Relapse of leukemia remains a common event after allogeneic bone marrow transplantation, despite potential donor antihost alloreactivity present in most transplants. This work examined posttransplant relapse of the DBA/2 P815 mastocytoma in a murine model of MHC-matched, minor histocompatibility antigen (mHAg)-mismatched bone marrow transplantation (BALB/c donors into DBA/2 recipients). Antihost alloreactivity was associated with reduction of posttransplant tumor burden and prolongation of survival, but posttransplant relapse commonly occurred. No evidence of acquired resistance to immune control was found in 12 relapse reisolates. Relapse tumors remained sensitive to donor antihost CTLs in vitro, suggesting continued expression of mHAgs. Reisolates also continued to express Fas. However, loss of posttransplant alloreactivity was observed at 3 weeks. This was temporally associated with the time of relapse. Antihost alloreactivity could be reactivated in stable graft-versus-host disease-free recipients by immunization with host cells. The results of this study suggest that one mechanism for relapse after bone marrow transplant is acquired tolerance of allogeneic minor histocompatibility antigens and that posttransplant immunotherapy directed against mHAgs may induce antitumor activity.
Modifying the diet can have a favorable impact on the course of chronic inflammatory bowel disease. In contrast, nutrition plays no major role in the development of the disease or in provoking an acute attack. During an acute attack of Crohn's disease, the application of oral (drinks) or tube feeding (enteral nutrition) can result in a substantial clinical improvement. The remission rate of this side effect-free therapy is, however, lower than that seen with steroids. The data available for diet therapy in ulcerative colitis are less unequivocal, but the effects are probably slight at best. During the remission phase, the patient should eat a balanced, vitamin-rich and varied normal diet that excludes all poorly tolerated foods: lactose intolerance in particular appears to be increased in patients with Crohn's disease. Known vitamin or mineral deficiencies should be corrected by appropriate dietary measures or supplementation. There is no such thing as a "Crohn's diet" or "colitis diet". The patient can be allowed to eat anything that is tolerated.
Canada's Royal Commission on New Reproductive Technologies rejects all forms of surrogacy arrangement under the rubric of objecting to commercial surrogacy. Noncommercial surrogacy arrangements, however, can be defended against the commission's objections. They can be viewed as cases of giving a benefit or service to another in a way that expresses benevolence, and establishes a relationship between surrogates and prospective 'social' parents that allows mutual understanding and reciprocal personal interaction between them.

Several morphological and biochemical techniques are in use for identification of apoptotic and necrotic cells in a studied cell population. It is essential to define not only the type of cell death but also to identify the apoptotic process itself, which represents a multistage, active process, requiring activation of a molecular event cascade. In the present study, we have examined and discussed effectiveness of the selected techniques detecting apoptosis in lymphocytes exposed to incubation at an elevated temperature. The appraisal involved detection of caspase-3 active form, detection of Bcl-2, TUNEL reaction, the comet assay and electrophoresis of DNA.

Among the most contested aspects of medical research is the randomized clinical trial (RCT). While the majority of arguments justifying the RCT and its use in medical research rest within a utilitarian framework, many Kantians claim that a deontological ethical framework is prohibitive of the use RCTs in medical research. This paper argues that, in fact, the RCT is permissible within a deontological framework.

Sympathetic neuronal apoptosis after nerve growth factor (NGF) deprivation requires the activation of two events: a protein synthesis-dependent, Bax-dependent release of mitochondrial cytochrome c and a protein synthesis-independent, Bax-independent development of competence. Unlike in most cells, cytosolic cytochrome c is not sufficient to induce cell death in NGF-maintained sympathetic neurons but can do so in neurons that have developed competence. We report that cytosolic cytochrome c-induced apoptosis in competent sympathetic neurons is completely dependent on caspase-9. In addition, the neuroprotective agents KCl and chlorophenylthio-cAMP are potent inhibitors of the development-of-competence pathway in NGF-deprived sympathetic neurons. We also find that the development of competence is reversible. Readdition of NGF reverses competence, and neurons can regain their resistance to cytosolic cytochrome c. Importantly, we examined the mechanism of development of competence and report that the inability of cytochrome c to activate caspases in NGF-maintained sympathetic neurons can be overcome with exogenous Smac that inhibits the inhibitor of apoptosis (IAP) family of proteins. Microinjection of cytochrome c and Smac, but neither alone, induces rapid cell death in NGF-maintained neurons. These data suggest that development of competence may be the result of the loss of the function of one or more members of the IAP family of caspase inhibitors that is needed before cytochrome c can activate caspases and induce cell death in neurons.
A theoretical method is derived for computation of the interactin of pressure and a weak anesthetic such as nitrogen or other narcotic gases in a two-dimensional material so as to prevent signs and symptoms of the high pressure nervous syndrome. A ratio of one part nitogen to 9 parts helium (pressure) is derived, which is in excellent agreement with previous human experiments in men at 31 ATA, with PO2 = 0.5 ATA.
