Volatile and nonvolatile nitrosamines in beer.

The occurrence of N-nitroso compounds was investigated at different stages in the production of four different beer types and in 14 retail beer samples. Only two N-nitroso compounds, namely N-nitrosodimethylamine (0.17 +/- 0.18 microgram/kg; not detected to 0.60 microgram/kg) and N-nitrosoproline (1.51 +/- 1.01 micrograms/kg; range 0.5-3.60 micrograms/kg) were detected. For moderate beer drinkers, the N-nitrosodimethylamine exposure levels are unlikely to present a significant health risk while the presence of noncarcinogenic N-nitrosoproline does not pose any hazard to human health.

Further evidence of a positive correlation between exposure to nitrate fertilizers (NaNO3 and KNO3) and gastric cancer death rates: nitrites and nitrosamines.

Mean age-adjusted death rates for gastric cancer in Chile for 1960, 1962 and 1964 were associated with exposure to sodium nitrate and nitrates (NaNO3 and KNO3) used as fertilizers, by province. The general population exposure to NaNO3 and nitrates exhibited correlation coefficients of 0.745 and 0.718 with the death rates, respectively.

Inhibition of nitrosamine formation by ascorbic acid: participation of free radicals in its anaerobic reaction with nitrite.

The participation of semiquinone free radicals during the reaction of ascorbic acid with acidified sodium nitrite has been demonstrated by ESR spectroscopy unambiguously for the first time. Scavenging of the nitrosating agent, reflected by the observed free radical concentration, unexpectedly occurs with scarcely varying efficiency over the pH range 0.1--4.5.

Immunoassays for proteins alkylated by nicotine-derived N-nitrosamines.

Polyclonal antibodies recognizing the pyridyloxobutyl (POB) moiety of 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) were produced in rabbits immunized either with POB-bovine albumin or POB-Sepharose. The POB intermediates necessary to modify the protein were generated by alkaline (pH 9.0) treatment of the synthetic precursor 4-(carbethoxynitrosamino)-1-(3-pyridyl)-1-butanone. In a competitive enzyme linked immunoabsorbent assay (ELISA), 70 pmole NNK inhibited 50% of the binding of the anti-POB antibodies to POB-protein absorbed on microtiterplates. This 50% inhibition varied from 70 pmole to 200 nmole using a series of NNK analogues, depending on the integrity of the POB moiety. Immunological techniques initiated in this study detect NNK-protein conjugates or measure the quantity of POB groups liberated upon alkaline or acid treatment of NNK modified protein.
