[Nonprecipitability of the nucleic factor in the ultracentrifugation of tumor ascitic fluid].

Samples of cell-free ascitic liquid of mice and rats suffering from Ehrlich tumor and Zajdela's hepatoma, respectively, were centrifuged at 70,000 g and 200,000 g for 1 hour. Nucleic acid preparations were then isolated by phenol deproteinization and analysed by 8% polyacrylamide gel electrophoresis with subsequent densitometry. Under such conditions, DNA of nucleic factor as well as minor RNA fractions failed to precipitate. These results show that viral particles, tumor cell-produced vesicles and other large complexes do not incorporate DNA nucleic factor or minor RNA fractions.

Ultracentrifugal study of metabolic effects of probucol: plasma lipoproteins in patients with proven coronary artery disease.

The primary metabolic effect of probucol is characterized by significant lowering of plasma low-density lipoproteins (LDL2) (p less than 0.01), frequently connected with a decrease in concentration of very-low density lipoproteins (VLDL) (p less than 0.05). This main effect is correlated (r = 0.84-0.75) with a decrease of chylomicrons (CHY) and macromolecular very, very-low density lipoproteins (VVLDL). High density lipoproteins (HDL2,3) tended to decrease in the majority of cases, although a restoration of their levels was observed after the therapy. The strong significance of probucol's lowering action of LDL2 does not statistically relate to any other metabolic phenomenon, confirming its relative independence on remaining pathways.

Radioimmunoassay of human thyroglobulin with use of "thyroglobulin-free" plasma prepared by ultracentrifugation as diluent.

We describe the preparation and evaluation of a new diluent for use in preparing standard curves for the radioimmunoassay of human thyroglobulin (TG). This diluent, "TG-free human plasma, allows measurement of TG in lower concentrations than with previous assay systems because the components of the standards are more similar to those of the samples. TG-free plasma was prepared by a modification of the differential centrifugation method of Edelhoch (J. Biol. Chem. 235:1326, 1960). Standard curves prepared by use of TG-free plasma and plasma obtained from thyroidectomized patients were identical. In our assay, plasma from patients who had undergone total thyroidectomy produced no displacement when 25 to 100 microL of plasma was assayed. Twenty-five normal subjects all had detectable TG concentrations, ranging from 4.0 to 25.7 microgram/L. Only 27 of 2000 determinations performed by this new method had undetectable values and 26 of these were from patients with total or near-total thyroidectomies. TG concentration determined by this assay may be more sensitive for finding recurrent thyroid malignancy.

[Concentrated purified vaccine against tick-borne encephalitis prepared by means of zonal ultracentrifugation. Development of the preparation].

A concentrated and purified (lyophilized) preparation with a high immunological activity was obtained from formalin-inactivated tissue culture suspensions of tick-borne encephalitis (TBE) virus. Its high immunogenicity was demonstrated in mouse protection tests against fatal challenge with TBE and in the studies of antibody production in monkeys of two species. The preparation was shown to contain no infectious TBE virus, to be safe for small laboratory animals and monkeys. Trials of the preparation in 19 volunteers demonstrated it to be well tolerated and to induce virus-neutralizing antibody production in man. It also sensitized lymphoid cells when inoculated subcutaneously with Al2O3. These experimental materials indicate the possibility of developing a highly potent vaccine against tick-borne encephalitis by concentration and purification of TBE particle suspensions.
