[Supplementary report on the study of resistance to coumafene in Rattus ratus observed at Marseilles].

A resistance to coumafene has previously been observed among Rattus rattus in a foodstuffs warehouse in the port of Marseille. New tests are here reported. They concern rats captured in two warehouses situated at a distance of 1,000 m. from one another, and separated by a distance approximately similar to that of the first warehouse explored. The experiments which were carried out, although limited in number, have clearly shown a resistance to coumafene administered in high doses in a rat of the first group and in three rats of the second group. There is every reason to fear a resistance extended to all the territories where Rattus rattus are found in the harbour and in the neighbouring regions.

Symptomatic brodifacoum ingestion requiring high-dose phytonadione therapy.

We report the deliberate ingestion of a superwarfarin product, brodifacoum, in a 39-y-old male. He presented with prothrombin and partial thromboplastin times of 150 and 113 sec, respectively. His coagulopathy was corrected by administration of blood products and phytonadione. To maintain normal clotting studies this patient required the highest maintenance dose of phytonadione, 200 mg/d, reported to date. The patient was able to tolerate this dose for 5 mo without adverse effects.

[Mutagenic effect of pindone on D. melanogaster].

A commercial grade Pindone was tested for the induction of genetic damage in male germ cells of Drosophila melanogaster by adult ingestion. Adults were starved for 4 days and then fed pindone at a concentration of 1 mM in ethyl alcohol 15% containing sucrose 5%. Pindone was found to increase the frequency of sex chromosome loss. Results were significant at the 1% level (Kastenbaum-Bowman test). However no significant results were obtained by the sex linked recessive lethal assay.

Coumafuryl (Fumarin) toxicity in chicks.

Coumafuryl (Fumarin) toxicity was diagnosed in chickens less than 1 week of age. Mortality rate was 100%. Necropsy showed crops and gizzards to be full of feed. There was diffuse hemorrhage and unclotted blood in the abdomen and thorax. Histological examination showed congestion and hemorrhage. Chemical analysis of crop and gizzard contents contained approximately 340 ppm coumafuryl. The source of the coumafuryl was found to be the wood-straw mats in the chick boxes.

[The toxicological characteristics of the interaction of cholinolytics with aminostigmine--a new reversible cholinesterase inhibitor].

This investigation has been carried out on albino mice. In the first series of experiments, we determined the dependence of LD50 of aminostigmine on different doses of 10 cholinolytics, and conversely, the dependence of LD50 of cholinolytics on aminostigmine administration. The initial slopes of the dose-effect curves were calculated. This data form the basis for evaluation of the character and degree of interactions. We established that benactyzine, spasmolytin, ftoracizin, arpenal, atropine, ganglerone, and methacin at low doses exhibit antagonism (with decreasing activity), whereas at high doses they exhibit synergism to the toxic effect of aminostigmine. In the interaction with aminostigmine, pirenzepine and amitriptyline (M1--cholinolytics) reveal a slightly pronounced antagonism, whereas aetyrophene, a selective central N-cholinolytic, displays mutual synergism. In the second series of experiments we showed that a combination of M1-, M2-cholinolytic atropine with M1- cholinolytics does not change the efficacy of the prophylaxis of aminostigmine and physostigmine poisoning, whereas a combination with N-cholinolytic increases it.
