Calculation of 2,3,7,8-TCDD equivalent concentrations of complex environmental contaminant mixtures.

Sufficient toxicological data are now available to permit use of conventional risk assessment techniques to estimate the hazards associated with human exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD). However, many real-world exposures involve complex mixtures of dibenzodioxins, dibenzofurans, and related compounds. Historical approaches to risk assessment on such mixtures have ranged from ignoring all compounds except 2,3,7,8-TCDD itself to assuming that all compounds have potencies equal to 2,3,7,8-TCDD. An alternative approach which uses existing literature data and analytical results to calculate the "2,3,7,8-TCDD equivalent" concentration of a mixture in order to "predict" its biological potency relative to 2,3,7,8-TCDD itself is advanced here. Previously reported in vivo acute and subchronic studies and some recently obtained analytical chemistry data are integrated here to clarify the utility of this important approach and to assess the uncertainties associated with its use. This predictive approach, and various conceptually similar ones, have now found wide applicability to the risk assessment process associated with exposure to complex mixtures of dioxins, dibenzofurans, and related compounds.

Assessment of human exposure to chemicals from Superfund sites.

Assessing human exposure to chemicals from Superfund sites requires knowledge of basic physical, chemical, and biological processes occurring in the environment and specific information about the local environment and population in the vicinity of sites of interest. Although progress is being made in both areas, there is still a tremendous amount to be done. Participants at this meeting have identified several of the areas in need of greater understanding, and they are listed below. Movement of dissolved and volatile organics, especially NAPLs, in the subsurface environment. This includes study of the partitioning of compounds between NAPLs, air, water, and soil. Partitioning of volatilized chemicals between gaseous and aerosol components of the atmosphere. This includes understanding how these components influence both wet and dry deposition. Long-term movement from sediments into biota and how these affect chronic toxicity to sediment biota. Broad validation of PBPK models describing partitioning of compounds from sediment and water into fish. Reactions of chemicals sorbed to atmospheric particles. This includes application of laboratory models to real and varied atmospheric conditions. Interactions between biotic and abiotic transformations in soil and sediment. Applicability of physiological pharmacokinetic models developed in laboratory studies of experimental animals and clinical investigations of humans to environmental chemicals, concentrations, and routes of exposure in humans. Use of human and wildlife behavioral and biomonitoring information to estimate exposure. This includes better understanding of human variability and the applicability of information gathered from particular wildlife species. To successfully address these gaps in our knowledge, much more analytical data must be collected.(ABSTRACT TRUNCATED AT 250 WORDS)

Experiences of "not belonging" in collectivistic communities: narratives of gays in kibbutzes.

The kibbutz is a uniquely Israeli phenomenon which is currently undergoing major changes, both ideologically and practically. It has attracted worldwide sociological and psychological research attention due to its unique characteristics, particularly in relation to communal child-rearing, education, and family life. The purpose of this paper is to understand the life experiences of homosexuals who experienced their adolescence on kibbutzes during the 1970s. In this study, the life experiences often gay men who still live on the kibbutz or have spent significant periods of their lives on it are examined from their own perspectives in order to understand how they view the connection between homosexuality and kibbutz life. Researching such a topic by utilizing a qualitative research paradigm approach enriches our understanding of the meanings associated with being different and the various ways in which marginality is perceived and experienced within the communal life of the kibbutz. A purposive sample of 10 gay men was recruited, and the age of participants ranged from 17-45. At the time of the interviews, six interviewees still lived on the kibbutz. Each participant was interviewed in-depth, with interviews lasting between one-and-a-half to two hours. All interviews were recorded, transcribed, and analyzed. Two methods were used to analyze the data: analytic induction and constant comparison analysis. The findings suggest an interesting dynamic regarding the experiences of gay men in their transition from one total institution, the kibbutz, to another total institution, the army. Paradoxically, life in the army, which is undoubtedly a restricted system, is accompanied by experiences of freedom and actualization of self when compared to life in the kibbutz. The findings are discussed within the frameworks of differentness and marginality, homosexuality theory from both psychological and sociological perspectives, and the unique ideological and practical aspects of kibbutz life.

The evaporation path of seawater and the coprecipitation of Br- and K+ with halite.

Brines and salt were sampled at the Morton Bahamas solar salt production facility on Great Inagua Island in the Bahamas. The brines were analyzed by ion chromatography to define more precisely than heretofore the evaporation path of seawater to the end of the halite facies. At Inagua, calcium carbonate begins to precipitate at a brine concentration factor of 1.8 times that of seawater. Gypsum begins to precipitate at a brine concentration of 3.8 times seawater, and halite at a concentration factor of 10.6. Three of the most concentrated brines from Inagua (40 times seawater) were evaporated further in the laboratory. Magnesium sulfate first precipitated at brine concentrations about 70 times those of seawater, and potassium-bearing phases began to precipitate for these brines at concentrations greater than 90 times those of seawater. The distribution of coefficients of Br- and K+ between brines and halite were determined by combining analytical data for the Inagua brines with measurements of the Br- and K+ content of halites from Inagua and of halite which had precipitated from Inagua brines during storage. The observed average value of DBr- is 0.032, in good agreement with some of the previous measurements. The measured values of DK+ are highly variable (0.001 to 0.021); DK+ for halite precipitated early in the halite facies is in the vicinity of 0.015.

Pharmacokinetics of diphenhydramine in healthy volunteers with a dimenhydrinate 25 mg chewing gum formulation.

This pharmacokinetic study evaluated diphenhydramine in the plasma of healthy volunteers after a single 25 mg oral dose of dimenhydrinate (diphenhydramine theophyllinate), corresponding to 12.7 mg diphenhydramine, in a chewing gum formulation. Seven volunteers (4 men, 3 women; age: 26.3 +/- 1.2 years; body weight: 63.1 +/- 4.1 kg; height: 172.4 +/- 4.6 cm) chewed the gum for 1 h. Blood samples (10 ml) were collected at different time intervals up to 24 h. Blood plasma was subsequently processed and analyzed for diphenhydramine content using a GLC method and an NPD detector. Analytical data revealed the following kinetic parameters: AUC(0-24h): 155.2h x ng x ml(-1); AUC(0-infinity): 195.3 h x ng x ml(-1); Mean resident time: 16 h; t(1/2): 10 h; C(max): 14.5 ng x ml(-1); t(max): 2.6 h; and plasma clearance: 9.0 ml x min(-1) x kg(-1). This study indicates that the pharmaceutical formulation employed provided sustained plasma concentrations of diphenhydramine, presumably sufficient to support its clinical efficacy towards motion sickness owing to the almost complete (> 95%) release by the formulation of the active principle. Moreover, the maximal concentrations of diphenhydramine attained in plasma were much lower than the concentration threshold needed to produce drowsiness.

Fluorescence lifetime imaging and spectroscopy as tools for nondestructive analysis of works of art.

A system for advanced fluorescence investigation of works of art has been assembled and integrated in a characterization procedure that allows one to localize and identify organic compounds that are present in artworks. At the beginning of the investigation, fluorescence lifetime imaging and spectroscopy address a selective microsampling of the artwork. Then analytical measurements of microsamples identify the chemical composition of the materials under investigation. Finally, on the basis of fluorescence lifetime and amplitude maps, analytical data are extended to the whole artwork. In such a way, information on the spatial distribution of organic materials can be inferred. These concepts have been successfully applied in an extensive campaign for analysis of Renaissance fresco paintings in Castiglione Olona, Italy. Residue of various types of glue and stucco left from a restoration carried out in the early 1970s was localized and classified. Insight into the technique used by the painter to make gilded reliefs was also obtained.

Trace element analysis of scales from four populations of Norwegian Atlantic salmon (Salmo salar L.) for stock identification using laser ablation inductively coupled plasma mass spectrometry.

The pre-smolt stage of the scale of adult Norwegian Atlantic salmon from four populations, encompassing both farmed and wild hatchery stocks, has been analyzed by laser ablation inductively coupled plasma mass spectrometry (LA-HR-ICP-MS). The purpose of this study was to test for differences in elemental composition between stocks of Atlantic salmon that have lived in separate fresh water locations until the smoltification and natural run out or transportation to the sea-cages. The populations studied were from fish farms located at Bremanger and Sorfold, a cultivated stock from Mossa, and one wild local stock from the river Gaula. The following elements were included in the analytical protocol: Li, Mg, Ca, Cr, Mn, Fe, Zn, Sr, Ba, Pb, and U. Calcium was used as a natural internal standard. Classification of the analytical data is studied by multivariate statistical techniques such as principal component analysis (PCA). We have been able to delineate the population of Atlantic salmon (Salmo salar L.) from Gaula and Mossa from the Bremanger and the Sorfold stock based on the ten elements analyzed. The Bremanger and Sorfold stocks were partially delineated. The differences in elemental composition in the scales, which makes the delineating of the four stocks possible, probably reflects geological differences in the bedrock at the four fresh water locations experienced by the salmon during the pre-smolt stage.

Mycophenolate mofetil is safe, well tolerated, and preserves lung function in patients with connective tissue disease-related interstitial lung disease.

BACKGROUND: Interstitial lung disease (ILD) frequently complicates connective tissue diseases (CTDs). Glucocorticoids and immunomodulatory agents are regarded as mainstays of therapy for CTD-related ILD; however, apart from those studies that have evaluated certain medications for patients with scleroderma, few studies have been performed. In this study, our objectives were to examine the safety and tolerability of mycophenolate mofetil (MMF) and to determine its impact on lung function in patients with CTD-ILD. METHODS: In this retrospective observational study, we analyzed patients at our center who ever received MMF for CTD-ILD. We examined the frequency and severity of side effects associated with MMF and used longitudinal data analytic methods to determine the ability of MMF to maintain lung function over time. RESULTS: Twenty-eight patients were treated with MMF over 35.9 patient-years. The most common underlying CTD diagnosis was scleroderma (n = 9). The most common reason for initiating MMF was an adverse effect of a prior immunomodulatory agent. Six patients had clinically significant side effects related to MMF; all resolved with dose reduction. Compared to before MMF, the mean daily prednisone dose while patients were receiving MMF was lower (10 mg/d vs 15 mg/d, p = 0.09). In addition, since starting MMF, the average percentage of predicted forced vital capacity (FVC), average percentage of predicted total lung capacity, and average percentage of predicted diffusing capacity of the lung for carbon monoxide for the cohort increased by 2.3%, 4.0%, and 2.6%, respectively. CONCLUSION: MMF appears to be safe and well tolerated in patients with CTD-ILD. Larger-scale studies are needed to further evaluate the efficacy of MMF in this patient population.

Chemical studies on the structure of human serum high-density lipoprotein (HDL). Photochemical crosslinking of azido-labelled lipids in HDL.

The lipid classes of native human serum high-density lipoprotein (HDL) were exchanged against phosphatidylcholine, sphingomyelin and cholesteryl ester species substituted with photosensitive fatty acyl residues and against 25-azido-27-norcholesterol. The photosensitive fatty acyl residues were 5-and 16-azidopalmitic, 12-azidooleic and 18-azidolinoleic acid, all labelled with high tritium radioactivity. The lipid exchange method previously described was used. After UV irradiation and delipidation the apoproteins AI and AII, photocrosslinked with the radioactive lipids, were separated. The yield of covalently crosslinked lipid molecules amounted to 30% of the photosensitive lipid molecules incorporated into the HDL particle. ApoAI and apoAII were labelled by each of the photosensitive lipid classes, intergrated in the HDL particle although in very different stoichiometry. The regiospecific photochemical labelling of the lipopolypeptides AI and AII was established by cyanogen bromide cleavage and the separation of the four apoAI and two apoAII CNBr fragments. The analytical data prove the close steric relation and interaction of defined protein regions with hydrophobic regions of the lipid molecules by chemical means. These results are discussed with respect of the HDL structure.

Determination of serum triglycerides by capillary on-column gas chromatography.

We report an accurate method, based on capillary on-column gas chromatography, for determining triglycerides in human serum. After serum extraction and chemical hydrolysis, glycerol is directly measured by gas chromatography (GC). With our extraction method no free glycerol is extracted from serum. The accuracy of this method was compared with that of a method based on the original procedure of Carlson (J. Atheroscler. Res. 3, 334-336 (1963)) and which is standardized by the Centers for Disease Control (CDC, Atlanta, U.S.A.). Orthogonal regression analysis of the GC method (y) and the CDC reference method (X) resulted in y = 0.996 X + 0.000 with a correlation coefficient of 0.999. The variances of analytical data, collected over a two month period showed that, for the GC method, the within-day coefficient of variation (CV) was less than 1.43%; the between-day CV less than 1.35%. The data for the CDC method were less than 3.36% (within-day) and less than 6.38% (between-days). The CDC method is linear up to 3.7 mmol/l and the GC method to 22 mmol/l.

Comparison of the reflectance method (Reflotron reflectance photometer) with the absorbance method (automatic analysers) for the determination of cholesterol.

The European Atherosclerosis Society (1) and the Expert Panel of the US National Cholesterol Education Program (2) have issued detailed guide values for recognition and management of hyperlipidaemia in adults. In these guidelines, the diagnosis of dyslipidaemia based on the measurements of total cholesterol, triacylglycerols, HDL and LDL cholesterol plays an important role. A prerequisite for the desired success of interventive measures is the reliability of the analytical data. The aim of this study was to investigate the precision and accuracy of Reflotron Cholesterol, a method based on the dry chemistry principle. Accuracy was assessed by establishing the correlation with the standardized automated methods used in routine lipid diagnosis. In addition, it was also examined whether the Reflotron Cholesterol results in plasma and blood are comparable. The Reflotron cholesterol (sample: blood) showed a good correlation with the CHOD/PAP method on a Hitachi 737 instrument (sample: plasma). The median value of the differences of the test results was -0.4%. Similarly, the method comparison of Reflotron Cholesterol (sample: blood) versus CHOD/PAP method on a SMAC instrument (sample: plasma) showed that Reflotron produces slightly (1.8%) higher results. The Reflotron Cholesterol values obtained from blood samples were slightly lower than those from plasma samples (median value of the differences: -2.2%). The results suggest that for routine purposes Reflotron Cholesterol provides results which are in good agreement with those obtained by standardized wet chemistry methods.

Isoperformance curves in applied psychology.

Isoperformance is a technique for reading information out of a data-analytic model, comparable to expected mean square or omega squared analyses. It results in a trade-off function (an isoperformance curve) among the determinants of performance. The technique was developed primarily to generate trade-off functions between personnel aptitude and time in training or on the job. However, the technique is general and can be applied in any trade-off situation. In part, the purpose of this paper is to recall the antecedents of isoperformance in psychophysics and to recount the origins and development of the isoperformance readout. Its main purpose, however, is to present several examples of isoperformance curves in applied psychology and to make the case for their usefulness.

Anthropometric, haemodynamic, humoral and hormonal evaluation in patients with incidental adrenocortical adenomas before and after surgery.

OBJECTIVE: To compare clinical and humoral parameters before and after surgery in patients with incidental adrenocortical adenomas. DESIGN: Six patients with subclinical Cushing's syndrome and nine with non-functioning adenomas were investigated before and 12 Months after removal of the mass. METHODS: Anthropometric (body weight, body mass index and waist to hip ratio), haemodynamic (blood pressure and heart rate), metabolic (lipids and oral glucose tolerance test (OGTT)), hormonal (cortisol, plasma renin activity, aldosterone, androgens and catecholamines) and bone metabolism (hydroxyproline, parathyroid hormone, osteocalcin and ostase) parameters were evaluated. RESULTS: In the whole group, a significant decrease in body weight (69.7+/-3.5 vs 70.8+/-3.5 kg, P<0.03), in systolic (135.3+/-5.1 vs 145.6+/-4.9 mmHg, P<0.009) and diastolic (83.7+/-1.9 vs 91.0+/-3.5 mmHg, P<0.03) blood pressure and in glucose levels in response to OGTT (106.4+/-9.6 vs 127.5+/-6.5 mg/dl, P<0.05) was observed after surgery. All other parameters examined did not change significantly. This trend was also found in both groups separately. Analytical data showed a high frequency of overweight/obesity (66.6%), hypertension (66.6%) and impaired glucose profile (26.6%) in our patients, with a greater prevalence of these cardiovascular risk factors in the subclinical Cushing's syndrome group. After surgery, values normalized or improved in eight out of ten hypertensive patients and in three out of four patients with impaired glucose profile. CONCLUSIONS: Solid adrenocortical incidentalomas are associated with some cardiovascular risk factors which may be corrected after removal of the mass. Therefore, surgery may be an appropriate choice in patients with subclinical Cushing's syndrome but also in those with solid non-functioning adenomas and coexistent cardiovascular risk factors.

Chemical composition of cynomolgus monkey milk.

Milk samples from eight cynomolgus monkeys whose lactation period ranged 44-119 days were investigated for their gross chemical composition such as total solids, crude protein, lipids, lactose and ash. Nitrogen distribution, compositions of fatty acids, amino acids and triglycerides as well as major minerals were also determined. These analytical data were compared with those reported for man, simian and cow, and cynomolgus monkey milk was found to have close similarities to human milk.

CD11b+ cells are the major source of oxidative stress in UV radiation-irradiated skin: possible role in photoaging and photocarcinogenesis.

Exposure of skin to solar UV radiation induces oxidative stress and suppression of cell-mediated immune responses. These effects are associated with the greater risk of several skin disorders including photoaging and photocarcinogenesis. We have shown that UV-induced infiltrating leukocytes contribute in developing oxidative stress in UV-irradiated skin. The peak period of UV-induced infiltrating leukocytes lies between 48 and 72 h after UV exposure of the skin. In this study we demonstrated that UV (90 mJ/cm2)-induced infiltrating CD11b+ cells in C3H/HeN mice skin were the major source of oxidative stress. Hydrogen peroxide (H2O2) was determined as a marker of oxidative stress. Flow cytometric analysis of viable cells revealed that the number of CD11b+H2O2+ cells were significantly higher (31.8%, P < 0.001) in UV-irradiated skin in comparison with non-UV-exposed skin (0.4%). Intraperitoneal administration of monoclonal antibodies to CD11b (rat IgG2b) to C3H/HeN mice inhibited UVB-induced infiltration of leukocytes, as evidenced by reduction in myeloperoxidase activity (64-80%, P < 0.0005), concomitant with significant reduction in H2O2 production both in epidermis and dermis (66-83%, P < 0.001-0.0005) when compared with the administration of rat IgG2b isotype of anti-CD11b. Furthermore, CD11b+ and CD11b- cell subsets were separated by immunomagnetic cell isolation technique from total epidermal and dermal single cell suspensions obtained 48 h after UV irradiation of the skin and analyzed for H2O2 production. Analytical data revealed that CD11b+ cell population from UV-irradiated skin resulted in significantly higher production of total H2O2 in both epidermis and dermis (87-89%, P < 0.0001) in comparison with CD11b- cell population (11-13% of total H2O2). These data revealed that infiltrating CD11b+ cells were the major source of oxidative stress in UV-irradiated skin and thus may contribute to photoaging and promotion of skin tumor growth within the UV-irradiated skin. Together, these data suggest that reduction in UV-induced skin infiltration of CD11b+ cells may be an alternative and effective strategy to reduce solar UV light-induced oxidative stress-mediated skin disorders including photoaging and photocarcinogenesis.

[Are some Brazilian population groups subject to endocrine disrupters?].

Several chemical substances suspected of endocrine disruption are currently being used in industrial and agricultural activities in Brazil. This paper presents analytical data showing contamination of some population groups by these substances. Virtually nothing is known about the related health effects, highlighting the need to implement a national program to evaluate such impacts.

Amino acid metabolism in Wegener's granulomatosis.

The urinary excretion of amino acids and their derivatives in cases of Wegener's granulomatosis have been examined, and the excretion of urinary amino acids in such cases was different from normal and reticulum cell sarcoma. In Wegener's cases, the amino acids which were always found in readily detectable amounts totaled 33; urinary phenylalanine was 4-5 times higher than in normal subjects. From these experimental results, we believe that the observation of amino acid excretion into urine is an important indicator for the diagnosis of Wegener's granulomatosis. In addition, the results of quantitative examination on the urinary amino acid excretion were compared with normal and reticulum cell sarcoma. Comparison of the analytical data on urinary amino acids of the normal and patient groups indicated significant metabolic abnormalities of some amino acids at the earliest stage of Wegener's granulomatosis.
