Diode-array spectrophotometry for simultaneous measurement of hemoglobin pigments.

A prototype oxygen saturation meter was used to measure the concentrations of deoxygenated hemoglobin (rHb), oxyhemoglobin (HbO2), carboxyhemoglobin (HbCO), methemoglobin (MetHb), and sulfhemoglobin (SHb) in 35 microliter blood. Simultaneous absorbance measurements at 535, 560, 577, 622, 636, and 670 nm permitted the composition of any hemoglobin pigment mixture to be determined more accurately, precisely and easily than before. The inclusion of 670 nm, where the hemoglobin pigments have low absorption coefficients, allowed correction for turbidity.

Stopped-flow spectrophotometry monitoring the initial processes of platelet activation by platelet-activating factor.

The initial processes of platelet activation by platelet-activating factor (PAF) were observed by stopped-flow light scattering and fluorometry. The binding of PAF to rabbit platelets did not change the membrane fluidity, though it caused the removal of calcium from internal stores and induced concomitantly platelet shape changes. These results were quite in contrast to those by thrombin and ADP activation, where the membrane fluidity increased before the calcium release from the internal stores. The increased membrane fluidity in the latter system seemed to be used to transmit an external signal to a GTP-binding protein.

Intrinsic fluorescence, difference spectrophotometry and theoretical studies on tertiary structure of calf thymus histone H1.

Tyr-72 is included in the hydrophobic cleft which is formed in the histone H1 globular head. Tyr-72 is screened against polar aqueous environment and its intramolecular mobility is sharply retarded. This microenvironment causes a red shift (lambda max = 279 nm) and a sharpening of the longer wavelength shoulder of absorption spectra, a high fluoresence anisotropy value (A = 0,11), high quantum yield of fluoresence (approximately 0.2) and a decrease of the Stern-Volmer Constant during quenching of histone H1 fluorescence by acrylamide. It has been found that the change in the intensity of histone fluorescence at lambda excit = 265 nm, but not at lambda excit = 280 nm, is due to the changes in the quantum yield of fluorescence. The increase of fluorescence intensity at lambda excit = 280 nm depends on the changes in the quantum yield and molar extinction coefficient of histone H1 tyrosyl chromophore. The change in the ratio of fluorescence intensity exited at 280 nm (F280) to the fluorescence intensity excited at 265 nm (F265) corresponds to the change of delta epsilon 286 in difference absorption spectra. The introduction of the parameter Cf = F280/F265 allows one to go over to studying excitation spectrum shifts instead of histone absorption spectrum shifts, which is much more convenient methodologically since in this case it is possible to carry out research using lower protein concentrations and turbid solutions. The results make it possible to designate Tyr-72 of histone H1 as a special class of fluorescent tyrosyls whose properties differ from those of tyrosyls of other tryptophane-free proteins: RNAase, insulin, core histones--H2A, H2B, H3, H4 and some others.

Derivative spectrophotometry of dimer and monomer of enolase.

1. SDS causes significant polar exposure of aromatic amino acids of enolase. The alpha-helix content remains unchanged. The enzyme lost all its activity. 2. The presence of 1 M K Br in enzyme solution results in a smaller increase of polarity of aromatic amino acids residues environment. The amount of alpha-helix does not decrease in comparison to native enzyme. Enzyme lost nearly 80% of its initial activity. 3. The extreme pH values and the presence of 6 M Gnd.HCl influence the whole structure of enolase. It is accompanied by a large polar shift of aromatic amino acids and significant decrease of alpha-helix content of the protein.

Production of bovine rhodopsin by mammalian cell lines expressing cloned cDNA: spectrophotometry and subcellular localization.

Cloned cDNA encoding bovine rhodopsin has been recombined into an expression vector and cotransfected with an antibiotic resistance plasmid into cultured human embryonic kidney cells. The resulting cell lines produce 100-200 micrograms of bovine opsin per liter of saturated tissue culture medium (10(9) cells). Incubation in vitro with 11-cis retinal produces a photolabile pigment the absorbance spectrum of which is indistinguishable from that of bona fide bovine rhodopsin. Expressed rhodopsin accumulates in the plasma membrane as determined by immunoelectron microscopy.

Determination of cobalt in forages by Zeeman electrothermal atomic absorption spectrophotometry without preliminary extraction.

A simple and rapid method, avoiding preliminary extraction, is described for the determination of Co in plant samples. The factors influencing the mineralization steps and the interference of major elements are investigated. The described method gives a recovery in the range 100-110%. Use of the standard addition method avoids interference from Ca and Fe. In the range 0.1-1 ppm (dry basis), Co can be determined with a precision of 5%. The detection limit of the method is 0.02 ppm of Co in forages.

Microsampling technique and determination of blood lead by Zeeman atomic absorption spectrophotometry.

Quality control materials and participation in proficiency testing programmes were used for assessing the long-term performance of our blood lead analysis. The accuracy and precision of the blood controls, Metals 1 and 2 from the Behring Institute, were evaluated over a 1-year period. The accuracy of the method was also monitored by the Centers for Disease Control (CDC) Blood Lead Proficiency Testing Programme over a 3-year period. Nearly 85% (28 out of 33 samples) of our results were within +/- 5% of the CDC reference values. There were excellent correlations between our results and the target values of several proficiency testing samples, with correlation coefficient values of 0.9959-0.9970. The method was used for the screening of blood lead in children in conjunction with microsampling techniques.

Comparison of wet digestion procedures for the determination of cadmium and lead in marine biological tissues by Zeeman graphite furnace atomic absorption spectrophotometry.

Three wet digestion procedures, namely, nitric acid/sulphuric acid, nitric acid/hydrogen peroxide and nitric acid/perchloric acid were compared for their relative efficiency in determining cadmium and lead in marine biological tissues. The nitric acid/perchloric acid procedure was the most efficient, whereas the nitric acid/hydrogen peroxide procedure was the least efficient. The nitric acid/sulphuric acid procedure was also found to be effective but there was a problem with the method as the presence of sulphuric acid in the matrix markedly suppressed the absorbance signals especially for lead analysis and thus reduced sensitivity. The addition of ascorbic acid as matrix modifier improved sensitivity. However, this analysis still required the standard addition method. No apparent interferences were encountered for cadmium analysis.
