Urban and rural differences in mortality and causes of death in historical Poland.

The purpose of this paper is to document and interpret urban-rural differences in mortality in the past. To this end, we used data on mortality in Wielkopolska, Poland, in the 19th century and at the beginning of the 20th century. The data on mortality in rural areas (N = 1,173,910 deceased), small towns (N = 573,903 deceased), and Poznań, the capital of the Wielkopolska region (N = 86,352 deceased), were gathered from original Prussian statistical yearbooks (Preussische Statistik). Causes of death were also analyzed (rural areas, N = 449,576 deceased; small towns, N = 238,365 deceased; Poznań, N = 61,512 deceased). Mortality measures such as crude death rate (CDR), infant death rate (IDR), and neonatal and postneonatal death rates were calculated. Life tables were constructed for both stationary and stable population models and measures of the opportunity for natural selection calculated (Crow's index I(m), potential gross reproduction rate R(pot), and biological state index I(bs)). Relative frequencies of leading causes of death were computed. Stratification depending on the place of residence was evident in all mortality measures as well as in the values of the life tables and the measures of the opportunity for natural selection, but it was reverse of what is observed today in developed countries. In Poznań (a large industrial city), the mortality situation was the least favorable. It was caused by large population density, lack of water supply and sewage systems (up to 1896), and bad working conditions. The values of CDR ranged between 26.89-31.46, and IDR between 190.6-280.5. Newborn life expectancy (for a stable population model) was 31.6 years, I(m) = 0.79, R(pot) = 0.85, and I(bs) = 0.47. The most common causes of death were tuberculosis, other diseases of the respiratory and circulatory systems, dysentery and diarrhea, and cancer. These diseases were less common in rural areas, so they had the most favorable values of mortality measures (CDR between 22.87-27.32, IDR between 181.8-219.4, life expectancy of newborn e(0) = 42.12, I(m) = 0.55, R(pot) = 0.93, I(bs) = 0.60). Infectious diseases (other than tuberculosis), frailty at birth, and frailty in old age were the most frequent causes of death in rural areas. Small towns (population <20,000) had a mortality intermediate between city and rural areas.

Recent estimates of adult mortality from widowhood in Bangladesh: a critical comparison.

"The indirect techniques of demographic estimation available at the moment are often inadequate to estimate levels in the presence of trends. The generalised stable population relations have been applied to census and survey data collected in Bangladesh for the estimation of adult mortality. This method combines two sets of widowhood data and yields estimates for the intersurvey period. The generalised stable population relations are based on the assumption that the population is closed to migration between the censuses. The application of the method is simple whatever may be the length of the inter survey period. But the method may be found to be affected by age misreporting and other errors." A refined estimator of measures of location of the age at first marriage.

Because of bias of unknown sign and extent introduced by age misreporting when calculating the singulate mean age of marriage in the usual manner, Van de Walle has suggested a fairly robust estimator based on stable population structure. Unfortunately not much is known about the properties of this estimator. Various demographers have argued informally that it indeed estimates the SMAM; others feel that it instead estimates the mean age of marriage in a cohort, the mean age of marriage in the stable population, or the singulate median age of marriage. In this paper the properties of this estimator are examined. Further, extensions of the Van de Walle estimator based on regression are shown to be significantly superior to the estimator alone.

Time and trajectory of convergence to population stationarity with immigration and low fertility.

"Recent research aimed at extending classical stable population theory to include immigration has shown that a stationary population is the long-term equilibrium outcome if, starting from any initial configuration, a population is projected forward under conditions of constant below-replacement fertility, constant mortality, and a constant annual number of immigrants whose age-sex composition is also fixed. This paper addresses two related questions: (1) What path does the projected population follow on its way to a long-term stationary population equilibrium? and (2) How long does it take for a stationary population to be achieved? To answer these questions a formal theory of population dynamics in the below replacement case is developed and then illustrated with a projection of the 1980 U.S. population."

Incidence and outcome of pauci-immune rapidly progressive glomerulonephritis in Wessex, UK: a 10-year retrospective study.

BACKGROUND: The Wessex Renal Unit serves a large stable population (2.5 million). Pauci-immune rapidly progressive glomerulonephritis (RPGN) is a frequent cause of acute renal failure requiring admission to our unit ( approximately 8%). At a population level, little is known of the epidemiology and outcome of RPGN. METHODS: Between 1 April 1986 and 31 March 1996, 141 cases of biopsy proven pauci-immune RPGN were seen in the Wessex region. The records of 128 patients were reviewed. Median (range) follow-up was 1.8 (0.9-9.64) years from diagnosis. RESULTS: The incidence of 4 per million was stable throughout the period. No clustering was seen. The diagnosis was made (median, 25th, 75th centile) 78.5 (45, 166) days after symptom onset. Co-morbidity (mostly hypertension) was seen in 47% of patients. Other organs affected were lungs 63%, nose/sinuses 50%, joints 42%, muscle 33%, skin 22% and nervous system 14%. Anti-neutrophil cytoplasmic antibody (ANCA) was positive in 73%; cytoplasmic ANCA 34%, peri-nuclear ANCA 26% and undifferentiated 14%. Twenty-seven per cent tested ANCA negative. The differences between the groups were small; time to diagnosis was shorter in the ANCA negative (-ve) group (P=0.02) and there were more airway symptoms in the ANCA positive (+ve) group (P:<0.05). All biopsies demonstrated a necrotizing process; crescents were seen in 96% involving (mean+/-SD) 54+/-26% of the glomeruli. Creatinine concentration (mean+/-SD) at diagnosis was 806+/-540 micromol/l. Treatment followed established immunosuppressive regimens. Initial dialysis was required by 59%, 36% needing long-term dialysis. At 1 year 68% were alive. The need for dialysis (P=0.0004) and age (P=0. 004) were poor prognostic markers. Ten per cent were transplanted, graft survival was 90% at 1 year, no recurrence was seen. CONCLUSIONS: This study, looking at a large cohort, has established the incidence and outcome of ANCA +ve and ANCA -ve RPGN in a defined stable population. It stresses the similarities between ANCA +ve and ANCA -ve cases and supports the notion that pauci-immune RPGN is part of a continuum of vasculitic illness. In this series transplantation is a safe option.

Conditional dispersal under kin competition: extension of the Hamilton-May model to brood size-dependent dispersal.

I consider a site-based model with contest competition among siblings, and assume that dispersal is conditional on the number of offspring in the natal site. Evolutionarily stable populations contain threshold dispersal strategies, which retain a certain number of offspring in the natal site and disperse the rest (if the actual number of offspring is less than the threshold, then all offspring are retained). Due to the discrete nature of the strategy set (the threshold must be integer), the ESS may not be unique or may not exist. In the latter case, two neighboring threshold strategies coexist in the evolutionarily stable population. Dispersal first decreases and then increases as a function of dispersal mortality, such that all but one offspring should be dispersed both when dispersal mortality is very small or very high. Population-level dispersal fractions are often similar to the unconditional ESS, but differ strongly when fecundity is small and dispersal mortality is high.

The immunological basis of lichenoid tissue reaction.

An autoimmune attack by T cells on the epidermis is the primary pathological event in the lichenoid tissue reaction (LTR). The severity of epidermal damage in the LTR is dependent on the relative balance between the intensity and perpetuation of T cell activation and the capacity of epidermal cells to protect from the attack. In natural disease processes, T cells activated by some exogenous agents migrate from the blood to the skin sites and invade the epidermis. Although this epidermotropic migration of T cells resulting in epidermal destruction is a complicated multistep process, this can be bypassed in fixed drug eruption (FDE). Thus, FDE is the most simplified disease model for investigating the pathogenesis of the LTR, in that large numbers of effector CD8+ T cells persist as a stable population in the resting lesions without causing epidermal damage and activation of these T cells resulting in localized epidermal injury can only be induced after administration of the causative drugs. Based on the findings observed with these CD8+ skin-resident T cells, we hypothesize that CD8+ T cells primed during viral infections could evolve into long-lived effector memory phenotype T cells; they would be subsequently trapped either specifically or nonspecifically in the inflamed skin sites and eventually persist as a stable population. Once cross-reacted with exogenous stimuli, such as drug or self-antigens, however, they would become effectors of epidermal damage as seen in various lichenoid skin diseases.

Simplified birth rate estimates under nonstable conditions.

Coale's robust birth rate estimate, obtained by adjusting the birth rate of a stable population selected by matching the observed population of both sexes under the age of 15, C(15), and the probability of survival of births to age 5, l5, is shown to be equal to the birth rate obtainable by reverse surviving the proportion under the age of 15. Variations of matching the criterion of Coale's method to the rate of increase and C(15) or l5 are shown to lead to variants of Coale's birth rate estimate that are also nearly equal to the reverse survival birth rate based on C(15). A simplified birth rate estimate that does not require reference to models of life tables or stable populations is suggested and some of its applications are illustrated.

Generation and characterization of a stable cell population releasing fluorescent HIV-1-based Virus Like Particles in an inducible way.

BACKGROUND: The availability of cell lines releasing fluorescent viral particles can significantly support a variety of investigations, including the study of virus-cell interaction and the screening of antiviral compounds. Regarding HIV-1, the recovery of such biologic reagents represents a very hard challenge due to the intrinsic cytotoxicity of many HIV-1 products. We sought to overcome such a limitation by using a cell line releasing HIV-1 particles in an inducible way, and by exploiting the ability of a HIV-1 Nef mutant to be incorporated in virions at quite high levels. RESULTS: Here, we report the isolation and characterization of a HIV-1 packaging cell line, termed 18-4s, able to release valuable amounts of fluorescent HIV-1 based Virus-Like Particles (VLPs) in an inducible way. 18-4s cells were recovered by constitutively expressing the HIV-1 NefG3C mutant fused with the enhanced-green fluorescent protein (NefG3C-GFP) in a previously isolated inducible HIV-1 packaging cell line. The G3C mutation creates a palmitoylation site which results in NefG3C-GFP incorporation into virions greatly exceeding that of the wild type counterpart. Upon induction of 18-4s cells with ponasterone A and sodium butyrate, up to 4 mug/ml of VLPs, which had incorporated about 150 molecules of NefG3C-GFP per viral particle, were released into the culture supernatant. Due to their intrinsic strong fluorescence, the 18-4s VLPs were easily detectable by a novel cytofluorometric-based assay developed here. The treatment of target cells with fluorescent 18-4 VLPs pseudotyped with different glycoprotein receptors resulted in these becoming fluorescent as early as two hours post-challenge. CONCLUSION: We created a stable cell line releasing fluorescent HIV-1 based VLPs upon induction useful for several applications including the study of virus-cell interactions and the screening of antiviral compounds.

Schistosoma japonicum in the pig: the host-parasite relationship as influenced by the intensity and duration of experimental infection.

Parasitologic, clinicopathologic, and pathologic aspects of Schistosoma japonicum infections of varying durations and intensities were studied in growing pigs injected intramuscularly with a dose of either 0, 100, 500, or 2,000 cercariae and killed at 4, 11, 17, or 24 weeks postinfection (PI). The number of viable worm pairs decreased significantly in the high dose group after 11 weeks PI but not in the lower dose groups; however, a stable population of immature worms persisted throughout the study in all dose groups. Liver egg counts also tended to decrease in the high dose group after 11 weeks but not in the other groups. Fecal egg excretion began at six weeks PI, was highest at eight weeks PI with a pronounced peak occurring only in the high dose group, and then decreased to low levels by 14 weeks PI in all groups. Egg counts from the feces as well as the liver correlated strongly with worm pair numbers during the acute phase of infection. The only clinicopathologic abnormality observed was an increase in circulating eosinophils corresponding to cercarial dose in all infected pigs by week six with peak counts occurring between six and eight weeks PI. The pigs exhibited no clinical signs of disease aside from diarrhea at the onset of patency. However, lesions were present throughout the large intestine of all infected pigs from 11 weeks PI, gradually decreasing with time. Severe liver fibrosis occurred in the 500 and 2,000 dose groups mainly at 11 weeks PI and then decreased in severity. In the liver, but not in the intestine, the severity of lesions at all time points was proportional to the cercarial dose given. The results indicate that after several weeks of patency, pigs with high intensities of S. japonicum infection are able to effectively eliminate the majority of adult worms while maintaining a stable population of immature schistosomes.

Subsistence strategies of two "savanna" chimpanzee populations: the stable isotope evidence.

Twenty-two chimpanzee hair samples collected from night nests at two different "savanna" sites were analyzed for carbon and nitrogen stable isotope ratios represented as delta13C and delta15N values. The first at Ugalla, Tanzania is a miombo woodland with grass groundcover and small patches of forest. The second at Ishasha, Democratic Republic of the Congo is a habitat composed of riverine gallery forest, semideciduous thicket forest, wooded grassland, and grassland. Based on comparative data from other primates, Ugalla hair delta13C values suggest that the chimpanzees are feeding primarily in the woodland rather than in forest patches or on grassland foods (grasses or grammivorous fauna). Similar comparisons indicate that the Ishasha chimpanzees are feeding within the forests and not in more open areas. In addition, the Ugalla chimpanzees had delta15N values that indicate extensive ingestion of leguminous flowers, seeds, and/or leaves. The Ishasha samples show a range encompassing three trophic levels. Two samples with the most positive values may indicate a nursing signal or vertebrate-feeding. Three individuals with intermediate values are similar to those in omnivorous nonhuman primate species. The four individuals with the lowest values are very similar to those in herbivorous monkeys. Stable isotope ratios permit time-averaged and habitat-specific dietary comparisons among sites, even without habituation or detailed foraging observations.

Demographic trends and biological status of historic populations from Central Poland: the Ostrów Lednicki microregion.

The microregion of Ostrów Lednicki in the province of Wielkopolska was the center of the formation of the Polish State in Early Middle Ages. The analysis of skeletal remains and parish records from the region documented the biological status of inhabitants and its changes over a millennium. The study material comprised 424 human skeletons from an Early Medieval burial ground on Lake Lednica, records of 2,704 deaths from the registers of the Roman Catholic parish of Dziekanowice, made between 1818-1903, data on the deaths of 929,192 inhabitants of rural areas all over the province Wielkopolska obtained from Prussian statistical materials for the years 1865-1900, and comparative data from the literature. Assuming both a stationary population model and a stable population model with nonzero natural increase, parameters of life tables and measures of opportunity for natural selection (Crow's index I(m), potential gross reproductive rate R(pot), and the biological state index I(bs)) were calculated for the Early Middle Ages and for the two periods of the 19th century which were characterized by different laws of land ownership and thus different rural economies. In the first period, peasants were tenants, whereas in the second, they were given freehold of the land they cultivated. Causes of death were also analyzed. A distinct increase in longevity from the early Middle Ages to the end of the 19th and the beginning of the 20th century was found. This was related to a higher level of demographic development in the parish of Dziekanowice during the 19th century, which was achieved earlier than in other areas of Poland. This was confirmed by genetic measures: coefficients of exogamy and coefficients of kinship. The reasons were related to the historical prominence of this region and to its proximity to the first two capitals of the Polish state, Gniezno and Poznań.

Birth interval study in a culturally stable urban population.

Five hundred women were interviewed within 2 days of delivery to examine indigenous birth spacing among the urban and rural population of Ife township. The crude birth interval was between 30 and 40 months due mainly to cultural attitudes towards lactation and sexual abstinence. The women studied possessed considerable knowledge of Western contraceptive methods, but they rejected them. The possible cause of this rejection is examined and solutions to the problem are suggested.

Role of apoptosis in the regulation of virus-induced T cell responses, immune suppression, and memory.

Apoptosis is an important mechanism enabling the selection of the non-self-reactive T cell repertoire and for maintaining homeostasis in the immune system after it has expanded to combat infections. Highly activated, proliferating T cells become susceptible to apoptosis driven by a number of stimuli, and T cells activated during a viral infection become susceptible to "activation induced cell death" after repeated stimulation through the T cell receptor (TcR). This is a major mechanism for the immune deficiencies observed during many viral infections. During infections with a high antigen load this can lead to a selective deletion of virus-specific cytotoxic T lymphocytes (CTL) and to the establishment of persistent infection. More commonly, the CTL control the infection first, and high levels of apoptosis in the expanded lymphocyte population occur after antigen and growth factors become limiting. This cell death does not seem to depend on TcR specificity, as the residual population contains a remarkably stable population of memory CTL precursors that approximate the frequency per CD8 cell of that seen during the peak of the acute infection. Subsequent infections with heterologous viruses result in an expansion and then an apoptotic elimination of T cells, with the consequence being a reduction in precursor CTL specific for the first virus. Thus, apoptosis shapes the quality and quantity of T cell memory.

Molecular diversity of methanotrophs in Transbaikal soda lake sediments and identification of potentially active populations by stable isotope probing.

Soda lakes are an environment with an unusually high pH and often high salinity. To identify the active methanotrophs in the Soda lake sediments, sediment slurries were incubated with a 10% (v/v) (13)CH(4) headspace and the (13)C-labelled DNA was subsequently extracted from these sediments following CsCl density gradient centrifugation. This DNA was then used as a template for PCR amplification of 16S rRNA genes and genes encoding PmoA and MmoX of methane monooxygenase, key enzymes in the methane oxidation pathway. Phylogenetic analysis of 16S rRNA genes, PmoA and MmoX identified that strains of Methylomicrobium, Methylobacter, Methylomonas and 'Methylothermus' had assimilated the (13)CH(4). Phylogenetic analysis of PmoA sequences amplified from DNA extracted from Soda lake sediments before Stable Isotope Probing (SIP) treatment showed that a much wider diversity of both type I and type II methanotroph sequences are present in this alkaline environment. The majority of methanotroph sequences detected in the (13)C-DNA studies were from type I methanotrophs, with 50% of 16S rRNA clones and 100% of pmoA clones from both Lake Suduntuiskii Torom and Lake Gorbunka suggesting that the type I methanotrophs are probably responsible for the majority of methane oxidation in this environment.
