[Cellular subcutaneous tissue. Anatomic observations].

INTRODUCTION: We showed in a companion paper that the definition of the French "subcutaneous cellular tissue" considerably varied from the 18th to the end of the 20th centuries and has not yet reached a consensus. To address the anatomic reality of this "subcutaneous cellular tissue", we investigated the anatomic structures underlying the fat tissue in normal human skin. METHODS: Sixty specimens were excised from the surface to the deep structures (bone, muscle, cartilage) on different body sites of 3 cadavers from the Institut d'Anatomie Normale de Strasbourg. Samples were paraffin-embedded, stained and analysed with a binocular microscope taking x 1 photographs. Specimens were also excised and fixed after subcutaneous injection of Indian ink, after mechanic tissue splitting and after performing artificial skin folds. RESULTS: The aspects of the deep parts of the skin greatly varied according to their anatomic localisation. Below the adipose tissue, we often found a lamellar fibrous layer which extended from the interlobular septa and contained horizontally distributed fat cells. No specific tissue below the hypodermis was observed. Artificial skin folds concerned either exclusively the dermis, when they were superficial or included the hypodermis, but no specific structure was apparent in the center of the fold. India ink diffused to the adipose tissue, mainly along the septa, but did not localise in a specific subcutaneous compartment. DISCUSSION: This study shows that the histologic aspects of the deep part of the skin depend mainly on the anatomic localisation. Skin is composed of epidermis, dermis and hypodermis and thus the hypodermis can not be considered as being "subcutaneous". A difficult to individualise, fibrous lamellar structure in continuity with the interlobular septa is often found under the fat lobules. This structure is a cleavage line, as is always the case with loose connective tissues, but belongs to the hypodermis (i.e. fat tissue). No specific tissue nor any virtual space was observed below the skin. Thus, the commonly used term "subcutaneous cellular tissue" is inappropriate.

[Microvasculature of the dog's periodontal ligament and its immediate response to orthodontic forces].

The purpose of this study was to examine the microvascular architecture of dog's periodontal ligament (PDL) and its immediate response to horizontal orthodontic force. A force of 200 g was applied mesially to the mandibular premolar of a dog for 2 hours, the distal side served as the contrast side. Formalin ink was perfused into the carotid arteries bilaterally through the cannulas. After perfusion the mandible was decalcified, celloidin embedded, serially sectioned and stained with eosin. The sections were then observed by light microscope and analyzed by image analyzer. Vascular corrosion casts were prepared by the similar way with the formalin ink perfusion and examined by scanning electron microscope. The results showed: there were extensive interconnections among the PDL, dental pulp, gingiva, alveolus and the neighbour tooth PDL, which would benefit the maintenance of health and functions of the dental pulp and the periodontal tissues. Arterioles and venulae could be confirmed by the presence of characteristic differences of nuclear impressions in endothelial cell under the scanning electron microscope. The quantitative analysis showed that the tensioned PDL sites in the experimental group had a higher microvascular volume density than the corresponding sites in the contrast group, while the pressed PDL sites in the experimental group had a lower microvascular volume density than the corresponding sites in the contrast group. This should result from the hydraulic mechanism in the PDL.

Parallel-plate flow chamber for studies of 3D flow-endothelium interaction.

Flow induced shear stress influences vascular cellular biology and pathophysiology in numerous ways. Previous in vitro studies on interactions between flow and endothelial cells using parallel-plate flow chambers involve two-dimensional flows, whereas flows in larger vessels are commonly three-dimensional. We have constructed a parallel plate flow chamber with a backward facing step aligned oblique to the axis of the chamber. Flow visualisation by steady injection of ink through a hypodermic tube reveals swirling flow in the recirculation region downstream of the step. At given angles of the step, theta; (to the axis of the chamber), the pitch of the swirl and the width of the separation region, as measured in the direction perpendicular to the step, increase with the Reynolds number (Re). On the other hand, at given values of Re, reduction of theta; results in increases in the swirl pitch but decreases in the width of the separation zone. Furthermore, clearance time of ink from the separation region is shorter with an oblique step than a perpendicular one at given Re. Computer simulation confirms the 3D swirling flow created by the oblique step and provides detailed distribution of wall shear stresses in the flow chamber.

[The effect of anterior cruciate ligament rupture and reconstruction on the degeneration of articular cartilage in rabbit knee].

OBJECTIVE: To investigate the effect of rupture and reconstruction of the anterior cruciate ligament (ACL) on the degeneration of rabbit knee articular cartilage. METHODS: 14 mature New Zealand white rabbits were divided into four groups. In group I, the ACL of the right knees in 7 rabbits was resected and immediately reconstructed, and the contralateral ACL was resected only in controll f group I. In group II, the ACL of the right knees in 7 rabbits was reconstructed 3 weeks after the ACL was resected and the contralateral joints in control group II, in which only a medial arthrotomy was performed. The rabbits were killed 8 weeks after the operation. The methods of ink straining, histology and SEM were used to analyze the changes in articular cartilage of the joints. RESULTS: The results of ink method and HE straining were analyzed quantitatively. The degeneration of knee articular cartilage in group I was significantly weaker than that in control group I (Hc = 5.9889, P = 0.0144). The degeneration of knee articular cartilage in group II was as serious as that in control group I (Hc = 0.7143, P = 0.785). CONCLUSIONS: Immediate reconstruction of the ACL can effectively prevent articular cartilage from degeneration. Once the articular cartilage damaged moderately, delayed reconstruction of the ACL could not effectively reduce the development of degeneration. So once the ACL is ruptured, reconstruction should be performed in the early stage to restore the stability of knee joint to prevent the articular cartilage from degeneration.

[Our experience with the Soviet "Thromb-2" thromboelastograph].

The soviet thrombelastograph "Thromb-2" is four-channel, recording the processes of coagulation of whole and citrate blood at 37 degrees C. It is used for macro- and micro tests as micro tests are performed with peripheral blood as well. The recording of the process is achieved immediately by means of a moving diagram paper for a two-sided or one-sided record, using writing apparatus with ink. The velocity of the diagram paper is such that it assures a great precission of the recording. The charaeter of the thrombelastogram itself is determined by an electrical signal, proportional to the elasticity of the formed blood coagulum. Maintenance of the thrombelastograph with paper and ink is easy, available and cheap.

[Blood-brain barrier breakdown following gliotoxic drug injection in the brainstem of Wistar rats].

Ethidium bromide (EB) causes local astrocytic disappearance, with glia limitans disruption and supposed blood-brain barrier (BBB) breakdown The aim of this study was to investigate the BBB integrity after the injection of 0.1% EB (group E) or 0.9% saline solution (group C) into cisterna pontis of Wistar rats. Brainstem fragments were collected from 24 hours to 31 days post-injection for ultrastructural study and GFAP immuno-histochemical staining. Some animals received colloidal carbon ink by intravenous route at the same periods. In rats from group C, there was no sign of astrocyte loss and no leakage of ink from blood vessels in the injection site. In group E, astrocyte disappearance began at 48 hours and some areas were still devoid of astrocytic processes 31 days after. Leakage of carbon particles was seen from 48 hours to 7 days in the EB-induced lesions. Tight junctions did not show any detectable ultrastructural change due to the lack of perivascular astrocytes.

IBM correcting selectric typewriter: an analysis of the use of the correctable film ribbon in altering typewritten documents.

From the point of view of a document examiner, the advent of the Correcting Selectric typewriter has not changed any of the procedures ordinarily used in the examination of altered typewritten material. The same precautions should be observed in conducting examinations of such material as would be observed in making examinations for any other type of possible alteration on negotiable instruments. The security question raised with the Correcting Selectric is no different than the questions raised when an eraser was put on a pencil or a chemical bleach was found to remove ink on paper. The Correcting Selectric might even be compared to the introduction of ball-point pens on the market in the late 1940s. At that time, financial publications were suggesting that ball-point pens should not be used on any negotiable or valuable document as the ink was considered to be too readily transferable, and it was thought that they left little in the way of individual writing characteristics. However, it was not long before financial institutions were supplying such pens on their counters. Because of the complexities involved in using the Correcting Selectric to alter a document, it is felt that such problems will prove to be less prevalent than those involving other means of alterations.

[Experimental study on the reconstruction of vascular links between a pedicle skin flap with preserved subcutaneous vascular network and its implanting bed].

A comparative study of skin flap with a preserved subcutaneous vascular network and conventional skin flap was undertaken in rats with determination of skin temperature, laser Doppler, ink perfusion, microangiography and transparent specimen technics. It was found that all the skin flaps with a preserved subcutaneous vascular network survived after transplantation. Three hours postoperation, the flap microcirculation was poorest (LDF, 28% of the preoperative value). Six hours postoperation, it began to augment, and microvessels containing ink could be seen under microscope at 24 hours, as compared to 3 days postoperation in the controls. On the fifth day, LDF returned to the preoperative level, and the diameter and density of reformed microvessels were similar to the vessels of the adjacent skin. This phenomenon only occurred on the 7th day in the controls. After division of the pedicle on the fifth day, all the flaps survived, while the controls showed partial necrosis. The results indicate that skin flaps with a preserved subcutaneous vascular network can survive on the blood flow of pedicle in the early stage, at 24 hours postoperation flap microvessels begin to reform, and on the fifth day flaps can survive on the reformed microcirculation in spite of the severance of the pedicle.

The forensic analysis of thermal transfer printing.

Thermal transfer printing refers to printing processes that utilize heat to produce an image by either physical or chemical means or by a combination of both. As the technology has improved and the supplies have become less expensive, the use of thermal printing in the personal and business markets has increased significantly. Specifically, dye diffusion thermal transfer and thermal mass transfer have become predominant in the production of counterfeit credit cards, drivers' licenses, and other types of documents produced on plastic media. Chemical analysis by means of thin layer chromatography (TLC) has proven to be useful in characterizing various types of inks (e.g., writing and inkjet inks). In this study, the authors examined 81 different samples that included a total of 54 printer samples (43 photographic prints on paper and eleven plastic card samples) and 27 printer ribbons. A new TLC method was developed and tested utilizing a solvent system (80% n-hexane, 3% methyl ethyl ketone, and 17% ethyl acetate) that is capable of producing excellent resolution.

A crown reconstructing method for resin-embedded transparent teeth.

OBJECTIVE: Resin-embedded transparent teeth are devoid of the original crown morphology, because enamel is lost by demineralization. This study was designed to reproduce artificial enamel and to reconstruct the original crown morphology for resin-embedded transparent teeth. METHOD AND MATERIALS: The impression of the coronal portion of human permanent teeth was taken with a silicone impression material. After demineralization, drawing ink was injected into the pulp cavities. The ink-infiltrated teeth were made transparent with methyl salicylate and embedded with polyester resin. Urethane prepolymer was injected into the impression, and the resin-embedded teeth were reinserted into the impression. After polymerization of the urethane resin, the specimens and the urethane resin were removed from the impression. RESULTS: The original crown morphology of the resin-embedded transparent teeth could be precisely reconstructed with artificial and removable enamel. The resin-embedded teeth showed morphologic details of the black-stained pulp cavities through the transparent dentin and cementum. CONCLUSION: This study established a crown reconstructing method for resin-embedded transparent teeth. The specimens will be useful for demonstration of morphology of teeth and pulp cavities.

Comparison of rotary and manual instrumentation techniques on cleaning capacity and instrumentation time in deciduous molars.

PURPOSE: The aim of this study was to evaluate, in vitro, the cleaning capacity and time needed for instrumentation of root canals of deciduous molars by manual and rotary instrumentation. METHODS: Thirty-three deciduous molar root canals were injected with India ink and divided into 3 groups: group I--the root canal instrumented manually with K files; group II--the root canal instrumented with rotary Profile .04 instruments; group III--control group, (ie, root canals not instrumented). Instrumentation time was recorded. The teeth were cleared and the removal of India ink was measured in the cervical, middle, and apical thirds. RESULTS: There was no significant difference for cleaning capacity between manual and rotary techniques in the 3 root thirds (P>.05), but both techniques were different from the control group (P<.001). Significantly less time was needed for instrumentation with the rotary technique (3.46 minutes) than with the manual technique (9.06 minutes). CONCLUSIONS: Although no differences were found for cleaning capacity, the reduction of instrumentation time by the rotary technique was a relevant clinical factor for endodontic treatment.

Comparison of near infrared light photography and middle infrared light photography for deciphering obliterated writings.

There are obliterated writings that are undecipherable by conventional visible and near infrared monochromatic light photography. Some of them could be deciphered undestructively by middle infrared light photography. Characters were written on paper with 101 kinds of pens. Middle infrared light photographs and near infrared light photographs of them were taken and compared to recognize what kinds of inks is transparent at examined wavelength regions. Based on the results obtained, the combination of ink for drawing and obliterating (which is not deciphered by visible and near infrared light photography, but expected to be deciphered with middle infrared light photography) is selected. From the examined photographs, it is demonstrated that there exist obliterated writings which are undecipherable by conventional near infrared light photography, but decipherable by middle infrared light photography.

[An experimental study of the effect of biomechanical environment on the incorporation of cortical bone plates allografts].

Sixteen goats with fractures of right femur received cortical bone plates allografts on both the sides of femurs. The right allograft strut endured the stimulation of physiological stress, and the left allograft strut did not. Groups of goats were sacrificed and specimens were procured at 3, 6, 12, and 24 week after surgery for histology observation and image analysis of the vessels after Chinese ink perfusion, the rate of bone porosity, the integrated optical density (IOD) of tetracycline fluorescence labeling and new bone formation were investigated in order to evaluate the incorporation of the allograft strut. The allograft strut revascularized at 6 weeks after surgery in the fracture group, but at 3 weeks in the control group. The rate of area of vessels after Chinese ink perfusion, the rate of bone porosity, the integrated optical density (IOD) of tetracycline fluorescence labeling and new bone formation in the fracture group were worse than control from 3 weeks to 6 weeks, but the observed and measured values were better in the fracture group than in the control group beyond 6 weeks after surgery (P < 0.05). The stimulation of stress would be harmful to the allograft strut if the strut endured the stress at an earlier period postoperation. Yet, it would be beneficial to the revascularization, new bone formation, substitution, and internal re-building on the strut provided that the extremity was immobilized for 6 weeks; and if the cortical graft endured the stimulation of physiological stress from 6 weeks postoperation till cancellous conjunction between the ends of fractures, the revascularization on the allograft strut and the bone conjunction between partially allograft strut and host would be faciliated.

[The effect of rupture and reconstruction of posterior cruciate ligament on the degeneration of articular cartilage in rabbit knee].

OBJECTIVE: To investigate the effect of rupture and reconstruction of the posterior cruciate ligament (PCL) on the degeneration of rabbit knee joint. METHODS: Thirty-three mature New Zealand white rabbits were divided into 3 groups. In experiment group I, the PCL of the right knees in 21 rabbits were resected, the contralateral joints as control group, in which only a medial arthrotomy was performed. In experiment group II, the PCL of the right knees in 12 rabbits were immediately reconstructed after PCL were resected. In experiment group I and control group the rabbits were respectively killed 6, 12 and 26 weeks after the operation. In experiment group II the rabbits were respectively killed 12, 26 weeks after the operation. The methods of ink staining, histology, immunohistochemistry and SEM were used to analyze the changes of articular cartilage of the joints. RESULTS: In experiment group I, the knee joints had obvious degeneration of articular cartilage at 26 weeks. The quantitative analysis of the results of the ink staining method showed the degeneration of knee articular cartilage was more serious at 26 weeks than that at 26 weeks in control group. Histological results demonstrated: the large area of fibrosis of cartilage could be seen in the medial condyle at 26 weeks; the fibrosis was deep into the deep layer of cartilage and the number of cells decreased; the average Mankin's score was 7.7. At 26 weeks, type I and III collagen were detected in fibrotic cartilage, while the staining of type II collagen was slight. In experiment group I, the injuries of cartilage in medial condyle could be seen at every stage through SEM, while the surface of cartilage demonstrated disorder in control group through SEM. In experiment group II, only a few specimens had mild injuries of cartilage, the degeneration of cartilage in medial condyle and trochlea were slighter at 26 weeks than that in experiment group I. CONCLUSIONS: PCL rupture can result in articular cartilage degeneration, which develops as time goes on. The degeneration of articular cartilage is more serious in medial compartment and patellofemoral joint than those in other positions. Immediate reconstruction of PCL can effectively prevent articular cartilage degeneration.

[Cryptococcal meningitis and AIDS. Clinical description of 10 patients].

We have studied 10 patients with cryptococcal meningitis and AIDS. Nine of them were intravenous drug users and four have been previously diagnosed of AIDS. In 60% of them cryptococcal meningitis was the first opportunistic infection, and as group represented only 6.3% of our 158 patients with AIDS on the same period. The most common symptoms were: malaise (100%), headache (80%), fever (60%), meningeal signs (50%). Two of them had focal neurological disease. CSF culture and serum cryptococcal antigen test were positive in 90%, the Indian ink in 77% and blood cultures in 30% of the cases, while indian ink preparation did it in 77%. MRI showed bilateral small lesions, deeply located, in 3 cases; it was also useful to prove optical tract lesions in a patient with blindness as a result of cryptococcal meningitis. We had treatment successes in 80% of the cases, all patients being treated with amphotericin B, alone in 4 and amphotericin B plus fluorocytosine in 6. Two patients died within the first 2 weeks. Maintenance therapy with fluconazole was effective and well tolerated, with 3 patients dying from causes other than cryptococcal meningitis. We recorded a survival rate over 12 months in 33% of patients.

[Determination of the sequencing of sealing and writing with Fourier IR microscopic chemical image system].

Using Fourier-transform IR microscopic chemical image system, combined with surface scan and reflection mode, non-invasive fast identification of the sequencing of writing and sealing was carried out. Results indicate that sealing prior to writing leads to evident characteristic peaks of the ink in the IR spectra, while writing prior to sealing results in clear characteristic peaks of the stamp-pad ink in the IR spectra. The method features speediness, exactness, impersonality, and non-invasion to the sample.

Low-pressure perfusion results in effective microvascular perfusion of isolated rabbit hearts during hypothermic preservation for twenty-four hours.

Hypothermic, low-pressure coronary artery perfusion with oxygenated electrolyte solutions containing oncotic agents has resulted in successful orthotopic transplantation of hearts after extended preservation periods. Coronary flow may not, however, be consistently maintained during preservation. Previous research has demonstrated that coronary flow during preservation is related to contractile function after preservation. Specific mechanisms leading to reduced coronary flow during preservation remain undefined. This study was designed to determine whether low-pressure perfusion is a mechanism for decreased coronary flow and decreased microvascular perfusion during preservation. With India ink used as a marker of flow, microvascular perfusion was measured in rabbit hearts immediately after isolation (controls) or after 24 hours of hypothermic perfusion at 13 mm Hg (preserved hearts). There were no differences in the percentage of perfused microvessels in the control hearts perfused at either 13 or 80 mm Hg (94% +/- 2% and 99% +/- 1%, respectively). Nor was there a difference in the percentage of perfused microvessels in hearts perfused at either 13 or 80 mm Hg after 24 hours of hypothermic, low-pressure perfused preservation (67% +/- 9% and 74% +/- 6%, respectively). There was a significant difference, however, between the percentage of perfused microvessels in control hearts and in hearts preserved for 24 hours. These differences were independent of the pressure at which the India ink solution was administered. A perfusion pressure of 13 mm Hg is as effective as is 80 mm Hg in providing perfusion of the coronary microvascular beds during long-term hypothermic perfused preservation.

[Retrograde cerebral perfusion and deep hypothermic systemic circulatory arrest].

This study was carried out in order to find out whether retrograde cerebral perfusion has a beneficial effect on protection of the brain during deep hypothermic circulatory arrest. Six mongrel dogs weighing 11 to 18 Kg were placed on hypothermic cardiopulmonary bypass. After induction of circulatory arrest, retrograde cerebral perfusion was implemented by using the bypass connecting the arterial and venous lines of the extracorporeal circuit to reverse flow into the cranial vena cava in 2 dogs or selective cannulation of bilateral external jugular veins in remained 4 dogs. Retrograde cerebral perfusion was continued for 60 minutes. Each dog was rewarmed, and weaned from cardiopulmonary bypass. During the experiment, electroencephalography and somatosensory evoked potentials were monitored. In one dog, India ink with gelatin was infused retrogradely into bilateral retroarticular veins and the brain subjected to microscopic examination of the distribution of the pigment. The protective effect of retrograde cerebral perfusion for the brain in comparison with deep hypothermic circulatory arrest alone was not demonstrated clearly by electroencephalography and somatosensory evoked potentials. The reason was that the external jugular veins had many functioning valves that obstructed retrograde perfusion and the internal jugular veins were rudimentary in dogs. In the dog with selective infusion of India ink into retroarticular veins, however, wide range of the distribution of the pigment in subarchnoidal venulae was observed. It is suggested that retrograde cerebral perfusion via selected route depends on anatomical variations of the venous drainage from the brain is effective. It is also considered that retrograde cerebral perfusion via the superior vena cava is available for the surgery of the aortic arch and may be beneficial.
