The role of the lymphatic system and its specific growth factor, vascular endothelial growth factor C, for lymphogenic metastasis in prostate cancer.

OBJECTIVE: To compare prostate carcinoma, with and with no lymph node metastasis, to benign prostatic hyperplasia (BPH) tissue for lymphatic vessel density (LVD) and the expression of the lymph-endothelial specific growth factor, vascular endothelial growth factor C (VEGF-C), to determine their role in lymphogenic metastasis. PATIENTS, MATERIALS AND METHODS: Lymphatic vessels were stained using lymphatic vessel endothelial hyaluronan receptor 1 and assessed in standard areas. The expression of VEGF-C was assessed by the number of positive epithelial cells. The data were compared with the clinical staging. RESULTS: The lowest LVD was found in tumorous areas as opposed to periphery and nontumorous tissue (P = 0.007; P < 0.001). The highest LVD was in BPH tissue (P < 0.001). There was no correlation with clinical staging. There was more VEGF-C staining in pN1 than in pN0 and in BPH specimens (P = 0.002). CONCLUSION: LVD is not a prognostic variable for the process of lymphogenic metastasis in prostate cancer. VEGF-C is up-regulated in prostate cancer and its correlation with lymph node status suggests a role for the development of lymph node metastasis, e.g. via an increased permeability of lymphatic vessels.

The drainage of cerebrospinal fluid into the lymphatic system of the neck in humans.

A histo- and immunohistochemical examination of the superior deep cervical lymph nodes was performed in a group of 19 patients who died from intracerebral haemorrhage. For comparison two control groups without cerebral lesions were studied (n = 17, n = 13). Free iron deposits as shown within the lymph nodes by Prussian blue reaction were seen significantly more often following intracerebral haemorrhage than without bleeding. The expression of CD 68 (a marker for macrophages) or CR 3/43 (microglia) gave a strong reaction, but there are no significant differences between the three groups. Glial fibrillary acidic protein was rarely seen in the three groups. Raising the CSF pressure results in increased CSF drainage into the cervical lymph nodes. The clinical importance of this lymphatic drainage suggests a communication between the brain and the immune system of the head and neck, with a possible selective up- and down-regulation.

Route from the paraaortic lymphatic system to the tracheobronchial lymph nodes evidenced on lymphangiogram in a patient with gastric cancer.

Lung metastasis from gastric or colonic cancer, without liver metastasis, is seldom seen. However, its metastatic pathway has not been delineated on imaging. We present and discuss such a lymphatic route in a patient with gastric cancer. The appearance of the mediastinum on a lymphangiogram is discussed. To the best of our knowledge, the lymphangiographic demonstration of the route from the paraaortic lymphatics to the tracheobronchial lymph nodes has been not reported. Lymphatics running from the paraaortic to the tracheobronchial lymph nodes through the diaphragm may play an important part in direct metastasizing to lung or tracheobronchial lymph nodes in certain patients.

Replication and persistence of murine herpesvirus 72 in lymphatic system and peripheral blood mononuclear cells of Balb/C mice.

After intranasal inoculation of murine herpesvirus 72 (MHV-72) to Balb/c mice the virus persisted in adherent lung mononuclear cells (AMC). Infectious virus was occasionally detected during a period of eight months after infection in lymphatic organs (thymus, spleen, lymph nodes), bone marrow, alveolar and peritoneal AMC, and lymphocytes and macrophages of peripheral blood by indirect immunofluorescence and cocultivation with permissive VERO cells. Mouse B-cell lines NS0 and SP2/0 were permissive for MHV-72 infection when inoculated in vitro. Virus-neutralizing antibodies have been detected during one year period in the sera of infected mice, and their levels correlated with activation of the latent MHV-72.
