[Investigation of erythrocyte immune function of patients with endometriosis].

OBJECTIVE: To study erythrocyte immune function of patients with endometriosis and explore the pathogenesis of endometriosis. METHODS: 38 patients with endometriosis diagnosed by laparoscopy or laparotomy and 21 normal women as the control were studied. The patients were divided into three groups by revised American Fertility Society (RAFS) classification: group one (stage I, II; n = 13), group two(stage III, n = 15), and group three(stage IV, n = 10). Using rosette formation test and polyethyleneglycol sedimentation methods, the erythrocyte immune function including C3b receptor rosette rate (C3bRR), immune complex rosette rate(ICR), erythrocyte rosette forming enhancing rate (RFER) and inhibitory rate (RFIR), and circulatory immune complex (CIC) was determined. RESULTS: The C3bRR, ICR, and RFER of group one, two, and three were decreased (P < 0.05 or P < 0.01), and RFIR and CIC of group one, two, and three were increased (P < 0.05) when compared with those of the control. All parameters had no significant difference among group one, two and three. In addition, these parameters had no significant difference in infertile groups of endometriosis as compared with those of fertile group. CONCLUSION: The erythrocyte immune function of patients with endometriosis is depressed, resulting from decreased erythrocyte immune adherence enhancer and increased inhibitor. These changes of erythrocyte immune function may be related to the pathogenesis of endometriosis.

[Significance and correlation of T lymphocyte subgroup, erythrocyte immune function and argyrophilic protein in nucleolar organizer regions in tuberculosis patients].

OBJECTIVE: To study the significance and relationship of T lymphocyte subgroups, erythrocyte immune functions and argyrophilic protein in nucleolar organizer regions (AgNORs) in tuberculosis patients. METHOD: CD3+, CD4+, CD3+/CD4+ were determined using alkaline phosphatase-antialkaline phosphatase (APAAP) method, E-C3bRR and E-ICR were assayed by observing receptor rate. Granules area of AgNORs (GA) and nuclear area (NA) were studied by image analysis system. RESULT: CD3+, CD4+, CD4+/CD8+, E-C3bRR, GA/NA in tuberculosis patients were significantly lower than those in the healthy group (P < 0.001). However, the values of E-ICR, CD8+ were significantly higher than those of control group (P < 0.001). CONCLUSION: The results of T lymphocyte subgroup, erythrocyte functions and AgNORs are in accord, and might be usefull in assessing diagnosis and prognosis of tuberculosis.

Effects of moxa-cone moxibustion at Guanyuan on erythrocytic immunity and its regulative function in tumor-bearing mice.

In the sarcoma S180 ascitic mice, the effects of moxa-cone moxibustion at Guanyuan (CV 4) on erythrocytic immunity and its regulative function were investigated. The results indicated that moxibustion at Guanyuan (CV 4) could significantly increase the decreased erythrocytic C3b receptor rosette forming rate (RBC-C3bRR), lower the raised erythrocytic immunocomplex rosette forming rate (RBC-ICR, P < 0.05 or P < 0.01), increase the decreased activity of erythrocytic immuno-accelerative factor, and reduce the increased activity of erythrocytic immunosuppressive factor (P < 0.05) in the tumor-bearing mice. This suggests that moxibustion at Guanyuan (CV 4) can strengthen erythrocytic immunity and promote its regulative function.

Use of complement receptor 1 (CD35) assay in the diagnosis and prognosis of immune complex mediated glomerulopathies.

Complement Receptor 1 (CR1) is a polymorphic glycoprotein expressed on erythrocytes, leukocytes and glomerular podocytes and has a major role in immune complex processing. In addition, it regulates the complement cascade activation by preventing formation of classical and alternative pathway convertases and by acting as a cofactor for Factor I mediated cleavage of C3. In this study, we have examined the expression of erythrocyte CR1 (E-CR1) and glomerular CR1 (G-CR1) in different kinds of nephropathies using ELISA and immunofluorescence microscopy to understand their role in immune complex (IC) mediated renal diseases. E-CR1 was significantly reduced in all categories of lupus nephritis in comparison to normal subjects and non-IC renal diseases. However, other IC mediated diseases like IgA nephropathy and membranoproliferative glomerulonephritis had normal E-CR1 levels. G-CR1 showed distinct differences between IC and non-IC mediated diseases. G-CR1 was virtually absent in lupus kidneys. In other IC mediated diseases, there was a correlation of G-CR1 expression to the IC and complement fragment deposition. G-CR1 serves as a useful diagnostic marker for IC mediated diseases while E-CR1 is useful as a prognostic marker to monitor the course of disease after the treatment has initiated.

[Study on regulatory effect of composite taixian tablet on immune function of red blood cell in patients with oral lichen planus].

OBJECTIVE: To explore the regulatory effect of Composite Taixian tablet (CTXT) on red blood cell (RBC) immune function in patients with oral lichen planus (OLP) for the sake of providing the basis of clinical medication. METHODS: Sixty patients with OLP were assigned randomly into three groups and were treated by CTXT, Tripterygium hypoglaucum tablet and polyactin-A tablet respectively. And the changes of RBC-C3b receptor and immune circulating rosette complex on the surface of erythrocytes (RBC-ICR) were measured by saccharomycetic assay. RESULTS: Effect of CTXT was superior to that of Tripterygium hypoglaucum and polyactin-A tablets. CONCLUSION: CTXT is a relatively effective remedy with less side effect, it is worthy to be studied further.

Participation of alpha-smooth muscle actin-positive cells in renomedullary interstitial cell tumors.

Renomedullary interstitial cell tumors are benign lesions which are generally discovered in specimens nephrectomized for other malignant tumors or by autopsy. We examined the histologic features of eight tumors from four patients and investigated the appearance of alpha-smooth muscle actin (ASMA)-positive cells in these tumors using immunohistochemistry. We considered that five tumors are cellular type and the remaining three as fibrous. Characteristic hyalinization was observed in two of the three fibrous tumors. All the tumors except for one fibrous type contained entrapped tubular cells. CD35-positive cells (dendritic cells) and ASMA-positive cells were observed in all the tumors, with a more frequent occurrence in the cellular type than the fibrous type. CD35-negative spindle cells were considered as fibroblasts or activated fibroblasts (myofibroblasts). The number of CD35-positive cells was higher than that of ASMA-positive cells. Additionally, the entrapped tubular cells showed the transition to spindle cells and some of them expressed for ASMA. With double immunohistochemical staining, there were some cells showing positive reactions for both CD35 and ASMA. Furthermore, an ultrastructural examination confirmed the presence of ASMA-positive filaments in the dendritic cells and myofibroblasts. The expression of TGF-beta 1 was observed not only in the tumor cells and the collecting ducts surrounding the tumor but also in the entrapped tubular cells. In addition, the intensity of TGF-beta 1 was stronger in/around the tumor than in the areas distant from the tumor. The positive cells were more numerous in the cellular type than in the fibrous type. In conclusion, ASMA-positive cells appear in renomedullary interstitial cell tumors and some of the cells may originate in dendritic interstitial cells, fibroblasts including myofibroblasts, and entrapped tubular cells. Furthermore, TGF-beta 1 may contribute to the formation of fibrosis in the tumors.

Immune adherence of nascent hepatitis B surface antigen-antibody complexes in vivo in humans.

Upon i.v. injection into humans, pre-formed immune complexes bind complement and adhere to complement receptor type I (CR1, CD35) on erythrocytes (immune adherence). However, in most circumstances antigen and antibody react in the presence of complement; such nascent immune complexes may have properties different from pre-formed immune complexes. To define whether nascent immune complexes would also adhere to erythrocytes in vivo in humans, we studied immune complexes that formed upon i.v. injection of radiolabelled hepatitis B surface antigen (HBsAg) into immunized volunteers (eight subjects with anti-HBsAb levels ranging from undetectable to 50 U/ml.; and three control non-immune individuals). Immune complexes formed immediately in the subjects with detectable levels of specific antibody, and the clearance rate of these immune complexes correlated with the anti-HBsAb level (r = 0.78, P < 0.01). A fraction of the circulating immune complexes bound to erythrocytes in the three individuals with the highest antibody level (8-15% at 10 min). The effect of CR1 number per erythrocytes was analysed in two subjects with similar antibody levels and immune complexes clearance rates: immune adherence was higher in the subject with more CR1 per erythrocytes. The same immune complexes model studied in vitro provided similar results: a fraction of nascent immune complexes bound to human erythrocytes; this immune adherence was observed only when immune complexes formed in the presence of antibody excess, and correlated with CR1 number per erythrocytes (r = 0.99, P < 0.01). Finally, adherence of nascent HBsAg-antibody immune complexes to platelets was demonstrated in rabbits. Although immune adherence involves only a small fraction of nascent immune complexes at any given time, it may be essential for the safe disposal of large nascent immune complexes.

[Observation on the immune function of circulating red cells of patients with periodontal diseases].

OBJECTIVE: To find out the relation between the causes of periodontal diseases and the immune function of red blood cells (RBC). METHODS: The functional state of SOD activity, red blood cell C3b receptor rosstte (C3bR), red cell immune complex rosstte (RBCICR), RBC immune adherence enhance factor (RFER) and RBC immune adherence inhibitor factor (RFIR) of red cells from the systemic circulation of patients with periodontal diseases were determined and compared with the normal control group. RESULTS: The SOD activity C3bR and RFER of red cells were lower, while RFIR and RBCICR were higher for patients with periodontal diseases comparing to the normal control. CONCLUSION: It indicates that the compromised function of red cells may contribute to the initiation and development of periodontal diseases.

[Part of biological activity of peptidoglycan from Streptococcus lactis SB900].

Peptidoglycan of Streptococcus lactis SB900(LABPG) was isolated. It's chemical composition was analyzed and part of biological activity was examined. The PG contained 9.84% protein, 0.871 mumol/m NAG, 1.14 mumol/mg NAM. The amino acids of relatively high concentrations were Ala, Glu, Asp and their concentrations were 1.046, 0.775, 0.304 mumol/mg respectively. Using mice as subject, the animal experiment confirmed that LABPG was non-toxic and safe. Effect of i.p. LABPG 0.5 mg/mouse on the phagocytic function were studied. It was suggested that the phagocytic activity of PM phi had markedly enhanced, the activity of serum lysozyme was increased significantly. YC-Rosette experiment suggested that the activity of C3b receptors of PM phi were increased and the YC-Rosette forming ratio were higher than controls. The difference was significant by statistical analysis. Therefore, it is considered that LABPG was able to activate M phi and improve immune function in mice.

[Effect of folium ginkgo extract on the erythrocyte immunity function and serum lipid peroxide in asphyxia neonate].

OBJECTIVE: To observe the changes of erythrocyte immunity and serum lipid peroxide in asphyxia neonate, and to study the effect of Folium Ginkgo extract (FGE) on them. METHODS: Thirty asphyxia neonates were randomly divided into 2 groups, the treated group and the control group, 15 in each group. Erythrocyte C3b receptor rosette rate (E-C3bRR), erythrocyte immune complex rosette rate (E-ICR), blood superoxide dismutase (SOD) activity and serum lipid peroxide (LPO) level were determined at 24 hrs after birth. Conventional treatment was given to both groups and FGE (15 mg/kg.d) was given to the treated group additionally for 7-8 days, then the above-mentioned parameters were re-examined and neonatal behavioral neurological assessment (NBNA) was measured as well. RESULTS: E-C3bRR and SOD lowered, E-ICR and serum LPO increased in the asphyxia neonate significantly (P < 0.05). After treatment, comparison between the two groups showed that E-C3bRR and SOD were higher, E-ICR and serum LPO were lower in the treated group than those in the control group, and NBNA scoring was obviously higher in the former than that in the latter (all P < 0.05). CONCLUSION: Decrease of erythrocyte immunity in asphyxia neonate is related to the declined anti-oxidation ability and lipid peroxidase injury. FGE could suppress the free radical production, scavenge free radicals, antagonize the lipid peroxidation injury of cell membrane and up-regulate erythrocyte immunity. It displays the effects of nerve tissue protection and hypoxia-ischemic brain injury alleviation.

[Effect of hyperthermia on the red-cell immune function of rats and its teratogenicity on developing embryos].

After rats on gestation day 10 exposed to hyperthermia(40 degrees C) for 0.5, 1.0, 2.5 and 5.0 h, red-cell C3b receptor activity(aggregate), red-cell immune complex, circulating immune complex, Nile blue sulfate (NBS) vital staining, electronic microscope and histochemical techniques were used in investigate whether the hyperthermia was disturb red-cell immune functions of pregnant rats and played some role in teratogenesis. The results showed that hyperthermia increased the level of red-cell C3b receptor activity, red-cell immune complex and circulating immune complex. On the other hand, many of them also decreased to normal level after rats were placed in room temperature for 20 h. Following the extension of hyperthermic exposure, the teratogenic incidence increased from 0.9%(1/117) to 18.3%(22/120), in apparent time-effect relationship, when pregnant rats of gestation day 11.5 were killed. It was found that hyperthermia induced.

[The changes and significance of red cell nature-immune-adhesion function in liver diseases at different stages].

OBJECTIVE: To study the changes of red cells nature immune adhesion function (RNIAF) in liver diseases at different stages, and the feasibility of RNIAF in evaluating the severity of liver diseases. METHODS: Venous blood was extracted from 682 patients with liver disease, including cirrhosis, chronic hepatitis, acute hepatitis, hepatitis gravis, and 50 healthy blood donors as controls. Suspension of red cells in self-plasma was mixed with solution of mouse ascites carcinoma cells. A tumor cell attached with 5 red cells or 2 lymphocytes/granulocytes was counted as one rosette. The adhesion rate was calculated. Serum soluble complement receptor 1 (sCR1) was measured with a newly established sandwich enzyme-linked immunosorbent assay (ELISA). The CR1 expressed on erythrocytes has assayed by cell-ELISA. RESULTS: The expression of CR1 on erythrocytes was lower among the patients with liver diseases. Compared with that in healthy controls, RNIAF decreased among the patients with various liver diseases (all P < 0.01) in the order of acute hepatitis, chronic hepatitis, cirrhosis Child's A, cirrhosis Child's B, cirrhosis Child's C, and hepatitis gravis. CHE and PTA decreased and PT increased among the patients with cirrhosis and hepatitis gravis (all P < 0.05 or P < 0.01). RNIAF was positively correlated with CHE, PT, and PTA in patents with cirrhosis. CHE, PT and PTA remained at low levels while RNIAF almost returned to its normal level in the convalescents of hepatitis gravis. CR1 expressed on erythrocytes and sCR1 in serum in various liver diseases were decreased at different degrees, however, they changed significantly later than RNIAF. Serum sCR1 significantly increased in the patients with cirrhosis, particularly in those in grades Child's A to C. CONCLUSION: Low-cost, easy to operate and with early change and stable results, RNIAF may be used as an important indicator to evaluate the severity of liver diseases. of CHE, PT/PTA. It is an important to evaluate the severity of liver function in clinic.

Protective effects of soluble CR1 in complement- and neutrophil-mediated tissue injury.

Complement activation is an important step for triggering of acute inflammatory reactions. Soluble human recombinant complement receptor type 1 (sCR1) blocks complement activation by both classical and alternative pathways. In addition to glycogen-induced peritonitis, three models of complement-dependent acute inflammatory injury have been used to assess the protective effects of sCR1: lung and dermal injury after intraalveolar or intradermal deposition of IgG immune complexes; acute lung injury resulting from intravascular activation of complement after the i.v. injection of cobra venom factor; and acute skin and lung injury (at 4 h) after thermal trauma involving 25 to 30% total body surface area. Vascular injury was quantified by increases in vascular permeability, hemorrhage, neutrophil infiltration, and, as indicated, tissue water content. Intravenous infusion of sCR1 reduced lung and dermal vascular injury in all models studied. In glycogen-induced peritoneal exudates sCR1-reduced neutrophil accumulation by 79%. In animals undergoing IgG immune complex-induced alveolitis, sCR1 treatment reduced vascular permeability and hemorrhage by 72 and 71%, respectively, and tissue accumulation of neutrophils was reduced by 68%. After cobra venom factor injection, sCR1 reduced increases in lung vascular permeability by 67%, hemorrhage by 73%, and lung myeloperoxidase content by 55%. Four hours after thermal injury of skin, sCR1-treated animals demonstrated significant protection against lung injury; increases in vascular permeability and hemorrhage were reduced by 45 and 46%, respectively, and myeloperoxidase content was lowered by 39%. In thermal injury of the skin, sCR1 injection reduced dermal vascular permeability by 25% at 1 h (p = NS) and 44% at 4 h. Water content in skin biopsies was also decreased. There was a dose-response relationship between the amount of sCR1 infused and the extent of protection in each of the injury models. These data demonstrate that sCR1 offers significant protection against complement-dependent tissue injury in the animal models studied and that the protective effects are related to reduced neutrophil content.

Role of complement in endotoxin/platelet-activating factor-induced lung injury.

C receptor-1 is a protein involved in the regulation of C3 and C5-convertases. Recombinant human soluble C receptor-1 has recently been produced and shown to reduce infarct size in a rat model of myocardial ischemia/reperfusion injury. The present study aimed to investigate whether recombinant human soluble C receptor-1 exerts any protective effect on pulmonary injury produced in a rodent model of adult respiratory distress syndrome. In this model, Escherichia coli endotoxin (LPS, 0.1 microgram/kg) combined with platelet-activating factor (1 pmol/kg/min over 60 min, n = 10) caused microvascular lung injury characterized by elevation of myeloperoxidase activity, deposition of C3 and C5b-9 on the endothelium of pulmonary vessels, and pulmonary edema. Furthermore, bronchoalveolar lavage revealed increased neutrophil count and elevated protein concentration. These pulmonary responses were associated with elevated serum TNF-alpha. Pretreatment (10 min, i.v.) with recombinant human soluble C receptor-1 at 10 mg/kg (n = 13), but not at 1 mg/kg, prevented the LPS/platelet-activating factor-induced pulmonary edema (p less than 0.01) and changes in the bronchoalveolar lavage fluid cell count (p less than 0.01) and protein concentration (p less than 0.05), and attenuated the deposition of C3 and C5b-9 to lung vessels. There was no effect on lung myeloperoxidase activity and serum TNF-alpha. Also, C depletion by cobra venom factor (500 U/kg, i.v.) eliminated the pulmonary edema and elevated leukocyte count in bronchoalveolar lavage fluid, but had no effect on lung myeloperoxidase activity and serum TNF-alpha. These data suggest that C factors may play an important role in the pathophysiology of adult respiratory distress syndrome.

[Depression of the mononuclear phagocyte system caused by high doses of narcotics].

We evaluated in human monocytes the effect of high doses of alfentanyl on the expression of vimentin filaments, the phagocytic activity and the membrane display of HLA-DR molecules in the subjects undergoing surgery. The study was performed on 30 patients, ASAI-II. The patients received 100 mcg/kg i.v. of Alfentanil and the maintenance of anaesthesia was made with Alfentanil (2-3 mcg/kg/min.). The patients were randomized in two groups. The patients were ventilated with N2O:O2 (1:1) (Group I) or air: O2 (1:1) (Group II). After surgery, all patients of the Group II received Naloxone (0.2-0.4 mg). Central venous blood samples were obtained before induction, one and two hours after induction of anaesthesia and at the end of surgery. Separation of monocytes was performed according to Boyum technique. CD35 and HLA-DR molecules and vimentin filaments were studied by indirect immunofluorescence method using monoclonal antibodies. Percentage of positive cells were read with a cytofluorometer. The phagocytic function of monocytes was determined by ingestion of latex particles. Cortisol and ACTH plasma levels were determined by RIA. High doses of Alfentanyl depress phagocytic function and membrane display of CD35 and HLA-DR molecules in monocyte and induce marked changes in the organization of vimentin filaments in these cells in patients undergoing surgery. This monocytic depression was more marked in the patients ventilated with N2O. In our results there was uninhibition of ACTH and cortisol plasma levels responses to surgical stress by Alfentanil administration. Since the effects of Alfentanil were reversed by Naloxone, an opioid receptor mechanism seems to mediate these events.

[Determination of red blood cell immune functions in patients with pulmonary tuberculosis].

Red blood cell immune functions were determined in 106 patients with pulmonary tuberculosis (tbc) and in 37 blood donors as a control group. In control group, RBC-C3bRR was 17.2 +/- 6.5%, RBC-ICR was 4.0 +/- 2.2%. In the group, both RBC-C3bRR and RBC-ICR levels were elevated in 23 patients, only RBC-C3bRR level or RBC-ICR level rose in 40 or 31 cases respectively. so, 88.6% of patients showed elevation of RBC immune function. The pathological changes were also correlated with the level of RBC-ICR, RBC-C3bRR significantly (P < 0.01). The authors suggested that the measurement of RBC immune functions might be useful in diagnosis, assessment of the severity and outcome of pulmonary tuberculosis.
