O/W microemulsion as a vehicle for sunscreens.

In recent years, transparent dispersions or diluted milks have been used as sunscreens. These products contain water-soluble sunscreen agents, and quite frequently are washed away from the skin. However, O/W microemulsions are now being prepared as transparent vehicles for sunscreens. They are waterproof, nonsticky, and easily spreadable. The microemulsions are prepared by using pseudoternary diagrams, by combining lipids with surfactant blends and a polar phase. Soya lecithin and decylpolyglucose produce transparent systems with the lowest percentage of surfactants. These microemulsions contain 4-methylbenzilidene camphor or octylmethoxycinnamate as sunscreen agents. Cyclomethicone, menthol, and allantoin give products a good skin feel, and stearyl methicone gives the waterproof effect. These systems show a Newtonian flux. Little permeation of the sunscreens' trough lipophilic and hydrophilic membrane is evidenced.

[Appropriate duration of postoperative oral adjuvant chemotherapy with HCFU for colorectal cancer].

We conducted a joint study of different duration of drug administration for oral adjuvant chemotherapy using camphor (HCFU) with patients having advanced colorectal cancer who underwent curative resection. The patients were randomly divided into 2 groups, according to length of HCFU administration (6-month group and 2-year group), and followed up for 5 years postoperatively. In total, 239 patients were originally enrolled, out of which 155 were chosen as subjects for this study. There was significant difference in the overall cumulative 5-year survival rate between the short-term group and the long-term group (78.1% vs 89.6%). Between the respective subgroup that was defined by tumor location (colon or rectum), no differences were observed, but there was significant difference in the subgroup that was defined by the presence/absence of lymph node metastasis (59.4% vs 83.9%). It appears that oral adjuvant chemotherapy with HCFU is more effective when administered for 2 years than for 6 months.

[Pharmacognostic research of some species of Achillea. Note 1. Volatile oils analysis].

We established the hydrodistilled essential oil yields from the dried flower heads of A. collina J. Becker ex Reichenb. (collected in Austria and Romanina), A. millefolium L. (s.l.) (Romania) and of three mixtures of some species belonging to the Achillea millefolium L. group. Constitutents of the essential oils were studied by GC method. The main components of the monoterpene fraction of the oils were 1.8 cineol, camphor, and borneol. The main active principle of the volatile oil, chamazulene, was in highest concentration (53%) in the essential oil hydrodistilled from A. collina collected in Austria. The results obtained for a mixture of some species (Plafar Botoşani) differ from literature data in the presence of bisabolol, which have not been found so far in Achillea species.

Induction of apoptosis by beta-diketones in human tumor cells.

A variety of beta-diketones were evaluated for their cytotoxic profiles against oral human normal and tumor cells. Among 22 compounds (BD1-22) tested, the cytotoxicity of 3-formylchromone (BD17) (CC50=7.8 microg/mL) against human oral squamous cell carcinoma (HSC-2) cells was higher than that of curcumin (CC50=23.6 microg/mL). Tumor cell-specific cytotoxicity was also detected in BD17 which exhibited little cytotoxic activity against a normal human cell, gingival fibroblast (HGF). (-)-3- (BD13) (CC50=21.7 microg/mL) and (+)-3-(Trifluoroacetyl)camphor (BD12) (CC50=29.7 microg/mL) are enantiomers and showed cytotoxicity comparable to curcumin and dibenzoylmethane (BD2) (CC50=22.5 microg/mL). BD13 did not induce DNA fragmentation in HL-60 cells nor activate caspase 3, 8 and 9 in both HL-60 and HSC-2 cells, regardless of the presence or absence of FeCl3. On the other hand, BD17 was found to induce apoptosis in HSC-2 and HL-60 cells, as judged by internucleosomal DNA fragmentation, caspase 3, 8 and 9 activation and dysfunction of mitochondrial membrane potential. The cytotoxic activity of BD13, BD17 and curcumin was significantly reduced by chelation with FeCl3. The tumor-specific cytotoxicity and apoptosis-inducing activity of BD17 against human tumor cells undoubtedly warrant further studies of its efficacy as a cancer chemotherapeutic agent.

[Heterogeneity of molecular mechanisms the olfactor reception].

In experiments on the frog isolated olfactory epithelium by using vital fluorescent microscope, odorants with fruit, rank, flower and camphor smell were shown to involve intracellular signaling systems in olfactory transduction. The odorants with different qualitative smells have different messenger and activity mechanisms. Intracellular messengers do not participate in reception of odorants with piquant and rotten smells. Thus the perception of different odour substances is maintained by physical and chemical processes. Hence, not only taste, carotid, medullar, but olfactory reception as well are characterised by heterogeneity of biophysical mechanisms.

Quality assessment of Flos Chrysanthemi Indici from different growing areas in China by solid-phase microextraction-gas chromatography-mass spectrometry.

Flos Chrysanthemi Indici is a common traditional Chinese medicine (TCM). In this paper, headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS) was developed for quality assessment of Flos Chrysanthemi Indici from different growing areas in China. SPME parameters such as extraction fibers, extraction temperature, extraction time and sample mass were investigated to achieve identical results to those obtained by the steam distillation (SD). The selected SPME conditions were as follows: SPME fiber coated with 65-microm PDMS/DVB, extraction temperature of 60 degrees C, extraction time of 30 min and sample mass of 1.0 g. Furthermore, four active compounds (eucalyptol, camphor, borneol and bornyl acetate) presented in the TCM were applied to evaluating the quality of Flos Chrysanthemi Indici from 20 various areas. The quality assessment was successfully performed to compare the similarity value (S) between different sample vector of Flos Chrysanthemi Indici and the standard profile vector (SPV). The results showed that the proposed HS-SPME-GC-MS was an alternative technique for quality assessment of Flos Chrysanthemi Indici samples.

Determination of sixteen UV filters in suncare formulations by high-performance liquid chromatography.

Reversed-phase liquid chromatography (RP-LC) with UV detection was developed for the simultaneous determination of 16 organic UV filters worldwide authorized in suncare products. The filters determined were: 4-aminobenzoic acid, homosalate, benzophenone-3,2-phenylbenzimidazole-5-sulfonic acid, terephthalidene dicamphor sulfonic acid, 4-tert-butyl-4'-methoxy-dibenzoylmethane, octocrylene, 2-ethylhexyl-4-methoxycinnamate, isoamyl-p-methoxycinnamate, ethylhexyltriazone, drometrizole trisiloxane, diethylhexyl butamido triazone, 3-(4-methylbenzyliden) camphor, 2-ethylhexylsalicylate, 2-ethylhexyl-4-dimethylaminobenzoate and benzophenone-4. A C18 stationary phase and a gradient of ethanol-aqueous acetate buffer containing 0.2 mM of EDTA, was used with a flow-rate of 1.0 ml/min. UV detection was carried out at 313 and 360 nm. The analysis required 32 min and the limits of detection were between 30 and 4130 mg/kg in the original suncare product. Tween 80 was used to break down the different emulsions in order to procure a proper extraction of the UV filters. The method was validated for UV filters in three matrices, oil, water-in-oil emulsion and oil-in-water emulsion. Recoveries from spiked samples were 86-113% depending on the matrix used.

Re-evaluation of cytotoxicity and iron chelation activity of three beta-diketones by semiempirical molecular orbital method.

We investigated the chelating effect of FeCl3 on three beta-diketones, curcumin [1], (-)-3-(trifluoroacetyl)camphor [2] and 3-formylchromone [3], as judged by changes in their cytotoxicity and absorption spectra. Addition of an equimolar concentration of FeCl3 almost completely abrogated the cytotoxicity and changed the pattern of absorption spectra (decrease in the peak height at 430 nm) of [1], without affecting those of [2] and [3]. A semiempirical molecular orbital method (CAChe), based on these experimental data, proposed the estimated structure of [1] chelated with FeCl3 at 1:1 molar ratio. The present study suggests that [1], but not [2] and [3], is an iron chelator.

[Two forms of Ca2+-dependent ATPase from sarcoplasmic reticulum].

Two highly purified sarcoplasmic reticulum membrane fractiones differing in their sensitivities to the uncoupling action of caffeine were isolated from white skeletal muscles of the rabbit. The main protein component of both fractions is a catalytical polypeptide of Ca2+-dependent ATPase. Treatment of the caffeine-sensitive reticular fraction by trypsin or DTNB completely removes the effect of caffeine. It was found that similar effects on the caffeine-sensitive reticular fraction are exerted by bemegride, camphor, ethymizole and cordiamine. Isolation of Ca2+-dependent ATPase from both reticular fractions and reconstruction of Ca2+-transporting vesicles were carried out. Ca2+ transport by the vesicles enriched by ATPase from the caffeine-sensitive reticular fraction is uncoupled under the effect of caffeine; however, caffeine has no effect on the vesicles enriched by caffeine-insensitive reticular ATPase. The molecular weight of caffeine-sensitive and caffeine-insensitive ATPases determined in the presence of sedium dodecyl sulfate are found to be identical. Electrophoresis in the presence of digitonin revealed different electrophoretic behaviour of the two forms of ATPase.

The insecticidal activity of Eucalyptus globulus oil on the development of Musca domestica third stage larvae.

The insecticidal activity of Eucalyptus globulus oil (Camphor oil) against the larval maturation and adult emergency of the house-fly, Musca domestica 3rd stage was evaluated under controlled laboratory conditions. Concentrations of 100%, 70%, 50% 25%, 5%, 2%, 1%, 0.9 % and 0.7% showed 100% larval mortality rate. At concentrations of 0.5%, 0.3%, 0.2% and 0.1% the larval mortality rate was 90%. On the other hand, the developed pupae did not emerge to adults. Eucalyptus globulus oil is safe, cheap and widely used in Medicine. So, it is recommended to be use in controlling larvae of M. domestica.

[Determination of nucleic acids using toluidine blue as a fluorescence probe].

A new fluorimetric method has been developed for the determination of deoxyribonucleic acid (DNA) with toluidine blue (TB) as a fluorescence probe. It is based on the fluorescence quenching of toluidine blue in the presence of DNA. In Tris-HCl buffer solution (pH 8.5), the calibration graph was linear over the range 0.10-6.00 microg x mL(-1) for ctDNA, and the detection limit was 27 ng x mL(-1). The result of the determination of DNA of camphor tree's leaf by this method was satisfactory.

Histological study of therapy for infected nonvital permanent teeth with incompletely formed apices.

The histological response of periapical tissues to root canal fillings with calcium hydroxide (Ca(OH)2)-iodoform paste, Vitapex (Neo Dental Chemical Products Co., Ltd., Tokyo, Japan), and Ca(OH)2-camphorated paramonochlorophenol (CMCP) paste in nonvital permanent teeth with incompletely formed apices was investigated in 160 root canals from 90 dog premolar and incisor teeth. Periapical inflammatory lesions were initially induced in the experimental teeth. After debridement and cleaning, root canals were filled with one of the two materials. The animals were sacrificed after 30, 60, 120, or 180 days, and histological sections of each specimen were prepared with hematoxylin and eosin staining. Histological findings showed periapical repair and apical closure in both experimental groups. However, the differences in the level of inflammation, apical closure, and reparative process among the two groups were significant (p less than 0.05). It was concluded that the Ca(OH)2-iodoform paste, Vitapex, produced better results than Ca(OH)2-CMCP paste in treatment of infected nonvital permanent teeth with incompletely formed apices.

[HPLC-investigations of imidazoquinazolone derivatives. Part 2].

In a previous paper (2) authors, reported good correlation between the HPLC (log k') and TLC (RM) retention data and the 1-octanol/water partition coefficient (log l') of imidazoquinazolone derivatives synthetized in their laboratory. In the present work the behaviour of the compounds in HPLC systems containing ion-pairing agents (camphorsulfonic acid, Na-dodecyl sulfate) has been studied. While, in reversed phase systems no change of retention indicating the formation of ion-pairs or molecule complexes was observed, camphor-sulfonic acid caused significant increase of retention in the chromatographic systems with bare silica as stationary phase. This phenomenon is interpreted by the formation of camphorsulfonic acid double layer on the silica surface and as a consequence of mixed retention mechanism. The results of a detailed study on the latter subject will be published in a next paper. Double peak formation was observed in reversed phase system in case of C1-C3 disubstituted derivatives. This was interpreted with the formation of isomeric mixtures. Theoretical considerations as well as the identical spectra of the two peaks make plausible the appearance and separation of conformational isomers.

[Changes in sunscreening products in West Germany between 1988 and 1989/90].

We give a comparative survey of preparations absorbing UV light, based on data collected in 1988 (740 products) and in 1989/90 (988 products). Regarding the ingredients and the way of labelling of these preparations, our study yielded the following results: 1. With regard to the labelling of all ingredients according to CTFA, the number of sunscreen products increased from 11% in 1988 to 21% in 1989/90, that of cosmetics containing light filters from 26% to 28%. 2. Since, during the period studied, the ingredients of 50% of the preparations were changed, we call for the obligation to make a declaration. 3. Only 14 out of 42 UV filters registered in Europe are used in more than 1% of the preparations. In sunscreen products, UV-A filters increased from 51% to 61%, whereas in cosmetics, they decreased from 46% to 37%. In the 1990 list, the UV-B filters most frequently found in sunscreen products were cinnamates (70%), those least common were PABA and PABA derivatives (19%). 4. Since hazardous UV filters, such as 4-isopropyl-dibenzol methane and 3-(4-methyl-benzylidene) camphor, were eliminated from some of the preparations, the prevalences of individual filter substances have changed between 1988 and 1989/90. 5. Only those UV filters that are currently in use should be checked in the photopatch test. 6. Close cooperation between physicians and manufacturers resulted in making products absorbing UV light safer.

An in vitro study of the diffusibility of intracanal medicaments.

In this study, the diffusion properties of the vapors of four canal antiseptics were examined. Tricresol, camphore-chlorophenol, Cresophene, and povidone-iodine in clinical doses were tested against Staphylococcus aureus and Streptococcus faecalis. Forty-five extracted human maxillary central incisor teeth were used, and their root canals were prepared. A small amount of antiseptic was placed on a cotton pellet in the pulp chamber of each tooth, and the chamber was sealed with Cavit. Vapors from each of the test medicaments produced zones of inhibition on Staphylococcus aureus and Streptococcus faecalis. The study showed that vapors of the test medicaments do pass through the apex of a tooth and thus have a potential effect at the periapical area. The largest zone of inhibition resulted from the vapor of tricresol.

[Etiology of alveolitis and its therapy with indomethacin].

The local antiphlogistic therapy of alveolitis was founded from the importance of inflammation in the etiology. Indomethacin (3% age oily solution) was analysed on their effectiveness. In the control group was treated with chlorphenol-camphor-menthol-solution (ChKM) with addition of procain substance on the gauze strip. Indomethacin is significantly more active with regard to effectiveness, decrease of pain intensity and achievement of painlessness. The local therapy of alveolitis, is free of risk with solution out of Metindol--vials under dosage up to 6 milligram indomethacin substance.
