The suppressive effect of meperidine on PHA-stimulated transformation of human lymphocytes.

The effect of meperidine on the response of human peripheral lymphocyte in culture to the stimulation by the mitogen, phytohemagglutinin, was studied by measuring the incorporation of 3H-thymidine into DNA. The addition of meperidine at a high concentration (40-200 micrograms/ml) resulted in significant decrease in response to mitogen, the inhibition being dose-dependent. Meperidine may influence immune responses by altering DNA synthesis in the lymphocytes.

Microbial community analysis with a high PHA storage capacity.

Activated sludge was submitted to aerobic dynamic substrate feeding for the production of biodegradable plastics. Two sequencing batch reactors were operated with acetate or propionate as sole carbon substrates. With acetate a homopolymer of polyhydroxybutyrate (PHB) was obtained and with propionate a copolymer of hydroxybutyrate and hydroxyvalerate P(HB/HV) was produced. Three main morphotypes were identified in both sludges: two belong to the Alphaproteobacteria class and the third to the Betaproteobacteria class. Bacilli belonging to Betaproteobacteria were shown by FISH analysis, applied in combination with Nile Blue post-staining, to be the main responsible for PHAs storage. The latter were affiliated to Azoarcus genus within Betaproteobacteria.

Enhanced lymphocyte response to PHA among leukemia patients taking oral lithium carbonate.

Seven children receiving maintenance therapy for acute lymphoblastic leukemia (in remission) were given oral lithium carbonate for 12 weeks. Hematologic values, serum immunoglobulin levels, and responsiveness of peripheral blood mononuclear cells to mitogen stimulation were studied during the lithium treatment period, and compared to measurements made during a 12-week period when no lithium was administered (children served as their own controls). Changes attributable to treatment with lithium carbonate included increases in total white blood cell count, circulating neutrophil count, and response to phytohemagglutinin-P. No important toxicity to lithium carbonate was observed.

Effect of medroxyprogesterone acetate upon PHA, Con A and PWM stimulated lymphocytes and on E-rosette function.

We evaluated if Medroxyprogesterone acetate (MPA) had an immunosuppressive action as progesterone and other steroid hormones. Lymphocyte function was evaluated by the blastogenic response to mitogens and by the E-rosette test. MPA was tested at 7 ng/ml and 100 ng/ml concentrations. Cells were either preincubated for 30 min., 90 min., 180 min. and 12 hr or incubated with MPA for 72 hr, 18 hr and 16 hr. MPA showed a significative inhibitory effect only when used at the concentration of 100 ng/ml.

Suppression of lymphocyte PHA-responsiveness after rubella vaccination with Cendehill and RA 27/3 strains.

Two rubella vaccine strains, Cendehill and RA 27/3, were compared with regard to their immunosuppressive effect in vivo. 26 female vaccinees were studied. Both vaccines caused a slight transient suppression of lymphocyte responsiveness to mitogens on day 11 after vaccination. A whole blood culture method was used for lymphocyte stimulation tests, and the method appeared suitable for clinical studies of cell-mediated immunity.
