Influence of cholinergic and adrenergic blocking drugs on hyperglycemia and brain glycogenolysis in diazinon-treated animals.

Diazinon, an organophosphorous compound, produced hyperglycemia and reduced the glycogen content of the brain 2 h after its administration to rats (40 mg/kg, i.p.). The activities of the glycogenolytic enzymes, glycogen phosphorylase and phosphoglucomutase, were significantly increased, while that of glucose-6-phosphatase was not altered. Atropine (20 mg/kg, i.p.) given immediately after diazinon abolished the changes; tolazoline or propranolol (each at 10 mg/kg, i.p.) injected 30 min before the administration of diazinon significantly reduced the hyperglycemia and the increase in brain glycogenolysis. A combination of tolazoline and propranolol was more effective than either of them alone and completely abolished the hyperglycemia and the changes in brain glycogenolysis. It may be concluded that diazinon initially activates central cholinergic processes leading to hyperglycemia and increased cerebral glycogenolysis in animals.

Enzyme activities and maturation in unstimulated and exogenous gonadotropin-stimulated human oocytes.

With the advent of new techniques of human in vitro fertilization (IVF), identifying parameters of oocyte quality to allow selection of those most likely to fertilize becomes crucial. Morphology of oocytes, which correlates positively with biological performance, is the currently utilized classification criterion. However, biological links between form and function are tenuous, and underlying mechanisms remain elusive. We investigated whether biochemical activation is quantitatively associated with the stages of maturation in ova obtained from patients undergoing gynecologic surgery during unstimulated cycles and women undergoing IVF after exogenous gonadotropin stimulation. Changes in selected enzymes from protein, lipid, and carbohydrate metabolism (hexokinase, phosphoglucomutase, glycogen synthetase, uridine diphosphoglucose pyrophosphorylase, glucose-6-phosphate dehydrogenase, cytosolic thiolase, beta-hydroxyacyl-CoA dehydrogenase, alanine aminotransferase, and aspartate aminotransferase) were determined simultaneously, in individual oocytes, utilizing a highly sensitive biochemical methodology. Several enzyme activities paralleled maturation grade and were higher in stimulated oocytes after correction for grade. These biochemical findings quantify metabolic and functional changes that increase as ova mature, possibly contributing to their reproductive performance.

Renal metabolism of calcitonin.

The kidneys account for approximately two-thirds of the metabolism of calcitonin, but relatively little is known regarding the details thereof. To further characterize this process, we examined the renal handling and metabolism of human calcitonin (hCT) by the isolated perfused rat kidney. We also studied the degradation of radiolabeled salmon calcitonin (sCT) by subcellular fractions prepared from isolated rabbit proximal tubules. The total renal (organ) clearance of immunoreactive hCT by the isolated kidney was 1.96 +/- 0.18 ml/min. This was independent of the perfusate total calcium concentration from 5.5 to 10.2 mg/dl. Total renal clearance exceeded the glomerular filtration rate (GFR, 0.68 +/- 0.05 ml/min), indicating filtration-independent removal. Urinary calcitonin clearance as a fraction of GFR averaged 2.6%. Gel filtration chromatography of medium from isolated kidneys perfused with 125I-labeled sCT showed the principal degradation products to be low molecular weight forms eluting with monoiodotyrosine. Intermediate size products were not detected. In the subcellular fractionation experiments, when carried out at pH 5.0, calcitonin hydrolysis exclusively followed the activities of the lysosomal enzyme N-acetyl-beta-glucosaminidase. Typically, at pH 7.5, 42% of total degradation occurred in the region of the brush-border enzyme alanyl aminopeptidase and 29% occurred in the region of the cytosolic enzyme phosphoglucomutase. Although 9% of the calcitonin-degrading activity was associated with basolateral membrane fractions, most of this activity could be accounted for by the presence of brush-border membranes.(ABSTRACT TRUNCATED AT 250 WORDS)

Chromosome localization of three syntenic gene pairs in the American mink (Mustela vison).

Twenty eight American mink X Chinese hamster somatic cell hybrids were analysed for the expression of mink enzymes and for mink chromosomes. The results of this analysis made it possible to assign the genes for phosphoglucomutase-1 and phosphogluconate dehydrogenase to chromosome 2, those for lactate dehydrogenase A and glucose phosphate isomerase to chromosome 7, and those for lactate dehydrogenase B and triosephosphate isomerase to chromosome 9.

An analysis of red cell enzymatic markers in the province of Bologna (Italy).

About 280 unrelated individuals living in the province of Bologna (Northern Italy) have been studied for the following red cell enzymatic markers: phosphoglucomutase (PGM), adenylate kinase (AK), adenosine deaminase (ADA) and phosphohexose isomerase (PHI). 116 subjects from the same sample have also been analysed for red cell acid phosphatase (ACP). The observed gene frequencies are PGM21 = 0.280; AK2 = 0.030; ADA2 = 0.091; ACPa = 0.297; ACPb = 0.647; ACPc = 0.056. In the PHI system two individuals with the variant PHI 3-1 phenotype have been found.

Plasma protein and red cell enzyme groups among the Nepalese.

Nepalese blood samples were tested for two plasma proteins and seven red cell enzyme systems. Polymorphic variation was present for the haptoglobin protein system and for the acid phosphatase, phosphoglucomutase locus 1, adenylate kinase and 6-phosphogluconate dehydrogenase loci controlling red cell enzyme systems. Gene frequencies are compared between the eastern and western Nepalese tribes, and with the distribution pattern known in other South and South-East Asian population groups. The possible interpretations of these observations are discussed.

Phophoglucomutase first locus polymorphism as revealed by isoelectric focusing in Southern Africa.

Eleven Southern African populations (representing European, Asian and Negroid populations) have been typed for the first locus phosphoglucomutase (PGM1) using isoelectric focusing (pH range 5.0-8.0) in acrylamide gels. The gene frequencies of the four common alleles at this locus in these populations were compared to those found previously in European and Negroid populations. Marked differences in gene frequencies were observed: Negroes have a lower PGM1(2-) compared with Caucasoids due to a lower PGM1(2-) frequency, Indians a relatively high PGM1(2) due to a higher frequency of the PGM1(2+) allele. The Afrikaans and Ashkenazim do not differ appreciably from their European counterparts. The appearances of the rarer PGM1(6) and PGM1(7) alleles on isoelectric focusing are described and some kinetic properties examined. The PGM2(2-1), or 'Atkinson' phenotype, can also be detected with this technique.

Blood group, red cell enzyme and serum protein types in the Buka Islanders, Papua New Guinea.

Genetic marker studies on a sample of 80 speakers of the Petats and Tinputs families of languages, all pupils at a single high school, indicate a homogeneity among them which can be extrapolated to their areas of origin. Buka and its offshore islands and the northern part of Bougainville Island in the North Solomons Province of Papua New Guinea. Several markers systems, most notably first-locus phosphoglucomutase and liver acetyltransferase, reinforce the morphological evidence that these peoples are quite distinct from most other Papua New Guinea populations, with whom, however, there has been some gene exchange, probably through East New Britain. Their principal affinities are with the peoples of the Solomon Islands to the south.

Isoelectric focusing studies of human red cell PGM1 in Japanese, with special reference to the characterization of PGM17.

The distribution of phosphoglucomutase (PGM1) subtypes in human red cells was determined by isoelectric focusing in 218 Japanese samples. Nine common phenotypes were observed corresponding to the following frequencies of the four alleles at the PGM1 locus: PGM11+ 0.6560, PGM11- 0.1170, PGM12+ 0.1674 and PGM12- 0.0505. In addition, a characterization of the PGM17 allele was performed. Our results obtained in the present study revealed the possibility that the PGM17 allele may be differentiated in the two alleles of PGM17+ and PGM17- through an investigation of isoelectric focusing.

Distribution of polymorphic traits in Mazandaranian and Guilanian in Iran.

In 1972, over 2,000 blood specimens were obtained from two Iranian populations, Mazandaranians and Guilanians, who live on the southern coast of the Caspian Sea. All specimens were screened for hypocatalasemics and randomly selected samples were analyzed for several blood groups (A1A2BO, MNSs, Rh, P, Fy, K, Di, Jk, Lu, Nya), red cell enzyme systems (glucose-6-phosphate dehydrogenase, phosphoglucomutase, acid phosphatase) and serum protein groups (Hp, Tf). The genetic constitution of these Iranian populations was described, and the genetic relationship to Mongolians and Caucasians was discussed.

Blood genetic markers in Bengali Muslims of Bangladesh.

Serum protein (albumin, haptoglobin, ceruloplasmin, transferrin and group-specific component), haemoglobin, and red cell enzyme (glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, acid phosphatase, esterase D, adenylate kinase, glyoxalase I, phosphoglucomutase, lactate dehydrogenase, malate dehydrogenase, phosphohexose isomerase and superoxide dismutase) polymorphisms were studied among the Bengali Muslims of Bangladesh. In general, the gene frequencies of the polymorphic systems were similar to those in West Bengal and Assam. There appears to be a relatively strong Mongoloid influence in the present population as evidenced by the presence of HbE and TfDChi, higher frequencies of Hp1 and GcIF, and a lower AK2 frequency.

AK, PGM1 and 6PGD polymorphisms in Central Sardinia.

Adenylate kinase (AK), phosphoglucomutase (PGM1) and 6-phosphogluconate dehydrogenase (6PGD) polymorphisms were investigated in a sample of individuals from Central Sardinia. The gene frequencies were: AK1 = 0.973, PGM1(1) = 0.842 and PGDA = 0.969. The frequencies were compared with those of other Italian populations.

A PGM1*1A variant with a reduced activity.

An adverse homozygosity at the phosphoglucomutase-1 (PGM1) locus was detected in a family by electrophoresis on starch gels. Isoelectric focusing on polyacrylamide gels could clearly demonstrate a faint band at the same position as that of PGM1 1A in the mother and the child. Further analysis of other family samples and densitometric evaluation of the stained bands revealed the genetic transmission of a variant allele with a reduced activity. The molecular basis for the low activity is still unknown.

Red cell polymorphisms in Sardinia.

Acid phosphatase (ACP1), esterase D (ESD) and phosphoglucomutase 1 (PGM1) polymorphisms have been studied in Sardinia and the following gene frequencies have been found: ACP1*A = 0.235, ACP1*B = 0.684 and ACP1*C = 0.081; ESD*2 = 0.118 and PGM1*2 = 0.233. These findings confirm the genetic uniqueness of Sardinians compared to the other Italian and European populations.

Red-cell enzyme polymorphisms in Ibiza (Balearic Islands, Spain).

A sample of 390 individuals from Ibiza (Balearic islands, Spain) was studied for the red-cell enzyme polymorphisms: esterase D (ESD), acid phosphatase 1 (ACP1), glyoxalase I (GLO), 6-phosphogluconate dehydrogenase (PGD), phosphoglucomutase 1 (PGM1) and adenylate kinase 1 (AK1). The following allele frequencies were observed: ESD*2 = 0.181; ACP*A = 0.249; ACP*B = 0.719; ACP*C = 0.032; GLO*2 = 0.556; PGD*C = 0.018; PGM1*2 = 0.235, and AK1*2 = 0.032. These results were compared with those reported from other Balearic populations. There was a heterogeneity between island samples in the ESD and PGD systems. In spite of this, the whole Balearic population could be differentiated from most European and circum-Mediterranean populations.

Diversity of some gene frequencies in European and Asian populations. VI. Geographic patterns of PGM and ACP.

Genetic variation at the phosphoglucomutase-1 (PGM) and acid phosphatase (ACP) loci, in 160 and 120 European and Asian populations, respectively, is described by spatial autocorrelation statistics and directional variograms. Short-distance patterns of gene frequencies correspond to those predicted by models of isolation by distance, but long-range differentiation of populations is observed as well. A possible role of climatic selection in maintaining the PGM polymorphism is supported by the north-south orientation of the gradient for that locus, but not by biochemical evidence. By and large, the observed patterns of gene frequencies seem to reflect a combination of demographic processes, subdivision and isolation of local populations among them.
