Modified Feulgen staining of DNA with aqueous solution of pinacyanol.

The paper reports on the use of a quinoline dye, pinacyanol, towards staining of acid hydrolysed DNA. The dye as an aqueous solution can be used after treatment of mammalian tissue sections in concentrated phosphoric acid at 5 degrees C for 20 min followed by hydrolysis in 6N HCl at room temperature for 15 min, for staining DNA-aldehyde molecules. It has also been observed that staining of DNA-phosphate groups is also possible in sections treated with cold concentrated phosphoric acid after selective extraction of RNA. Both in situ absorption characteristics of stained nuclei as well as in vitro absorption data of an aqueous solution of the dye have been presented. It has been suggested that staining DNA-aldehydes with pinacyanol, without any primary amino group in its molecules is due to a modified Feulgen reaction.

[Acardiac fetus].

Acardiac fetus is a rare lethal fetopathy usually encountered in monozygous pregnancies. Ultrasound prenatal diagnosis has enabled an increasing number of observations and raised the need for an adequate therapeutic approach since the spontaneous prognosis for the healthy twin is unfavorable in half of the cases. An acardiac fetus was identified at 12 weeks gestation in a 36-year-old woman. Growth of the healthy fetus was carefully monitored and progressed normally to delivery by cesarean section of a 2.900 kg boy at 36 weeks. At delivery, the acardiac fetus was found to be totally free of any attachment, floating in the remaining fluids. Pathology examination showed a 16 g macerated fetus with a cephalic extremity, a ventral pedicle and a syrenomelic caudal extremity. The caryotype was not significative. Acardiac fetus occurs in less than 1% of multiple pregnancies and can develop in single pregnancies. Twin reversed arterial perfusion has been recognized as necessary for development of the perfused fetus. Genetic and immunologic theories have been proposed to explain the pathogenesis which remains unknown. Clinical management depends on the spontaneous development of the acardiac fetus and the deleterious consequences for the healthy twin resulting from heart failure, anasarca or prematurity. Medical management with digoxin, or selective extraction by hysterotomy may improve prognosis but results have been variable. Echoguided umbilical cord ligation has also been proposed to improve maternal mortality. Therapeutic abstention is no longer indicated at prenatal diagnosis of an acardiac fetus and a healthy twin despite the risk of invasive treatment.

Specific radioimmunoassay for amitriptyline and nortriptyline in plasma.

A specific radioimmunoassay (RIA) for the determination in plasma of the widely used tricyclic antidepressant amitriptyline (AT) and its major metabolite nortriptyline (NT) has been developed employing 3H-AT as the radioligand and a rabbit antiserum to a bovine serum albumin conjugate of N-succinyl-nortriptyline. Although the antiserum cross-reacts almost equally well with AT and NT, specificity is achieved by selective extraction of each compound from plasma at a different pH. A unique aspect of the assay is that at no time during the entire extraction procedure is the AT or NT taken out of solution. Both compounds are back extracted from the organic phase into 0.1 N HC1 and the acid fraction subjected to RIA directly. The method has a limit of sensitivity of about 2 ng/ml using a 0.5 ml sample of plasma. Satisfactory agreement was obtained for plasma levels of AT and NT when determined by the RIA and a specific GC/MS procedure. The correlation coefficients were 0.89 and 0.98 for AT and NT, respectively. THE RIA has been used to measure steady-state levels of AT and NT in man after chronic administration of AT and following a single oral 75 mg dose. The method also lends itself for the specific determination of NT alone in subjects receiving therapeutic doses of NT.

Detection of breast tumor associated mucin epitope on CAMA cell line using monoclonal antibody G3F1 generated against HMFG membrane.

The expression of breast tumor associated mucin epitope on CAMA cell line was detected employing the mouse MAB G3F1 generated against HMFG membrane. Immunocytochemical studies revealed that MAB G3F1 strongly reacted with 85% of breast cancer tissue sections with specific staining of apical cell membrane of malignant epithelial cells. The mucin antigen recognised by MAB G3F1 was detected by selectively extracting high molecular weight glycoprotein antigen from HMFG membrane using lectin affinity chromatography and gel filtration chromatography. Immunoprecipitation studies revealed that MAB G3F1 recognized a high molecular weight glycoprotein with an approximate molecular weight of 300kd. The expression of MAB G3F1 reactive antigen on CAMA cells was detected by immunocytochemistry and by immumoprecipitating 300kd antigen from 125I labelled Tx100 solubilized extract of CAMA cells. The results from these investigations suggest that CAMA cells express MAB G3F1 reactive antigen with tumor associated epitope, similar to tumor associated mucin epitope of HMFG membrane.

Treatment planning: teeth versus implants.

Attempts to save natural teeth have created numerous restorative, endodontic, and periodontal techniques it has always been considered the treatment of choice to save teeth whenever possible, and extreme efforts to save teeth are often considered because of the poor treatment alternatives for replacing them. Today osseointegration provides a predictable means of replacing teeth; therefore, retaining questionable teeth may not be as necessary as in the past. This article reviews situations in which the selective extraction of compromised natural teeth or even healthy natural teeth is considered because of alternative treatment options utilizing osseointegrated implants.

Collagen subtype pattern in normal and varicose saphenous veins in humans.

Our present study was to determine the collagen subtype pattern in the greater saphenous vein of the lower limb, obtained from 21 normal (macroscopically and ultrastructurally non-varicose vein segments from non-varicose subjects) and 37 varicose subjects, and to compare affected (macroscopically and ultrastructurally varicose segments from varicose veins) vs. non-affected (macroscopically and ultrastructurally non-varicose segments from varicose veins) segments (16). After elastase pretreatment and partial pepsin digestion, types I, III & V collagens (CI, CIII, CV) were extracted selectively by differential salt precipitation and measured quantitatively in samples obtained from normal and varicose saphenous veins-either affected or unaffected segments. Significant elevations of water (p < 0.05) and collagen type I [CI] (p < 0.01) content in varicose veins (both affected and unaffected segments) as compared with normal saphenous veins were observed. The collagen type III (CIII) and collagen type V (CV) content of varicose veins were found to be slightly reduced as compared to normal veins and consequently the CI/(CIII+CV) ratio in varicose veins increased significantly (p < 0.02) as compared to normal veins. Elevation of the CI/(CIII+CV) ratio in varicose veins may cause considerable weakening of the venous wall, further supporting the "weak wall" theory of varicose vein etiology.

Biliary mucin secreted by cultured human gallbladder epithelial cells carries the epitope of CA 19-9.

Serum CA 19-9 is increased in patients with different gastrointestinal malignancies. Unfortunately, CA 19-9 is also detected in high concentrations in normal bile causing unspecific serum elevations during inflammatory disease of the biliary tract and cholestasis. In order to identify the source of CA 19-9 in bile, the capacity of cultured human gallbladder epithelial cells (HGBEC) to secrete CA 19-9 was investigated. Cells were harvested from gallbladders removed by cholecystectomy and cultured for up to 14 days in collagen I coated 24-well culture dishes. CA 19-9 was measured in the culture medium by a solid-phase CA 19-9 EIA (Boehringer). In addition, culture medium was separated by Sepharose 4B-Cl, Concanavalin-A (Con-A) and CA 19-9 affinity chromatography. Significant CA 19-9 activity was measured in the culture medium after a 24 hour incubation period. Following separation of the culture medium by Sepharose 4B-Cl and Con-A affinity chromatography, 90% of the CA 19-9 activity was recovered in the exclusion volume (> 2000 kDa) from which 90% were identified as Con-A negative. A close correlation was found between CA 19-9 and concentrations of mucin purified from human gallbladder bile. Furthermore, CA 19-9 affinity chromatography selectively extracted mucins from the culture medium of HGBEC. Finally, addition of the mucin secretagogue bethanechol (6 mM) to the culture medium increased CA 19-9 activity in the medium. In conclusion, normal HGBEC secrete mucins carrying the epitope of CA 19-9. During inflammatory biliary disease unspecific elevation of CA 19-9 in serum may reflect both inflammatory hypersecretion and leakage of biliary mucins into serum.

[Conditions for isolation of cholesterol oxidase from Actinomyces lavendulae mycelia].

The isolation conditions, composition and certain properties of cholesterol oxidase from the mycelium Actinomyces lavendulae were studied. Selective extraction of the enzyme occurred upon the mycelium extraction by the Tween-80 containing buffer. The specific activity of cholesterol oxidase thus obtained exceeded seven-fold that of the enzyme isolated via other methods. The optimal concentration of the detergent upon extraction was 0.15%. By disc-electrophoresis it was demonstrated that the preparation isolated from disrupted cells contained, in addition to cholesterol oxidase, 16 other protein components that had no enzymatic activity whereas the Tween-80 extracted preparation included no more than 7 fractions. The study of the relationship between activity of the resultant cholesterol oxidase and the substrate concentration and pH demonstrated that optimal conditions for the enzyme action were at pH 7.0-7.5 and cholesterol oxidase concentration of 0.0316 mM.

[Isolation of cationic proteins from rabbit thrombocytes and study of their bacteriostatic activity].

Cation-containing proteins, which possessed the bacteriostatic activity against Grampositive microorganism Bacillus subtilis SHGW, were isolated from rabbit thrombocytes by means of selective extraction. These substances were of heterogenous nature (as shown by polyacrylamide gel disc electrophoresis), including two groups of proteins with molecular weight about 8=10-10(3) and 20=30-10(3) daltons. The bacteriostatic activity of each group of these proteins was studied after gel filtration of the total preparation on Sephadex G=75. The group of proteins with molecular weight 8=10-10(3) daltons was responsible for bacteriostatic activity of the total cation-containing protein from thrombocytes.

[Nuclear matrix-lamina-intermediate filament system in PtK 2 cells].

Selective extraction, whole mount cell preparation and DGD embeddment-free section were involved in visualizing the nuclear matrix-lamina-intermediate filament system in PtK 2 cells. After extraction the anaphase chromosome residues adjoined to the cytoplasmic intermediate filament in some areas. Immunofluorescent staining showed us that the intermediate filament reacted with AE 1 and AE 3; McAb 223 could be localized specifically on the lamina while McAb C 23 could crossreact with cytoplasmic intermediate filament beside the lamina location, monoclonal antibody against lamin A (C) could also bind to chromosome residues. Antibody to 280 kD nuclear matrix protein which were positive stained in HeLa cells could not react with the nuclear matrix components of PtK 2 cells. 2-D electrophoresis demonstrated that there were some differences in the composition of the nuclear matrix-lamina-intermediate filament system of HeLa and PtK 2 cells. TdR treatment could lead to alteration of nuclear matrix proteins.

Different patterns of p53 mutations in prostatic intraepithelial neoplasia and concurrent carcinoma: analysis of microdissected specimens.

Prostatic intraepithelial neoplasia (PIN) is characterized by intraluminal proliferation of epithelial cells and can be divided into high-grade (HGPIN) and low-grade (LGPIN) lesions. HGPIN is regarded as the most likely precursor of prostatic carcinoma (PCA). Microdissected DNA selectively extracted from paraffin-embedded sections of 29 cases of PCA and 1 benign prostatic hypertrophy were analyzed for p53 mutation by single-strand conformation polymorphism (SSCP) of polymerase chain reaction (PCR)-amplified DNA fragments followed by direct sequencing. These patients had received total prostatectomy (27 cases) or transurethral resection (3 cases). Under direct microscopic observation, DNA was microdissected from 108 lesions: 67 lesions from 22 cases of PIN (55 HGPIN and 12 LGPIN), 29 from 22 cases of PCA, and 12 from 11 cases of adjoining benign glands. Analysis revealed 13 mutations in 10 lesions from six cases. All 13 were point mutations: 7 missense, 5 silent, and 1 nonsense. Mutations were detected in 3 cases (14%) of PIN and 5 cases (25%) of PCA. PIN lesions with p53 mutations were all categorized as HGPIN. Neither LGPIN nor benign glands adjoining PIN and/or PCA had mutations. Two PIN and one PCA lesion in each of two cases had mutations that were different from each other. G-to-A transition was the commonest mutation pattern. The current findings showed that HGPIN, but not LGPIN, and PCA are similar with regard to p53 mutation. The diverse patterns of p53 mutation among HGPIN and PCA lesions suggested multiclonal development of prostatic precancerous lesions.

Impermeant maleimides. Oriented probes of erythrocyte membrane proteins.

Maleimides impermeant to human erythrocyte membranes have been synthesized and applied to studies of the sulfhydryl groups of the membrane. Reaction of radioactive dextran-maleimide and glutathione-maleimide with either intact erythrocytes or ghosts yields sulfhydryl titers for the outer (exofacial) and inner (endofacial) surfaces, respectively, of 1.5 to 1.7 and 27 to 28 amol/cell. Corresponding values for sulfhydryl groups within the membrane interior, as estimated with radioactive N-ethylmaleimide, are 16 to 22 amol/cell. After exofacial labeling of intact cells with [35S]glutathione-maleimide, autoradiography of sodium dodecyl sulfate-polyacrylamide gels demonstrates four bands (alpha, beta, gamma, and delta) containing, respectively, 13%, 63%, 11%, and 13% of the radioactivity. The major beta-band corresponds in position to polypeptides of molecular weight 40,000 to 70,000 and to Coomassie brilliant blue-stained Band 5. Selective extraction demonstrates that the major Band 5 protein is not identical with the labeled beta-band polypeptides. Following endofacial labeling of ghosts with [35S]glutathione-maleimide, autoradiography reveals radioactivity in all of the major Coomassie brilliant blue bands. The impermeant maleimides described are also applicable to studies of discrete functional proteins of the erythrocyte membrane, including the hexose transport mechanism and the major Rho antigenic site.

Rapid and specific RIA of serum estrone sulfate with selective solid phase extraction.

The methods commonly used to evaluate conjugated steroids require hydrolysis and chromatographic purification. To avoid these steps, a simple method involving selective solid phase extraction and RIA using a highly specific antiserum for estrone sulfate (E1S) has been evolved. A Bond-Elut C2 cartridge was used for solid phase extraction of estrone (E1) and E1S; recoveries were 80 and 90% respectively. The intra- and inter assay precision of the assay at 3 serum levels, were 6.5, 10.4 and 4.4 and 12.7, 13.9 and 7.4% respectively. Accuracy, tested by linearity and recovery tests, was acceptable. A good correlation exists between a conventional enzymatic method and the proposed method. The latter is less time consuming and more reliable, thus providing a rapid assay to evaluate E1 and E1S in the same serum sample.

Tumour grading from magnetic resonance spectroscopy: a comparison of feature extraction with variable selection.

Magnetic resonance spectroscopy (MRS) provides a non-invasive measurement of the biochemistry of living tissue. However, signal variation due to tissue heterogeneity causes considerable mixing between different disease categories, making accurate class assignments difficult. This paper compares a systematic methodology for classifier design using multivariate bayesian variable selection (MBVS), with one based on feature extraction using independent component analysis (ICA). We illustrate the methodology and assess the classification performance using a data set comprising 41 magnetic resonance spectra acquired in vivo from two grades of brain tumour, namely low- and medium-grade astrocytic tumours, labelled astrocytomas (AST), and high-grade gliomas and glioblastomas labelled glioblastomas (GL). The aim of this study is threefold. First, to describe the application of the alternative methodologies to MRS, then to benchmark their classification performance, and finally to interpret the classification models in terms of biologically relevant signals derived from the spectra. The classification performance is assessed using the bootstrap method and by application to a test sample in a retrospective study.

In vitro binding experiments with a Valerian, hops and their fixed combination extract (Ze91019) to selected central nervous system receptors.

The fixed valerian-hops extract combination Ze91019 is used as a sleep aid. Although its exact mechanism of action is not well understood, earlier studies indicate that the CNS effect of valerian might occur through interaction with the GABA, melatonin and/or the adenosine systems in the brain. The use of hops in sleep remedies, however, is mainly based on traditional use and scarce scientific information. In this report, the binding of Ze91019, and the component valerian and hops extracts within, was tested on 14 subtypes of five classes of central receptors (dopamine, serotonin, melatonin, MCH and neuropeptide-Y). Binding affinities could be demonstrated at some of the screened melatonin (ML1 and ML2) and serotonin (5-HT4e, 5-HT6 and 5-HT7) receptor subtypes.

Introducing selective supercritical fluid extraction as a new tool for determining sorption/desorption behavior and bioavailability of persistent organic pollutants in sediment.

This review article intends to introduce the possibility of utilizing selective supercritical fluid extraction (SFE) as a tool to study sorption/desorption processes and bioavailability of persistent organic pollutants (POP) in sediment. Sorption/desorption behavior and bioavailability studies of POPs is a large research area, but still many unsolved problems exists. Therefore novel approaches to investigate mechanistic behavior of POPs in sediments are needed. Present literature on SFE points to the fact that selective SFE measurements can improve our knowledge, and recent investigations have been performed that demonstrate this. Results obtained with selective SFE can be connected to desorption of POPs in sediments under natural conditions in aquatic ecosytems. The ultimate goal is to use selective SFE as a way to determine the bioavailable fraction present within a matrix. A few preliminary results are presented here which may serve as a starting point for future studies.

Determination of catechins and catechin gallates in tissues by liquid chromatography with coulometric array detection and selective solid phase extraction.

Catechins levels in organ tissues, particularly liver, determined by published methods are unexpectedly low, probably due to the release of oxidative enzymes, metal ions and reactive metabolites from tissue cells during homogenization and to the pro-oxidant effects of ascorbic acid during sample processing in the presence of metal ions. We describe a new method for simultaneous analysis of eight catechins in tissue: (+)-catechin (C), (-)-epicatechin (EC), (-)-gallocatechin (GC), (-)-epigallocatechin (EGC), (-)-catechin gallate (CG), (-)-epicatechin gallate (ECG), (-)-gallocatechin gallate (GCG) and (-)-epigallocatechin gallate (EGCG) (Fig. 1). The new extraction procedure utilized a methanol/ethylacetate/dithionite (2:1:3) mixture during homogenization for simultaneous enzyme precipitation and antioxidant protection. Selective solid phase extraction was used to remove most interfering bio-matrices. Reversed phase HPLC with CoulArray detection was used to determine the eight catechins simultaneously within 25 min. Good linearity (>0.9922) was obtained in the range 20-4000 ng/g. The coefficients of variance (CV) were less than 5%. Absolute recovery ranged from 62 to 96%, accuracy 92.5 +/- 4.5 to 104.9 +/- 6%. The detection limit was 5 ng/g. This method is capable for determining catechins in rat tissues of liver, brain, spleen, and kidney. The method is robust, reproducible, with high recovery, and has been validated for both in vitro and in vivo sample analysis.
