Retention of ionizable compounds in high-performance liquid chromatography. IX. Modelling retention in reversed-phase liquid chromatography as a function of pH and solvent composition with acetonitrile-water mobile phases.

The influence of pH and solvent composition of acetonitrile-water mobile phases on the retention of acids and bases on a polymeric stationary phase is studied. Very good relationships between retention and mobile phase pH are obtained if the pH is measured in the proper pH scale. The fit of retention to pH for a particular solvent composition provides the pKa values of the equilibria between the different acid-base species and the retention parameters of these species at this solvent composition. Several models are tested that relate these parameters to solvent composition and properties in order to propose a general model to predict retention for any mobile phase pH and composition.

Gas-liquid chromatography and gas-liquid chromatography combined with mass spectrometry of a butyl ester formulation of (2,4-dichlorophenoxy) acetic acid.

A method is presented for the analysis of (2,4-dichlorophenoxy)acetic acid (2,4-D) in commercial formulations, together with concurrent detection and quantitation of related compounds. The method is based on direct gas-liquid chromatographic (GLC) analysis of the formulation to estimate the various esters present, and on an analysis of the total 2,4-D present involving a prior cleanup, alkaline hydrolysis, acidification and n-butylation steps. This procedure also quantitates any other related acids or phenols present. A step which would extract dioxin impurities is also included. The method was checked by the accepted acid-base estimation of 2,4-D content, GLC, and by subjecting radioactive-labelled herbicide to the above procedures.

High-performance liquid chromatography of amino acids, peptides and proteins. LXVII. Evaluation of bandwidth relationships of peptides related to human beta-endorphin, separated by gradient-elution reversed-phase high-performance liquid chromatography.

The bandwidths of several polypeptides related to human beta-endorphin have been investigated with different n-alkylsilica stationary phases and different elution gradients of 0.1% trifluoroacetic acid-water-acetonitrile mobile phases. In particular, we have examined the influence of changes in the gradient steepness parameter, b, on peakwidth with five different octadecylphases, chemically bonded to porous spherical silica particles of nominally 4 micron and 6 micron average particle diameter, respectively. The effect on the zone dispersion of these polypeptide solutes as the average pore diameter of the silica matrix was increased from 7.3 nm to 30 nm with stationary phases of similar ligand densities packed into columns of identical configuration has been further documented. The experimental data on solute bandwidths and peak capacities are comparable with the corresponding bandwidth and peak capacity values calculated from analytical equations, derived from the general plate height theory and from gradient elution theory. These comparisons clearly demonstrate that anomalous bandbroadening phenomena may occur when polypeptides are eluted with steep gradients, i.e. with gradients of large b values. Moreover, as the relative chromatographic residence times of beta-endorphin peptides capable of forming a C-terminal amphiphilic secondary structures is increased, i.e. as the dwell times and median capacity factors, k, for such peptides are increased, significant divergencies arise between the observed peakwidth behaviour and the behaviour predicted by analytical relationships which describe either the dependency of peak bandwidth (as 4 sigma v) on the gradient steepness parameter, b, or the dependency of peak capacity on gradient time, tG, median capacity factor, k, and the Knox parameter, C, respectively. The importance of these divergences from the predicted bandwidth and peak capacity behaviour for polypeptides separated on reversed phases, and for resolution optimisation in particular, is evaluated. These investigations thus enable further assessment of the quantitative relevance of current models that describe polypeptide zone migration under gradient elution reversed-phase chromatographic conditions in which solute-dependent slow equilibria, mediated by conformational or solvation effects, may still occur.

Determination of beta-carbolines in foodstuffs by high-performance liquid chromatography and high-performance liquid chromatography-mass spectrometry.

A high-performance liquid chromatographic method combined with fluorimetric detection is described for the determination of beta-carboline (norharman) and 1-methyl-beta-carboline (harman). The analysis of foodstuffs for the identification of beta-carbolines is facilitated by clean-up samples using Bond Elut PRS cartridges. Recoveries were excellent. Further, a high-performance liquid chromatographic-mass spectrometric method was also developed for their identification. The concentration of beta-carboline among the foodstuffs and alcoholic beverages varied greatly. Also, norharman and harman were observed in uncooked foodstuffs, whereas acetaldehyde was found in most fermented food. The toxicological implication of beta-carbolines in foodstuffs is discussed.

High-performance liquid chromatography of amino acids, peptides and proteins. XLVIII. Retention behaviour of tryptic peptides of human growth hormone isolated by reversed-phase high-performance liquid chromatography: a comparative study using different chromatographic conditions and predicted elution behaviour based on retention coefficients.

The retention behaviour of the tryptic peptides of human growth hormone under a variety of reversed-phase chromatographic conditions has been investigated. By using water-acetonitrile gradients containing phosphate, bicarbonate, trifluoroacetate, or heptanesulphonate buffers, the influence of these ionic modifiers on peptide selectivity with several different alkylsilicas has been examined. Comparisons of observed and predicted retention behaviours for the various tryptic peptides in the different chromatographic systems are described. Application of these systems to the isolation of deamidated forms of human growth hormone is presented.

Liquid chromatography-mass spectrometry of trace compounds with a moving-belt interface and multi-dimensional chromatography.

A high-performance liquid chromatographic method with on-line mass spectrometric detection is described for the structural analysis of a number of synthetic impurities, present at trace levels in almitrine. To obtain mass spectra with various ionization methods and high-resolution mass measurements, a moving-belt liquid chromatograph-mass spectrometer interface is used. A two-column switching system allows the injection of large amounts of almitrine, from which the trace compounds are trapped on a second column, while discarding the major component. This permits the introduction of the impurities into the mass spectrometer by elution of the second column, without the risk of introducing too large an amount of the major compound into the mass spectrometer. The mass spectra thus obtained are of sufficient quality to permit a correct structural assignment of the impurities.

High-performance liquid chromatography of amino acids, peptides and proteins. LXIX. Evaluation of retention and bandwidth relationships of myosin-related peptides separated by gradient elution reversed-phase high-performance liquid chromatography.

The gradient elution behaviour of ten peptide analogues encompassing the primary amino acid sequences adjacent to the serine-19 phosphorylation site of myosin light chain has been investigated using a 30-nm pore diameter n-butylsilica stationary phase and 0.1% trifluoroacetic acid-water-acetonitrile mobile phases. Quantitative expressions derived from linear solvent strength theory and general plate height theory have been used to assess the influence of gradient time and flow-rate on the relative retentions and bandwidths of these peptides. The retention behaviour of these peptide analogues was found to closely mirror the predictions of linear solvent strength gradient elution theory. Although the experimentally observed band-width changes over a conventional range of gradient conditions (i.e. tG = 30-60 min) also correlated with bandwidth changes predicted on the basis of plate theory derived for low molecular weight organic molecules, for very steep and very shallow gradients divergencies from theory were observed. The significance of these discrepancies is discussed in relation to the influence of amino acid sequence changes rather than composition of these peptide analogues. These data allow the importance of sequential effects on the chromatographic behaviour and in particular, the resolution optimisation of these closely related peptides to be evaluated.

High-performance liquid chromatography of amino acids, peptides and proteins. C. Characterisation of coulombic interactive regions on hen lysozyme by high-performance liquid anion-exchange chromatography and computer graphic analysis.

The molecular characteristics of the dominant anion-exchange binding site of hen egg white lysozyme (HEWL) has been investigated using a combination of high-performance liquid chromatographic techniques and computer graphic analysis of the X-ray crystallographic structure. These studies have indicated that the site of highest electrostatic potential, in terms of the density of negatively charged amino acid side chains, is located around the catalytic cleft area. The four residues tentatively identified to be involved in the electrostatic binding domain were aspartic acid 48, 52, 101 and glutamic acid 35. The number of these charged groups correlated with the maximum value of the chromatographically determined retention parameter (Zc value). Variations in the range of experimental Zc values obtained under different elution conditions have been interpreted in terms of conformational flexibility of the structural domains of HEWL which result in the opening or closure of the catalytic cleft during the retention process.

High-performance liquid chromatography of amino acids, peptides and proteins. LVII. Analysis of radioiodinated thyrotropin polypeptides by reversed-phase high-performance liquid chromatography.

The chromatographic behaviour of bovine and human thyrotropin, radiolabelled stoichiometrically with lactoperoxidase, on octadecylsilicas and other reversed-phase n-alkylsilicas has been investigated. As part of this investigation the effects of a variety of elution systems on resolution and recovery have been examined. Analysis of the tryptic peptides of radioiodinated bovine thyrotropin (bTSH) preparations by reversed-phase high-performance liquid chromatographic mapping methods resulted in the separation of more than fourteen major radioactive peptide components. The data indicate that bTSH is radioiodinated with unequal incorporation into both the alpha- and beta-subunits. Further, assessment of the level of microheterogeneity of radiolabelled bTSH preparations can be achieved with these reversed-phase techniques.
