Cyclic nucleotide independent casein kinase (G type) in bovine adrenal cortex: purification and properties of two molecular forms.

Two soluble cyclic nucleotide independent protein kinase (ATP: protein-phosphotransferase, EC 2.7.1.37) activities have been purified from bovine adrenal cortex cytosol. Both purified enzymes exhibit the best affinity for acidic substrates such as casein and can use GTP as well as ATP as phosphoryl donor. They can thus be classified as casein kinase of the G type as previously proposed (Cochet C. et al., (1980) Endocrinology 106, 750-757). Whereas the two moieties could be separated using their different affinities toward a phosphocellulose resin, both purified enzymes appeared indistinguishable on the basis of several molecular and catalytic properties. Both G type casein kinase moieties have an identical sedimentation behavior (5.5 S in the presence of 0.5 M NaCl), yield similar patterns upon electrophoresis under denaturing conditions with three major protein components (42 000, 38 000 and 27 000), and show an ability to undergo self-phosphorylation mostly on the 27 000 component. Both enzymes have the same protein and nucleotide (ATP and GTP) substrate specificity, show similar increases in activity in the presence of polyamines and Mg2+ (optimum at 50 mM) and similar inhibition by NaCl above 0.2 M. The only difference between the two forms of casein kinase (i.e., affinity for phosphocellulose) could not be explained by a different degree of self-phosphorylation or by a limited proteolytic process during handling and purification. These results suggest that the two active moieties may represent isoenzymatic forms of the G type casein kinase activity in bovine adrenal cortex cytosol.

Ultrastructural changes of the adrenal cortex in Cushing's syndrome treated with aminoglutethimide (Elipten Ciba)

The main ultrastructural feature of human adrenal cortices following administration of aminoglutethimide (Elipten CIBA), a steroid biosynthesis blocking agent, is a striking intracellular accumulation of lipids, also seen in the light microscopy. The lipids accumulate in the adrenocortical cells but are also stored in the cytoplasm of pericapillary histiocytes. The lipids are present in the adrenocortical cells mostly as rounded liposomes of variable size, whereas those in the histiocytes usually show up as an amorphous debris with abundant myelin figures and cholesterol crystals. In some areas of the adrenal cortex activated histiocytes changing into lipophages may become the prevalent cell. They also take the place of destroyed adrenocortical cells and can form compact cell aggregates of variable size or even columnar strands between neighbouring capillaries, thus resembling columns of adrenocortical cells. In some of the adrenocortical cells focal cytoplasmic degradation, turning into coagulation necrosis, was observed. In others initial cytolysis was found. In contrast with earlier findings in experimental animals, mitochondrial changes in humans were not a prominent feature. The only changes occasionally observed were a focal disappearance of the vesicular cristae and a concomitant fibrillary loosening of the mitochondrial matrix.

[Functional morphology of the adrenal cortex and incretory kidney structures in the salt-losing form of adrenogenital syndrome in children].

The adrenals and kidney incretory structures (the juxtaglomerular apparatuses and renomedullary interstitial cells) were studied in 12 cases of a desalinization form of the adrenogenital syndrome in children who died at the age of 1-6 mos of acute water-electrolytic disturbances in order to assess function of the structures involved in the regulation of the water-salt equilibrium. The adrenals and kidneys from 7 children of the same age who died of mechanical asphyxia, were taken as controls. The depth of adrenocortical zones, volumes of nuclei and nucleoli in different cortical zones were determined; the juxtaglomerular index, cell count (including vacuolized cells), the area of the juxtaglomerular apparatus and mesangium were determined in the juxtaglomerular apparatus. The amount of cell lipid granules (the interstitial-cellular index), the mean granule volume and the total granule volume were counted in renomedullary interstitial cells. The results were statistically processed, and a paired correlation analysis of all the studied parameters was performed. In the desalinization form of the adrenogenital syndrome morphometric investigation of the glomerular and fascicular zones revealed compensatory processes, not of hyperplastic nature but mainly of hypertrophic nature; morphological and morphometric characteristics of the kidney incretory structures indicated tension of the renin-angiotensin system and probably a decrease in prostaglandin synthesis in salt and water loss. In the control group parameters of structures related to the regulation of the water-salt equilibrium, especially in the adrenal cortex, showed good correlation. In the desalinization form of the adrenogenital syndrome some other relationships between these structures developed.

Precocious breast development: a case of unilateral hyperplasia of the adrenal cortex.

The case of a 14-month-old girl presenting with precocious breast development due to adrenal hyperplasia is reported. The endocrine studies revealed a slight elevation of the plasma progesterone, 17 alpha-hydroxyprogesterone, testosterone and oestrone levels, and an increased urinary oestrogen excretion. The findings of the ultrasound examination and the evidence that the oestradiol concentration in the left adrenal vein was higher than in the right adrenal vein confirmed the diagnosis of left adrenal hyperfunction. Surgical exploration revealed unilateral hyperplasia of the adrenal cortex. After adrenalectomy the plasma hormone levels and urinary oestrogen excretion fell to normal, with concomitant regression of breast tissue. The girl shows no signs of recurrence of the disorder in a 2 year follow-up. This adrenal hyperplasia might be a benign adrenal disorder causing precocious breast development.

Level of cortisol and reactivity of adrenal cortex to exogenous ACTH at neonatal period in calves.

The studies were carried out on calves from 1st to 21st day of life. Cortisol level and reactivity of adrenal cortex to exogenous ACTH were analysed. The highest level of cortisol in blood was observed in first days of life in the calves. Then the level became considerably decreased. In the experimental group two subgroups differing in cortisol level were distinguished. High or low level occurred on the first day and the difference kept over two weeks of their life. Adrenal cortex just on the day of birth showed full functional maturity of molecular receptors binding ACTH.

Characterisation of endothelin-1-related protein in human adrenal cortex and in cortical lesions.

Endothelin (ET)-1 is a 21-amino acid peptide with potent vasopressor and vasoconstrictive properties. Biochemical studies suggest that this peptide occurs in the adrenal cortex, where it appears to influence steroid hormone production and catecholamine release. Concomitant with our previous immunohistological study, we found ET-1 immunoreactive (IR) cells in human adrenal cortex and cortical neoplasms, but not in the medulla. These ET-1 IR cells were numerous in adenomas, but were seen only occasionally in some of the carcinomas. In the present study, the ET-1 IR protein was extracted from normal (n = 5) and hyperplastic (n=3) human cortex as well as from cortical adenomas (n = 10) and carcinomas (n = 5). Its molecular weight, determined by SDS-polyacrylamide gel electrophoresis and immunoblotting, was 9 kD, which is lower than that of prepro-ET-1 (21 kD), but larger than that of pre-ET-1 (4.3 kD) and ET-1 (2.5 kD). The normal cortical specimens, hyperplasias, adenomas and three of the five carcinomas all contained this distinct band. The two carcinomas lacking it were associated with Conn's syndrome. The protein may constitute a protein not previously described, but further studies are needed to determine its complete structure.
