[Effect of maixinkang capsule on Ca2+ and mitochondrial membrane potential in liver cells of ApoE(-/-) mice].

OBJECTIVE: To observe the effect of Maixinkang Capsule (MXK) on Ca2t concentration and mitochondrial membrane potential in liver cells of ApoE(-/-) mice. METHODS: Liver cells from ApoE(-/-) mice were separated using collagenase digestive method. After the primary cells were cultured for 8 days in vitro, the concentration of 10% MXK contained rat's serum was added into the culture fluid. The Ca2+ concentration and mitochondrial membrane potential in liver cells after 48-hr culture were measured by confocal laser scanning microscopy with Flou-3 and Jc-1 as probes. RESULTS: MXK could decrease Ca2+ concentration in liver cells, which was significantly different to that in the control group (P < 0.01). Meanwhile, MXK could significantly improve mitochondrial membrane potential in liver cells (P < 0.01). There was no obvious dose-effect relationship shown in both effects of MXK. CONCLUSION: MXK can decrease Ca2+ concentration and improve the mitochondrial membrane potential in liver cells of ApoE(-/-) mice so as to regulate the lipids and prevent the occurrence and development of hyperlipemia and atherosclerosis.

[Removal of two calcium hydroxide preparations: S.E.M. study].

Calcium hydroxide is one of the most often medication used for temporary treatment in endodontics. Different forms of preparation are available. The aim of this study is to evaluate in vitro the elimination of the pharmaceutical and the commercial forms of presentation. The aspect of the dentinal walls were analysed using SEM and X-ray microanalysis. The results show that Ca(OH)2 removal is difficult but there is no significant difference of elimination between the two forms.

Zinc sulfide in intestinal cell granules of Ancylostoma caninum adults.

A source of confusion has existed since the turn of the century about the reddish brown, weakly birefringent "sphaerocrystals" located in the intestines of strongyle nematodes, Strongylus and Ancylostoma. X-ray diffraction and energy dispersive spectrometric analyses were used for accurate determination of the crystalline order and elemental composition of the granules in the canine hookworm Ancylostoma caninum. The composition of the intestinal pigmented granules was identified unequivocally as zinc sulfide. It seems most probable that the granules serve to detoxify high levels of metallic ions (specifically zinc) present due to the large intake of host blood.

[Pathogenesis of disorders of transparency of the optic media of the eye. III. Quantitative detection of copper in relation to the ultrastructure of the vitreous body in chalcosis].

The authors elaborated a new analytical micromethod for the detection of elements in morphologically investigated ultrastructures of the vitreous body. For the investigation of quantitative ultrastructural changes electron microanalysis (EMA) was used, combined with raster electron microscopy (REM). Using this method, the authors proved ultrastructural changes of the vitreous body by selective uptake of copper in the trabecular collagen fibres of the vitreous body in experimental chalcosis. From the results ensues that copper does not diffuse in the vitreous body in a homogeneous manner but aggregates in irregular agglomerations of precipitates and crystals. The different quantitative detection of copper in different parts of the same specimen is according to the authors due to the heterogeneous affinity of ultrastructural portions of the vitreous body for copper.

Histochemistry of colloidal iron stained crystal associated material in urinary stones and experimentally induced intrarenal deposits in rats.

Organic material associated with the calcium oxalate crystals in urinary stones and experimentally induced nephrolithiasis was stained with colloidal iron and analysed by energy dispersive x-ray microanalysis using standard techniques. Iron was positively identified in the stained specimens indicating that some of the organic material is an acidic mucosubstance. The results also indicate that some of the organic material of urinary stones may originate in the kidneys.

X-ray microanalysis of cultured chondrocytes.

Methodological aspects of X-ray microanalysis of cultured chondrocytes from rabbit auricular cartilage were investigated. The cells were grown on a Formvar-film covered carbon specimen holder with a hole, which allowed examination in STEM. Several different fluids for washing away the culture medium from the cells were used. The effect of these fluids on the elemental composition of the cells was studied. The composition of cultured chondrocytes was compared with that of auricular chondrocytes in situ. Washing with 0.3 M sucrose gave the most satisfactory results. Subsequently, the specimen holders could be frozen in liquid nitrogen and the cells freeze-dried. The effects of human growth hormone on rabbit chondrocytes were investigated by X-ray microanalysis and transmission electron microscopy. Although no ultrastructural effects of hormone treatment could be observed, marked changes in the elemental concentrations in nucleus and cytoplasm of the chondrocytes were detected.
