ToF-SIMS analysis of elemental distributions in human hair.

Elemental distributions on whole and longitudinal sections of hairs plucked from the scalp were studied with the surface sensitive technique time-of-flight secondary ion mass spectrometry (ToF-SIMS). Endogenous and environmental influences on the distributions of elemental species were identified. The cuticle scales appear to play the major role in the accumulation of exogenous products. The functionality of the outer surfaces and scale edges each preferentially bind different elemental species. The majority of elements considered accumulated longitudinally on the outer surface of the hair above the scalp level. Internally, most elemental signals (especially Al) decreased longitudinally once exposed to the environment with the exception of Si, which showed an increase. Images of elemental distributions within the medulla suggest that regions of different reactivity exist and show a variable ability to accumulate elemental species. The greatest signal intensities were observed in the cuticle and medulla regions rather than the cortex. The cuticle is continually exposed to environmental contamination and the medulla may, or may not, exist in a hair. Therefore, the components of a hair that potentially contribute the most to the elemental concentrations (i.e. the cuticle and medulla) are also the most variable, and as such greatly complicate the interpretation of elemental concentrations in hair. Results also suggest that bleaching hair can enhance the accumulation of contaminants.

Effect of pH on element release from dental casting alloys.

STATEMENT OF PROBLEM: Dental casting alloys are subjected to transient acidity in the oral environment, yet most studies have not investigated the effects of these transient environments on elemental release from alloys. Elemental release is important because it plays a significant role in alloy biocompatibility. PURPOSE: It was hypothesized that acidic environments would increase elemental release from dental alloys during exposure and after the acidic environment was removed. This hypothesis was based on the known increase in release of nickel from nickel-based alloys in an acidic environment. MATERIAL AND METHODS: High-noble, noble, and base metal casting alloys were exposed for 30 minutes to solutions with pH ranging from 1 to 7. Elemental release of representative elements was measured by means of atomic absorption spectrometry during the exposure and in the week after the exposure. This release was compared with elemental release in the week before the exposure. RESULTS: High-noble and noble alloys were resistant to acidic environments. A pH of 4 did not increase elemental release during or after exposure. A pH 1 environment slightly elevated release of Ag, Cu, and Pd in some alloys. However, a Ni-based alloy released large amounts of Ni during the acidic exposure of pH 1 or 4, and more importantly, in the week after the exposure as well. Increased time of exposure to acid did not alter elemental release from noble or high-noble alloys, but markedly increased release from the Ni-based alloy. CONCLUSIONS: Transient exposure of casting alloys to an acidic oral environment is likely to significantly increase elemental release from Ni-based alloys, but not from high-noble or noble alloys.

Long range interactions between oriented texture elements.

Long range interactions between texture elements (short, oriented line segments) were examined. Specifically, we studied the influence of a background array of texture elements on the detectability of a target element (separated from the background by an intermediate textured region) using textures like those of Caputo (Vis. Res. 1996, 36, 2815-2826). We found that, in general, when the background elements were oriented orthogonally to the target element, detection of the target element was better than when the background elements had the same orientation as the target element. We discuss these interactions in terms of inhibitory and excitatory connections between orientation and spatial frequency selective linear filters (e.g. filters which mimic V1 simple cells) which would respond to the individual texture elements.

Comparison of the element composition in several plant species and their substrate from a 1500000-km2 area in Northern Europe.

Leaves of 9 different plant species (terrestrial moss represented by: Hylocomium splendens and Pleurozium schreberi; and 7 species of vascular plants: blueberry, Vaccinium myrtillus; cowberry, Vaccinium titis-idaea; crowberry, Empetrum nigrum; birch, Betula pubescens; willow, Salix spp.; pine, Pinus sylvestris and spruce, Picea abies) have been collected from up to 9 catchments (size 14-50 km2) spread over a 1500000 km2 area in Northern Europe. Soil samples were taken of the O-horizon and of the C-horizon at each plant sample site. All samples were analysed for 38 elements (Ag, Al, As, B, Ba, Be, Bi, Ca, Cd, Co, Cr, Cu, Fe, Hg, K, Li, Mg, Mn, Mo, Na, Ni, P, Pb, Rb, S, Sb, Sc, Se, Si, Sn, Sr, Th, Tl, U, V, Y, Zn and Zr) by ICP-MS, ICP-AES or CV-AAS (for Hg-analysis) techniques. The concentrations of some elements vary significantly between different plants (e.g. Cd, V, Co, Pb, Ba and Y). Other elements show surprisingly similar levels in all plants (e.g. Rb, S, Cu, K, Ca, P and Mg). Each group of plants (moss, shrubs, deciduous and conifers) shows a common behaviour for some elements. Each plant accumulates or excludes some selected elements. Compared to the C-horizon, a number of elements (S, K, B, Ca, P and Mn) are clearly enriched in plants. Elements showing very low plant/C-horizon ratios (e.g. Zr, Th, U, Y, Fe, Li and Al) can be used as an indicator of minerogenic dust. The plant/O-horizon and O-horizon/C-horizon ratios show that some elements are accumulated in the O-horizon (e.g. Pb, Bi, As, Ag, Sb). Airborne organic material attached to the leaves can thus, result in high values of these elements without any pollution source.

Elemental composition and sources of fine and ultrafine ambient particles in Erfurt, Germany.

We present the first results of a source apportionment for the urban aerosol in Erfurt, Germany, for the period 1995-1998. The analysis is based on data of particle number concentrations (0.01-2.5 microm; mean 1.8 x 10(4) cm(-3), continuous), the concentration of the ambient gases SO(2), NO, NO(2) and CO (continuous), particle mass less than 2.5 microm (PM(2.5)) and less than 10 microm (PM(10)) (Harvard Impactor sampling, mean PM(2.5) 26.3 micro/m(3), mean PM(10) 38.2 microg/m(3)) and the size fractionated concentrations of 19 elements (impactor sampling 0.05-1.62 microm, PIXE analysis). We determined: (a) the correlations between (i) the 1- and 24-h average concentrations of the gaseous pollutants and the particle number as well as the particle mass concentration and (ii) between the 24-h elemental concentrations; (b) Crustal Enrichment Factors for the PIXE elements using Si as reference element; and (c) the diurnal pattern of the measured pollutants on weekdays and on weekends. The highly correlated PIXE elements Si, Al, Ti and Ca having low enrichment factors were identified as soil elements. The strong correlation of particle number concentrations with NO, which is considered to be typically emitted by traffic, and the striking similarity of their diurnal variation suggest that a sizable fraction of the particle number concentration is associated with emission from vehicles. Besides NO and particle number concentrations other pollutants such as NO(2), CO as well as the elements Zn and Cu were strongly correlated and appear to reflect motor vehicle traffic. Sulfur could be a tracer for coal combustion, however, it was not correlated with any of the quoted elements. Highly correlated elements V and Ni have similar enrichment factors and are considered as tracers for oil combustion.

Two different gene elements are required for glucose regulation of S14 transcription.

Carbohydrate feeding increases the transcriptional activity of the hepatic S14 gene. The region of the S14 promoter between -1384/-1275 contributes to the transcriptional regulation by carbohydrate. A previously identified element (-1303/-1289) within this region is required but is not sufficient for the carbohydrate effect. Therefore, we ligated -1384/-1275 to a heterologous promoter and created mutants in this region to identify other potential responsive sequences. We found that mutation within -1365/-1350 eliminated the response to high glucose (27.5 mM). However, three copies of this element ligated to a mouse mammary tumor virus-luciferase vector did not respond to glucose indicating the -1365/-1350 element is insufficient to confer a glucose response in isolation. Nevertheless. mutating the -1365/-1350 element in the native promoter led to a loss of response to glucose, proving this element is necessary. Electrophoretic mobility shift assays (EMSA) using three copies of the element showed significant binding to rat hepatic nuclear extracts, but no difference between the dietary states. Competition EMSA studies showed that the previously identified element at -1303/-1289 was unable to compete for proteins that bind to the -1365/-1350 element. Therefore, we have demonstrated two separate elements within the -1384/-1275 region of the S14 gene that bind different proteins and interact to elicit the carbohydrate effect.

An antisilencer element is involved in the transcriptional regulation of the human vimentin gene.

Vimentin is an intermediate filament protein normally expressed in cells of mesenchymal origin. The promoter of the human vimentin gene was previously reported to contain two positive-acting regions, separated by a negative region (Rittling, S.R., Baserga, R., 1987. Functional analysis and growth factor regulation of the human vimentin promoter. Mol. Cell. Biol. 7, 3908-3915). Here, detailed studies reveal two additional regulatory elements, a new positive transcriptional element located between -717 and -757, and a new repressor element at -780 to -821. In transient transfections, the positive-acting element is able to completely override the effect of different silencer elements when fused to a heterologous promoter. However, this element does not enhance gene activity when the silencer element is absent and thus cannot be viewed as a true enhancer. Since it appears to overcome the effect of a silencer element, we refer to it as an antisilencer element. Gel mobility shift assays, UV-cross-linking experiments, and Southwestern blots reveal that a 105-kDa protein specifically binds to this region.

A study on carbothermal reduction of sulfur dioxide to elemental sulfur using oilsands fluid coke.

Experiments and reaction equilibrium calculations were carried out for the SO2 gas and oilsands fluid coke system. The goal was to develop a coke-based sulfur-producing flue gas desulfurization (SP-FGD) process that removes SO2 from flue gases and converts it into elemental sulfur. The conversion of SO2 to elemental sulfur proceeded efficiently at temperatures higher than 600 degrees C, and the sulfur yield reached a maximum (> 95%) at about 700 degrees C. An increase of temperature beyond 700 degrees C enhanced the reduction of product elemental sulfur, resulting in the formation of reduced sulfur species (COS and CS2), which lowered the sulfur yield at 900 degrees C to 90%. Although equilibrium calculations suggest that a lower temperature favors the conversion of SO2 as well as the yield of elemental sulfur, experiments showed no formation of elemental sulfur at 600 degrees C and below, likely due to hindered kinetics. Faster reduction of SO2 was observed at a higher temperature in the range of 700-1000 degrees C. A complete conversion of SO2 was achieved in about 8 s at 700 degrees C. Prolonging the product gas--coke contact, the yield of elemental sulfur decreased due to the formation of COS and CS2 while the SO2 conversion remained complete. Equilibrium calculations suggest that the ultimate yield of elemental sulfur maximizes at the C/SO2 ratio of 1, which represents the stoichiometry of SO2 + C-->CO2 + S. For the C/SO2 ratio < 1, equilibrium calculations predict elemental sulfur and CO2 being major products, suggesting that SO2 + C-->CO2 + S is the predominant reaction if SO2 is in excess. Experiments revealed that elemental sulfur and CO2 were only major products if the conversion of SO2 was incomplete, which is in agreement with the result of the equilibrium modeling.

Norbornyl cations of group 14 elements.

Norbornyl cations of the group 14 elements Si --> Pb have been synthesized from substituted 3-cyclopentenemethyl precursors by intramolecular addition of transient cations to the C=C double bond of the 3-cyclopentenemethyl substituent (pi-route to norbornyl cations). The norbornyl cations 4a (E = Si, R = Me), 4e (E = Si, R = Et), 4f (E = Si, R = Bu), 4g (E = Ge, R = Bu), 4h (E = Sn, R = Bu), and 4i (E = Pb, R = Et) have been identified by their characteristic NMR chemical shifts (4a,e,f, delta((29)Si) = 80-87, delta((13)C)(CH=) = 149.6-150.6; 4g, delta((13)C)(CH=) = 144.8; 4h, delta((119)Sn) = 334, delta((13)C)(CH=) = 141.5; 4i, delta((207)Pb) = 1049, delta((13)C)(CH=) = 138). The significant deshielding of the vinylic carbon atoms (Deltadelta((13)C)) relative to those of the precursor (Deltadelta((13)C) = 19.3-20.3 (4a,e,f), Deltadelta((13)C) = 14.6 (4g), Deltadelta((13)C) = 11.1 (4h), Deltadelta((13)C) approximately 8 (4i)) and the small J coupling constants between the element and the remote vinyl carbons in the case of 4h and 4i (J(CSn) = 26 Hz, J(CPb) = 16 Hz) give experimental evidence for the intramolecular interaction and the charge transfer between the positively charged element and the remote C=C double bond. The experimental results are supported by quantum mechanical calculations of structures, energies, and magnetic properties for the norbornyl cations 4a,b (E = Ge, R = Me), 4c (E = Sn, R = Me), 4d (E = Pb, R = Me), and 4e,f at the GIAO/B3LYP/6-311G(3d,p)//MP2/6-311G(d,p) (Si, Ge, C, H), SDD (Sn, Pb) level of theory. The calculated (29)Si NMR chemical shifts for the silanorbornyl cations 4a,e,f (delta((29)Si) = 77-93) agree well with experiment, and the calculated structures of the cations 4a-f reveal their bridged norbornyl cation nature and suggest also for the experimentally observed species 4a,e-i a formally 3 + 1 coordination for the element atom with the extra coordination provided by the C=C double bond. This places five carbon atoms in the close vicinity of the positively charged element atom. The group 14 element norbornyl cations 4a,e-i exhibit only negligible interactions with the aromatic solvent, and they are, depending on the nature of the element group, stable at room temperature in aromatic solvents for periods ranging from a few hours to days. In acetonitrile solution, the intramolecular interaction in the norbornyl cations 4a,e-h breaks down and nitrilium ions with the element in a tetrahedral environment are formed. In contrast, reaction of acetonitrile with the plumbyl cation 4i forms an acetonitrile complex, 10i, in which the norbornyl cation structure is preserved. The X-ray structure of 10i reveals a trigonal bipyramidal environment for the lead atom with the C=C double bond of the cyclopentenemethyl ligand and the nitrogen atom of the acetonitrile molecule in apical positions. Density functional calculations at the B3LYP/6-311G(2d,p)//(B3LYP/6-31G(d) (C, H), SDD (Si, Ge, Sn, Pb)) + DeltaZPVE level indicate that the thermodynamic stability of the group 14 norbornyl cations increases from Si to Pb. This results in a relative stabilization for the plumbanorbornyl cation 4d compared to tert-butyl cation of 52.7 kcal mol(-)(1). In contrast, the intramolecular stabilization energy E(A) of the norbornyl cations 4a-d decreases, suggesting reduced interaction between the C=C double bond and the electron-deficient element center in the plumbacation compared to the silacations. This points to a reduced electrophilicity of the plumbacation compared to its predecessors.

Changes of the metal composition in German white wines through the winemaking process. A study of 63 elements by inductively coupled plasma-mass spectrometry.

Elemental patterns are often used for the classification or identification of the origin of wines. A prerequisite is that the concentration of the elements is not strongly influenced by the addition of different substances such as yeast and fining products during the winemaking process. Inductively coupled plasma-mass spectrometry (ICP-MS) has been used in this study to determine in total 63 elements (including some nonmetals and the rare earth elements) in five German white wines from five regions of origin. The whole winemaking process was studied, from the must to the ready wine. Microwave acid digestion was used for sample preparation, and indium was added as internal standard for a semiquantitative analysis. Two winemaking processes were compared: with the addition of clarifying agents (bentonites) before and after the fermentation. The concentration of only a few elements such as Li, B, Mg, Ca, Rb, Cs, and Pb seems to be constant throughout the whole winemaking process (changes of <+/-50%) and are independent of the time of addition of the bentonites. When bentonites are added before fermentation, the concentration of other elements, such as V, Co, and Fe, remains constant. If bentonites are added after fermentation, the concentration of some other elements such as Sr, Zn, and Mn is nearly unaffected. These elements are therefore robust elements for origin studies in German white wines.

The organization of serotonin-, dopamine-, and FMRFamide-containing neuronal elements and their possible role in the regulation of spontaneous contraction of the gastrointestinal tract in the snail Helix pomatia.

The distribution of serotonin-, tyrosine hydroxylase-, and FMRFamide-immunoreactive neuronal elements, as well as the concentrations of serotonin and dopamine in the different parts of the gastrointestinal tract, were studied in the snail Helix pomatia. The sensitivity of the spontaneous contractions of the alimentary tract to serotonin, dopamine, and FMRFamide was also tested. Serotonin-, tyrosine hydroxylase-, and FMRFamide-immunoreactive elements could be demonstrated in each part of the gastrointestinal tract, but they showed different innervation patterns. Serotonin- and tyrosine hydroxylase-immunoreactive elements were dominant in the submucosal layer, whereas FMRFamide-immunoreactive elements were dominant in both the mucosal and submucosal layers. Tyrosine hydroxylase-immunoreactive elements were confined to the longitudinal muscle trabeculae of submucosa, whereas serotonin-immunoreactive elements were distributed throughout the submucosal layer. No serotonin-immunoreactive cell bodies, but only fibers, could be detected in the gastrointestinal tract, and therefore they represent extrinsic elements. Tyrosine hydroxylase- and FMRFamide-immunoreactive cell bodies represent intrinsic elements of the tract. The occurrence and density of the serotonin- and tyrosine hydroxylase-immunoreactive elements showed significant differences in the different parts of the alimentary tract, in accordance with HPLC assays, which revealed a significant frontocaudal decrease in both the serotonin (from 2.11 to 1.21 pM/mg) and dopamine (from 3.28 to 0.52 pM/mg) contents of the different parts of the alimentary tract. Dopamine at 10(-5) M concentration proved to be effective only on the longitudinal muscles by increasing the tone and frequency of contractions, but was ineffective on the circular muscles. Serotonin affected both the longitudinal and circular muscles. Serotonin at 10(-5) M concentration decreased the tone and increased the frequency of low-amplitude contractions of the longitudinal muscles of the esophagus and the gizzard but increased both the tone and frequency of the crop. Serotonin at 10(-9) M concentration slightly decreased the tone and blocked the contractions of the circular muscles in the crop but at 10(-5) M concentration induced contractions of the circular muscles in the gizzard. FMRFamide at 10(-6) M concentration decreased the tone and was shown to block the contractions of both the longitudinal and circular muscles.

Historical variation of elements with respect to different geochemical fractions in recent sediments from Pigeon Lake, Alberta, Canada.

Geochemical analysis of elements and organic matter were conducted on vertical profiles of the recent sediments from Pigeon Lake, Alberta, Canada, to determine historical variations in elemental content of the sediments as related to their geochemical fractions. The elements are grouped according to their affinity with different geochemical fractions, by using cluster analysis and sequential extraction experiments. As a result, four elemental fractions were identified: clastic mineral detritus; carbonate; organic; and elements that show less similarity to the previous groups perhaps due to anthropogenic input or the influence of other fractions, such as oxyhydroxides. Following the identification of geochemical fractions in the sediments, a three-step normalizing method was applied using parameters that represent each geochemical fraction. These normalizing techniques appear to be important in verifying whether the variation of elements is indeed the result of anthropogenic and/or natural activities. The normalized data are correlated with the recent history of human activity and natural events near Pigeon Lake. Given the age of the lake sediments, this correlation indicates that the depth profiles of elements after being normalized to the organic and carbonate fractions reflect the variation of detrital input into the lake. However, the former mainly corresponds to the coarse-grained clastic minerals originating from high-energy erosion as the result of natural events (e.g., flooding), whereas the latter corresponds to the low-energy erosion of the fine particles (enriched in lithophile elements) due to deforestation in the drainage basin. Normalizing to the clastic mineral detritus fraction results in the increase of heavy metals in the uppermost part of the sediment profiles, which coincides with industrial activities during the past two decades in central Alberta. However, the concentration of these elements is negligible, as compared to the quantities released by geogenic processes (erosion).

Identification and characterization of DNA elements implicated in the regulation of CYP4A1 transcription.

We have identified a peroxisome proliferator response element (PPRE) approx. 4300 nucleotide upstream of the rat cytochrome P-450 CYP4A1 gene. Two members of the steroid-hormone-receptor superfamily, the peroxisome proliferator-activated receptor-alpha (PPAR alpha) and the retinoid X receptor-alpha (RXR alpha), bind specifically to this element as a heterodimer, and this element confers responsiveness to the peroxisome proliferator Wyeth-14,643 when tested in co-transfection assays. A second element, located 35 nucleotides further upstream, fails to bind PPAR alpha/RXR alpha heterodimers and is unresponsive to Wy-14,643 in co-transfection assays. Both elements are, however, responsive to 9-cis-retinoic acid in the presence of RXR alpha, when tested in the co-transfection assay. As RXR alpha fails to bind to either element as a homodimer, we suggest that RXR alpha interacts with PPAR alpha to regulate transcription via the proximal element, and interacts with some other cellular factor to regulate transcription via the more distal element. This is consistent with previous reports that a number of peroxisome proliferator-regulated genes contain PPRE-like elements as part of their regulatory sequences, which may be recognized by several receptor combinations. This provides further evidence that PPARs and their co-factors are important in mediating the pleiotropic action of peroxisome proliferators.

Cell-type specificity of the Anabaena fdxN-element rearrangement requires xisH and xisI.

The fdxN element, along with two other DNA elements, is excised from the chromosome during heterocyst differentiation in Anabaena sp. strain PCC 7120. Previous work showed that rearrangement of the fdxN element requires the xisF gene, which encodes a site-specific recombinase, and suggested that at least one other heterocyst-specific factor is involved. Here we report that the xisH and xisI genes are necessary for the heterocyst-specific excision of the fdxN element. Deletion of a 3.2 kb region downstream of the xisF gene blocked the fdxN-element rearrangement in heterocysts. The 3.2 kb deletion was complemented by the two overlapping genes xisH and xisI. Interestingly, extra copies of xisHI on a replicating plasmid resulted in the xisF-dependent excision of the fdxN element in vegetative cells. Therefore, xisHI are involved in the control of cell-type specificity of the fdxN rearrangement. The xisHI genes had no effect on the two other DNA rearrangements. The xisHI-induced excision of the fdxN element produced strains lacking the element and demonstrates that the 55 kb element contains no essential genes. xisH and xisI do not show similarity to any known genes.

Monitoring of RNA polymerase-DNA UP element interaction by a fluorescent probe conjugated to alpha subunit.

The carboxy-terminal domain (CTD) of Escherichia coli RNA polymerase alpha subunit was specifically modified by a reporter label, fluorescein mercuric acetate (FMMA), conjugated to Cys269 on the surface of UP element recognition helix. The modified enzyme was used to investigate RNA polymerase interaction with different promoters, either with or without an UP element. In a single-round transcription assay, the activity of modified RNA polymerase was found to decrease as measured with rrnBP1, trpP and lacP2 promoters but not with many other promoters including mutant rrnBP1 without the UP element, supporting the idea that Cys269 or the domain including Cys269 is involved in UP element recognition. Both trpP and lacP2 have sequence similarity to the rrnBP1 UP element. The chemical modification of RNA polymerase, however, did not affect an apparent equilibrium dissociation constant with rrnBP1, as measured by gel-retardation assays, indicating that the DNA-binding ability is retained even after FMMA conjugation. Interaction with the rrnBP1 UP element led to substantial alterations in the spectral parameters of the reporter label, which are different from those induced by complex formation with promoters without UP elements. A pronounced spectral blue shift suggests that the labeled surface of alphaCTD closely approaches the charged UP DNA helix. These observations imply that the fluorescent labeling at Cys269 can be used as a good tool for monitoring the presence or absence of an UP element in a given promoter. Spectral parameters of the label displayed the spectral blue shift when the modified RNA polymerase interacted with trpP, supporting the prediction that this promoter carries an rrnBP1-type UP element.

Functional erythroid promoters created by interaction of the transcription factor GATA-1 with CACCC and AP-1/NFE-2 elements.

We have investigated interactions between the erythroid transcription factor GATA-1 and factors binding two cis-acting elements commonly linked to GATA sites in erythroid control elements. GATA-1 is present at all stages of erythroid differentiation, is necessary for erythropoiesis, and binds sites in all erythroid control elements. However, minimal promoters containing GATA-1 sites are inactive when tested in erythroid cells. Based on this observation, two erythroid cis elements, here termed CACCC and AP-1/NFE-2, were linked to GATA sites in minimal promoters. None of the elements linked only to a TATA box created an active promoter, but GATA sites linked to either CACCC or AP-1/NFE-2 elements formed strong erythroid promoters. A mutation of T to C at position -175 in the gamma-globin promoter GATA site, associated with hereditary persistence of fetal hemoglobin (HPFH), increased expression of these promoters in both fetal and adult cells. A construct bearing the beta-globin CACCC element was more active in adult and less active in fetal erythroid cells, when compared with the gamma-globin CACCC element. These studies suggest that erythroid control elements are formed by the interactions of at least three transcription factors, none of which functions alone.

Glucocorticoid and progestin receptors are differently involved in the cooperation with a structural element of the mouse mammary tumor virus promoter.

We have previously characterized a regulatory element located between -294 and -200 within the mouse mammary tumor virus (MMTV) long terminal repeat (LTR). This element termed AA element cooperates with the glucocorticoid response elements (GREs) for glucocorticoid activation. Here we show that in a MMTV LTR wild type context, the deletion of this element significantly reduces both glucocorticoid and progestin activation of the promoter. Deletion of the two most distal GREs forces the glucocorticoid receptor (GR) and the progestin receptor (PR) to bind the same response elements and results in a dramatic decrease in the inducibility of the MMTV promoter by the two hormones. The simultaneous deletion of the two distal GREs and of the AA element abolishes completely the glucocorticoid-induced activation of the promoter. In contrast it restores a significant level of progestin-induced activation. This different effect of the double deletion on glucocorticoid- and progestin-induced MMTV promoter activation is not cell specific because it is also observed, and is even stronger, when either GR or PR is expressed in the same cell line (NIH 3T3). This is the first description of a mutated MMTV promoter that, although retaining GREs, is activated by progestins and not by glucocorticoids. This suggests a different functional cooperation between protein(s) interacting with the AA element and GR or PR. Cotransfections with constructs containing wild-type or mutated MMTV LTR with either PR lacking its C-terminal domain or GR/PR chimeras in which the N-terminal domains have been exchanged demonstrate that the N-terminal domains of the receptors specify the different behavior of GR and PR regarding the AA element.

Cell cycle-dependent transcription of CLN1 involves swi4 binding to MCB-like elements.

Two promoter elements have been defined that activate G1/S-specific transcription in Saccharomyces cerevisiae. SCB elements (CACGAAA) are activated by the Swi4-Swi6 complex, and MCB elements (ACGCGTNA) are activated by the Mbp1-Swi6 complex. CLN1 encodes a cyclin which is expressed during this interval, and requires Swi4 and Swi6 for peak transcription, but it has no consensus SCB elements in its promoter. Two SCB-like sequences had been previously noted and suggested to be the functional promoter elements. Our studies indicate that these sequences are unable to activate transcription of a lacZ reporter construct, or to bind Swi4-Swi6 complexes in vitro. However, a cluster of three sequences resembling MCB sequences are active promoter elements, sufficient to confer G1/S-specific transcription to a reporter. These sites are the predominant activation elements in the CLN1 promoter, and despite their resemblance to MCB elements, they bind Swi4-Swi6 complexes in vitro and require Swi4 and Swi6 for their activity in vivo. This indicates that the sequences that promote Swi4/Swi6 binding have not been fully defined, or that there are multiple Swi4- and Swi6-containing complexes with distinct DNA binding specificities. In addition to these novel Swi4/Swi6-binding sites, these studies also show that there must be at least one novel promoter element that can confer G1/S-specific transcription to CLN1, because when all the potential SCB- and MCB-like sequences are eliminated the transcript is still cell cycle regulated.
