Primary nerve cell cultures derived from embryonic rat central nervous system form [3H]ACh from exogenous [3H]Ch, and release it upon potassium depolarization. Pretreatment of the cultures with botulinum-A toxin or tetanus toxin diminishes the cellular accumulation of [3H]ACh. Poisoning the cultures during the period of [3H]Ch uptake fails to lower [H]ACh formation. Dependent on dosage, both toxins suppress the release of [3H]ACh upon potassium depolarization. Heat-denaturated toxins as well as tetanus toxin preincubated with tetanus antitoxin were without effect.
Lytic corynebacteriophage betahv64(tox+) has been characterized, and methods for studying the expression of its tox(+) gene in nontoxinogenic Corynebacterium diphtheriae strain C7(s)(-)(tox-) described. During one cycle of viral growth there was a 1 million-fold increase in extracellular toxin. Both the conditions of the experiment and the use of purified phage, free from toxin, support the conclusion that all of the toxin was newly formed. This toxin was immunochemically indistinguishable from standard toxin produced by the PW8(r)(Pdi)(tox+) strain. Chloramphenicol was found to be an effective agent for synchronizing the initiation of viral growth. Once chloramphenicol was removed, intracellular toxin appeared and continued to increase throughout the latent period. Proflavine, added early in the latent period, blocked phage maturation without similarly affecting yields of toxin. Iron exerted a limited inhibitory effect on final toxin levels attained.

Newborn guinea pigs (less than 24 hours old) are highly susceptible to the lethal effect of endotoxin and susceptibility decreases with age. The vulnerability to endotoxin has not been attributed to hypofunction of either the adrenal gland or the reticuloendothelial system of the newborn, since prior injection of corticosteroids provides only partial protection to endotoxin and newborn guinea pigs can efficiently remove colloidal substances from the circulation. In contrast to adults, newborns do not develop a delayed inflammatory reaction following intradermal injection of endotoxin, and after intravenous challenge frequently fail to display neutropenia and sustained fever.
