Pattern selection at medium and high nonlinearity is investigated. While in the former the transient time levels off for large system sizes, in the latter it diverges exponentially giving rise to supertransients. In both cases, the final attractors are quite stable with as a consequence that even at high nonlinearity an attractor can easily be reached by means of a parameter sweep.

The clinical and pharmacokinetic risk factors for toxicity after high-dose methotrexate (MTX) in children with acute lymphoblastic leukemia were evaluated using a multivariate statistical analysis. Plasma samples were collected after 44 24-h infusions of MTX (5 or 8 g/m2) in 13 children (age 3.3-12.9 years) and subsequently analyzed by HPLC to determine the MTX and 7-hydroxymethotrexate (7-OHMTX) concentrations. Toxicity was evaluated according to the WHO criteria. Severe toxicity was not observed. Oral mucositis (WHO grade > or = 1) was significantly related to a high plasma MTX concentration at 28 h after starting the infusion (p = 0.013), a low ratio of plasma 7-OHMTX/MTX at 66 h after starting the infusion (p = 0.049), and a slow clearance of MTX (p = 0.048). The risk of leukopenia (WHO grade > or = 2) increased significantly with the number of courses (p = 0.02). Increasing age and a long exposure to a high MTX concentration in plasma (AUC) were significant risk factors (p = 0.047 and p = 0.009, respectively) for developing elevated liver enzymes (ALAT) (WHO grade > or = 2). This study shows how a statistical model can be used to identify clinical and pharmacokinetic factors that may influence MTX-induced toxicity. The therapeutic ratio could thereby potentially be improved.
This paper presents a mathematical model that depicts the relationship between the possibility of occurrence of common health problems and factors leading to Sick Building Syndrome symptoms in domestic interiors in Mauritius. The prevalence of upper respiratory symptoms (dry eyes, runny nose), central nervous system symptoms (headache, nervousness), and musculoskeletal symptoms (pain/stiffness in shoulders/neck) were found to be elevated when responses were statistically regressed to type of building and age of respondents. The model presented here will be useful in helping to identify and quantify the relative role of factors that contribute to Sick Building Syndrome. Thus it may be possible to evaluate the effectiveness of current building operation practices and to prioritise allocations of resources for reduction of risk associated with Indoor Environmental Air Quality.
Early-phase clinical trials, conducted to determine the appropriate dose of an experimental drug to take forward to later trials, are considered. The objective is to find the dose associated with some low probability of an adverse event. A Bayesian model is presented, and a decision-theoretic procedure for finding the optimal doses for each of a series of cohorts of subjects is derived. The procedure is flexible and can easily be conducted using standard statistical software. The results of simulations investigating the properties of the procedure are presented.

The hourly changes in the Ca2+ content of various physiological compartments in relation to shell formation has been calculated for the domestic hen throughout a 4-d ovulatory sequence. These calculations consider the circadian rhythm in Ca2+ intake when the hen is fed a complete feed or a diet containing oyster shell, the effect of the stage of the ovulatory sequence and the stage of shell formation. The calculations indicate that medullary bone accrues Ca2+ during the day and that Ca2+ is drawn from the bone reserve during the scotophase if shell calcification is active. When fed Ca2+ as an identifiable feedstuff, the hen will ingest large amounts of Ca2+ at the end of the photophase. Some of this Ca2+ is temporarily stored in medullary bone and the remainder is metered into the lower digestive tract throughout the early part of the scotophase. It is suggested that the preovulatory surge of estrogens sensitizes medullary bone to the osteoclastic effect of parathyroid hormone, the production of which is stimulated by a decline in the total and ionized concentrations of Ca2+ during shell formation.
