Studies on viridenomycin, a novel 24-membered macrocyclic polyene lactam antibiotic.

A new antitumor antibiotic, designated AL081, was obtained from the culture filtrate of an actinomycete identified as Streptomyces gannmycicus, and found to be identical with viridenomycin by direct comparison. The structure of the antibiotic was determined by NMR spectral analysis including a variety of two-dimensional techniques to be a novel 24-membered macrocyclic polyene lactam. Viridenomycin prolonged the survival periods of mice bearing P388 leukemia and B16 melanoma cells.

A family of novel macrocyclic lactones, the saccharocarcins produced by Saccharothrix aerocolonigenes subsp. antibiotica. I. Taxonomy, fermentation, isolation and biological properties.

A nocardioform actinomycete, SCC 1886, isolated from a soil sample collected in Ohio was found to produce, in fermentation, six novel macrocyclic lactones, the saccharocarcins. The producing culture was identified as Saccharothrix aerocolonigenes subsp. antibiotica based on the formation of fragmenting substrate mycelia, aerial mycelia that coalesce to form aerial colonies, whole-cell hydrolysates that contain meso-diaminopimelic acid, galactose and rhamnose and physiological comparisons to type species of the genus. Peak production of the saccharocarcins occurred after 95 hours of fermentation in a starch rich medium. The compounds were isolated from the fermentation broth by solvent extraction and purified by HPLC. Isolated compounds were active against Micrococcus luteus, Staphylococcus aureus and Chlamydia trachomatis; none were cytotoxic at concentrations up to 1.0 microgram/ml.

New insights into rifamycin B biosynthesis: isolation of proansamycin B and 34a-deoxy-rifamycin W as early macrocyclic intermediates indicating two separated biosynthetic pathways.

Proansamycin B, the formerly postulated intermediate of rifamycin B biosynthesis, was isolated from cultures of the Amycolatopsis mediterranei mutant F1/24. The structure was determined using UV, IR, NMR and MS techniques. Biotransformation studies demonstrate that proansamycin B is an intermediate of a shunt pathway, a 8-deoxy variant, of rifamycin B biosynthesis leading to 8-deoxy-rifamycin B as the final product. In addition, 34a-deoxy-rifamycin W, the direct precursor of rifamycin W, could be isolated representing the earliest macrocyclic intermediate obtained so far in the biosynthetic route to rifamycin B. Furthermore, the new rifamycin W-28-desmethyl-28-carboxy and rifamycin W-hemiacetal, intermediates in the transformation sequence of rifamycin W to rifamycin S, were isolated. Application of proton NMR measurements (double resonance and ROESY experiments) on the latter compound indicated that the stereochemistry at the chiral center C-28 is R.

A macrocyclic reporter ligand for Mg2+: analytical implications for clinical magnesium determinations.

A new approach is presented for measuring Mg in plasma using the macrocyclic reporter ligand, NOTMP (1, 4, 7-triazacyclononane- 1, 4, 7-tris (methylene methylphosphinate)) and 31P nuclear magnetic resonance spectroscopy (NMR). By virtue of its intermediate binding constant for Mg (Kd = 0.35 mM), measurements of Mg using NOTMP allows one to discriminate between Mg bound to the high and low affinity ligands present in plasma, when combined with more conventional measurements of Mg. We used this approach in conjunction with measurements of total Mg using atomic absorption spectroscopy (AAS) and ionized Mg using an ion selective electrode (ISE) to characterize the distribution of Mg in the plasma of 16 normal adults. The percentage of Mg distributed among high and low affinity ligands and in an ionized fraction was 31, 14 and 55 per cent respectively. Similar measurements on plasma following equilibrium dialysis suggest that the high and low affinity ligands in plasma correspond to high and low molecular weight compounds, respectively. Measurement of Mg by NOTMP, AAS and ISE were not affected by the storage of blood samples for up to 48 h at 4 degrees C. The addition of MgSO4 to plasma and its subsequent analysis by these three methods suggests that the added Mg is primarily distributed among the high affinity ligands (mostly likely proteins) and ionized fractions. The approach presented here may offer novel insights into assessing the distribution of Mg in clinical samples.

Binding of a macrocyclic bisacridine and ametantrone to CGTACG involves similar unusual intercalation platforms.

The binding of a macrocyclic bisacridine and an antitumor intercalator ametantrone to DNA has been studied. We carried out X-ray diffraction analyses of the complexes between both intercalators and CGTACG. We have determined the crystal structure, by the multiple-wavelength anomalous diffraction (MAD) method, of bisacridine complexed with CGTA[br(5)C]G at 1.8 A resolution. The refined native crystal structure at 1.1 A resolution (space group C222, a = 29.58 A, b = 54.04 A, c = 40.22 A, and R-factor = 0.163) revealed that only one acridine of the bisacridine drug binds at the C5pG6 step of the DNA, with the other acridine plus both linkers completely disordered. Surprisingly, both terminal G.C base pairs are unraveled. The C1 nucleotide is disordered, and the G2 base is bridged to its own phosphate P2 through a hydrated Co(2+) ion. G12 is swung toward the minor groove with its base stacked over the backbone. The C7 nucleotide is flipped away from the duplex part and base paired to a 2-fold symmetry-related G6. The central four base pairs adopt the B-DNA conformation. An unusual intercalator platform is formed by bringing four complexes together (involving the 222 symmetry) such that the intercalator cavity is flanked by two sets of G x C base pairs (i.e., C5 x G8 and G6 x C7) on each side, joined together by G6 x G8 tertiary base pairing interactions. In the bisacridine-CGTACG complex, the intercalation platform is intercalated with two acridines, whereas in the ametantrone-CGTACG complex, only one ametantrone is bound. NMR titration of the bisacridine to AACGATCGTT suggests that the bisacridine prefers to bridge more than one DNA duplex by intercalating each acridine to different duplexes. The results may be relevant in understanding binding of certain intercalators to DNA structure associated with the quadruplet helix and Holliday junction.

In vitro and in vivo anticancer activities of synthetic macrocyclic ketone analogues of halichondrin B.

Halichondrin B is a highly potent anticancer agent originally found in marine sponges. Although scarcity of the natural product has hampered efforts to develop halichondrin B as a new anticancer drug, the existence of a complete synthetic route has allowed synthesis of structurally simpler analogues that retain the remarkable potency of the parent compound. In this study, we show that two macrocyclic ketone analogues of halichondrir B, ER-076349 and ER-086526, have sub-nM growth inhibitory activities in vitro against numerous human cancer cell lines as well as marked in vivo activities at 0.1-1 mg/kg against four human xenografts: MDA-MB-435 breast cancer, COLO 205 colon cancer, LOX melanoma, and NIH: OVCAR-3 ovarian cancer. ER-076349 and ER-086526 induce G2-M cell cycle arrest and disruption of mitotic spindles, consistent with the tubulin-based antimitotic mechanism of halichondrin B. This is supported further by direct binding of the biotinylated analogue ER-040798 to tubulin and inhibition of tubulin polymerization in vitro by ER-076349 and ER-086526. Retention of the extraordinary in vitro and in vivo activity off halichondrin B in structurally simplified, fully synthetic analogues establishes the feasibility of developing halichondrin B-based agents as highly effective, novel anticancer drugs.

Analysis of the structure-anticancer activity relationship in a set of Schiff bases of macrocyclic 2,6-bis(2- and 4-formylaryloxymethyl)pyridines.

Quantitative estimation of the structure-anticancer activity relationship in a series of macrocyclic Schiff bases of 2,6-bis(formylaryloxymethyl)pyridines was carried out by the topological approach. Correlation equations describing the relationship between the anticancer activity and structural parameters of the molecules studied and descriptors characterizing their structure were obtained on the basis of in vitro screening data. The influence of structure of the investigated substances as reflected by the parameters studied on the anticancer activity, was established.

The analysis of structure-anticancer and antiviral activity relationships for macrocyclic pyridinophanes and their analogues on the basis of 4D QSAR models (simplex representation of molecular structure).

A new 4D-QSAR approach has been considered. For all investigated molecules the 3D structural models have been created and the set of conformers (fourth dimension) have been used. Each conformer is represented as a system of different simplexes (tetratomic fragments of fixed structure, chirality and symmetry). The investigation of influence of molecular structure of macrocyclic pyridinophanes, their analogues and certain other compounds on anticancer and antiviral (anti-influenza, antiherpes and antiadenovirus) activity has been carried out by means of the 4D-QSAR. Statistic characteristics for QSAR of PLS (partial least squares) models are satisfactory (R = 0.92-0.97; CVR = 0.63-0.83). Molecular fragments increasing and decreasing biological activity were defined. This information may be useful for design, and direct synthesis of novel anticancer and antiviral agents.

[Studies on correspondence between catalytic activity and fluorescence spectra character of macrocyclic dioxotetramine complexes].

In this paper, the conventional fluorescence spectra and laser induced fluorescence spectra of a novel macrocyclic dioxotetramine, 1,4,7,10-tetraazacyclotridecane-11,13-dionato and its Cu(II), Co(II), Ni(II) complexes at different pH are studied. The results show that the complexes fluorescence quantum yields have the relation of phi CuL5 < phi CoL5 < phi NiL5. At the same time, the reasonable explanation is given. The correspondence between their catalytic activities and fluorescence spectra character as minic superoxide dismutase metalloenzynes are also analyzed. It has been found that the fluorescence quantum yield of CuL5 decreases when the pH gets higher. But it is on the contrary for CoL5 and NiL5. It has been inferred that catalytic activities may be CuL5 > CoL5 > NiL5 according to their fluorescence quantum yield.

Sample preparation for determination of macrocyclic lactone mycotoxins in fish tissue, based on on-line matrix solid-phase dispersion and solid-phase extraction cleanup followed by liquid chromatography/tandem mass spectrometry.

A new method based on matrix solid-phase dispersion (MSPD) on-line with a solid-phase extraction (SPE) cleanup process followed by liquid chromatography with tandem mass spectrometry (LC/MS/MS) is presented for the determination of 3 macrocyclic lactone mycotoxins in fish tissues: zearalenone, alpha-zearalenol, and beta-zearalenol. The sample was prepared in a device that used a reversed-phase material (C18) or a normal-phase material (neutral alumina) as a matrix dispersing agent, and a graphitized carbon black cartridge was used for sequential cleanup by SPE. LC/MS/MS was used for selective determination. Isocratic elution with acetonitrile-methanol-water was used for LC separation; for MS/MS, 2 types of interfaces (a pneumatically assisted electrospray ionization interface or an atmospheric pressure chemical ionization interface) were evaluated and compared in terms of the intensity of the total ion current produced by each analyte. The use of highly selective MSPD on-line with SPE for sample preparation before analysis allowed the removal of interfering matrix compounds present in tissue extracts that would otherwise cause severe ionization suppression of zearalenone and its metabolites during the ionization process. Average recoveries at 100 ng/g were between 83 and 103% with C18 and > or = 67% with neutral alumina; the relative standard deviations were < 11% with C18 and < 18% with alumina. The limits of detection ranged from 0.1 to 1.0 ng/g. Sample preparation is simple to perform, no special technical equipment is required, and solvent volumes are minimal.

Selective separations of peptides with sequence deletions, single amino acid polymorphisms, and/or epimeric centers using macrocyclic glycopeptide liquid chromatography stationary phases.

Separating closely related peptides (those differing by one or two amino acids or the chirality of a single amino acid) can be challenging using reversed-phase liquid chromatography (LC), ion-exchange LC, or using ion-pairing agents. Also, the mobile phases that give the best separations in these modes may not be electrospray ionization mass spectrometry (ESI-MS) compatible. Forty-two peptides from 11 peptide families were separated on three macrocyclic glycopeptide stationary phases in reverse-phase mode using ESI-MS-compatible mobile phases. The peptide classes studied were angiotensin, bradykinin, alpha-bag cell factor, beta,gamma-bag cell factor, beta-casomorphin, dynorphin, enkephalin, leucokinin, lutinizing hormone releasing hormone, neurotinsin, substance P, and vasopressin. High selectivity was observed for single amino acid substitutions (achiral and chiral) regardless of the position of the substitution in the sequence. Mobile phase optimization, its effect on peptide elution behavior, and chromatographic efficiency is also discussed. Using LC-ESI-MS, a 2 ng limit of detection was obtained, two orders of magnitude lower than the UV detection limit.

Macrocyclic polyesters. I. A novel class of 1,3-butandiol derivatives as potential antimicrobial agents.

This paper reports the synthesis of a novel class of macrocyclic tetraesters containing 1,3-butandiol sub-units by reacting the stannolan derivative of the diol with diacyl chloride. The structure of the possible isomers was assigned by spectroscopic data and comparison with samples otherwise prepared. Preliminary screening revealed that these compounds have low antimicrobial activity, although it is higher than that of the starting diol.
