Expression of cyclin-dependent kinase inhibitors in vascular disease.

Arterial lesions in cardiovascular diseases are characterized by proliferation and migration of smooth muscle cells as well as deposition of connective tissue matrix. Factors that stimulate vascular smooth muscle cell (VSMC) proliferation are well described; however, the role of proteins that limit intimal hyperplasia is not well understood. To examine the function of Kip/Cip and INK cyclin-dependent kinase inhibitors (CKIs) in vascular diseases, the expression of p27Kip1 and p16INK was examined in VSMCs in vitro and in porcine arteries and human atherosclerosis in vivo. Western blot and fluorescence activated cell-sorting analysis demonstrated that levels of p27Kip1, but not p16INK, increased during serum deprivation of primary VSMC cultures and caused G1 arrest. p27Kip1 inhibited Cdk2 activity, suggesting that Kip CKIs promote G1 arrest in VSMCs by binding cyclin E/Cdk2. In porcine arteries, p27Kip1, but not p16INK, was constitutively expressed at low levels. Immediately after balloon injury, cell proliferation increased as p27Kip1 levels declined. Three weeks after injury, p27Kip1 was strongly expressed in intimal VSMCs when VSMC proliferation was < 2%, suggesting that p27Kip1 functions as an inhibitor of cell proliferation in injured arteries. In contrast, p16INK expression was detected only transiently early after injury. CKI expression was examined in 35 human coronary arteries, ranging from normal to advanced atherosclerosis. p27Kip1 expression was abundant in nonproliferating VSMCs and macrophages within normal (7 of 8) and atherosclerotic (25 of 27) arteries. p21Cip1 levels were undetectable in normal arteries but were elevated in atherosclerotic (19 of 27) arteries. p16INK could not be detected in normal or atherosclerotic arteries (0 of 35). Thus, the Kip/Cip and INK CKIs have different temporal patterns of expression in VSMCs in vitro and in injured arteries and atherosclerotic lesions in vivo. In contrast to p16INK, p27Kip1 likely contributes to the remodeling process in vascular diseases by the arrest of VSMCs in the G1 phase of the cell cycle.

Corneal neovascularization after excimer keratectomy wounds in matrilysin-deficient mice.

PURPOSE: Matrilysin, matrix metalloproteinase (MMP)-7, is upregulated in the corneal epithelium during wound healing after excimer keratectomy wounds. The purpose of this study was to determine the role of matrilysin in maintaining corneal avascularity during wound healing. METHODS: Matrilysin-deficient mice (n = 17) and their age-matched wild-type littermates (n = 18) were treated with 193 nm argon-fluoride excimer keratectomy (experiment I). The percentage of corneal surface occupied by neovascularization was measured with a computer image-analysis program adjusted for parallax. In another experiment (experiment II), epithelial closure was monitored with slit lamp biomicroscopy and fluorescein staining, and corneal neovascularization was confirmed by india ink perfusion, electron microscopy, and immunolocalization of CD31 and type IV collagen. Corneal micropocket assays were performed to compare the area of corneal neovascularization in matrilysin-deficient mice and wild-type littermates (experiment III). To determine whether the differences in corneal neovascularization were related to differences in angiogenic factors, the levels of basic fibroblast growth factor (bFGF) were compared with those of vascular endothelial growth factor (VEGF) in matrilysin-deficient and wild-type mouse corneas (experiment IV). RESULTS: The percentages of the corneal surface occupied by neovascularization after excimer laser keratectomy in the matrilysin-deficient mice measured 21.3% +/- 5.2% and 18.7% +/- 5.8% at days 3 and 7, respectively, compared with 5.3% +/- 2.4% and 5.5% +/- 3.4% in the wild-type littermates at days 3 (P < 0.01) and 7, respectively (P < 0.05; experiment I). No significant differences in the rates of epithelial closure of corneal wounds were observed between matrilysin-deficient and wild-type mice after wounding. Corneal neovascularization in the matrilysin-deficient mice was confirmed by india ink present in the corneal stromal blood vessels (extending from the limbus to the wound), immunohistochemical staining, and electron microscopy. Gram, Giemsa, calcofluor white, and acridine orange stains and electron microscopy showed no evidence of corneal infection (experiment II). The area of corneal neovascularization in matrilysin-deficient mice was not significantly different from that of wild-type littermates after implantation of bFGF pellets (0.91 +/- 0.55 mm(2) and 0.77 +/- 0.34 mm(2), respectively; experiment III). The levels of bFGF and VEGF (VEGF, VEGF-B, and VEGF-C) in corneal epithelial cells were not elevated in matrilysin-deficient mice compared with the wild-type mice (experiment IV). CONCLUSIONS: Matrilysin may play an important role in maintaining corneal avascularity during wound healing. The differences in corneal neovascularization between matrilysin-deficient mice and wild-type littermates seem unrelated to the bFGF and VEGF levels in the corneal epithelium.

Early glial responses after acute elevated intraocular pressure in rats.

PURPOSE: To study the responses of glial cells to a short-term elevation in intraocular pressure (IOP) in rats. METHODS: Adult Sprague-Dawley albino rats, 45 to 55 days old, were given India ink intracamerally. After 7 days, 200 spots of laser burn over 360 degrees were delivered by an Argon laser (620-637 nm; 200 mW; 200 mm; 0.2 seconds) aimed at the ink deposits in the trabecular meshwork. IOP was recorded and eye tissues at 12 hours and 1, 3, 5, 7, or 14 days after laser were examined by immunohistochemistry with antibodies against glial fibrillary acidic protein (GFAP), vimentin, S-100, ED1, and OX42. To evaluate neuronal loss, the number of cells in the retinal ganglion cell layer (RGCL) was counted on flat preparations of retinas at various times after elevation of IOP. RESULTS: Significant elevation of IOP from 1 to 7 days and loss of cells in the RGCL from 3 days onward were noted after trabecular laser photocoagulation. In the inner retina, there was a gradual and sustained increase in GFAP and S-100 immunoreactivity, but only a transient increase in vimentin immunoreactivity. No remarkable changes in GFAP, vimentin, and S-100 immunoreactivity were noted at the optic nerve head (ONH). ED1- and OX42-labeled cells were noted in the choroidal plexus, the parapapillary region of the optic nerve, and the ONH from 3 days onward, whereas expression in the retina was unremarkable. CONCLUSIONS: There is differential expression of glial cell markers in the retina and the ONH, with early loss of cells in the RGCL in response to the elevation of IOP. Macroglia such as astrocytes and Müller cells may be involved in the pathophysiology of retinal ganglion cell death or retinal repair, and activated microglial/phagocytic cells may play an important role in modulating the changes in the ONH that occur with the elevation of IOP.

Malignant atypical cell in urine cytology: a diagnostic dilemma.

AIMS: The aim of this study was to find out the characteristic morphology of malignant atypical cells which were missed on routine cytology of urine. MATERIALS AND METHODS: In this retrospective study, we examined detailed cytomorphology of 18 cases of atypical urinary cytology which were missed on routine examination and were further proved on histopathology as transitional cell carcinoma (TCC) of bladder. The cytological features of these cases were compared with 10 cases of benign urine samples. RESULTS: There were 11 cases of high grade TCC and 7 cases of low grade TCC on histopathology of the atypical urine samples. Necrosis in the background and necrosed papillae were mostly seen in malignant atypical cells. The comet cells and cells with India ink nuclei (single cells with deep black structure-less nuclei) were only observed in malignant atypical cells. The most consistent features in malignant atypical cells were: i) high nuclear and cytoplasmic (N/C) ratio ii) nuclear pleomorphism iii) nuclear margin irregularity iv) hyperchromasia and v) chromatin abnormalities CONCLUSION: The present study emphasizes that nuclear features such as high N/C ratio, hyperchromasia and chromatin abnormalities are particularly useful for assessing the malignant atypical cells. Other cytological features such as comet cells and cells with India ink nuclei are also helpful for diagnosis but have limited value because they are less frequently seen.

Vascular permeability alterations induced by arsenic.

The impact of arsenic on the integrity of blood vessels in vivo via in situ exposure (local injection) of arsenic was investigated. Vascular permeability changes were evaluated by means of the Evans blue assay and the India ink tracer techniques. Rats were intravenously injected with Evans blue followed by intradermal injections of various doses of sodium arsenite on the back skins of the animals. Evans blue at different time points was extracted and assayed as indices of vascular leakage. Skin at various time point injection sites was sampled for arsenic measurement via graphite furnace atomic absorption spectroscopy. Our time course study with Evans blue technique demonstrated a biphasic pattern of vascular permeability change: an early phase of permeability reduction and a later phase of permeability promotion at all dose levels tested. The India ink tracer technique also demonstrated a time-correlated increase in vascular labelling in the tissues examined, signifying an increase in vascular leakage with time. Moreover, we found that despite an early increase in tissue arsenic content at time of injection, tissue arsenic declined rapidly and returned to near control levels after 30-60 min. Thus, an inverse correlation between tissue arsenic content and the extent of vascular permeability was apparent. This study provides the first demonstration that in situ exposure to arsenic will produce vascular dysfunction (vascular leakage) in vivo.

Processing of defensive pigment in Aplysia californica: acquisition, modification and mobilization of the red algal pigment, r-phycoerythrin by the digestive gland.

The marine snail Aplysia californica obtains its purple defensive ink exclusively from the accessory photosynthetic pigment r-phycoerythrin, which is found in the red seaweeds of its diet. The rhodoplast digestive cell, one of three types of cell lining the tubules of the digestive gland, appears to be the site of catabolism of red algal chloroplasts (rhodoplasts) since thylakoid membranes, including phycobilisome-sized membrane-associated particles, were found within the large digestive vacuoles of this cell. Immunogold localization showed that there was a statistically significant occurrence of the red algal phycobilisome pigment r-phycoerythrin within these rhodoplast digestive vacuoles, but not in other compartments of this cell type (endoplasmic reticulum, mitochondria, nucleus) or in other tissues (abdominal ganglion). Immunogold analysis also suggested that the rhodoplast vacuole is the site for additional modification of r-phycoerythrin, which makes it non-antigenic: the chromophore is either cleaved from its biliprotein or the biliprotein is otherwise modified. The hemolymph had spectrographic absorption maxima typical of the protein-free chromophore (phycoerythrobilin) and/or r-phycoerythrin, but only when the animal had been feeding on red algae. Rhodoplast digestive cells and their vacuoles were not induced by the type of food in the diet: snails fed green seaweed and animals fed lettuce had characteristic rhodoplast cells but without the large membranous inclusions (rhodoplasts) or phycobilisome-like granules found in animals fed red seaweed. Two additional cell types lining the tubules of the digestive gland were characterized ultrastructurally: (1) a club-shaped digestive cell filled with electron-dense material, and (2) a triangular 'secretory' cell devoid of storage material and calcium carbonate. The following model is consistent with our observations: red algal rhodoplasts are freed from algal cells in the foregut and then engulfed by rhodoplast digestive cells in the tubules of the digestive diverticula, where they are digested in membrane-bound vacuoles; r-phycoerythrin is released from phycobilisomes on the rhodoplast thylakoids and chemically modified before leaving the digestive vacuole and accumulating in the hemolymph; the pigment then circulates throughout the body and is concentrated in specialized cells and vesicles of the ink gland, where it is stored until secreted in response to certain predators.

[Biological activities of natural resources around us are now in the limelight].

There are various kind of natural resources around us, and they must contain a lot of unknown bioactive substances. Some may be structurally very strange for us, and some are very familiar. But their biological activity was not unfortunately investigated in detail. Accordingly, exploring new types of pharmaceutical resources may give lead compounds of the drugs in the future. Among those natural resources, I examined squid ink and scallop soup. From squid ink, an antitumor glycoconjugate was obtained. Its polysaccharide moieties, illexin A, illexin B and illexin C, were isolated, and the spectral data and chemical transformation of their acid hydrolysate revealed to bear a unique branched repeating unit, [-3GlcA beta 1-4(GalNAc alpha 1-3)Fuc alpha 1-]n. Moreover, scallop soup gave antitumor glycogen by the action of protease. The fine structure of glycogen was investigated by the sequential enzyme digestion method using beta-amylase and pullulanase, while the unit chain was analyzed by high performance anion exchange chromatography. The results showed that the antitumor active glycogen was highly branched with shorter chain than glycogens without antitumor activity.

The time course of lymph drainage from the peritoneal cavity in beagle dogs.

Lymph drainage routes from the abdominal and pelvic cavities in beagle dogs were observed serially by following the time course of India ink administered intraperitoneally. Four systems of lymph drainage routes from the peritoneal cavity were observed in this study. The earliest drainage returned to the cranial mediastinal lymph nodes via the sternal lymph vessels; subsequently, the sternal lymph nodes located along the internal thoracic artery became involved. Then, a drainage route via the lymph vessel along the left vagus nerve was observed. The final drainage route flowed into the lateral lymph vessel through the thoracic duct located on the vertebra. These results show that India ink is absorbed from the peritoneal cavity, and that the lymph drainage first flows mainly towards the cranial mediastinal lymph nodes through the ventral lymphatic channels. Our serial observations suggest that, over time, the lymph drainage routes changed from the ventral abdominal to the dorsal thoracic lymphatic channels in the thorax.

Emergency illumination panel that displays different pictures depending on room brightness.

We fabricated an emergency illumination panel by stacking two translucent pictures, one of which was painted with ink and the other with phosphors. When the panel was illuminated by backlight or room light, only the ink-printed picture was visible to the human eye and only the phosphorescent picture was visible in darkness. A two-way mirror was inserted between the two pictures both to prevent super-imposition of the two images and to enhance phosphorescence. This phosphorescent emergency sign acted as an illuminator independently of electric power sources and provided sufficient brightness for reading newspaper headlines.

Revised Kubelka-Munk theory. II. Unified framework for homogeneous and inhomogeneous optical media.

We extend the applicability of the recently revised Kubelka-Munk (K-M) theory to inhomogeneous optical media by treating inhomogeneous ink penetration of the substrate. We propose a method for describing light propagation in either homogeneous or inhomogeneous layers using series representations for the K-M scattering and absorption coefficients as well as for intensities of the upward and downward light streams. The conventional and matrix expressions for spectral reflectance and transmittance values of optically homogeneous media in the K-M theory are shown to be special cases of the present framework. Three types of ink distribution-homogeneous, linear, and exponential-have been studied. Simulations of spectral reflectance predict a depression of reflectance peaks and reduction of absorption bands characteristic of hue shifts and significant reduction of saturation and, in turn, color gamut.

Real-time bronchoscope tip localization enables three-dimensional CT image guidance for transbronchial needle aspiration in swine.

STUDY OBJECTIVE: To determine the feasibility of using real-time bronchoscope position technology coupled with previously acquired three-dimensional CT data to enhance transbronchial needle aspiration (TBNA). DESIGN: Eight swine were given percutaneously created target lesions for TBNA. A miniature position sensor was placed at the tip of a bronchoscope, and real-time position information during bronchoscopy was presented on a monitor simultaneously displaying previously acquired three-dimensional CT data. INTERVENTIONS: TBNA of target lesions and submucosal ink-spot injection of computer-generated targets. MEASUREMENTS AND RESULTS: TBNA specimens revealed successful aspiration of target material. Distances between ink marks made at computer-generated tracheal targets varied, on average (+/- SD), 4.2 mm +/- 2.6 mm from predetermined computer-distance coordinates. CONCLUSION: Real-time bronchoscope position technology coupled with previously acquired CT images may aid with TBNA of nonvisible extrabronchial lesions.

Microfluidic ethanol biobatteries on a microchip.

This chapter outlines the methods and procedures for making a microfluidic and microfabricated biofuel cell. Commercially available screen-printing carbon inks are employed as electrodes by micromolding them onto glass microchips. The carbon ink electrodes are modified with methylene green and alcohol dehydrogenase immobilized within a modified Nafion membrane to act as bioanodes in the microfluidic system. The complete biofuel cell produces power using an external platinum cathode and an integrated microfluidic bioanode. Miniaturization of power sources, such as biofuel cells, is important in applications for implementation in small technologies (i.e., sensors, bioreactors, and lab-on-a-chip technology).

Enhancement of enzymatic digestibility of recycled newspaper by addition of surfactant in ammonia-hydrogen peroxide pretreatment.

The effect of surfactant on enzymatic digestibility was investigated during the pretreatment stage. Newspaper was pretreated with an ammonia-hydrogen peroxide mixture on a shaking bath at 40 degrees C and 130 strokes/min for 3 h. Two kinds of nonionic surfactants, NP series and Tween series, were utilized. The effect of hydrophile-lipophile balance (HLB) value of both series surfactants on digestibility was found to be negligible, even though de-inking efficiency was improved as HLB value was increased. The effect of surfactant loading on digestibility was small, below 0.5 wt%, and negligible above 0.5 wt% at 60 international filter paper units (IFPU). The percentage improvement in digestibility increased as enzyme loading decreased. Digestibility of NP-5-added sample relative to control sample, increased significantly at an enzyme loading <60 IFPU, i.e., 19 and 13% at 15 and 30 IFPU, respectively. Such an increase in digestibility was not explained clearly from the experimental results. It was also found that ink removal before enzymatic hydrolysis is very important to enhance digestibility.

Preovulatory follicular events in sheep.

To investigate the number, size and fate of the follicles greater than 2 mm in diameter involved in the differentiation of the preovulatory follicle in-vivo monitoring of growth and regression of individual follicles was performed at different times after a PG-induced follicular phase. In Exp. 1, the 3 largest follicles of each ovary were labelled with ink in the peripheral stroma at 0, 4, 8, 12, 24 or 48 h after a PG injection. At a second laparotomy, it was assessed which follicles had ovulated. In Exp. 2, a similar procedure was applied in 12 ewes which underwent repeated laparotomies at 6, 30 and 54 h after PG and after ovulation. At all times, there was a huge variability in the size at the time of ink labelling of the follicles that finally ovulated ('ovulatory' follicles). A single measurement of follicle size is therefore a poor prediction of the ovulatory follicles. However, a significant reduction in the proportion of non-ovulatory follicles within the size range of ovulatory follicles occurred at 12-24 h after PG, together with a change in follicle hierarchy at 8-24 h after PG. The results of repeated laparotomies showed that only 60% of follicles greater than 2 mm at 6 h after PG had grown by 30 h. Selection occurred between 30 and 54 h after PG as indicated by a significant drop in the number of growing follicles to a level at 54 h that equalled the ovulation rate. The growth rate of the ovulatory follicles was 1.4 and 0.4 mm per day between 6 and 30 h and 30 and 54 h after PG respectively. The shrinkage rate of non-ovulatory follicles was 1.6 mm/day.

Variations in patterns of follicle development in prolific breeds of sheep.

Prolific breeds of sheep (Romanov, Finn and Booroola Romanov crosses heterozygous for the Booroola gene (F+) were compared with breeds of lower prolificacy (Ile-de-France, Finn X Scottish Blackface, Merino X Blackface and Booroola X Romanov not carrying a copy of Booroola gene (++] by in-vivo monitoring of follicular kinetics by ink labelling during the late luteal phase and follicular phase of the oestrous cycle followed by histological examination of the ovaries or follicle dissection. At each of 3 successive laparotomies, the 3 largest follicles of each ovary were measured and ink labelled. At the final laparotomy, around the beginning of oestrus, all ewes were ovariectomized. High ovulation rate was not associated with the total number of antral follicles in any of the breeds. However, there were more follicles greater than 2 mm in diameter in Romanov and Booroola X Romanov crosses (F+) compared to their respective controls. Such a feature was not observed in Finnish Landrace compared to Finn X Blackface and Merino X Blackface ewes. A more numerous population of recruitable follicles, together with a similar incidence of selection through atresia, were the features associated with the high ovulation rate of Romanov compared to Ile-de-France ewes. The high ovulatory potential of the Finn ewes resulted from a markedly reduced incidence of selection through atresia. Booroola X Romanov ewes carrying a copy of the Booroola gene (F+) appeared to possess features of both parental breeds, including high numbers of recruitable follicles, smaller follicular size when recruitment occurs and an extended time for recruitment. Booroola X Romanov (++) ewes, not carrying the gene, appeared to have lost part of the 'Romanov characteristics' of a more numerous population of recruitable follicles. The variability in the kinetics of preovulatory enlargement, seen in these breeds of sheep, demonstrates that there are a number of pathways through which high ovulation rate can be achieved and hence through which ovulation rate might be manipulated.

[Study on the repeated oral administration method in infant rats].

The purpose of this study was to establish a new technique for repeated oral administration to infant rats. To determine the maximum volume which could be administered to infant rats the following amounts of the Chinese ink were given by metal gastric zonde for mice: 0.01, 0.04 and 0.08ml/g B. W. General conditions and the arrival distance of the Chinese ink in the gastro-intestinal tract were also observed in infant rats. The best way of administration to infant rats was decided as follows: infant rats were isolated from the dums for one hour before administration and held tenderly by their neck to sustain their mouth upward, then a metal gastric zonde for mice 2 cm long was inserted to their mouth and drug solution was injected slowly. From the observation of general conditions and pathological examination, the maximum volumes for single or repeated administration were considered to be 0.04ml/g B. W. and 0.01 ml/g B. W. respectively. Daily oral administration of 0.1ml/g B. W. of distilled water, 1% CMC solution or 1% tragacantha gum suspension for 44 days caused no effects in infant rats when administration was begun 4 days after birth. These results show that the new method for administration to infant rats is useful to evaluate the toxicity or pharmacological activity of drugs.

Three-dimensional arrangement of ductular structures formed by oval cells during hepatocarcinogenesis.

The three-dimensional arrangement of ductular structures formed by oval cells in rats fed 2-acetylaminofluorene (2-AAF) was studied by scanning electron microscopy (SEM) of biliary tract casts and light microscopy of sections of liver injected with india ink via the biliary tract. Both resin and india ink were well injected up to bile ductules, and the findings of each method correlated with each other. By the second week after 2-AAF administration, a few oval cells appeared in the periportal areas forming ductular structures which connected with the portal bile ducts. At the 4th week, increased ductular structures occupied two thirds of the lobule and formed networks communicating with each other, and with the portal bile ducts. At the 8th week, such ductular structures were compressed around hyperplastic nodules and appeared like a basket in biliary casts examined by SEM. Although a histochemical study of gamma-glutamyl transpeptidase revealed activity both on the luminal side of the ductular structures and hepatocytes in hyperplastic nodules, no transition was observed between these two cell populations. These results suggest that oval cells have characteristics more similar to those of biliary epithelia than of hepatocytes, and have no relation to the development of hyperplastic nodules.

Laser removal of tattoos.

Tattoos are placed for different reasons. A technique for tattoo removal which produces selective removal of each tattoo pigment, with minimal risk of scarring, is needed. Nonspecific methods have a high incidence of scarring, textural, and pigmentary alterations compared with the use of Q-switched lasers. With new advances in Q-switched laser technology, tattoo removal can be achieved with minimal risk of scarring and permanent pigmentary alteration. There are five types of tattoos: amateur, professional, cosmetic, medicinal, and traumatic. Amateur tattoos require less treatment sessions than professional multicolored tattoos. Other factors to consider when evaluating tattoos for removal are: location, age and the skin type of the patient. Treatment should begin by obtaining a pre-operative history. Since treatment with the Q-switched lasers is painful, use of a local injection with lidocaine or topical anaesthesia cream may be used prior to laser treatment. Topical broad-spectrum antibacterial ointment is applied immediately following the procedure. Three types of lasers are currently used for tattoo removal: Q-switched ruby laser (694 nm), Q-switched Nd:YAG laser (532 nm, 1064 nm), and Q-switched alexandrite laser (755 nm). The Q-switched ruby and alexandrite lasers are useful for removing black, blue and green pigments. The Q-switched 532 nm Nd:YAG laser can be used to remove red pigments and the 1064 nm Nd:YAG laser is used for removal of black and blue pigments. The most common adverse effects following laser tattoo treatment with the Q-switched ruby laser include textural change, scarring, and pigmentary alteration. Transient hypopigmentation and textural changes have been reported in up to 50 and 12%, respectively, of patients treated with the Q-switched alexandrite laser. Hyperpigmentation and textural changes are infrequent adverse effects of the Q-switched Nd:YAG laser and the incidence of hypopigmentary changes is much lower than with the ruby laser. The development of localized and generalized allergic reactions is an unusual complication following tattoo removal with the Q-switched ruby and Nd:YAG lasers. Since many wavelengths are needed to treat multicolored tattoos, not one laser system can be used alone to remove all the available inks and combination of inks. While laser tattoo removal is not perfect, we have come a long way since the advent of Q-switched lasers. Current research is focusing on newer picosecond lasers, which may be more successful than the Q-switched lasers in the removal of the new vibrant tattoo links.
