[Distribution of zymodemes of Entamoeba histolytica from different areas of China].

Using agarose gel electrophoresis, the electrophoretic patterns of four enzymes: malate NADP oxidoreductase (malic enzyme) (ME), glucosephosphate isomerase (GPI), phosphoglucomutase (PGM) and hexokinase (HK), were compared and analysed for five strains of Entamoeba histolytica, which had been obtained from patients with acute amebic dysentery or asymptomatic cyst carrier in Beijing, Tianjin and Fujian Province. The results showed that the electrophoretic patterns of all the five strains of E. histolytica belong to pathogenic zymodeme, zymodeme XIV, which is suggested to be the main pathogenic zymodeme in our country.

[Isoenzymatic studies of Mediterranean populations of sandflies of the subgenus Larroussius].

Isoenzymes of hexokinase, phosphoglucomutase and glucosephosphate isomerase were examined, using agarose gel isoelectrofocusing, in populations of Phlebotomus ariasi (France), P. neglectus (Greece), P. perfilewi (Italy & Greece) and P. tobbi (Greece). Comparison between enzymatic profiles by numerical analysis separated Larroussius from Phlebotomus and Paraphlebotomus subgenera and brought together P. ariasi and P. neglectus. Nei genetic coefficients indicated similarities between populations of P. perfiliewi from Corfu and central Italy and between Corfuan and Cretan P. neglectus samples.

[The determination of the phenotypes of 3 enzyme systems by the polyacrylamide gel electrophoresis method].

Possibility of combined determination of phenotypes of three enzyme system (glyoxylase-1, phosphoglucomutase and 6-phosphogluconate dehydrogenase) in the apparatus with two vertical blocks of polyacrilamide gel (PAAG) was shown. The method is based on the principle of combination of differently composed blocks of PAAG and single universal electrode buffer. High productivity and economy of the method equally with good quality of resulting phoregrams make this method useful for medicolegal practical work.

[Presence of enzymes catalyzing UDP-glucose biosynthesis in a low density Golgi fractions of cat hepatocytes].

A Golgi-rich fraction is prepared from cat hepatocytes by the means of a four-step sucrose density gradient. The material applied to this gradient is composed either of smooth microsomes prepared from healthy animals, or of total microsomes prepared from cat treated by 50 per cent ethanol (0.6 g/100 g body weight, administered by stomach tube). A light fraction (d : 1.10) is obtained by the two procedures. It does not show any glucose-6-phosphatase activity, but is enriched in sialyltransferase, known as a marker enzyme for Golgi apparatus. It also contains the three enzymes implicated in the biosynthetic pathway for UDP-glucose (glucokinase, phosphoglucomutase and UTP : glucose-1-phosphate uridylyltransferase). UDP-glucose being the ultimate substrate in membranous glucosylation reactions, these results could support the hypothesis that sugar-nucleotides necessary for the glycoprotein biosynthesis are produced in the Golgi vesicles directly.

The examination of vaginally inserted plastic tampon applicators for genetic markers and evidence of prior sexual intercourse.

Vaginally inserted plastic tampon applicators were obtained from 42 female volunteers. The applicators were examined for the presence of ABH blood group substances, phosphoglucomutase (PGM), amylase, acid phosphatase, P30, and intact spermatozoa. Each applicator was accompanied by a control blood sample, a saliva specimen, a brief sexual and menstrual history, and method of birth control of the donor. Eight of the male sexual partners of the donors submitted blood and saliva samples. One male sexual partner submitted only a saliva sample. ABH blood group substances corresponding to the donor were recovered from 36 of the 42 applicators. The remaining 6 applicators revealed a combination of the donor's and sexual partner's ABH substances. The female's PGM type was recovered from 34 of the applicators. The remaining 8 applicators failed to show PGM activity. Of the applicators, 15 indicated evidence of prior sexual intercourse by the detection of ABH substances not consistent with the applicator donor (6 samples), high levels of acid phosphatase (11 samples), or recovery of spermatozoa (8 samples) or some combination of these. All applicator samples failed to show the presence of either P30 activity or PGM factors foreign to the female.

Population data of casework in West Virginia on six genetic marker systems.

Blood specimens and stains submitted from all geographic regions of West Virginia were analyzed for six genetic markers: International ABO, phosphoglucomutase (PGM), esterase D (ESD), erythrocyte acid phosphatase (EAP), adenylate kinase (AK), and adenosine deaminase (ADA). The four-year study indicates that markers identified were distributed in Hardy-Weinberg equilibrium and are consistent with population data previously reported.

Electrophoretic and isoelectric focusing studies in Brazilian Indians: data on four systems.

Three-hundred ninety-nine individuals living in seven populations of two Brazilian Indian tribes (Macushi and Içana River Indians) were tested for the phosphoglucomutase 1 (PGM1), properdin factor B (BF), haptoglobin (HP), and alpha-1-antitrypsin (PI) systems. We observed significant internal heterogeneity in the two tribes for the PGM1 alleles and in the Macushi for the HP markers. Frequencies in three of the four systems (the exception being BF) also show clear differences in the Macushi and Içana River Indians. Compared with other ethnic groups, South American Indians generally present high frequencies of PGM1*1B, BF*S, HP*1S, and PI*M3. On the other hand, PGM1*1A, PI*M1, and PI*M2 are reduced, and HP*1F is absent or rare. This is the first report about HP subtypes among American Indians.

Examination of the correlation of groupings in blood and semen.

The grouping of blood/saliva samples from a male so as to predict his semen groups is only justified if there is a strict correlation between the groupings in these body fluids. This correlation has been examined in the ABO, phosphoglucomutase (PGM1) and glyoxalase I (GLO) grouping systems in blood and semen samples collected from more than 250 individuals. Though no results proved inconsistent with this correlation, a number of semen gave inconclusive grouping results. Reasons for this are discussed as well as the relevance of the results to semen stain analysis. Semen amylase activities are also reported.

Erythrocyte enzyme allotypes in the X-linked recessive disorders, Duchenne muscular dystrophy and haemophilia-A hemizygotes and heterozygotes.

The erythrocyte enzyme-systems acid phosphatase, phosphoglucomutase, glutamate pyruvate transaminase, adenosine desaminase, adenylate kinase, glyoxase, glucose-6-phosphate dehydrogenase and esterase-D-isoenzyme phenotypes were studied for their percentile distribution and were compared with their incidence in the diseases with X-linked recessive heredity, Duchenne muscular dystrophy (DMD) and haemophilia-A, in hemizygous male children and heterozygous mothers. Considering the frequency distribution of the above mentioned isoenzyme phenotypes of the enzyme-systems in DMD, the phenotypes proved to be homogeneous, only the X transmitted 6-phosphogluconate dehydrogenase (6-PGD) isoenzyme types were found to be genetic markers in DMD hemizygotes and heterozygotes. In these genotypes the 6-PGD A phenotype showed a decrease while the phenotypes 6-PGD AB and B were significantly increased. The adenylate kinase (AK) 2-1 isoenzyme phenotype was increased to 25% against the population frequency of 6.34%, while the AK 1-1 phenotype occurred in 75% against its population frequency of 93.59%, showing a significantly decreasing tendency in haemophilia-A hemizygotes and heterozygotes.

5-Iodoribose 1-phosphate, an analog of ribose 1-phosphate. Enzymatic synthesis and kinetic studies with enzymes of purine, pyrimidine, and sugar phosphate metabolism.

The 5'-deoxy-5'-iodo-substituted analogs of adenosine and inosine are cytotoxic to tumor cells that have high activities of 5'-methylthioadenosine phosphorylase and purine nucleoside phosphorylase, respectively (Savarese, T.M., Chu, S-H., Chu, M.Y., and Parks, R. E., Jr. (1984) Biochem. Pharmacol. 34, 361-367). 5-Iodoribose 1-phosphate (5-IRib-1-P), the common intracellular metabolite of these 5'-iodonucleosides, has been synthesized enzymatically from 5'-deoxy-5'-iodoadenosine via adenosine deaminase from Aspergillus oryzae and human erythrocytic purine nucleoside phosphorylase. The purification and chemical properties of 5-IRib-1-P are described. The analog sugar phosphate inhibited purine nucleoside phosphorylase from human erythrocytes, phosphoglucomutase from rabbit muscle, and 5'-methylthioadenosine phosphorylase from Sarcoma 180 cells with Ki values of 26, 100, and 9 microM, respectively. Enzymes that react with 5-phosphoribosyl 1-pyrophosphate (P-Rib-PP), P-Rib-PP amidotransferase, hypoxanthine-guanine phosphoribosyltransferase, adenine phosphoribosyltransferase, and orotate phosphoribosyltransferase-orotidylate decarboxylase from extracts of Sarcoma 180 cells, were inhibited with Ki values of 49, 465, 307, and 275 microM, respectively. 5-IRib-1-P had no effect on P-Rib-PP synthetase. Since the Ki values of the analog sugar phosphate for 5'-methylthioadenosine phosphorylase and P-Rib-PP amidotransferase are much lower than the Km values of the natural substrates, Pi or P-Rib-PP which are reported to be present at nonsaturating concentrations under physiological conditions, these enzymes could be significantly inhibited by 5-IRib-1-P in intact cells.

Isoenzymes of Trypanosoma rangeli stocks and their relation to other trypanosomes transmitted by triatomine bugs.

Culture forms of three trypanosome species, Trypanosoma rangeli, T conorhini and T. cruzi, have been compared isoenzymatically by isoelectrofocusing. Of 9 enzymes examined, three: nonspecific esterase (NSE), phosphoglucomutase (PGM) and glucosephosphate isomerase (GPI), showed species specific enzyme profiles. Little isoenzymatic variation was found amongst the 14 T. rangeli stocks examined. Two of them were identified as T. cruzi of Group I demonstrating the necessity of a clear identification of trypanosome populations isolated from bugs in areas endemic for T. rangeli and T. cruzi. In extracts of artificially mixed cultures of T. rangeli and T. cruzi only the patterns of NSE showed clearly the specific bands of both species. The NSE should be tested for the detection of mixed infections under natural conditions.

The relevance of glucose 1,6-bisphosphate formation and degradation to human red blood cell metabolism.

In human erythrocytes, in the absence of specific enzymes, G1,6P2 synthesis and degradation are carried out by phosphoglucomutase PGM2 isoenzymes. The results presented, obtained by using partially purified preparations of these enzyme forms, suggest that erythrocyte G1,6P2 may play a crucial role in the physiological interconversion of several important sugar monophosphates.

The concentrations of free Mg2+ and free Zn2+ in equine blood plasma.

The enzyme phosphoglucomutase can be used as a metal ion indicator to measure the concentrations of free Mg2+ and free Zn2+ in physiological fluids. In horse plasma, the concentration of free Mg2+ is close to 0.5 mM, whereas that of free Zn2+ is about 2 X 10(-10) M, although numerous physiological roles for Zn2+ have been postulated that would require free Zn2+ concentration orders of magnitude higher than this. A titration of plasma with Zn2+ shows that the fractional increase in free Zn2+ is essentially the same as the fractional increase in total exchangeable Zn2+, and the results are consistent with a model in which essentially all of the Zn2+ in plasma is bound to albumin. Regardless of the model, the buffering capacity of plasma for free Zn2+ is intrinsically low; however, its capacity relative to the total (exchangeable) Zn2+ present is maximal. The implications of this type of buffering for homeostasis of plasma Zn2+ are considered. Treatment of plasma with a strong reducing agent such as dithiothreitol (0.1 mM) substantially increases the apparent binding of Zn2+ and thus reduces the free Zn2+ concentration. However, the concentration of free Zn2+ appears to be insensitive to decreases in the physiological concentrations of reduced glutathione and cysteine. The concentrations of free Zn2+ and free Mg2+ in plasma are similar to those that have been reported for muscle tissue (rabbit). Their ratio is about 4 X 10(-7). The physiological implications of these concentrations are considered. In some cases, if the Zn2+ and Mg2+ complexes of an uncharacterized vertebrate protein exhibit significantly different properties, their relative importance under physiological conditions can be approximated by evaluating those of the mixed complexes present in a solution that contains the physiological concentration of free Mg2+, plus Zn2+ buffered with histidine, at the appropriate pH and ionic strength. Other metal ion/chelon systems that come close to reproducing the concentrations of free Mg2+ and free Zn2+ in horse plasma also are considered.

Foetal macrosomia in diabetic pregnancy. Further data on the association with maternal PGM1 genotype.

The pattern of associations between maternal phosphoglucomutase locus 1 (PGM1) and foetal macrosomia in diabetic pregnancy has been compared with that observed in normal pregnancy. In diabetic pregnancy a substantial increase of macrosomic infants, as compared to normal pregnancy, is observed only among women homozygous for PGM1(1) allele. Also neonatal hypoglycemia is much more frequent in newborns from PGM1(-1) mothers than in newborns from other mothers. Since enzymatic activity of PGM1(-1) phenotype is lower as compared to other PGM1 phenotypes, this may influence glycaemic level and/or its stability in diabetic pregnant women with unfavourable effects on the metabolic and developmental patterns of the foetus.

Distributions of genetic markers in United States populations: II. Isoenzyme systems.

All published and unpublished population frequency data that could be located for U.S. populations is tabulated and presented for the isoenzyme systems phosphoglucomutase, esterase D, adenylate kinase, acid phosphatase, glyoxalase I, adenosine deaminase, 6-phosphogluconate dehydrogenase, glutamic-pyruvic transaminase, carbonic anhydrase II, and glucose-6-phosphate dehydrogenase. Results obtained by combining data for comparable racial/ethnic groups are also presented. The results obtained with combined data may give better information on frequencies for the U.S. population at large than is obtainable from studies conducted in restricted geographic areas.

First results of the application of isozyme analysis to study of Culicoides (Diptera: Ceratopogonidae).

Results of a biochemical study on the genus Culicoides are reported. A method of analysis using six discriminant enzymatic systems was used and the results obtained on zymograms are detailed. A genetic study of C. nubeculosus, based on phosphoglucomutase was carried out. The processing of the data obtained, by means of correspondence analysis, provides an interesting comparative study between morphologically related species.
