[Effects of urso-deoxycholic acid (UDCA) on alpha-naphthyl-isothiocyanate (ANIT) induced intrahepatic cholestasis in rats].

Using the ANIT induced model of cholestasis in rats, the therapeutic effects of UDCA to the intrahepatic cholestasis were evaluated by changes of serum chemistry and liver histology. ANIT was administered once at a dosage of 40 mg/kg b.w. per os and UDCA was given ad libitum for 7 days by a drinking water containing UDCA at 0.5 and 5.0% solution. In the period of bile duct epithelial degeneration and necrosis, effects of UDCA for jaundice was not detected, but hepato-cellular disturbances were appeared histologically. Moreover, the elevation of serum levels of chenodeoxy-cholic acid, deoxycholic acid and lithocholic acid was accompanied. On the other hand, in the recovery stage of the bile duct epithelium, serum bilirubin was decreased significantly in the UDCA group which seemed to be related with the potent choleretic effect of UDCA. These results may indicate that UDCA is effective for the intrahepatic cholestasis in the case with no bile duct epithelial damage but in the presence of it hepato-cellular injury is introduced by the accumulated toxic bile acids in the blood.

Immunological characterization of L-triiodothyronine and L-thyroxine labelled with rhodamine B isothiocyanate.

In the present work the label of L-T3 and L-T4 with RBITC have been developed by means of a relatively easy procedure. A purification by TLC, previously optimized, offers the possibility of obtaining a good recovery of conjugates T3 and T4-RBITC. The immunoreactivity of conjugates was calculated by a formulas especially designed for this purpose. This formulas could be applied to any molecule labelled with fluorescent compounds.

[Histochemical study of colorectal adenocarcinoma by fluorescein isothiocyanate (FITC) conjugated lectins].

Binding of FITC conjugated lectins to mucin in benign and malignant colon lesions was studied by fluorescence microscopy. In 145 cases of colonic adenocarcinoma, PNA (peanut agglutinin) was found to be bound to 75% of cases, while DBA (dolichos biflorus agglutinin) to 13% only. By contrast, DBA was bound to 94% of normal colon mucosa, while PNA to 15% only. Chronic colitis, simple adenoma and inflammatory polyps had the same binding pattern as normal mucosa, but villous adenoma, mixed polyps and multiple polyposis which are considered as premalignant lesions in colon had high positive rate of PNA binding and low binding percentage of DBA. These results indicate that an exposed carbohydrate structure Gal B1-2----GlNAc is expressed in the mucin produced by colonic adenocarcinoma. Meanwhile, some normal mucin component disappears from the malignant colonic epithelium. The mechanism of alteration of human colonic mucin present in malignant transformation is briefly discussed.
