Quantitative selective PCR of 16S ribosomal DNA correlates well with selective agar plating in describing population dynamics of indigenous Pseudomonas spp. in soil hot spots.

We used a quantitative PCR method targeting 16S ribosomal DNA using competitive PCR for specific detection of indigenous Pseudomonas DNA in soil hot spots. The amount of Pseudomonas DNA corresponded to the number of culturable Pseudomonas bacteria on Gould's S1 agar. This represents the first use of PCR for quantification of indigenous bacteria in more than one sample of soil.

Antimicrobial activity of 8-quinolinols, salicylic acids, hydroxynaphthoic acids, and salts of selected quinolinols with selected hydroxy-acids.

Seventy-seven compounds were screened by the disc-plate method against strains of five bacteria and five fungi. A new constant was proposed to describe the antimicrobial activity of a compound in a defined system of organisms. This constant includes not only the inhibitory level of activity of the material but also the number of organisms inhibited. This constant, the antimicrobial spectrum index, was compared with the antimicrobial index of Albert.

Selective and non-selective beta receptor blockade in the reduction of portal pressure in patients with cirrhosis and portal hypertension.

To elucidate the mechanisms by which beta receptor blockade leads to a reduction of portal pressure, 18 patients with cirrhosis and portal hypertension were given comparable doses of propranolol or metoprolol. The fall in portal pressure was more marked with propranolol together with a significant reduction in hepatic blood flow, which was not seen with metoprolol. No correlation between the reduction in cardiac output and the decrease in portal pressure or changes in hepatic blood flow could be elicited in each group, but there was a direct relationship between the decrease in hepatic blood flow and fall in portal pressure in the propranolol treated patients. The difference observed may be related to blockade of beta 2 vasodilator receptors in the splanchnic circulation which will occur only with propranolol and lead to a greater fall in splanchnic blood flow than will be produced by a reduction in cardiac output alone. Metoprolol, by maintaining effective hepatic blood flow, may be preferable to propranolol in patients with severely impaired liver function.

Effects of selective and non-selective beta-adrenergic blockade on coronary dynamics in man assessed by rapid atrial pacing.

The effects on coronary dynamics of propranolol and atenolol were studied in 12 patients undergoing cardiac catheterisation for suspected coronary artery disease. Myocardial blood flow was measured using the coronary sinus continuous thermodilution technique. Data were obtained immediately after drug administration and during rapid atrial pacing. The immediate effects were similar for both drugs. A significant reduction in heart rate was accompanied by a small reduction in myocardial oxygen consumption. Changes in coronary sinus flow induced by rapid pacing were closely related to changes in tension-time index. This relation was not modified by propranolol or atenolol. Neither propranolol nor atenolol therefore has significant coronary vasoconstrictor properties. Cardioselectivity appears to be unimportant with respect to beta-adrenergic blockade and the coronary circulation.

Y2 receptor-selective agonist delays the estrogen-induced luteinizing hormone surge in ovariectomized ewes, but y1-receptor-selective agonist stimulates voluntary food intake.

Neuropeptide Y (NPY) plays a major role in the regulation of food intake, regulation of homeostasis, and neuroendocrine function. We have previously shown that third ventricular infusion of this peptide delays the estradiol benzoate-induced surge in LH secretion in ovariectomized ewes. To determine the receptor subtype that transmits this effect, we have now used the same model to infuse a Y1 receptor agonist [NPY Leu31 Pro34], a Y2 receptor agonist (PYY3-36), and a Y4 receptor agonist (pancreatic polypeptide). We monitored the surges in animals given these agonists or artificial cerebrospinal fluid by measuring plasma LH levels, and we also measured daily voluntary food intake (VFI). A low (7 microg/h) dose of Y2 agonist delayed the surge but did not affect VFI, whereas a higher dose (14 microg/h) stimulated VFI. A dose of 18 microg/h of the Y1 agonist did not affect surge generation but also stimulated VFI. A dose of 24 microg/h of Y4 agonist affected neither surge generation nor VFI. These specificities are different from those reported for the rat and human (in which a Y2 agonist causes reduction in VFI). We conclude that, in sheep, the negative regulation of the reproductive axis by NPY and Y-receptor agonists is effected via the Y2 receptors, whereas the orexigenic effects are most likely effected via the Y1 receptors.

Selective surface modification technique for improvement of chromatographic separation selectivity for sugar derivatives.

Highly cross-linked macroporous polymers were prepared utilizing ethylene dimethacrylate as a cross-linking agent, in the presence or absence of methyl-alpha-D-glucoside as a kind of template molecule with methacrylic acid as a functional monomer. After the preparation of the polymers, we applied a high temperature to the cross-linked polymers to study the changes of adsorption properties of the polymers for sugar derivatives including the template molecule utilized. Interestingly, the heat treatment up to 250 degrees C afforded improvement of relative adsorption affinity for several sugar derivatives including the template molecule, while heat treatment up to 150 degrees C did not afford those improvements. The detailed studies including polymers prepared using acrylic acid as a functional monomer instead of methacrylic acid prove that temperatures higher than the Tg temperature of the polymer derived from a functional monomer such as methacrylic acid and higher than the melting point (mp) of the sugar template are necessary to afford the observed improvement of relative affinity based on the surface modification effects through the heat treatment to cross-linked polymers.

Selective extraction of calcium on tri-n-butyl phosphate plasticized selective extraction of calcium on tri-n-butyl phosphate plasticized polyurethane foam for its spectrophotometric determination in glass and ceramics.

The present paper describes the application of a solid phase extraction system in order to separate traces of calcium from glass and ceramics for its spectrophotometric determination. The method is based on the extraction of calcium from sodium hydroxide solution by tri-n-butyl phosphate (TBP) loaded polyurethane foam (PUF), followed by its elution in hydrochloric acid. The spectrophotometric measurement of the absorbance of calcium complex with calconcarboxylic acid (2-hydroxy-1-(2-hydroxy-4-sulfo-1-naphthylazo)-3-naphthoic acid) takes place at pH 12. The following parameters were studied: effects of sodium hydroxide concentration and temperature on the extraction of calcium, time of equilibration for quantitative calcium extraction, effect of TBP concentration, effect of hydrochloric acid concentration for quantitative elution of calcium from PUF, effect of pH and concentration of calconcarboxylic acid for quantitative formation of the complex with calcium, effect of acetone on the stability of calcium-calconcarboxylic acid complex and influence of diverse ions on calcium sorption by TBP-loaded PUF. The results show that calcium traces can be separated onto TBP-loaded PUF from 0.25 mol L(-1) NaOH at 30 +/- 5 degrees C within 30 min. PUF was loaded with TBP in CCl4 (40% v/v). Elution of calcium was done in 1.0 mol l(-1) HCl. The calcium formed a complex with calconcarboxylic acid at pH 12 and absorbance was measured at 560 nm in acetone-water medium. Molar absorptivity was found to be 1.082 x 10(4) l mol(-1) cm(-1). The method obeys Beer's law from 0.10 to 5.0 microg ml(-1) Ca. The validity of the method was established by its successful application in NIST standard reference materials. The method proposed was applied to determine calcium in glass and ceramic materials. The results of the proposed method are comparable with the results of ICP-AES analysis and they are found to be in good agreement.
