[Epidermal proliferation rate after skin expansion in the dog model].

The aim of this study was to evaluate the time course of the epidermal proliferation under the influence of skin expansion. Therefore 99 expanders were implanted in 18 beagles. The expansion times were between 2 hours and 4 months. Cells in the S-phase were marked by in vivo application of bromodesoxyuridine (BrdU). The tissue specimens were immunohistochemically stained and positive nuclei counted. The proliferation index increased markedly about 32 hours after the onset of the expansion and decreased afterwards slowly during the first week. Because the dog is the most appropriate species for experimental expansion studies (4,5) it can be assumed that skin expansion leads to a stimulation of the epidermal proliferation during the first few days also under clinical conditions. Nevertheless, the amount of skin that can be surgically used depends not only on the characteristics of the epidermal layer, but also on the dermis with its strong fibre system. Own studies published elsewhere (8) have shown that the dermal fibres react much slower to the expansion stimulus, so that even though there is an early epidermal proliferation increase new surgically applicable skin can only be produced by long-term expansion.

Advanced mercuric iodide detectors for X-ray microanalysis.

We first present a brief tutorial on Mercuric Iodide (HgI2) detectors and the intimately related topic of near-room temperature ultralow noise preamplifiers. This provides both a physical basis and technological perspective for the topics to follow. We next describe recent advances in HgI2 applications to x-ray microanalysis, including a space probe Scanning Electron Microscope (SEM), Synchrotron x-ray detectors, and energy dispersive detector arrays. As a result of this work, individual detectors can now operate stably for long periods in vacuum, detect soft x-rays to the oxygen K edge at 523 eV, or count at rates exceeding 2x10(5)/sec. The detector packages are small, lightweight, and use low power. Preliminary HgI2 detector arrays of 10 elements with 500eV resolution have also been constructed and operate stably. Finally, we discuss expected advances in HgI2 array technology, including improved resolution, vacuum operation, and the development of soft x-ray transparent encapsulants. Array capabilities include: large active areas, high (parallel) count rate capability and spatial sensitivity. We then consider areas of x-ray microanalysis where the application of such arrays would be advantageous, particularly including elemental microanalysis, via x-ray fluorescence spectroscopy, in both SEMs and in scanning x-ray microscopes. The necessity of high count rate capability as spatial resolution increases is given particular attention in this connection. Finally, we consider the possibility of Extended X-ray Absorption Fine Structure (EXAFS) studies on square micron sized areas, using detector arrays.

Safety and efficacy of 2% topical minoxidil in the management of male pattern baldness in Chinese.

Topic 2% minoxidil solution has been proved to be effective for hair regrowth in 30-40% of male pattern baldness in the United States and Europe. There was no report concerning topical minoxidil in Chinese patients, therefore, we selected 40 patients to investigate the efficacy and safety from December 1988 to January 1989. Those selected patients were consistent with male pattern baldness without systemic disease, and had never used topical hair regrowth agents. Patients applied topical medication 1 ml twice daily and were examined at 1 month intervals. Thirty two patients completed 6 to 12 months (average 9.6 months) of the study, moderate hair growth was found in 22% while minimal growth in 59% of the studied patients. All patients noted their shedding decrease after the application of drugs for 1 to 3 months. Scalp dryness and mild scaling appeared in 2 patients, but were tolerable. Before and after the trial, all patients had a chest roentgenogram, EKG, complete blood counts, urinalysis and blood chemistry studies, and no apparent abnormality was found. The efficacy of 2% minoxidil solution application in our study was less than that in the United States. which might be because some patients did not follow the application order regularly and a more strict evaluation standard was adopted in our series. However, the safety of no systemic adverse effect and the potential of inhibiting progressive shedding of hair were valuable as a topical drug.

The effect of the topical administration of bioactive glass on inflammatory markers of human experimental gingivitis.

Recent studies demonstrated that bioactive glass attenuated inflammatory reactions and bacterial growth in vitro. The aim of the present clinical study was to evaluate the effects of local bioactive glass-administration in vivo in subjects with experimental gingivitis. In each individual, contralateral teeth served as test and control over a 21-day non-hygiene (preventive phase) and a 7-day therapeutic phase. A 45S5 bioactive glass (10% solution) was applied daily (2 x 1 h) on the test teeth during the preventive and therapeutic phase of the study. Inflammation assessment was based on the plaque index record (PI), on the bleeding frequency (BOP) and the gingival crevicular fluid volume (GCF). Interleukin-1beta levels (IL-1beta) in the GCF were measured by ELISA. Bacterial deposits on teeth increased during the 21-days non-brushing period, with no difference in plaque accumulation between test and control sides. BOP levels were significantly reduced during the therapeutic phase by the application of bioactive glass. GCF-values showed a significant reduction on the test compared to control side during the preventive and therapeutic phase. IL-1beta counts decreased during the last 7 days of the study on the test side, but no significance was given. The topical application of 45S5 bioactive glass in humans with experimental gingivitis attenuated the clinical signs of inflammation, although the bacterial accumulation was not inhibited in this clinical trial.

[Genotoxic activity test of sodium fluoride in vitro].

The artificial drinking water fluoridization provided as a mass preventive measure against dental decay poses another substantial problem which is a possibility of genotoxic action. Up to this date, this matter remains still obscur and is discussed with more or less intensity from time to time. The mentioned fact supported authors to study the impact of short-term 24 hrs sodium fluoride (NaF) action in concentration range of 0-500 mg.1(-1) drinking water in the frame of so-called minimal testing set (analysis of chromosomal aberrations in human peripheral lymphocytes, Ames test). For the initial NaF concentration applied, the reference value of 1 mg per 1 liter artificial fluoridization was estimated. The use of Ames test with TA 98 and TA 100 Salmonella typhimurium (+/- S9) strains showed no significant increase in revertants responsible of NaF mutagenic activity in any of applied concentrations (0-1300 mg per 1 Petri dish; = 0-520 mg.l-1 converted to the basic supplementative dose). Cytogenetic analysis of peripheral lymphocytes showed more sensitivity than prototrofic salmonella test. Yet one order higher NaF concentration than its application norm 1 mg.l-1 (i.e. 11 mg.l-1) has induced the occurrence of 3.8% ABB after single addition for 24 hrs to the "healthy" blood cultivated in vitro at short-term. This accounts for a value close to the level of statistically significant difference with regard to the application norm recommended. This level has been even exceeded as for a total count of fragments and exchanged parts. Thus the two orders higher NaF concentration (110.0 mg.l-1) resulted in a strong increase of cells with chromosomal aberrations for all of indicators observed; e.g. 27.5% ABB. Based on literary sources, obtained results and properly experience in practical proceeding artificial fluoridation, the authors concluded that the latter is not adequate to the up-to-date status of knowledge. Besides of economical and technical problems, those scientific are mainly concerned with making doubtful the auto-presumed genotoxic inertness for chronic users of fluoridated drinking water. Author's opinion is that when necessarily provided, the artificial fluoridization of drinking water should be proceeded selectively (in accord with real requirements of an appropriated population group, its age structure and location), temporarily and with intermittent checkout of fluoridization application regimen. To conclude, authors recommend further observation with use of biological model situations in vivo.

Changes in aqueous flare and cells after mydriasis.

The influence of a commonly used short-acting mydriatic agent, tropicamide-phenylephrine, on aqueous flare intensity and cell number was studied both in postsurgical cases and normal subjects. Measurements with a laser flare-cell meter were carried out in 40 patients who had undergone planned extracapsular cataract extraction with posterior chamber intraocular lens implantation. On the second postoperative day, 20 patients received one drop of a combined solution of 0.5% tropicamide and 0.5% phenylephrine, and the remaining patients received no mydriatic agents. Flare-cell meter measurements in the non-mydriasis group were rather difficult, and measurements in 12 out of 20 (60%) cases were discontinued. In the mydriasis group, flare intensity became significantly lower 2 hours after the application (P less than 0.01) and continued to decrease throughout the rest of the measurement period. The cell counts tended to decrease throughout the period after mydriasis and showed significantly lower values compared to the initial value (P less than 0.01). It was considered that measurements with this instrument in postoperative cases are best performed between 30 minutes to 1 hour after the application of the short-acting mydriatic agents. In normal human eyes, the effects of 0.5% phenylephrine-0.5% tropicamide, 0.4% tropicamide, and 5% phenylephrine on aqueous flare and cells were tested separately. Aqueous flare intensity was significantly decreased by tropicamide-phenylephrine at 1, 1.5 and 2 hours (P less than 0.05), by tropicamide at 1 and 1.5 hours (P less than 0.01), and by phenylephrine at 3 hours (P less than 0.01) after the application, respectively. Use of 4 per cent chlorhexidine detergent solution (Hibiscrub) and other methods of skin disinfection.

In a comparison of three antiseptic detergent preparations for hand washing, Hibiscrub, a 4% chlorhexidine detergent solution, caused a significantly greater estimated immediate reduction of skin flora (86.7% +/- 3.0) than was obtained with Dermofax, a 0.75% chlorhexidine detergent solution (55.5% +/- 5.1), or with Disadine scrub, a povidone iodine detergent preparation (68% +/- 6.8). After six applications the mean estimated reductions of skin flora were 99.2% +/- 0.2 for Hibiscrub, 97.7% +/- 0.7 for povidone iodine, and 91.8% +/- 1.6 for Dermofax.After a series of hand washings with Hibiscrub, as with a hexachlorophane detergent preparation, a further large reduction of skin flora, shown by bacterial counts of hand sampling, was obtained by a second phase of disinfection consisting of two minutes' application on gauze swabs of 0.5% chlorhexidine digluconate in 70% ethanol; a further wash with Hibiscrub, in place of alcoholic chlorhexidine, for the second phase of disinfection caused an increase rather than a reduction in the yield of bacteria on skin sampling. Unlike this "two-phase" disinfection, the application for 30 minutes of compresses soaked in 10% aqueous povidone iodine or in 0.5% aqueous chlorhexidine digluconate did not cause a greater reduction in skin flora than that obtained by the conventional two minutes' application on gauze of 0.5% chlorhexidine in 70% ethanol.Chlorocresol (0.3%) liquid soap (the base used for Ster-Zac liquid hexachlorophane soap) caused a mean reduction of skin flora when used for hand washing of 29% after one application and 72% after six applications spread over two days. This formulation, though less active and more variable as a detergent skin antiseptic than chlorhexidine, hexachlorophane, or povidone iodine detergent preparations, is an inexpensive disinfectant soap which could be useful in catering establishments. Alcoholic cetrimide applied as for disinfection of an operation site caused a reduction of skin flora greater than that shown by aqueous cetrimide but comparable to that shown by 70% ethyl alcohol in previous experiments.

Modelling the thermal inactivation of Salmonella typhimurium using bioluminescence data.

Inactivation of micro-organisms by heat is a traditional food processing technique used to reduce or eliminate the microbial load in foods thus preventing bacterial associated disease and food spoilage. Models of thermal death kinetics are routinely used to predict the amount of heat required but such models are limited by the acquisition of accurate thermal death data for bacteria in situ and in complex microflora. In vivo bioluminescence from lux recombinant bacteria is an important alternative to traditional plate counts for examining bacterial injury and stress but the thermal instability of luciferase has appeared to preclude its application in heating studies. We have developed a procedure which overcomes the thermal instability of luciferase and demonstrate that computer generated models of the thermal injury of Salmonella typhimurium show equivalence between bioluminescence and viable count data.

Distribution of MCA-coated grits in maize fields after high wheel tractor application for disrupting orientation of Diabrotica virgifera virgifera LeConte.

High wheel tractor applications of 4-methoxycinnamaldehyde (MCA)-coated corn granules ('grits') were conducted in Ruski Krstur (Serbia) in summer 2001 in a 5 ha corn field. Grits are a by-product after corn is harvested and separated from the cob and used as a carrier medium to disseminate MCA into the corn field. MCA is a kairomone mimic derived form Cucurbita maxima (Duchesne) used to disrupt orientation of Diabrotica virgifera virgifera LeConte towards different MCA and pheromone baited traps. The ultimate goal is to investigate the use of MCA as a mating disruptant. MCA was dissolved in an organic solvent and mixed in a cement machine with the grits. Grits were applied at rates of 17.39, 17.1 and 12.45 kg/ha on July 4th, July 19th and August 3rd. Before the impact of MCA as a disruptant can be addressed, the distribution patterns of MCA coated grits have to be thoroughly investigated. They were evaluated by counting girts deposited in 16 or 20 plastic dishes of 30-cm diameter positioned along 2 rows through the field directly after the grit application by tractor. Additionally, grits deposited on corn plant surface such as leaves, leaf axils and corn cobs were counted. Total number of grits collected in plastic dishes revealed even application rates at the first and second application but not on the third application date. Number of grits collected on plant surfaces were significantly different from each other regarding each application date. Altogether, grit distribution in the dishes as well as on the plant surface was variable. However, distribution patterns achieved so far hold promise to disseminate MCA coated grits into corn fields for orientation disruption or mating disruption of D. virgifera virgifera.

Application of the sum peak method to determine the perturbed angular correlation parameters in biological specimens.

The counting rate of a sum peak formed by two gamma-ray emissions in radioactive decay of a nuclide depends on its integral attenuation factor G22. Therefore, in the "sum peak method", the G22 value for the specimen in question can be obtained merely from sum peak measurements using one radiation detector when the resolution time and the geometry are fixed. The procedure to determine the G22 values is described for specimens containing 111In, together with the detailed relationships between the sum peak counting rates and the values of G22.

A proposed method to minimize waste from institutional radiation safety surveillance programs through the application of expected value statistics.

Institutional radiation safety programs routinely use wipe test sampling and liquid scintillation counting analysis to indicate the presence of removable radioactive contamination. Significant volumes of liquid waste can be generated by such surveillance activities, and the subsequent disposal of these materials can sometimes be difficult and costly. In settings where large numbers of negative results are regularly obtained, the limited grouping of samples for analysis based on expected value statistical techniques is possible. To demonstrate the plausibility of the approach, single wipe samples exposed to varying amounts of contamination were analyzed concurrently with nine non-contaminated samples. Although the sample grouping inevitably leads to increased quenching with liquid scintillation counting systems, the effect did not impact the ability to detect removable contamination in amounts well below recommended action levels. Opportunities to further improve this cost effective semi-quantitative screening procedure are described, including improvements in sample collection procedures, enhancing sample-counting media contact through mixing and extending elution periods, increasing sample counting times, and adjusting institutional action levels.

Application of a hybrid pixel detector to powder diffraction.

Results obtained using a hybrid pixel photon-counting detector in powder diffraction experiments are presented. The detector works at room temperature and its dynamic response ranges from 0.01 photons pixel(-1) s(-1) up to 10(6) photons pixel(-1) s(-1). The pixel sizes are 0.33 mm x 0.33 mm for a total area of 68 mm x 68 mm. On recording high-resolution diffraction patterns of powders, a reduction of the experimental time by more than a factor of 20 is obtained without loss of data quality. The example of an X-zeolite shows that such detectors can be used for very demanding anomalous experiments. In situ experiments of quenching liquid oxides show that frames of 0.01 s can be achieved for studying such processes.

A modified Lucas cell for leakage measurement from encapsulated radium sources.

A new method to determine the leakage rate from encapsulated radium sources is proposed. The method is based on the principle of enclosing the source to be tested in a hermetically sealed container and measuring the radon radioactivity growth in the container as a function of time. A cylindrical modified Lucas cell, designed and built in our laboratory, is used for measuring the radon which are done by ZnS(Ag) alpha scintillation counting and can be easily compared with existing standard for leak tests. Results obtained show the applicability of the method for leak testing for radon leakage from sealed radium sources.

Long-term treatment of diseased, FIV-seropositive field cats with azidothymidine (AZT).

The long-term effectiveness of Azidothymidine (AZT, Zidovudine) was evaluated in FIV-seropositive cats with clinical symptoms (n = 9; Group 1) compared with conventional symptomatical therapy (n = 5; Group 2). The oral administration of Azidothymidine at a dosage of 5 mg/kg BW yielded a mean peak plasma concentration of 4.59 microM one hour after application (median: 3.74 microM). Elimination half time was 1.5 h. The permanent oral application of Azidothymidine at a dosage of 5 mg/kg BW TID led to a total recovery from clinical symptoms in six of nine FIV-seropositive cats (Group 1) 4-6 weeks after the onset of therapy. One cat clinically improved with only sporadical recurrence of disease; therapy with Azidothymidine was not effective in two cats. All the FIV-seropositive cats treated symptomatically responded well to antibiotics and immunomodulators within 10-14 days (n = 5; Group 2). Recurrence of clinical symptoms was noticed in three of five patients within 2 years after therapy and one cat died. During the treatment with Azidothymidine hyperproteinemia and abnormal albumin-to-globulin ratio became normal within 6 months in four FIV positive cats (Group 1), whereas hyperproteinemia did not change in Group 2. The following adverse effects were noticed in Group 1: a transient decrease of red blood cell count (RBC), packed cell volume (PCV) and haemoglobin until the 4th week of application of Azidothymidine (5/8), but the haemogram was within the normal range after 3 months. In one cat with hyperglycemia the anaemia remained until the administration of Azidothymidine was terminated. Heinz (Schmauch) bodies in the erythrocytes appeared in two FIV-positive cats 2 weeks after the onset of the therapy.

Distinct roles for glycine and GABA in shaping the response properties of neurons in the superior paraolivary nucleus of the rat.

The superior paraolivary nucleus (SPON) is a prominent periolivary cell group of the superior olivary complex. SPON neurons use gamma-aminobutyric acid (GABA) as their neurotransmitter and are contacted by large numbers of glycinergic and GABAergic punctate profiles, representing a dense inhibitory innervation from the medial nucleus of the trapezoid body (MNTB) and from collaterals of SPON axons, respectively. SPON neurons have low rates of spontaneous activity, respond preferentially to the offset of pure tones, and phase-lock to amplitude-modulated tones. To determine the roles of glycine and GABA in shaping SPON responses, we recorded from single units in the SPON of anesthetized rats before, during, and after application of the glycine receptor antagonist strychnine, the GABA(A) receptor antagonist bicuculline, or both drugs applied simultaneously. Strychnine caused a major increase in spike counts during the stimulus presentation, followed by the disappearance of offset spikes. In half of the recorded units, bicuculline caused moderately increased firing during the stimulus. However, in 86% of units bicuculline also caused a large increase in the magnitude of the offset response. Application of the drug cocktail caused increased spontaneous activity, dramatically increased spike counts during the stimulus presentation, and eliminated the offset response in most units. We conclude that glycinergic inhibition from the MNTB suppresses SPON spiking during sound stimulation and is essential in generating offset responses. GABAergic inhibition, presumably from intrinsic SPON collaterals, plays a subtler role, contributing in some cells to suppression of firing during the stimulus and in most cells to restrict firing after stimulus offset.
