggregation point, not at electrostatic PP-COL1 equivalence. As the ionic strength was raised, the PP-COL1 aggregates became smaller but the binding and asymmetric fibrillar aggregation persisted. In EM, the PP appear as dense spheres. Along the surfaces of the collagen aggregates, the PP are larger and more open or extended, suggesting that COL1-bound PP may undergo a conformational change, opening up so that a single PP molecule might interact with and electrostatically link several COL1 molecules. This might have important implications for dentin structure, stability, and mineralization.

Electrostatic interactions stabilizing ferredoxin electron transfer complexes. Disruption by "conservative" mutations.

Mitochondrial ferredoxins mediate electron transfer from NADPH:ferredoxin oxidoreductase to cytochrome P450 enzymes. Previous studies on human ferredoxin, in which acidic residues were replaced with neutral amino acids, established that Asp-76 and Asp-79 are are important for binding to both reductase and P450 (Coghlan, V. M., and Vickery, L. E. (1991) J. Biol. Chem. 266, 18606-18612). Here we report that replacement of Asp----Glu at position 76 or 79, whereas maintaining negative charge at these positions also results in dramatic decreases in binding affinity for both electron transfer partners (5-100-fold, delta(delta G) approximately 1.0-2.8 kcal/mol). These results imply that the active electron transfer complexes in these systems are dominated by a stable form which requires specific pairwise electrostatic interactions of fixed geometry for recognition and binding. This mechanism contrasts with that proposed for other electron transfer systems (as exemplified by cytochrome c) in which electrostatic interactions are believed to function primarily in precollisional orientation leading to "encounter complexes" having multiple geometries of similar free energy.

Electrostatic interaction influences cell adhesion?

The effect of electrostatic forces on the adhesion of LEP-19 diploid embryonal fibroblasts, Hep-2 laryngeal carcinoma cells, Raji lymphoblastoma cells and Sp 2/0 myeloma cells was examined in vitro. Adhesivity of all tested cell lines was higher on the cationized glass than on untreated or anionized glass. The negatively charged sialic acids on the cell surface play a role in cell adhesion. The participation of electrostatic interaction is independent of the energy metabolism in serum-free conditions.

[On the methods of calculation of the electrostatic potential in a protein channel].

The profiles of the electrostatic potential along the axis of a gramicidin channel were calculated using two quantum chemistry methods (EHT and CNDO/2) and three methods of force fields (AMBER, CHARMM, and OPLS). The calculations were performed without taking into account the contribution of water. A comparative analysis of the calculated profiles indicated that the electrostatic field of point charges of the CHARMM force field is close to that obtained by the quantum chemistry methods. It was concluded that there is no need to calculate the electric field using laborious quantum-mechanical methods since the less laborious method of the CHARMM force field gives approximately the same results.
