[The discovery of naked cluster particles of Parachlamydia and its developmental mechanism].

OBJECTIVE: To study the survival and developmental morphology of Parachlamydia (BN9) within Acanthamoeba. METHODS: The morphology of BN9 within Acanthamoeba was studied by inverted phase contrast microscope, electron microscope, Gimenez and AO-staining with amoebal co-culture. RESULTS: The endosomal maturation-blocked were formed after the egress of BN9. Two developmental stages-elementary and reticulate bodies, were both observed within the vacuoles. The reticulate bodies, multiplicated by binary fission, were located mainly within the vacuoles, while the elementary bodies can also be located in the plasma individually. The naked cluster particles were observed after the trophozoites cytolysis with Gimenez-staining. The light infectious trophozoites could encyst, and elementary bodies could survive within the mature cysts. CONCLUSION: The egress of BN9 could form the endosomal maturation-blocked, which was presented in two developmental stages-elementary and reticulate bodies. It exhibited the cytolysin activity that could lyse the infectious trophozoites and were expelled in the vesicles. A few light infected amoeba could encyst with survival elementary bodies in the plasma.

Chlamydia pneumoniae in atherosclerotic middle cerebral artery.

BACKGROUND AND PURPOSE: Atherosclerotic middle cerebral arteries are frequent sites of thrombosis, leading to stroke. Previous studies have suggested a role for Chlamydia pneumoniae in the pathogenesis of atherosclerosis. However, the presence of this pathogen in atherosclerotic middle cerebral arteries has heretofore not been documented. In the present study, we analyzed atheromatous plaques from middle cerebral arteries for the presence of C pneumoniae. METHODS: Atherosclerotic middle cerebral arteries from 15 cadavers who died of natural causes and corresponding nonatherosclerotic arteries from 4 otherwise healthy trauma victims were examined. Assays for C pneumoniae DNA were carried out by nested polymerase chain reaction (nPCR) specific for the C pneumoniae ompA gene. The presence of the bacterium was assessed by transmission electron microscopy. RESULTS: Five of the 15 atherosclerotic arterial samples and none of the control tissues were positive for C pneumoniae by nPCR. Particles similar in morphology and size to C pneumoniae elementary bodies were detected by transmission electron microscopy in 4 of the 5 nPCR-positive atherosclerotic samples. CONCLUSIONS: The demonstration of C pneumoniae in atherosclerotic middle cerebral arteries is consistent with the hypothesis that this bacterium is involved in acute and chronic cerebrovascular diseases.

The elementary RNP fiber--not the higher order structure--determines the all-or-none disintegration behaviour of Balbiani ring pre-messenger RNP particles upon RNase A treatment.

Balbiani ring premessenger ribonucleoprotein (RNP) particles are built from a 7-nm RNP fiber which is tightly folded into a ring-shaped RNP ribbon. Isolated particles are known to disintegrate in all-or-none fashion upon RNase A treatment. In the present study we investigated whether this mode of disintegration is dependent on an intact particle structure or is inherent in the 7-nm fiber. When treated at low ionic strength, the Balbiani ring (BR) particles lost their higher order structure and the 7-nm fiber was unpacked, as evidenced by sucrose gradient sedimentation and electron microscopy. When treated with RNase A, unfolded as well as intact particles disintegrated in the all-or-none fashion, with similar kinetics and without apparent intermediates. Proteinase K treatment, however, obliterated this pattern: the protein-free particle RNA degraded progressively. As the typical disintegration pattern of the particles was not altered by unfolding, but was lost by deproteinization, the all-or-none mode of disintegration is likely to be a property of the 7-nm RNP fiber.

Personal and microenvironmental concentrations of particles and microbial aerosol in relation to health symptoms among teachers.

A total of 81 randomly selected elementary school teachers participated in two sampling campaigns conducted 2 weeks apart during the winter. A 24-h sample collection was performed using personal and microenvironmental sampling from homes, and an 8-h sample collection was performed from workplaces of the studied subjects. Filters were analyzed for particle mass, absorption coefficient of the filter, and for both total and viable microorganisms. Comprehensive questionnaire responses were collected from the teachers concerning weekly occurred symptoms during the previous 12-month period, and they filled in symptom diaries immediately after each sampling campaign concerning symptoms during the previous 24-h and 7-day periods. The effect of different recall periods on agreement between questionnaire responses was assessed. Factor analysis was used in order to identify factors explaining the pattern of correlations within the personal, home, and work measurements. Moreover, associations between personal, home, and work measurements of pollutants and symptoms were analyzed using general estimation equations. The recall period of 7 days seemed to provide the most reliable data for the health effect assessment. Information from the factor analysis may allow reduction of variables related to the exposure assessment, and better interpretation of results. Both personal exposure and concentrations of pollutants at home were more frequently associated with health symptoms than concentrations at work. In multipollutant analyses, absorbance coefficient was positively associated with eye symptoms, and total bacteria with both cough and blocked nose.

The discovery of the 27-nm Norwalk virus: an historic perspective.

In 1972, a 27-nm virus-like particle was discovered by use of immune electron microscopy (IEM) in an infectious stool filtrate derived from an outbreak of gastroenteritis in an elementary school in Norwalk, Ohio. IEM enabled the direct visualization of antigen-antibody interaction, as the particles were aggregated and coated by specific antibodies. This allowed the recognition and identification of a 27-nm virus-like particle that did not have a distinctive morphology, was low-titered, and was among the smallest viruses known. Serum antibody responses to the 27-nm particle were demonstrated in key individuals infected under natural or experimental conditions; this and other evidence suggested that this virus-like particle was the etiologic agent of the Norwalk gastroenteritis outbreak. The fastidious 27-nm Norwalk virus is now considered to be the prototype strain of a group of noncultivatable viruses that are important etiologic agents of epidemic gastroenteritis in adults and older children.

Coherence of elementary excitations in a disordered electron system.

The localization properties of the single-particle and collective electron excitations were investigated in the intentionally disordered GaAs/AlGaAs superlattices by weak-field magnetoresistance and Raman scattering. The localization length of the individual electron was found to be considerably larger than that of the collective excitations. This suggests that the disorder has a weaker effect on the electrons than on their collective motion and that the interaction which gives rise to the collective effects increases localization.

Some observations on the electrical characteristics of radioactive (222Rn progeny) and nonradioactive aerosols.

The electrical characteristics of 222Rn, 222Rn progeny atmospheres have been investigated. Experiments were conducted in a 222Rn/220Rn Test Facility (RTTF) of the walk-in type using an electrical elutriator of the split-flow variety, originally designed by Johnston. Experiments were carried out with the "undisturbed" atmosphere, and when this atmosphere was exposed to a source of electrically charged particles produced by a negative ion-generator. Under normal conditions, the average electrical charge of the 222Rn, 222Rn progeny atmosphere was substantially less than one elementary unit. Under the influence of the ion-generator, however, the electrical charge exceeded two elementary units, and the electrical charge distribution was non-symmetrical. It was found that the particle concentration in the RTTF substantially decreased with the operation of the ion-generator.

Personal exposures to particles and microbes in relation to microenvironmental concentrations.

UNLABELLED: The aim of this study was to compare the personal exposures of particles and microbes with the exposure being assessed by stationary samplers in the main microenvironments, i.e. home and the workplace. A random sample of 81 elementary school teachers in eastern Finland were selected to perform the two wintertime 24-h measurement periods. Particle mass concentration, black smoke (BS) concentration and concentrations of viable and total microorganisms on the sampled filters were determined using personal exposure sampling and microenvironmental measurements in homes and workplaces. In this paper, the correlations between different pollutants in each environment and correlations between personal exposures and home and work concentrations are presented. The results show that personal BS exposures correlated with both home and work BS concentrations. Furthermore, the concentrations of viable fungi and bacteria were related between personal and home concentrations. The time weighted microenvironmental model underestimated the personal exposures of particle mass, viable fungi, total fungi and total bacteria concentrations but the model might satisfactorily assess personal exposure to concentrations of BS and viable bacteria. The mass concentration of total fungi and bacteria was <1% of the total particle mass concentration. PRACTICAL IMPLICATIONS: Stationary samples are only surrogate measures of personal exposures. Personal exposure measurements conducted on individuals' breathing zone are needed to assess the exposure to particles and microbes. The time weighted microenvironmental model is a useful method to assess personal exposure to combustion related particles and viable bacteria concentrations but the model underestimates personal exposures of particle mass, viable fungi, total fungi and total bacteria concentrations.

Transport on adaptive random lattices.

In this paper, we present a method for the solution of those linear transport processes that may be described by a master equation, such as electron, neutron, and photon transport, and more exotic variants thereof. We base our algorithm on a Markov process on a Voronoi-Delaunay grid, a nonperiodic lattice which is derived from a random point process that is chosen to optimally represent certain properties of the medium through which the transport occurs. Our grid is locally translation and rotation invariant in the mean. We illustrate our approach by means of a particular example, in which the expectation value of the length of a grid line corresponds to the local mean free path. In this example, the lattice is a direct representation of the "free path space" of the medium. Subsequently, transport is defined as simply moving particles from one node to the next, interactions taking place at each point. We derive the statistical properties of such lattices, describe the limiting behavior, and show how interactions are incorporated as global coefficients. Two elementary linear transport problems are discussed: that of free ballistic transport, and the transport of particles through a scattering medium. We also mention a combination of these two. We discuss the efficiency of our method, showing that it is much faster than most other methods because the operation count does not scale with the number of sources. We test our method by focusing on the transport of ionizing radiation through a static medium, and show that the computed results for the classical test case of an ionization front expanding in a homogeneous medium agree perfectly with the analytic solution. We finish by illustrating the efficiency and flexibility of our method with the results of a simulation of the reionization of the large scale structure of the Universe.

Ultrastructural studies of the nucleoids of the pleomorphic forms of Chlamydia psittaci 6BC: a comparison with bacteria.

The nucleoids of the various pleomorphic forms of Chlamydia psittaci have been examined by direct observation of infected cells and by observations on isolated particles. The fixation and staining methods used were the same as those routinely used for the examination of bacteria to facilitate the comparison of chlamydial fine structure with that of bacteria. The nucleoids of reticulate bodies were composed of fine fibrils which extended throughout these particles. The nucleoids of intermediate bodies are characterized by an electron-dense mass with which the fibrous elements are associated in a structurally coherent manner. As condensation of the intermediate bodies proceeds, the electron-dense mass becomes eccentrically located and the fibers form a distinct radiating structure. Large elementary bodies have a few fibers associated with their condensed electron-dense nucleoids but the more condensed mature elementary bodies have a very discrete and homogeneous electron-dense nucleoid which is separated from the cytoplasmic elements of these particles by a very distinct electron-transparent space. These highly condensed elementary body nucleoids are usually ovoid, but may be elongated or irregular, and a small number of these structures react very strongly with ruthenium red. While the nucleoid structure of reticulate bodies resembles that of the bacterial cell, both the condensation process and the nucleoid morphologies which result from it in intermediate and elementary bodies have no parallels among the bacteria. Thus we conclude that major differences in nucleoid organization exist between the chlamydia and the bacteria.

A universe for molecular modeling of self-replication.

An abstract two-dimensional universe is presented, in which self-replication can be modeled and studied by computer simulation. Two types of permanent elements (atoms) occupy squares of the universe. Atoms sharing a common square form a particle, with properties determined by its component atoms. Atoms, particles, and complexes of particles move and collide according to rules like those of classical mechanics. At a higher level of organization, the string of atoms in a particle is viewed as a program, whose execution can affect the space around the particle in ways similar to the operation of enzymes. Two systems that exhibit self-replication when embedded in the universe are demonstrated. The first is very simple showing elementary self-replication activity. The second is relatively elaborate and intended to show "on paper" how a collection of particle complexes could behave in a manner resembling mitosis in a simple cell.

Comparison of atomic-level and coarse-grained models for liquid hydrocarbons from molecular dynamics configurational entropy estimates.

Molecular liquids can be modeled at different levels of spatial resolution. In atomic-level (AL) models, all (heavy) atoms can be explicitly simulated. In coarse-grained (CG) models, particles (beads) that represent groups of covalently bound atoms are used as elementary units. Ideally, a CG model should reproduce the thermodynamic and structural properties of the corresponding AL model after mapping to the lower-resolution scale. In the present work, two such models are investigated: (i) the classical GROMOS atomic-level model; (ii) a CG model recently proposed by Marrink et al., which maps approximately four non-hydrogen atoms to one bead [J. Phys. Chem. B 2004, 108, 750]. The study is restricted to n-alkanes whose aliphatic fragments are abundantly found in lipids of biological interest. Additionally, cis-9-octadecene is included, as a template chain of the lipid dioleoylphosphatidylcholine (DOPC). The two representations of molecules in the liquid phase are compared in terms of average molecular structures, extent of configurational space sampled, and single-molecule entropies. An approximate method is used to estimate the rotational contributions to the absolute configurational entropy. Good correspondence between the AL and CG representations is found. The loss in configurational entropy due to the reduction in degrees of freedom upon coarse-graining of the model is estimated.

Indoor/outdoor relationships for ambient PM2.5 and associated pollutants: epidemiological implications in Lindon, Utah.

Outdoor and indoor fine particulate species were measured at the Lindon Elementary School in Lindon, Utah, to determine which components of ambient fine particles have strong indoor and outdoor concentration correlations. PM2.5 mass concentrations were measured using tapered element oscillating microbalance (TEOM) monitors and by gravimetric analysis of Teflon filter samples. Gas-phase HNO3, sulfur dioxide, particulate nitrate, strong acid, and particulate sulfate were measured using annular denuder samplers. Soot was measured using quartz filters in filter packs. Total particulate number was measured with a condensation nucleus counter (CNC). Total particulate number and fine particulate sulfate and soot were correlated for ambient and indoor measurements. Indoor PM2.5 mass showed a low correlation with outdoor PM2.5 mass because of the influence of coarse material from student activities on indoor PM2.5. Fine particle acidity and the potentiation of biological oxidative mechanisms by iron were not correlated indoors and outdoors.

Adiabatic Jacobi corrections for H2+-like systems.

The Coulomb three-body problem in Jacobi coordinates was solved by treating the distance of the particles having equal charge as a parameter. This method allows computation of electronic energies with finite nuclear masses while maintaining the notion of a potential energy curve. The rotationless ground-state electronic and the so-called adiabatic Jacobi correction (AJC) energies are presented for H2+, D2+, and HD+ at fixed internuclear separations. The AJCs are defined as the difference between the results obtained from calculations using proper finite and infinite nuclear masses. Except at the united atom limit, the AJCs are smaller than the traditional first-order diagonal Born-Oppenheimer corrections. Expectation values of proton-electron, p-e, and deuteron-electron, d-e, distances for HD+ have been computed as a function of internuclear separation. Similarly to the fully nonadiabatic approach, the present method is able to follow the symmetry breaking in HD+. Exact and approximate analytical and numerical results are given for counterfactual systems as well. In these cases changes are allowed for the values of the electron rest mass or the elementary charge, as well as for the mass or charge of the unique particle (electron).

Highly charging of nanoparticles through electrospray of nanoparticle suspension.

Patterned deposition of nanoparticles is a prerequisite for the application of unique properties of nanoparticles in future nanodevices. Recent development of nanoxerography requires highly charged aerosol nanoparticles to avoid noise deposition due to random Brownian motion. However, it has been known that it is difficult to charge aerosol nanoparticles with more than two elementary charges. The goal of this work is to develop a simple technique for obtaining highly charged monodisperse aerosol nanoparticles by means of electrospray of colloidal suspension. Highly charged aerosol nanoparticles were produced by electrospraying (ES) and drying colloidal suspensions of monodisperse gold nanoparticles. Size and charge distributions of the resultant particles were measured. We demonstrate that this method successfully charges monodisperse nanoparticles very highly, e.g., 122 elementary charges for 25.0 nm, 23.5 for 10.5 nm, and 4.6 for 4.2 nm. The method described here constitutes a convenient, reliable, and continuous tool for preparing highly charged aerosol nanoparticles from suspensions of nanoparticles produced by either wet chemistry or gas-phase methods.

Electron microscopy of the in vivo internalization of virulent Chlamydia psittaci 6BC strain.

The internalization of virulent Chlamydia psittaci 6BC particles by wandering mononuclear phagocytes in the peritoneal cavity of intraperitoneally inoculated mice occurred asynchronously, i.e., fragile reticulate bodies (RB) appeared to be more readily phagocytized than the rigid elementary bodies (EB). Early damage of mononuclear phagocytes occurred after internalization of chlamydiae. This was followed by a decreased uptake of particles, and may explain the relatively long persistence (up to 6 h after inoculation) of free, extracellular, "swollen", and RB-like particles. Internalized particles within phagolysosomes showed varying degrees of disintegration. The subsequent influx of polymorphonuclear phagocytes and monocytes into the inflammed peritoneal cavity may explain the rapid disappearance of chlamydiae and their antigens from the peritoneal fluid. The alteration in ultrastructure of peritoneal cells and chlamydial parasites during the inflammatory process are discussed.

Stochastic particle formulation of the vesicle hypothesis. Relevance to short-term phenomena.

Based on the vesicle hypothesis, the modes of elementary quantum-vesicle interactions have been formulated in terms of probabilities of induced and spontaneous interstate quanta transitions and generalized within the framework of the previously developed theory of the double barrier synapse. Among the three allowed states for a quantum, the transition state is a novel formulation for the fraction of immediately available quanta governed by both vesicle and presynaptic membranes. The parameters of the model were determined by fitting solutions to the experimental curves representing effects of single pulse and short train activation on transmitter release at the frog neuromuscular junction. Qualitative differences in particle physics of transmitter release found under low quantal outputs on one hand and normal transmission on the other allowed the formulation of the uncertainty hypothesis and the quantum condition of synaptic homeostasis.

Personal exposures and microenvironmental concentrations of particles and bioaerosols.

The aim of this study was to compare the personal exposure to particles and bioaerosols with that measured by stationary samplers in the main microenvironments, i.e., the home and the workplace. A random sample of 81 elementary school teachers was selected from the 823 teachers working for two councils in eastern Finland for the winter time measurement period. Bioaerosol and other particles were collected on filters by button samplers using personal sampling and microenvironmental measurements in homes and workplaces. The 24-hour sampling period was repeated twice for each teacher. Particle mass, absorption coefficient of the filter and the concentration of viable and total microorganisms were analyzed from each filter. In this paper, the study design, quality assurance principles and results of particle and bioaerosol exposure are described. The results show that particle mass concentrations, absorption coefficient and fungi were higher in personal exposure samples than in home and workplace samples. Furthermore, these concentrations were usually lower in the home than in the workplace. Bacterial concentrations were highest in heavily populated workplaces, while the viable fungi concentrations were lowest in workplaces. The fungi and bacteria results showed high variation, which emphasises the importance of quality assurance (duplicates and field blanks) in the microbial field measurements. Our results indicate that personal exposure measurements of bioaerosols in indoor environments are feasible and supplement the information obtained by stationary samplers.
