Effect of chemical modification with p-bromophenacyl bromide on the enzymatic and lethal properties of phospholipase A2 from Bothrops alternatus (Víbora de la Cruz) venom.

The effects on lethal potency and enzymatic activity were determined following alkylation, with p-bromophenacyl bromide, of the acidic toxic phospholipase A2 from Bothrops alternatus. The modified B. alternatus enzyme, which lost its enzymatic activity, retained considerable toxicity. Histopathologic studies on mice have demonstrated features similar to those of the native enzyme. However, the distribution of the damage was different and the survival time was longer. It is concluded that the enzyme activity is not important for the lethal action of the enzyme although it influences the distribution of the damage and survival time.

Contribution of bee venom phospholipase A2 contamination in melittin fractions to presumed activation of tissue phospholipase A2.

Melittin from bee venom has been suggested to activate tissue phospholipase A2 (PLA2) activity, and subsequently has been used as a specific PLA2 probe. The melittin in most cases was obtained commercially and used without further purification or treatment. To test the hypothesis that commercially obtained melittin specifically activates tissue PLA2, we radiolabeled the lipids of immortalized epithelial cells by incubating the cells for 22 hr with 14C-linoleic acid. The cells were then incubated with 2 microM melittin, 2nM bee venom PLA2, 2 microM melittin treated with p-bromophenacyl bromide (p-BPB) or PLA2 plus p-BPB-treated melittin. Lipids were extracted and separated by thin-layer chromatography. The radioactivity in each lipid fraction was then quantitated. The melittin-stimulated PLA2 activity observed in cells was primarily associated with phosphatidylcholine. Fatty acid release was decreased by 75% when the melittin fraction was pretreated with p-BPB to reduce contaminating venom PLA2 activity. Adding PLA2 to the p-BPB-treated melittin at an amount about equal to the original contamination (0.1%) resulted in the same PLA2 activity in cell as observed with the untreated melittin fraction. These findings suggest that bee venom PLA2 contamination, even at very low levels, can account for approximately 75% of the PLA2 activity in cells treated with commercial melittin fractions.

p-Bromophenacyl bromide modification of Bothrops asper myotoxin II, a lysine-49 phospholipase A2, affects its pharmacological activities.

Modification of Bothrops asper myotoxin II, a lysine-49 phospholipase A2 variant, was carried out with p-bromophenacyl bromide. Modified toxin did not show changes in its charge and immunological properties but two of its pharmacological activities were modified. Myotoxic activity, measured by histology and by increment of creatine kinase levels in plasma of mice, was significantly reduced after toxin modification. In addition, liposome disruption activity was also significantly lower with the modified toxin both at 3 and 24 hr of incubation with the alkylating reagent. Some of the implications of these results on the structure-function relationship of myotoxins are discussed.

Cardiotoxin 1 from cobra (Naja naja atra) venom causes necrosis of skeletal muscle in vivo.

Cardiotoxin 1 from cobra (Naja naja atra) venom was tested for its ability to cause necrosis of skeletal muscle cells after i.m. injection into mice. Light and electron microscopic examination of tissue indicated that the toxin caused necrosis of skeletal muscle as early as 30 min after injection. The plasma membranes of affected cells were ruptured in the area of delta lesions, and the myofibrils were condensed into dense clumps alternating with clear areas containing elements of the sarcotubular system and damaged mitochondria. By 24 hr the affected cells appeared as empty 'bags' containing only remnants of myofibrils and swollen mitochondria. To eliminate the possibility that the necrosis was due to contaminating phospholipase A2 (PLA2) activity of the sample, the sample was treated with p-bromophenacyl bromide (p-BPB), a known inhibitor of PLA2 activity. The p-BPB-treated preparation caused myonecrosis in vivo in mice, and the treatment caused a significant decrease in the release of fatty acids and no detectable lysophospholipid in human muscle cell cultures treated in vitro with the preparation, indicating the lack of PLA2 activity. Additionally, purified PLA2 from the same venom failed to cause myonecrosis in vivo at doses equal to or ten times the estimated contaminating concentration. Thus, it is concluded that cardiotoxin 1 from Naja naja atra venom causes necrosis of skeletal muscle cells in vivo upon i.m. injection.

Immunochemical relatedness between secretory phospholipase A2 and intracellular phospholipase A2 activity linked with arachidonic acid mobilization in macrophages.

To determine whether low mol. wt phospholipase A2 (PLA2) is involved in the mechanism of arachidonic acid mobilization induced by zymosan in mouse peritoneal macrophages, we developed an immunoblot analysis and ELISA assay using the polyclonal antibodies anti-PLA2 type I and type II previously characterized. We also measured the effect of low mol. wt PLA2 inhibitors such as p-bromophenacyl bromide, aristolochic acid, nordihydroguaiaretic acid, all trans-retinal and all trans-retinoic acid on the action of these enzymes. The antibodies antitype I PLA2 bound to mouse platelet protein fraction, while the antibodies antitype I or antitype II did not recognize components of the macrophages. Furthermore, low mol. wt PLA2 inhibitors did not inhibit arachidonic acid release produced during the phagocytosis of zymosan. This suggests that low mol. wt PLA2 are not present in cells such as macrophages. These results are consistent with a role for high mol. wt PLA2 in arachidonic acid mobilization.

Biosynthesis of platelet-activating factor (PAF-ACETHER). II. Involvement of phospholipase A2 in the formation of PAF-ACETHER and lyso-PAF-ACETHER from rabbit platelets.

The role of phospholipase A2 (PLA2) in the formation of platelet-activating factor (PAF-acether) by rabbit platelets is supported by several pieces of evidence. First, the release of PAF-acether was accompanied by that of its deacetylated analog, lyso-PAF-acether. Second, EDTA, EGTA, db-AMPc, p'-bromophenacylbromide and 874 CB, which, in spite of their structural diversity, are all PLA2 blockers, inhibited the release of both PAF-acether and of the lyso-compound. Third, addition of hog pancreas PLA2 to platelets as well as platelet lysis resulted in the release of lyso-PAF-acether, thus mimicking the metabolic events initiating formation of PAF-acether. These results indicate that PLA2 activation triggers both the second and the third pathway of platelet activation.

Studies on the mechanism of antiaggregatory effect of Moutan Cortex.

The effects of Moutan Cortex and one of its major components, paeonol, on platelet aggregation and arachidonic acid (AA) metabolism in human platelets were studied. One week oral administration of water extract of Moutan Cortex [Moutan Cortex (w), 3 g/day] significantly reduced platelet aggregation and thromboxane B2 (TXB2) formation induced by collagen, epinephrine and ADP. Paeonol dose-dependently inhibited ADP and collagen induced platelet aggregation in vitro. Moutan Cortex (w) and paeonol dose-dependently inhibited the conversion of exogenous [14C]AA to [14C]heptadecatetraenoic acid [( 14C]HHT) and [14C]TXB2 by washed human platelets, while both of them increased its conversion to [14C]12-hydroxy eicosatetraenoic acid [( 14C]12-HETE). High dose of Moutan Cortex (w) inhibited the release of [14C]AA from prelabeled platelets in vitro, while paeonol did not. These results suggest that a reduction in platelet aggregation by the oral administration of Moutan Cortex might be ascribed to a decrease in thromboxane synthesis and that paeonol might play an important role in the antiaggregatory effect of Moutan Cortex because of its potent inhibitory effect on platelet aggregation and thromboxane formation.

Effects of acid proteinase inhibitors on human neutrophil chemotaxis and lysosomal enzyme release. II. Bromphenacyl bromide and 1,2-epoxy-3-(p-nitrophenoxy)propane.

Two active site inhibitors of acid proteinases were tested for their effects on human neutrophil chemotaxis and lysosomal enzyme release. Both bromphenacyl bromide (BPAB) and epoxy-p-nitrophenoxypropane (EPNP) inhibited chemotaxis and chemotaxin-induced enzyme release elicited by pepstatin and formylmethionyl peptides, which share membrane receptors, and also by zymosan-activated serum, the major active component of which (C5a) occupies a different receptor. In contrast to the previously tested acid protease inhibitor diazoacetylnorleucine methyl ester, neither BPAB nor EPNP blocked binding of [3H]fMLP to neutrophils. Thus BPAB and EPNP inhibit chemotaxin-mediated neutrophil functions, but not by interaction with the chemotaxin receptor.

Effects of inhibitions of thromboxane synthesis on reactions of the cat bronchus in situ to prostaglandins.

Cats were anesthetized by chloralose, relaxed by suxamethonium, and ventilated. The influence of 3 inhibitors of thromboxane synthesis, N.0096 (+-alpha -benzyl-alpha-p-chlorobenzyl-4-hydroxyacetophenon phenylhydrogen-phosphonate), imidazole and dipyridamole were studied on bronchial reaction to prostaglandins E2 and F2 alpha and arachidonic acid. During a bronchoconstriction maintained by infusion of 5-HT, N.0096 and imidazole dose-dependently potentiated the intensity and duration of the bronchodilator effect of PGE2. This effect was maximal at a total dose of 17 mg/kg N.0096 or 25-50 mg/kg imidazole, but decreased after higher doses which are supposed to inhibit cyclo-oxygenase also. Bronchoconstrictor reactions to single injections of arachidonic acid were inhibited by N.0096 in the same dose range. Reactions to PGF2 alpha were unaltered by both drugs. Dipyridamole was devoid of a comparable activity.

The effect of an orally active leukotriene D4/E4 antagonist, LY171883, on antigen-induced airway responses in allergic sheep.

Leukotriene (LT) D4 is a putative mediator of allergic asthma: inhaled LTD4 produces early and late increases in specific lung resistance (SRL) and slows tracheal mucus velocity (TMV) similar to inhaled antigen. In this study we examined the effects of an orally active LTD4/LTE4 antagonist, LY171883 [1-less than 2-Hydroxy-3-propyl-4-less than 4-(1H-Tetrazol-5-yl) Butoxy greater than Phenyl greater than Ethanone], on early and late changes in SRL and TMV following airway challenge with Ascaris suum antigen in conscious allergic sheep. SRL and TMV were measured before and up to 8 h and 24 h after antigen challenge after either LY171883 (30 mg/kg, p.o. 2 h before challenge) or placebo pretreatment. After placebo pretreatment antigen challenge resulted in significant early (483% over baseline) and late (221% over baseline) increases in SRL (n = 9). LY171883 pretreatment, however, significantly reduced the early increase in SRL (163% over baseline) and blocked the late response. LY171883 did not prevent the antigen-induced fall in TMV from 5-8 h post challenge (n = 6), but TMV recovered more rapidly in the drug trial returning to baseline values by 24 h. These results suggest that the generation of LTD4, and its metabolite LTE4, during airway anaphylaxis contributes to the early increase in SRL and is important for eliciting the late increase in SRL as well as contributing to the fall in TMV.

Separation and quantification of prostaglandins E1 and E2 as their panacyl derivatives using reverse phase high pressure liquid chromatography.

Separation and quantification of prostaglandin E1 (PGE1) and prostaglandin E2 (PGE2) were achieved using reverse phase high performance liquid chromatography (HPLC). Panacyl bromide (p-(9-anthroyloxy)phenacyl bromide) (PAB) derivatives of PGE2 and PGE1 were prepared. Reverse phase HPLC using a linear gradient of 56% to 80% acetonitrile in water containing 0.10% acetic acid gave baseline resolution of the two derivatives. A 3 um diameter particle, C18 column provided good resolution and reproducible recoveries. Human synovial tissue cells were incubated with the precursor fatty acids for PGE1 or PGE2 and stimulated with a crude Interleukin 1 (IL-1) preparation. Cells grown in the presence of dihomogammalinolenic acid (DGLA), the precursor for PGE1, made significantly more PGE1 than cells grown in control medium or in the presence of arachidonic acid, precursor for PGE2. PGE2 synthesis was reduced when DGLA was added to cells (resting or IL-1-stimulated).

Pharmacological characterization using selected antagonists of the leukotriene receptors mediating contraction of guinea-pig trachea.

The effects of six leukotriene (LT) antagonists on LTC4-, D4- and E4-induced contraction of isolated guinea-pig tracheal spirals were examined. Concentration-response effects of the leukotrienes were determined by cumulative addition in the presence of indomethacin (5 microM) alone for LTE4, or with 10 mM of either glutathione or L-cysteine to inhibit metabolism of LTC4 or LTD4, respectively. Concentration-response curves to the LTs were obtained in the absence and presence of Wy-45,911, Wy-44,329, FPL-55,712, Ly-171,883, Wy-48,252 and ICI-198,615 representing three structurally different chemical groups of LT antagonists. At 30 microM, the antagonists produced little or no antagonism of LTC4-induced contractions. Analysis of the Schild plots for antagonism of LTD4 and E4 suggested two receptors for the agonist effects of LTD4 and a single receptor for the agonist effects of LTE4. Comparison of pA2 values for Wy-45,911, FPL-55,712, LY-171,883 and Wy-48,252 provided evidence that LTE4 is acting at the antagonist high affinity LTD4 receptor to produce contractile effects. From the data, we conclude that there are three LT receptors (one for LTC4 and two LTD4 subtypes) through which exogenously applied LTs evoke contraction of the isolated guinea-pig trachea.

Effects of 3,4-dihydroxyacetophenone on the biosynthesis of TXA2 and PGI2 in human placental villus and umbilical artery segments in vitro.

In this paper, the effects of 3,4-dihydroxyacetophenone, DHAP (Qingxintong), an active constituent of traditional Chinese medicine, on the biosynthesis of TXA2 and PGI2 in human placental villi and umbilical artery segments of normal term pregnancy in vitro were studied by a perifusion technique. The collected fractions were assayed by radioimmunoassay for TXB2 and 6-keto-PGF1 alpha. The results showed that DHAP inhibited TXA2 and PGI2 production by umbilical artery segments in a dose dependent fashion and in both tissues TXA2 was more sensitive to inhibition than was 6-keto-PGF1 alpha. According to these data it is suggested that DHAP might be useful in treatment of pathologic pregnancies with chronic defective utero-placental circulation such as PIH and IUGR to improve this circulation.

Leukotrienes cause mesenteric vasoconstriction and hemoconcentration in rats without activating thromboxane receptors.

Thromboxane A2/prostaglandin H2 (TP)-receptor activation has been reported to participate in some of the responses to peptide leukotrienes (LT). We examined the effect of TP-receptor antagonism on LT-induced mesenteric vasoconstriction and hemoconcentration in anesthetized rats. The antagonist used in these studies, SQ 30,741, was shown to have high selectivity and potency for vascular TP-receptors in the rat. Arterial (i.a.) injection of LTC4 and D4 elicited dose-dependent and transient reductions in mesenteric blood flow without changes in arterial blood pressure. These responses were unaffected by a dose of SQ 30,741 which produced approximately 99% inhibition of similar responses to U-46,619. In contrast, LT-induced mesenteric vasoconstriction was inhibited approximately 90% by two LT antagonists, LY 171,883 and SKF 104,353. In other experiments i.v. infusion of LTD4 caused increases in hematocrit and reductions in arterial blood pressure that were not influenced by SQ 30,741. These data suggest that increases in mesenteric vascular resistance and hemoconcentration in response to LTs are not the result of TP-receptor activation.

Pharmacologic characterization of the contractile activity of peptide leukotrienes in guinea-pig pulmonary artery.

The actions of the peptide leukotrienes (LT) LTC4, LTD4 and LTE4 and phenylephrine (PE) were studied in isolated left branches of the guinea-pig pulmonary artery (GPPA). Indomethacin 5 x 10(-6) M enhanced both the potency and maximal response of all agonists, but the effect on LTD4 and LTE4 was larger. The influence of indomethacin suggests the release of an endogenous vasodilating cyclooxygenase product in GPPA. In the presence of indomethacin the rank-order of potency was LTC4 greater than LTD4 greater than LTE4 greater than or equal to PE with respective pD2 values of 7.65, 7.39, 6.35 and 6.26. All further studies were carried out in the presence of 5 x 10(-6) M indomethacin. Removal of the endothelium further increased both potency (greater than 3-fold) and the maximal response of all agonists tested, indicating that a non-cyclooxygenase endothelium-dependent relaxing factor may be present in GPPA. In separate studies, GPPA was demonstrated capable of metabolizing 3H-LTC4 to 3H-LTD4 by an L-serine borate inhibitable gamma-glutamyl transpeptidase. In contrast, relatively little formation of 3H-LTE4 was apparent either from 3H-LTC4 or 3H-LTD4. The LTD4-selective antagonists, LY 171,883 and ICI 198,615 had -log molar KB values of 6.07 +/- 0.14 and 9.38 +/- 0.32, respectively, against LTD4 in the absence of endothelium. The ability of LY 171,883 to antagonize LTC4 was eliminated in the presence of 45 mM serine borate in endothelium denuded tissues. LT receptors in GPPA appear to be heterogeneous and similar to guinea pig airway receptors.
