[Induced pemphigus caused by occupational contact with Basochrom].

The appearance and development of a vesiculobullous eruption at the capillitium, on the body and on the mucous membrane of mouth and nose following occupational contact with basochrom (basic chromium sulfate) on a 25-year-old man is described. The clinical symptoms, the histological and immunological findings as well as the course of the disease led to the result that it concerns an induced pemphigus.

Phosphorus-32-colloidal chromic phosphate: treatment of choice for malignant pericardial effusion.

A 68-yr-old male with agnogenic myeloid metaplasia was given phosphorus-32-colloidal chromic phosphate intrapericardially for the treatment of malignant pericardial effusion. Technetium-99m-sulfur colloid was used to verify catheter placement and to visualize distribution within the pericardium. Estimated dosimetry for this mode of therapy is presented, and it is suggested that pericardial administration of phosphorus-32-colloidal chromic phosphate is the treatment of choice for malignant pericardial effusion.

Uptake and distribution of chromium in isolated rat hepatocytes and its relation to cellular injury.

In order to study the difference of uptake and distribution between hexavalent (Cr6+) and trivalent (Cr3+) chromium in isolated rat hepatocytes, the cells were incubated with Cr6+ or Cr3+ (1 mM Cr) at 37 degrees C for up to 60 min. Leakage of lactate dehydrogenase from the hepatocytes into the suspension medium as an indicator of cellular injury was facilitated by Cr6+ (K2Cr2O7) at 60 min of incubation, whereas Cr3+ [Cr(NO3)3] had no effect. After 60 min of incubation with Cr6+, about 33% of the added Cr was found in the hepatocytes, whereas incubation with Cr3+ resulted in transfer of about 66% of the added Cr to the cells. After 20 and 40 min of incubation with Cr6+, about 39% of cellular Cr was found in the cytosolic fraction of hepatocytes, followed by a reduction to about 35% after 60 min of incubation. However, Cr detected in the cytosolic fraction of hepatocytes incubated with Cr3+ was about 1% of cellular Cr. Cr-binding substances in the cytosolic fraction of hepatocytes incubated with Cr6+ were eluted with two Cr peaks by Sephadex G-200 chromatography. These Cr-binding substances in the low-molecular-weight fractions were separable into at least two substances by thin-layer chromatography. These results suggest that Cr6+ readily passes through the cell membrane and combines with substances already present in the cytosolic fraction of hepatocytes, unlike metallothionein induced by cadmium, followed by detoxification. Consequently, cellular injury might be induced by Cr6+ which could not combine with LMCr in the cytosolic space of the hepatocytes.

Use of MagniSep chromium dioxide particles in an immunoassay system.

The use of chromium dioxide particles as a solid support for very sensitive and rapid immunoassays, is the result of the combination of large surface area (40 m2) and high protein uptake capacity (40 mg/g) allowing rapid capture kinetics and high binding capacity. Magnetic and physical properties of these particles give a rapid separation, a complete resuspension and a rapid high-efficiency washing, highly desirable characteristics for efficient automation of immunoassays. Good precision and accuracy, exemplified by excellent recovery, parallelism and correlation were demonstrated. Test results prove that the technology is highly flexible and applicable to a variety of assay formats.

[Determination of the nucleic acids in pig embryonic kidney cells by magnetic cytaphoresis].

Gallocyanine-chrome alum-stained pig embryonic kidney cells have paramagnetic properties. They move under the influence of gradient magnetic field (magnetophoresis). The velocity of magnetophoresis is proportional to the content of nucleic acids in cells. This allows to estimate the content of nucleic acids per cell dry weight by magnetophoresis and analytical centrifugation.

Ultrastructure of the predentinal-dentinal border zone of human teeth exposed to basic chromium (III) sulfate. A method for contrasting of collagen fibrils.

An electron microscopic investigation was undertaken to study a method for contrasting predentine collagen fibrils by their cross-striations after exposure of tissue to chromium solution for various periods. After 1 day of treatment, the cross-striations of collagen fibrils in predentine were clearly visible. The contrast of the cross-striations increased with longer periods of exposure to chromium. This method makes it possible to study the orientation of crystallites as well as collagen fibrils.

[Morphological studies of the effect of chromium-III-chloride on the islet cell organ in rats under the conditions of high and low fat diets].

It is well known, that the glucose tolerance factor amplifies the peripheral action of insulin. This factor contains trivalent chromium. This investigation deals with the influence of trivalent chromium on morphometric and histological parameters of pancreatic islets of wistar rats. Animals were given chromium-III-chloride for 8 weeks (perorally, daily 15 ppm or 3 ppm) under conditions of high (HFD) and low (LFD) fat diet. Staining of pancreatic slices was performed with Victoria blue and acid Fuchsin. The relative volume density of the endocrine cells was obtained as the ratio of the number of islet cells to the number of exocrine pancreatic cells. The statistical evaluation was performed by using Students t-test. In HFD animals (n = 12) chromium supplementation caused an increase by 41% of volume of islets compared to HFD controls. A lot of small islets were found, which may be newly formed. Moreover, polynesia and hyperplasia of islets were observed. LFD animals (n = 12) do not show any morphometric changes in comparison with LED controls; but macronesia, micronesia and polynesia of islets were observed. In conclusion we suggest, that under special conditions trivalent chromium has growth stimulatory effects on the endocrine pancreas.

Application of novel chromium dioxide magnetic particles to immunoassay development.

We have used chromium dioxide magnetic particles as the solid support in developing a series of immunological tests. The high surface area (greater than 40 m2/g) available on the magnetic particles and their easy dispersion throughout a solution allow for rapid and complete capture of the target antigen. The magnetic responsiveness of the particles allows for rapid, high-efficiency washing to reduce nonspecific binding, which often limits the sensitivity of serological assays. These features form the basis of extremely rapid and flexible assays for several hormones and markers of cancer and infectious disease. Most of the assays involve monoclonal antibodies. Here we describe specific performance characteristics for thyroxin, follitropin, creatine kinase isoenzyme MB, and antibody to human immunodeficiency virus (HIV). All of the assays are performed in less than 90 min, many in 30 to 45 min. The technology is highly flexible and is suitable for a variety of formats, from manual to fully automated.

Colloidal chromic phosphate 32P synovectomy in antigen-induced arthritis in the rabbit.

Radioisotopes have been employed in the therapy of chronic arthritis, in particular, rheumatoid arthritis for many years. A variety of isotopes have been popularized, and in the last ten years a colloidal solution of radioactive chromic phosphate 32P has been in use apparently with equivalent efficacy to others such as 169erbium, 90yttrium, and 165dysprosium. No controlled studies on this modality have been reported and few animal studies were found. The efficacy of therapeutic doses of 32P as a medical synovectomy and its effect on rabbit joints with antigen-induced arthritis were observed in 62 arthritic knee joints in 31 adult rabbits treated on one side with 0.1 microCi of 32P, the opposite serving as control. The animals were observed over a period of 11 months and examined by histologic and biochemical means. The synovium showed no evidence of radiation necrosis in treated joints. Cartilage of treated and control joints showed similar changes consistent with chronic arthritis, persistent synovitis, progressive chondrocyte degeneration, and decreased matrix metachromasia. The radiosynovectomy had neither removed synovium nor protected the cartilage. Its efficacy in humans is therefore questionable.
