[A study on taxonomy of Rhizobia isolated from Astragalus sp].

Thirty-six strains isolated from root nodule of Astragalus spp., in comparison with 31 reference strains of Rhizobium, Bradyrhizobium and Sinorhizobium species, and some other strains isolated from legumes in Xinjiang and Hainian Province, were classified by performing numberical taxonomy, DNA-DNA hybrization and partial 16S rRNA gene sequencing. Results of herichical analysis showed most of trains isolated from Astragalus spp. fell into subgroups 8 and 9, Also the DNA homolgy among strains of subgroups 8 and 9 are more than 70%, with the exception of strains CA8593 and SX044, and the DNA homology between strain CA8561, JL84 and type strains of all described rhizobial species are less than 56%. These results indicated that these two subgroups 8 and 9 were unique DNA homologous groups, distinguishing from all described rhizobial species. Sequencing of partial 16S rRNA gene showed that cetrostrain CA8561 of subgroup 8 is phylogenetically far from all know species of Rhizobium, Bradyrhizobium, Sinorhizobium, Azorhizobium and Agrobacterium, and it is a unique geneline. The cetrostrain JL84 of subgroup 9 has a unique position in the phylogenetic branch consisted of species of Rhizobium and Agrobacterium.

[Effects of astragalus (ASI, SK) on experimental liver injury].

The saponins (ASI, SK) used in this study was extracted from the root of Astragalus membranaceous Bge and Astragalus sieversianus Pull. ASI and SK were found to protect liver from chemical injury induced by CCl4, D-galactosamine and acetaminophen in mice. The two saponins were shown to impede the elevation of SGPT level, decrease the MDA content and increase the GSH concentration in mouse liver. Obvious improvement of histological changes were also observed. The protective action of ASI and SK against the hepatotoxicity was also shown in experiments using primary cultured rat hepatocytes. The average value of GPT in the medium treated with different concentration of ASI and SK (0.00075-0.18 mmol/L) was lower than that in control. Analyzing through multiple linear correlation, we showed that the lowering of SGPT was negatively related to the increase of GSH, positively related to the decrease of MDA in mice given CCl4 or acetaminophen in combination with ASI or SK. These results indicate that the hepato-protective effects of ASI and SK may be due to their anti-oxidation activities, since the content of liver protein in mice given ASI or SK was more than that in the controls. Moreover, the level of hepatic microsomal cytochrome P-450 in all mice given the two saponins were significantly increased, the liver metabolism and immunoregulating action produced by ASI and SK may be also involved in their hepato-protective effects.

Toxicosis in pigs fed selenium-accumulating Astragalus plant species or sodium selenate.

Three groups of 5 pigs each were fed a high selenium (Se) diet by mixing either Astragalus praelongus (31.6 ppm Se in feed), A bisulcatus (31.7 ppm Se in feed), or sodium selenate (26.6 ppm Se in feed) with commercial hog feed. Ten control pigs were fed only commercial hog chow containing trace selenium (0.44 ppm Se). Pigs were fed for 9 weeks and necropsied when they had ataxia or paralysis. Blood was collected for hematologic and serum biochemical determinations, and samples of various tissues were collected and fixed in neutral-buffered 10% formalin for histologic evaluation or frozen for determination of selenium concentration. All forms of selenium induced clinical signs of weight and hair loss, with cracked hooves and inflamed coronary bands developing in all Na2SeO4-fed pigs and 1 A praelongus-fed pig, but not in A bisulcatus-fed pigs. Serum calcium, phosphorus, and albumin concentrations were unchanged or significantly decreased from prefeeding values in groups fed selenium. Serum aspartate transaminase (AST) activities in Astragalus species-fed groups, and amylase activities and PCV in all groups of pigs fed selenium, were increased. Serum alkaline phosphatase and creatine kinase activities were significantly increased in the A praelongus-fed pigs and significantly decreased in Na2SeO4-fed pigs. Terminal tissue and body fluid selenium concentrations were determined in all groups of pigs fed selenium and compared with values in control pigs. Urine and bile concentrations were increased by the greatest factor (40 to 100x), with tissue concentrations of selenium increased by a lesser factor (6 to 17x).(ABSTRACT TRUNCATED AT 250 WORDS)

Aliphatic nitro compounds in Astragalus lusitanicus Lam.

Astragalus lusitanicus Lam lusitanicus has been reported to be toxic for mammals. Annually, many sheep are poisoned and die because of feeding on this legume. Aliphatic nitro compounds were detected using two different methods. Colormetry with Griess-ilosvay reagent showed that nitro-compounds occurred in frozen plant samples. Two different compounds in aqueous extract and 1 or 2 in methanolic extracts were detected by thin-layer chromatography. Nitro compounds concentrations were similar to those reported in toxic species of North American Astragalus.

Astragalus bisulcatus--a cause of selenium or locoweed poisoning?

Certain plants accumulate selenium at levels that render them toxic. One of the principle groups of selenium-accumulating plants is the selenium-accumulating Astragalus. Reports in early literature suggest that selenium is the only toxin in these plants. Sheep fed Astragalus bisulcatus, a selenium-accumulating plant, had signs similar to those observed in locoweed poisoning, and the microscopic lesions from tissues collected from these sheep were similar to those of locoweed poisoning rather than selenium poisoning. This suggests this group of plants contains toxin compounds in addition to selenium.

Regulatory action of Astragalus saponins and buzhong yiqi compound on synthesis of nicotinic acetylcholine receptor antibody in vitro for myasthenia gravis.

With overall analysis of symptoms and signs of myasthenia gravis (MG) basing on "Pi (Spleen) Deficiency" [symbol: see text] theory and with years of our clinical experience in treating MG, we performed a pharmacological study of Astragalus saponins and Buzhong Yiqi Compound (Tonic granulae invigorating vital energy) in 14 peripheral blood mononuclear cell (PBMNC) cultures from 10 MG patients. PBMNG from two groups of patients given dexamethasone (Dxm) and cobalt 60 (60Co) treatment were used as controls. The results showed that water soluble Astragalus saponins significantly reduced the titer of nicotinic acetylcholine receptor antibodies (nAchR-Ab) in the cell culture supernatants, from 418.8-2328 to nil in 6 cases, and from 1143-1235 to 43-157 fmol/ml in 2 cases, and that Buzhong Yiqi compound also had inhibitory immunoregulatory action.

Tissue and serum swainsonine concentrations in sheep ingesting Astragalus lentiginosus (locoweed).

Locoweed intoxication or locoism results when animals continuously graze certain plants of the general Astragalus or Oxytropis. The locoweed toxin, swainsonine, is water soluble and is rapidly absorbed and eliminated. The purpose of this study was to determine the distribution of swainsonine in tissues of sheep eating locoweed and to determine if the tissue swainsonine concentrations change with continued locoweed ingestion. Fifteen cross-breed whethers were divided into 3 groups of 5 each and fed alfalfa pellets (Group 1) or alfalfa pellets with 10% Astragalus lentiginosus for 13 d (Group 2) or for 21 d (Group 3). After the feeding periods, the animals were slaughtered and tissues were collected, frozen and later analyzed for swainsonine using an in vitro, alpha-mannosidase inhibition assay. Significant alpha-mannosidase inhibitory activity (interpreted as ng/ml of swainsonine) was detected in whole blood, skeletal muscle, brain, kidney, liver, thyroid and urine. The swainsonine concentrations in tissues were significantly correlated with daily swainsonine intake (r = 0.58 to 0.96). With the exception of kidney, longer exposure did not result in significant increases in the swainsonine concentrations in blood, muscle, brain, liver or thyroid. Liver had the highest swainsonine concentrations with 3049 +/- 1952 and 3947 +/- 457 ng/ml (mean +/- SD) in Groups 2 and 3 respectively. Swainsonine concentrations varied widely within the groups suggesting individual animal variability in swainsonine absorption, metabolism and excretion. These findings suggest that swainsonine is present in tissues of animals eating locoweed and that in most tissues the amount was directly correlated to the swainsonine dose ingested, but not to the length of exposure.

[False positive with Tl-201 in the study of Ewing's sarcoma of the astragalus caused by reflex sympathetic dystrophy].

We describe a 14 year old male, diagnosed of Ewing sarcoma in left astragalus. We performed two 201-Thallium scan for evaluating the response to chemotherapy (before and after the treatment). The scan performed after chemotherapy showed patchy uptake in astragalus, calcaneus and some joints of the foot. The following radiology and scintigraphy diagnosed a reflex sympathetic dystrophy in the left foot due to disuse of the limb. The 201-Thallium overestimated viability of the tumour for this reason. We must discard the presence of this kind of associated pathology in order that they could be cause of false positives.

Chinese herbs: a clinical review of Astragalus, Ligusticum, and Schizandrae.

Although Astragalus, Ligusticum and Schizandrae have a long history of medicinal use within the traditional Chinese system, only recently has the West begun to understand their pharmacological possibilities and clinical applications. Astragalus has demonstrated a wide range of immunopotentiating effects and has proven efficacious as an adjunct cancer therapy. Ligusticum, and its active components, have been investigated for enhancement of the immune system, treatment of ischemic disorders, and as an anti-inflammatory. Clinically, the hepato-protective and antioxidant actions of Schizandrae have proven beneficial in the treatment of chronic viral hepatitis.

Suppressive effect of Astragalus membranaceus Bunge on chemical hepatocarcinogenesis in rats.

Astragalus membranaceus (AM) has been widely used for treating liver diseases in traditional Chinese medicine. Experimental evidence indicates that it has antitumor potential. In this study, the effect of AM on hepatocarcinogenesis induced by diethylnitrosamine (DEN), two-thirds partial hepatectomy, and 2-acetylaminofluorene (2-AAF) (DEN-PH-AAF) was evaluated using glutathione S-transferase placenta form (GST-P) as marker. First, rats were injected intraperitoneally (i.p.) with DEN (200 mg/kg in saline), a two-thirds partial hepatectomy was carried out 2 weeks later, and the rats were then placed on a basal diet containing 0.02% AAF from week 3 to week 8 to induce hepatocarcinogenesis. The rats were given AM (90 mg/kg or 180 mg/kg body weight) by gavage from week 3 to week 8 (treatment groups). The formation of GST-P-positive foci and the expression of GST-P protein and mRNA caused by DEN-PH-AAF were reduced in the treatment groups, which clearly suggests that AM is effective in delaying DEN-PH-AAF-induced hepatocarcinogenesis.

Preparative isolation and purification of calycosin from Astragalus membranaceus Bge. var. mongholicus (Bge.) Hsiao by high-speed counter-current chromatography.

Calycosin was purified from an ethyl acetate extract of the root of Astragalus membranaceus Bge. var. mongholicus (Bge.) Hsiao by high-speed counter-current chromatography. The separation was performed in two steps with a two-phase solvent system composed of n-hexane-chloroform-methanol-water (1:3:3:2, v/v). From 200 mg of the crude extract, 14.8 mg of calycosin was obtained at over 99% purity as determined by HPLC analysis, and its chemical structure was confirmed by MS, 1H and 13C nuclear magnetic resonance.

Flavonoids and phenol carboxylic acids in the oriental medicinal plant Astragalus membranaceus acclimated in Poland.

Astragalus membranaceus (Fisch.) Bunge has been was successfully acclimated in Central Europe. We report the content of isoflavones and some other polyphenolic compounds in roots and aerial parts that have been analyzed by means of TLC and HPLC. The total amount of isoflavones in leaves, was 0.55 mg g(-1) dry weight, and of the flavonols--up to 3.54 mg g(-1). In the roots isoflavonoid content was extremely variable, but reached 3.04 mg g(-1), whereas flavonols content was 0.49 mg g(-1).

[Influence of flavonoid of Astragalus membranaceus's stem and leaves on the function of cell mediated immunity in mice].

OBJECTIVE: To investigate the immune regulation of flavonoid of Astragalus membranaceus's stem and leaves (FAM). METHODS: Changes of total T cell count and subsets in mice were determined by monoclonal antibody assay before and after treatment with FAM, and the LAK activity was tested simultaneously by isotope technique. RESULTS: FAM could promote the proliferation of lymphocytes induced by ConA, raise the T cell count and regulate the T cell subsets disorder, elevate the LAK activity induced by IL-2. CONCLUSION: FAM possesses the effect of immune stimulation and immune regulation in treating immunosuppressive mice. This study provides the experimental basis for clinical application of FAM.

[Experimental studies on improving heart preservation effect of Astragalus saponins].

OBJECTIVE: To observe the effect of heart preservation of Astragalus saponins (AS) in modified Euro-Collins solution (mEC) containing AS. METHODS: Wistar rats were randomly divided into mEC and AS group. After preserving isolated rat hearts 6 hours by simple cold storage with mEC and mEC containing AS respectively, Langendorff's isolated rat heart model was used to reperfuse for 30 minutes to study cardiac function after preservation and the effect of preventing oxygen free radical injury. RESULTS: To compare AS group with mEC group, cardiac function and coronary flow of the hearts after preservation were better than mEC group (P < 0.05), in AS group the water content of myocardium was decreased very significantly (P < 0.01), myocardial lactate dehydrogenase and creatine phosphokinase release were reduced greatly (P < 0.05 and P < 0.01), myocardial superoxide dismutase activity was increased very significantly (P < 0.01) and the content of lipid peroxide was decreased significantly (P < 0.05). CONCLUSION: AS could improve the effect of heart preservation of mEC solution, its mechanism might be associated with the effect of negative contractile strength and preventing oxygen free radical injury.

[Effect of Astragalus Angelica Mixture on lipoprotein lipase and lecithin cholesterol acyltransferase of nephrotic rats].

OBJECTIVE: To investigate the mechanism of Astragalus Angelica Mixture (AAM) effect of regulating lipid metabolism disturbance in nephrotic rats. METHODS: To examine the effects of AAM on serum albumin, lipid levels, and activities of lipoprotein lipase (LPL) and lecithin-cholesterol acyltransferase (LCAT), which are key enzymes for catabolism of lipid, in immune-induced nephrotic hyperlipidemia rats and exogenous hyperlipidemia rats. RESULTS: Serum albumin was reduced, lipid increased significantly, LPL activity decreased significantly and the LCAT activity was relatively insufficient in nephrotic rats. Activities of LPL and LCAT increased significantly in AAM treated nephrotic rats. There were no change of activities of LPL and LCAT in exogenous hyperlipidemia rats and AAM showed no effect on the activities of these two enzymes. CONCLUSION: The effect of AAM in regulating lipid metabolism might be due to enhancing the clearance of triglyceride and cholesterol-rich lipoprotein by improving the activities of LPL and LCAT.

[Study on the anti-herpes simplex virus activity of a suppository or ointment form of Astragalus membranaceus combined with interferon alpha 2b in human diploid cell culture].

A study on the anti-herpes simplex virus activity of the suppository or ointment form of Astragalus membranaceus(AM) combined with recombinant human interferon alpha 2b(IFN) was carried out in human diploid cell culture. AM is a Chinese herb medicine and have been used in China as a tonic for thousands of years and the IFN was produced from E. coli with 95% purity. Obtained results indicated that the placebo suppository and ointment(without AM and IFN) seemed not to decrease markedly the anti-viral activity of IFN in WISH/VSV assay system. The anti-herpes simplex virus activities of suppository and ointment forms of AM and IFN were shown to be significantly higher than that of IFN alone. It is well known that chronic cervicitis is closely related to papillomavirus, cytomegalovirus as well as herpes virus infections. The AM-IFN suppository is suggested to be used in the treatment of cervicitis and the ointment in the treatment of skin herpes.

[Effect of Astragalus membranaceus injection on megakaryocyte hematopoiesis in anemic mice].

OBJECTIVE: To assess the effect of Astragalus membranaceus injection (AMI) on megakaryocyte hematopoiesis in anemic mice and explore its mechanism. METHODS: Anemic models of mice were randomly divided into two groups: treatment group and anemic control group. Intraperitoneal doses of AMI (20 mg/(ml.20 g) q.d x 6) were given to the treatment group, and equal doses of physiological saline were given to the anemic control group. On days 8, 11 and 14 after treatment, blood platelet and bone marrow cells were determined, and the numbers of CFU-Meg (colony forming unit-megakaryocyte) and Meg-CSA (megakaryocyte colony-stimulating activity) were determined by using technique of hematopoietic progenitor cells culture in vitro. RESULTS: Serum Meg-CSA of the treatment group was significantly higher than that of the anemic control group. The abovementioned indices of the treatment group recovered to normal by day 11, which was markedly earlier than the day of recovery observed in the anemic control group. CONCLUSION: AMI can increase serum Meg-CSA of anemic mice and accelerate the recovery of megekaryocyte hematopoiesis after bone marrow suppression.
