A retrospective review of gram-negative bacteraemia in Papua New Guinean adults revealed four cases of primary gram negative pneumonia over a two-year period. Patients with primary gram negative pneumonia constituted less than 1% of all pneumonias admitted to the medical ward. Pseudomonas pseudomallei, Haemophilus influenzae and Klebsiella pneumoniae were the aetiologic agents. Three of four patients died following a fulminant course. It is concluded that primary gram-negative pneumonia in Papua New Guinea is rare but carries a high mortality.
Sixteen goats were inoculated subcutaneously or intraperitoneally with cultured Pseudomonas pseudomallei isolated from a goat. They showed either acute or chronic phase of disease. When inoculated intraperitoneally, animals suffered from a septicemic illness of short duration accompanied with micro-abscesses scattered widely throughout the body. In animals subcutaneously inoculated no clinical signs were exhibited, but the disease was of rather long duration, and lesions were large and localized in the spleen and lungs. Coincident with lesions, a large number of Pseudomonas pseudomallei organisms were isolated from abscesses in the spleen and lungs. Ultrastructurally, numerous bacilli could be detected in the cytoplasm of macrophages in the abscesses. They had an electron-dense cell wall. Most of them were surrounded by electron-dense substance.

Indirect immunofluorescence microscopy was used as a colony identification method of Pseudomonas pseudomallei isolates. The antisera against lipopolysaccharide and protein fractions of P. pseudomallei were prepared in guinea pigs and rabbits. With these antisera and fluorescence-labelled anti-guinea pig IgG and anti-rabbit IgG prepared in sheep (goat), indirect immunofluorescence microscopy was conducted on the colonies of P. pseudomallei and other species of bacteria. The overall results indicated that this method is efficient, rapid and specific for identification of P. pseudomallei colonies from clinical specimens.
