Radiocarbon dating of archaeological samples (sambaqui) using CO(2) absorption and liquid scintillation spectrometry of low background radiation.

Sambaqui means, in the Tupi language, a hill of shells. The sambaquis are archaeological sites with remains of pre-historical Brazilian occupation. Since the sambaqui sites in the Rio de Janeiro state region are older than 10,000 years, the applicability of CO(2) absorption on Carbo-sorb and (14)C determination by counting on a low background liquid scintillation counter was tested. In the present work, sambaqui shells were treated with H(3)PO(4) in a closed vessel in order to generate CO(2). The produced CO(2) was absorbed on Carbo-sorb. On saturation about 0.6g of carbon, as CO(2), was mixed with commercial liquid scintillation cocktail (Permafluor), and the (14)C activity determined by counting on a low background counter, Packard Tricarb 3170 TR/SL, for a period of 1000 mins to enable detection of a radiocarbon age of 22,400 BP. But only samples with ages up to 3500 BP were submitted to the method because the samples had been collected in the municipality of Guapimirim, in archaeological sambaqui-type sites belonging to this age range. The same samples were sent to the (14)C Laboratory of the Centro de Energia Nuclear na Agricultura (CENA/USP) where similar results were obtained.

The neurotrophins NGF and NT-3 reduce sensory neuronal loss in adult rat after peripheral nerve lesion.

The effect of three different neurotrophins on axotomy-induced death of adult rat sensory neurons was examined. The ventral branch of the 13th spinal nerve was transected and the corresponding neurons in the 13th thoracic (T13) dorsal root ganglion (DRG) were pre-labelled with Fast Blue (FB). For a period of 4 weeks, animals received either no treatment, continuous intrathecal infusion of phosphate buffer, nerve growth factor (NGF), neurotrophin-3 (NT-3), or brain-derived neurotrophic factor (BDNF). Labelled neurons remaining after this period were counted. Inert, or no treatment, resulted in extensive loss of the DRG neurons. BDNF application was virtually non-effective, while NGF or NT-3 resulted in a greater number of FB-labelled neurons compared to normal controls. This suggests that NGF and NT-3 are survival factors for adult sensory neurons with a therapeutic potential in peripheral nerve injuries.

A TV system for image recording and processing in conventional transmission electron microscopy.

An image processing system for a CTEM is described which is particularly suited for the imaging of radiation-sensitive objects. It consists of an image intensifier device for single electron counting, a digital storage unit and a video recorder for intermediate storage of low-dose image series. During on-line operation the information can be transferred to a computer for further processing. The output can be transferred onto a second digital storage unit and observed on a monitor. The influence on the DQE of the pulse height distribution of the single electron video signals and of a multiple counting are eliminated by means of special electronic components. Various modes of operation and application possibilities of the image processing system are discussed.

Recent developments in ultra low-level gamma-ray spectrometry at IRMM

IRMM performs ultra low-level gamma-ray spectrometry in the underground laboratory HADES, located at the SCK-CEN site in Mol, Belgium. Recently a third low-background coaxial HPGe detector was put into operation. The detector has 60% relative efficiency and better background characteristics than the two previous detectors: The shield comprises 14 cm of lead with 21OPb specific activity of 20 Bq kg(-1) and 14 cm of freshly produced electrolytic copper. The background count rate integrated from 40 to 2700 keV is 394 +/- 2 counts per day. Details of the background and the shield are presented together with applications to nuclear safeguards and material selection.

Do disputes over priority tell us anything about science?

Conflicts between scientists over credit for their discoveries are conflicts, not merely in, but of science because discovery is not a historical event, but a retrospective social judgment. There is no objective moment of discovery; rather, discovery is established by means of a hermeneutics, a way of reading scientific articles. The priority conflict between Roger Guillemin and Andrew Schally over the discovery of the brain hormone, TRF, serves as an example. The work of Robert Merton, Thomas Kuhn, Augustine Brannigan, and Grygory Markus shows that scientists read scientific articles by means of the application of a set of pragmatic rules that subtend the normative requirements of what counts as a scientific discovery. In other words, there is a hermeneutics of science, but it is internal to that form of life. Recategorization of priority conflicts has an impact on our view of scientific controversy generally. The impact is the revision of the boundary lines of scientific controversy and the further specification of its fine-structure.

Simple selection criteria for drug-like chemical matter.

A simple pharmacophore point filter has been developed that discriminates between drug-like and nondrug-like chemical matter. It is based on the observation that nondrugs are often underfunctionalized. Therefore, a minimum count of well-defined pharmacophore points is required to pass the filter. The application of the filter results in 66-69% of subsets of the MDDR database to be classified as drug-like. Furthermore, 61-68% of subsets of the CMC database are classified as drug-like. In contrast, only 36% of the ACD are found to be drug-like. While these results are not quite as good as those obtained with recently described neural net approaches, the method used here has clear advantages. In contrast to a neural net approach and also in contrast to decision tree methods described recently, the pharmacophore filter has been developed by using "chemical wisdom" that is unbiased from fitting the structural content of specific drug databases to prediction models. Similar to decision tree methods, the pharmacophore point filter provides a detailed structural reason for the classification of each molecule as drug or nondrug. The pharmacophore point filter results are compared to neural net filter results. A statistically significant overlap between compounds recognized as drug-like validates both approaches. The pharmacophore point filter complements neural net approaches as well as property profiling approaches used as drug-likeness filters in compound library analysis and design.

Objectively measured intensity of physical activity and adiposity in middle-aged women.

OBJECTIVE: The aim of this study was to evaluate the relationship between objectively measured physical activity (ACT), particularly intensity of physical activity (iACT), and body fat percentage (BF%) in 278 middle-aged women. Secondary purposes were to ascertain the association between physical activity duration (dACT) and BF% and the extent to which potentially confounding factors, including total ACT, energy intake, body weight, and season of assessment, influenced the relationship between iACT and BF%. RESEARCH METHODS AND PROCEDURES: A cross-sectional design was used. Subjects were apparently healthy, 35 to 45 years old, premenopausal, nonsmokers, with BMIs < 30 kg/m2. Approximately 90 percent were white, 81% were married, and 37% were college graduates. ACT was assessed using Computer Science and Application accelerometers worn for 7 consecutive days. The accelerometers recorded movement continuously, and activity counts were collapsed into 10-minute epochs. Intensity was indexed using seven activity count cut-off points, and duration was based on the number of 10-minute epochs at each intensity level. BF% was assessed using multiple measurements in the Bod Pod. Energy intake was measured using 7-day weighed diet records during the same week subjects wore the accelerometers. RESULTS: BF% was strongly and inversely associated with iACT and dACT. Controlling for energy intake and body weight strengthened the relationships among iACT, dACT, and BF%. Control of total ACT weakened the association. DISCUSSION: Engaging in higher intensity and/or longer duration ACT is associated with lower BF% compared with lower intensity and/or shorter durations of activity.

Dynabeads plus 3 M Petrifilm HEC versus Vitek Immunodiagnostic Assay System for detection of E. coli O157 in minced meat.

The potentially low infective dose of Escherichia coli O157 makes it necessary to be able to detect low numbers in food, and the lack of sensitivity of direct plating has led to the development of various enrichment and detection methods. Until now, the most selective procedure for detection of E. coli O157 isolates was the immunomagnetic separation (IMS) method. The number of sorbitol non-fermenting micro-organisms other than E. coli O157 that adhere non-specifically to the magnetic beads hampers the application of IMS. The use of IMS in conjunction with 3 M Petrifilm-HEC yielded EHEC O157 in 21 of 165 samples of minced meat (12.7%). Without advance application of IMS, Petrifilm plates often yield confluent growth and colonies too numerous to count. The Vitek Immunodiagnostic Assay System (VIDAS-ECO) showed good sensitivity when testing artificially contaminated beef samples, but only four of 21 naturally contaminated samples were recognized. The addition of 3 M Petrifilm to IMS resulted in less growth of contaminants and eliminated much of the need to test presumed colonies for confirmation. The combination of IMS and 3 M Petrifilm-HEC is a fast and efficient screening procedure for E. coli O157 in minced meat.

Model applicable to NMR studies for calculating flux rates in five cycles involved in glutamate metabolism.

Based on the same principles as those utilized in a recent study for modeling glucose metabolism (Martin, G., Chauvin, M. F., Dugelay, S., and Baverel, G. (1994) J. Biol. Chem. 269, 26034-26039), a method is presented for determining metabolic fluxes involved in glutamate metabolism in mammalian cells. This model consists of five different cycles that operate simultaneously. It includes not only the tricarboxylic acid cycle, the "oxaloacetate --> phosphoenolpyruvate --> pyruvate --> oxaloacetate" cycle and the "oxaloacetate --> phosphoenolpyruvate --> pyruvate --> acetyl-CoA --> citrate --> oxaloacetate" cycle but also the "glutamate --> alpha-ketoglutarate --> glutamate" and the "glutamate --> glutamine --> glutamate" cycles. The fates of each carbon of glutamate, expressed as ratios of integrated transfer of this carbon to corresponding carbons in subsequent metabolites, are described by a set of equations. Since the data introduced in the model are micrograms of atom of traced carbon incorporated into each carbon of end products, the calculation strategy was determined on the basis of the most reliable parameters determined experimentally. This model, whose calculation routes offer a large degree of flexibility, is applicable to data obtained by 13C NMR spectroscopy, gas chromatography - mass spectrometry, or 14C counting in a great variety of mammalian cells.

Specific enumeration of the probiotic strain Enterococcus faecium NCIMB 10415 in the intestinal tract and in faeces of piglets and sows.

The intestinal bacterium Enterococcus faecium NCIMB 10415 (E. faecium SF68) has been used for more than a decade as a probiotic strain in animal nutrition as well as in the prevention and treatment of diarrhoea in humans. Beneficial effects have been shown in feeding and clinical trials. However, the strain has no selective growth markers and monitoring in the intestinal tract is impossible by cultivation. Using specific nucleotide sequences, in this study a probe for colony hybridization was constructed in order to quantify this probiotic strain in feed and intestinal and faecal samples from piglets and sows. The probiotic strain showed almost constant amounts in sow faeces (1.8 x 10(5) cfu/g wet weight), while contents in digesta and piglet faeces varied on a lower level depending on gut section and piglet age. The ratio of specific probiotic counts and total enterococci was much lower than in sow faeces however the strain could be detected reliably in faeces already on the 14th day of life. The application of the colony hybridization method enables for the first time the selective detection of the widely used probiotic E. faecium NCIMB 10415 strain among total Enterococcus spp. counts of digesta, faeces and feed. It is now possible to monitor the presence of the probiotic in the intestinal tract and faeces. Results of this study have implications for the proposed modes of action of probiotics in animal nutrition.

Evaluation of culture conditions used for isolation of Chlamydia pneumoniae.

As only a few viable strains of the frequent respiratory pathogen Chlamydia pneumoniae have been isolated worldwide, improvement and standardization of isolation procedures is mandatory for adequate diagnosis and recovery of strains for further investigation. Growth rates of eight C pneumoniae strains were assessed for five host cell lines, for the growth-promoting effect of host cell cytostasis, for infection of adherent or suspended host cells, for optimal first passage incubation time, and for use of multiple blind passages. Cycloheximide-treated HEp-2 or NCI-H 292 monolayers appeared most sensitive for isolation of C pneumoniae. No significant improvement of sensitivity was achieved by extending first passage incubation periods from 3 to 7 days. In contrast, continuous propagation in 4 culture passages yielded on average a 7-fold enhanced chlamydial recovery. Additional comparative isolation of two wild-type strains showed highest inclusion counts in HEp-2 cells, but subculture was necessary for optimal sensitivity. A widely applicable protocol using tissue culture plates for isolation of C pneumoniae is suggested.

Modeling tumor onset and multiplicity using transition models with latent variables.

We describe a method for modeling carcinogenicity from animal studies where the data consist of counts of the number of tumors present over time. The research is motivated by applications to transgenic rodent studies, which have emerged as an alternative to chronic bioassays for screening possible carcinogens. In transgenic mouse studies, the endpoint of interest is frequently skin papilloma, with weekly examinations determining how many papillomas each animal has at a particular point in time. It is assumed that each animal has two unobservable latent variables at each time point. The first indicates whether or not the tumors are in a multiplying state and the second is the potential number of additional tumors if the tumors are in a multiplying state. The product of these variables follows a zero-inflated Poisson distribution, and the EM algorithm can be used to maximize the observed-data pseudo-likelihood, based on the latent variables. A generalized estimating equations robust variance estimator adjusts for dependency among outcomes within individual animals. The method is applied to testing for a dose-related trend in both tumor incidence and multiplicity in carcinogenicity studies.

The value of a 51Cr platelet lysis assay as crossmatch test in patients with leukaemia on platelet transfusion therapy.

Current platelet crossmatch testing still results in a significant percentage of false positive or false negative results in oncological patients during platelet support. A 51Cr platelet lysis assay was used for the detection of platelet alloantibodies. We determined the predictive value of this assay as crossmatch procedure on 28 occasions in 14 patients who received random single donor platelet transfusions. To deal with the problem of spontaneous lysis we included a panel of 10 control sera from normal individuals and applied several methods of statistical analysis to these data. It appeared that the use of only one control serum was sufficient when the percentage relative counts was used as the criterion variable, which is of advantage in the practical application of the test. The test values were retrospectively compared with the clinical transfusion response, determined as the 1 h post-transfusion platelet recovery. The 51Cr platelet lysis crossmatch showed false negative results in 1/21 cases and false positive results in 1/7 cases. These data indicate that the 51Cr platelet lysis assay adds a useful dimension to the solution of the problem of selecting compatible platelet donors. Spontaneous lysis of target platelets appeared not to be a problem in the interpretation of test results.

Serum eosinophil cationic protein and interleukin-5 in children with bronchial asthma and acute bronchiolitis.

The aim of our study was to evaluate the clinical applicability of serum eosinophil cationic protein (ECP), interleukin-5 (IL-5) and total eosinophil counts in childhood asthma and bronchiolitis. These parameters were measured in 44 children aged 12-84 months with moderate and mild asthma during symptomatic and asymptomatic phases of disease. Fifteen of the patients were included at the time of admission to hospital due to an acute asthmatic attack, and ten of these were also examined one month after discharge. None of the patients were treated with glucocorticoids or cromoglycate at any time during the study. Serum ECP was significantly increased in the children with acute asthma compared to children with stable moderate asthma, stable mild asthma, as well as to controls. There was no difference between the groups with stable asthma or between stable asthma and controls, and there was large overlap between all groups of asthmatics and controls. Detectable levels of circulating IL-5 were demonstrated in eight of 15 children with acute asthma, with significantly higher levels in atopic children, whereas all samples from children with stable asthma and controls were negative. The results suggest that even though serum ECP and IL-5 increases during acute asthmatic attacks, these parameters cannot alone be used to discriminate between different groups of young children with stable asthma, nor between asthmatics and healthy controls. In addition, the same parameters of eosinophil inflammation were examined in serum samples from 25 children aged 1-17 months undergoing their first episode of acute bronchiolitis. Children with acute respiratory syncytial virus (RSV) bronchiolitis had significantly higher levels of serum ECP than those with RSV negative disease, whereas the total eosinophil counts were significantly decreased in all patients with acute bronchiolitis. Serum IL-5 was only detected in two children with acute bronchiolitis. The results suggest that the inflammation in RSV bronchiolitis differs from that induced by other viruses.

Metal Interactions with Microbial Biofilms in Acidic and Neutral pH Environments.

Microbial biofilms were grown on strips of epoxy-impregnated filter paper submerged at four sites in water contaminated with metals from mine wastes. At two sample stations, the water was acidic (pH 3.1); the other sites were in a lake restored to a near neutral pH level by application of a crushed limestone slurry. During a 17-week study period, planktonic bacterial counts increased from 10 to 10 CFU/ml at all sites. Biofilm counts increased rapidly over the first 5 weeks and then leveled to 10 CFU/cm in the neutral pH system and 10 CFU/cm at the acidic sites. In each case, the biofilms bound Mn, Fe, Ni, and Cu in excess of the amounts adsorbed by control strips covered with nylon filters (pore size, 0.22 mum) to exclude microbial growth; Co bound under neutral conditions but not under acidic conditions. Conditional adsorption capacity constants, obtained graphically from the data, showed that biofilm metal uptake at a neutral pH level was enhanced by up to 12 orders of magnitude over acidic conditions. Similarly, adsorption strength values were usually higher at elevated pH levels. In thin sections of the biofilms, encapsulated bacterial cells were commonly found enmeshed together in microcolonies. The extracellular polymers often contained iron oxide precipitates which generated weak electron diffraction patterns with characteristic reflections for ferrihydrite (Fe(2)O(3) . H(2)O) at d equaling 0.15 and 0.25 nm. At neutral pH levels, these deposits incorporated trace amounts of Si and exhibited a granular morphology, whereas acicular crystalloids containing S developed under acidic conditions.

Effect of immersion solutions containing enterocin AS-48 on Listeria monocytogenes in vegetable foods.

The effect of immersion solutions containing enterocin AS-48 alone or in combination with chemical preservatives on survival and proliferation of Listeria monocytogenes CECT 4032 inoculated on fresh alfalfa sprouts, soybean sprouts, and green asparagus was tested. Immersion treatments (5 min at room temperature) with AS-48 solutions (25 microg/ml) reduced listeria counts of artificially contaminated alfalfa and soybean sprouts by approximately 2.0 to 2.4 log CFU/g compared to a control immersion treatment in distilled water. The same bacteriocin immersion treatment applied on green asparagus had a very limited effect. During storage of vegetable samples treated with immersion solutions of 12.5 and 25 microg of AS-48/ml, viable listeria counts were reduced below detection limits at days 1 to 7 for alfalfa and soybean sprouts at 6 and 15 degrees C, as well as green asparagus at 15 degrees C. Only a limited inhibition of listeria proliferation was detected during storage of bacteriocin-treated alfalfa sprouts and green asparagus at 22 degrees C. Treatment with solutions containing AS-48 plus lactic acid, sodium lactate, sodium nitrite, sodium nitrate, trisodium phosphate, trisodium trimetaphosphate, sodium thiosulphate, n-propyl p-hydroxybenzoate, p-hydoxybenzoic acid methyl ester, hexadecylpyridinium chloride, peracetic acid, or sodium hypochlorite reduced viable counts of listeria below detection limits (by approximately 2.6 to 2.7 log CFU/g) upon application of the immersion treatment and/or further storage for 24 h, depending of the chemical preservative concentration. Significant increases of antimicrobial activity were also detected for AS-48 plus potassium permanganate and in some combinations with acetic acid, citric acid, sodium propionate, and potassium sorbate.

Effects of stimulus modality and response mode on the P300 event-related potential differentiation of young and elderly adults.

The P300 event-related brain potential (ERP) was examined in 14 young (20 - 29 years of age) and 16 elderly (60 - 82 years of age) adult subjects during the performance of auditory and visual discrimination tasks requiring silent counting or key pressing in response to target stimuli. P300 latencies were longer in elderly (vs young) adults and in visual (vs auditory) tasks, and visual tasks elicited larger P300 amplitudes than auditory tasks in both age groups. Neither stimulus modality nor response mode affected P300 differentiation of young and elderly subjects. Steeper P300 anterior-posterior scalp amplitude gradients were seen in the young (vs elderly) adults, regardless of stimulus or response type. Examination of inter-subject variability with the coefficient of variation (CV) statistic found the lowest (i.e., best) CV values to be exhibited in the visual task requiring the counting of target stimuli. Implications of the findings are discussed in relation to P300 applications in the clinical assessment of dementia and aging-associated cognitive alterations.
