Promoting glutathione synthesis after spinal cord trauma decreases secondary damage and promotes retention of function.

The study aimed to 1) quantify oxidative stress in spinal cord after crush injury at T6, 2) determine whether the administration of the procysteine compound L-2-oxothiazolidine-4-carboxylate (OTC) would up-regulate glutathione (GSH) synthesis and decrease oxidative stress, and 3) determine whether decreased oxidative stress results in better tissue and function retention. We demonstrate that spinal cord compression (5 s with a 50 g aneurysm clip) at T6 in rats results in oxidative stress that is extensive (significant increases in oxidative stress seen at C3 and L4) and rapid in onset. Indices of oxidative stress used were GSH content, protein carbonyl content, and inactivation of glutathione reductase. Administration of OTC resulted in a marked decrease in oxidative stress associated with a sparing of white matter at T6 (16+/-1.9% retained in OTC-treated animals vs. less than 1% in saline-treated). Behavioral indices in control, saline-treated, and OTC-treated animals after 6 wk were respectively: angle board scores (59 degrees, 32 degrees, and 42 degrees ), modified Tarlov score (7, 2.4, and 4.1), and Basso-Beattie-Bresnahan score (21, 5.3, and 12.9). We conclude that administration of OTC after spinal cord trauma greatly decreases oxidative stress and allows tissue preservation, thereby enabling otherwise paraplegic animals to locomote.

1-(5-oxohexyl)-3,7-Dimethylxanthine, a phosphodiesterase inhibitor, activates MAPK cascades and promotes osteoblast differentiation by a mechanism independent of PKA activation (pentoxifylline promotes osteoblast differentiation).

We have investigated the effect of 1-(5-oxohexyl)-3,7-dimethylxanthine or pentoxifylline (PeTx), a nonselective phosphodiesterase inhibitor, on osteoblastic differentiation in vitro by using two mesenchymal cell lines, C3H10T1/2 and C2C12, which are able to acquire the osteoblastic phenotype in the presence of bone morphogenetic protein-2 (BMP-2). PeTx induced the osteoblastic markers, osteocalcin and Osf2/Cbfa1, in C3H10T1/2 and C2C12 cells and enhanced BMP-2-induced expression of osteocalcin, Osf2/Cbfa1, and alkaline phosphatase. This activity was partially attributed to the fact that PeTx is able to enhance BMP-2-induced Smad1 transcriptional activity. Although PeTx clearly stimulates PKA in these cells, neither pretreatment of cells with the PKA inhibitor H89 nor transfection with the specific PKA inhibitor PKI prevented the induction or enhancement of osteoblast markers by PeTx, demonstrating that these effects were independent of PKA activation. On the other hand, PeTx induced the activation of ERK1/2 and p38 kinase pathways independently of the activation of PKA. Selective inhibitors of these MAPK cascades prevented the induction of osteoblastic markers in cells treated with PeTx, suggesting that the activation of these two pathways plays a role in the effect of PeTx on osteoblastic differentiation.

Beneficial effects and side effects of DHEA: true anti-aging and age-promoting effects, as well as anti-cancer and cancer-promoting effects of DHEA evaluated from the effects on the normal and cancer cell telomeres and other parameters.

The author evaluated the effects of DHEA (Dehydroepiandrosterone) on the amount of telomeres of normal cells and cancer cells and found the following: Contrary to the literature, which often recommended 25-50 mg of DHEA daily for the average adult human being, the author found that, depending on the individual, the maximum increase of normal cell telomere was obtained by a single optimal dose of 1.25-12.5 mg. This was examined in 50 people, both males and females, between the ages of 20-80 years old. When one optimal dose was given to each individual, the average telomere amount in normal tissues, measured in Bi-Digital O-Ring Test units, often increased from anywhere between 25-300 ng to between 500-530 ng. Cancer cell telomere reduced from higher than 1100 ng to less than 1 yg (=10(-24) g) with equally significant normalization of abnormal cancer parameters (such as Integrin alpha5beta1, Oncogen C-fosAb2, Acetylcholine, etc.). Circulatory improvement and an increase in grasping force of up to 25% were also detected, along with the changing of a few white hairs to black hairs. The beneficial effects of one optimal dose of DHEA generally lasted between 1 to 4 months, though in some individuals it lasted for a much shorter period of time due to a number of negative factors such as excessive stress/work, excessive exposure to low temperatures and toxic substances, or use of common pain medicines. On the other hand, if a patient took an excessive dose of DHEA, the amount of normal cell telomere decreased, while there was an increase in cancer cell telomere. It was found that those who took an overdose of 25-50 mg daily for more than 3 months had a high incidence of cancer of the prostate gland, breast, colon, lung, and stomach. Also, when the average normal cell telomere levels were less than 110 ng, compared with a normal value of 120-130 ng, and when DHEA in different parts of the body was also extremely low (less than 1-2 ng), one could suspect the possible presence of a malignant tumor somewhere in the body. When normal cell telomere was less than 110 ng, most individuals felt very weary with marked tiredness in the eyes, and grasping force was often reduced.

Problem-oriented training promotes spontaneous analogical transfer: memory-oriented training promotes memory for training.

Spontaneous analogical transfer is the use of information from one problem to solve another problem, without an explicit hint to use the previous information. The results of five experiments were that if subjects tried to solve a training problem before hearing its solution, or tried to explain a training story's solution before hearing the correct explanation, spontaneous transfer was more likely than it was if subjects had studied the same training passage for memory before hearing its solution or explanation. The advantage of problem-oriented processing over memory-oriented processing occurred even though solution attempts nearly always failed, and the advantage was not reduced if the target problem was tested 15 min later rather than immediately after training. We propose that problem-oriented processes performed at study are appropriate processes to use at test. Further support for the account comes from subjects' memory for the training passages; the advantage for problem-oriented processing on solution tasks was mirrored by an equally substantial advantage for the memory-oriented subjects on a recall task.

[The "rehabilitation sciences" promotional emphasis--a promotional program and its implementation].

The German Federal Research Ministry and the German pension insurance have set up a special funding programme for medical rehabilitation research. Eight regional research networks with about 70 projects receive funds for a first three year period. Rehabilitation experts from several countries selected the promising networks and studies from more than 100 proposals. In these networks medical and social scientists from universities and other research units cooperate with rehabilitation centers and pension insurance institutes in order to combine forces for the development of German rehabilitation science. The programme aims at improving the research structures and at tackling the major challenges of medical rehabilitation research. The following subjects represent the main fields of the ongoing research: a) etiology, course and prognosis of chronic diseases and their consequences to rehabilitation, b) diagnostics in rehabilitation, c) evaluation and improvement of rehabilitation therapy, d) the patient's perspective in rehabilitation and e) rehabilitation system research. As yet funding is limited to the year 2001. Further support of the networks will depend on the results of a thorough evaluation of the progress of their research and their structural perspective.
