Methylparathion, carbaryl and aldrin impact on nitrogen metabolism of prawn, Penaeus indicus.

Changes in hepatopancreas, muscle and gill tissue nitrogen metabolic profiles were studied in a penaeid prawn, Penaeus indicus, following its exposure to sublethal concentrations of methylparathion, carbaryl and aldrin. In all the insecticide exposed prawn tissues, Ammonia levels were significantly increased and a shift in the nitrogen metabolism towards the synthesis of urea and glutamine was observed. Inhibition of glutamate oxidation to ammonia and alpha-ketoglutarate by glutamate dehydrogenase suggests a mechanism whereby hyperammonemia is reduced by minimizing the addition of further ammonia to the already existing elevated ammonia pool. Increased alanine and aspartate aminotransferases demonstrates the onset of gluconeogenesis. Mechanisms to detoxify the ammonia by enhancing the synthesis of urea and glutamine at the cellular level was observed in the selected tissues pave way for the survivability of prawns in insecticide polluted environs.

Alterations in the behavior of young and adult rats exposed to aldrin during lactation.

The effects of the administration of aldrin, an organochlorinated pesticide, during the period of lactation were studied on behavior of the litters. The general activity of the young and adult animals was tested in an open-field. In both situations, activity was higher in experimental than in control rats. Experimental animals also showed a higher performance in a hole-board apparatus and in the shuttle-box of two-way active avoidance learning. The results suggest that the insecticide is involved in alterations occurring in the nervous/endocrine systems of the newborn.

Inhibition of gap junctional-mediated intercellular communication in vitro by aldrin, dieldrin, and toxaphene: a possible cellular mechanism for their tumor-promoting and neurotoxic effects.

Several mechanisms have been postulated to be responsible for the pleiotropic effects of toxic chemicals. Although the cytotoxicity and mutagenicity of chemicals are well studied and relatively easily detected, the noncytotoxic and nonmutagenic (i.e., epigenetic) mechanisms of chemical toxicity are less well understood. An in vitro assay, using cocultures of Chinese hamster cells to measure metabolic cooperation between V79 6-thioguanine-sensitive (6TGs) and resistant (6TGr) cells, has been developed to detect noncytotoxic and nonmutagenic chemicals that inhibit, quantitatively, gap junctional communication. The insecticides aldrin, dieldrin, and toxaphene, known to have pleiotropic toxic effects in animals, were shown to inhibit gap junctional communication. Interpretation of results suggests that chemical inhibition of gap junctional communication could be a possible mechanism to explain their tumor-promoting and neurotoxic effects.

Comparison of carcinogenicity studies with aldrin and dieldrin.

Laboratory testing of animals is the principal method used to identify chemical carcinogens. However conflicting results are often obtained. This is emphasized in a cross-comparison of 6 aldrin-dieldrin studies; 3 were positive for tumor induction and 3 were negative. Four components of the assays are examined: husbandry, compound administration, observation, and interpretation of results.

[Determination of aldrin, dieldrin and endrin in the air by gas chromatography].

A gas chromatographic method of determination of aldrin, dieldrin and endrin in air has been developed. The method involves trapping in ethylene glycol, extraction with hexane and determination by electron capture gas chromatography on a Gas Chrom Q column coated with 15% QF-1/10% DC-200. The determinability of the method amounts to 0,0015-0,003 mg/m3.

Aldrin epoxidase and 7-ethoxyresorufin-O-deethylase (EROD) activity in livers from lambs of three different breeds of Norwegian sheep.

The concentration of aldrin epoxidase was significantly greater in the livers of Norwegian Pelt lambs than in those of Rygja lambs but did not differ significantly between either of these two breeds and Spael lambs. Conversely, although there was no detectable EROD activity in the livers of many of the lambs in all three breeds, the median activity was greater in the livers of Rygja lambs than was the case for Spael lambs.

Analyses of lindane, vinclozolin, aldrin, p,p'-DDE, o,p'-DDT and p,p'-DDT in human serum using gas chromatography with electron capture detection and tandem mass spectrometry.

A sensitive, selective and reliable procedure was developed and validated to determine organochlorinated compounds, which present endocrine-disrupting effects, lindane, vinclozolin, aldrin, p,p'-DDT, o,p'-DDT and p,p'-DDE, in human serum. The analytical methodology combines serum extraction with organic solvents, clean-up of the organic extract using acid treatment with H2SO4, elution of the cleaned-up extract through liquid column chromatography system and analysis of the fraction eluted by gas chromatography (GC) with electron capture detection (ECD) and tandem mass spectrometry (MS-MS) detection mode. Performance characteristics, such as linearity, sensitivity, precision, accuracy or recovery, of both chromatographic methods were studied. The proposed analytical methodology was applied to analyse the target compounds in serum samples from women living in agricultural areas of Almería (Spain). Comparison of the MS-MS and the ECD results was made and the advantage of the MS-MS operation mode to determine endocrine disrupting compounds in complex matrices is also presented.

The effect of alcoholic cirrhosis on the activities of microsomal aldrin epoxidase, 7-ethoxycoumarin O-de-ethylase and epoxide hydrolase, and on the concentrations of reduced glutathione in human liver.

Activities of the microsomal mono-oxygenases 7-ethoxycoumarin O-de-ethylase (EOC) and aldrin epoxidase (AE), together with microsomal epoxide hydrolase (EH) activity and concentrations of reduced glutathione (GSH) have been measured in liver from patients with alcoholic cirrhosis and in normals. Activities of both mono-oxygenases were significantly reduced in alcoholic cirrhosis. EOC activity (pmol 7-OH coumarin formed/mg microsomal protein/min) was 108.0 +/- 10.6 (n = 8) in normals and 60.9 +/- 11.6 (n = 8) in alcoholic cirrhosis (P less than 0.01). AE activity (pmol dieldrin formed/mg microsomal protein/min) was 58.9 +/- 9.5 (n = 11) in normal liver biopsies and 29.9 +/- 8.6 (n = 9) in alcoholic cirrhosis (P less than 0.05). Microsomal EH activity (nmol styrene glycol formed/mg microsomal protein/min) was similar in normals (39.2 +/- 4.4, n = 11) and alcoholic cirrhosis (40.5 +/- 9.1, n = 6). GSH concentrations (microgram GSH/g liver tissue) were lower (P less than 0.01) in alcoholic cirrhosis (792 +/- 73, n = 10) compared to normals (1182 +/- 76, n = 6).

[Dieldrin, aldrin, endrin, isodrin, endosulfan 1 and 2 on fish in the Gulf of Gdańsk].

The residues of dieldrin, aldrin, endrin, isodrin, endosulfan 1 and 2 has been determined in a several species of fish caught in the Gulf of Gdańsk in 1992. The method of measurement was capillary gas chromatograph and low resolution mass spectrometry (HRGC/LRMS) after a nondestructive extraction and clean-up step with a further fractionation of the extract on Florisil column. Apart from dieldrin no other cyclodiene pesticides studied were found in fishes in detectable amounts, and for dieldrin concentrations ranged from 0.84 to 6.6 ng/g wet weight.

Genetics and linkage of aldrin resistance in the German cockroach, Blattella germanica (L.).

The inheritance mechanism and linkage association of cyclodiene resistance were studied in 2 cyclodiene-resistant strains of the German cockroach. The resistant strains were crossed to each other, to an unmarked susceptible strain, and to 6 susceptible marker strains. The progeny from succeeding generations were tested for resistance to aldrin using a dipping method or a time-exposure method, and were examined for markers in linkage tests. The results show that cyclodiene resistance (R-Cyclo) is inherited as a simple autosomal semidominant trait which is allelic in the 2 resistant strains. Linkage studies indicate that R-Cyclo is independent of linkage groups II-VI, but is linked with the group VII trait curly wing at a distance of 3.4+/-1.0-6.2+/-1.4.
