Inhibition of ACAT by avasimibe decreases both VLDL and LDL apolipoprotein B production in miniature pigs.

An orally bioavailable acyl coenzyme A:cholesterol acyltransferase (ACAT) inhibitor, avasimibe (CI-1011), was used to test the hypothesis that inhibition of cholesterol esterification, in vivo, would reduce hepatic very low density (VLDL) apolipoprotein (apo) B secretion into plasma. ApoB kinetic studies were carried out in 10 control miniature pigs, and in 10 animals treated with avasimibe (10 mg/kg/d, n = 6; 25 mg/kg/d, n = 4). Pigs were fed a diet containing fat (34% of calories) and cholesterol (400 mg/d; 0.1%). Avasimibe decreased the plasma concentrations of total triglyceride, VLDL triglyceride, and VLDL cholesterol by 31;-40% 39-48%, and 31;-35%, respectively. Significant reductions in plasma total cholesterol (35%) and low density lipoprotein (LDL) cholesterol (51%) concentrations were observed only with high dose avasimibe. Autologous 131I-labeled VLDL, 125I-labeled LDL, and [3H]leucine were injected simultaneously into each pig and apoB kinetic data were analyzed using multicompartmental analysis (SAAM II). Avasimibe decreased the VLDL apoB pool size by 40;-43% and the hepatic secretion rate of VLDL apoB by 38;-41%, but did not alter its fractional catabolism. Avasimibe decreased the LDL apoB pool size by 13;-57%, largely due to a dose-dependent 25;-63% in the LDL apoB production rate. Hepatic LDL receptor mRNA abundances were unchanged, consistent with a marginal decrease in LDL apoB FCRs. Hepatic ACAT activity was decreased by 51% (P = 0.050) and 68% (P = 0.087) by low and high dose avasimibe, respectively. The decrease in total apoB secretion correlated with the decrease in hepatic ACAT activity (r = 0.495; P = 0.026). We conclude that inhibition of hepatic ACAT by avasimibe reduces both plasma VLDL and LDL apoB concentrations, primarily by decreasing apoB secretion.

[Prospective lung function determination using an electronic miniature spirometer for detection of acute obstructive respiratory changes in diving students during occupational diving training].

BACKGROUND: Changes in lung function have been shown in experienced divers. The purpose of this study was to measure the pulmonary function of diving trainees who were not exposed to a hyperbaric environment before. METHODS: We measured the lung function in a sample of fifteen randomized selected young healthy non-smoking diving trainees parallel to fourteen open water dives. Five subjects used common compressed air diving apparatuses (breathing gas: 21% O2/78% N2) and ten subjects used closed circuit diving apparatuses (breathing gas compositions depended to the diving depths; 24 m: 60% O2/40% N2, 42 m: 40% O2/60% N2, 54 m: 32.5% O2/67.5% N2). The values (FVC, FEV1, PEF, MEF 25, MEF 50, MEF 75) were obtained by an electronic miniature-spirometer directly before descent, upon surfacing, and after one hour and four hours. RESULTS: The lung function values of the subjects who used compressed air showed no relevant impairment of lung function after the diving exposures. Significant (p < 0.05) post dive lung function reductions (FVC, FEV1, PEF, MEF 25, MEF 50, MEF 75), were evident in subjects using closed circuit diving apparatuses, after deep dives and shallow water dives. CONCLUSIONS: The decreased lung function of the used diving apparatuses. The impaired lung functions in subjects using closed circuit diving apparatuses may be induced by a subclinical lung edema generated by pressure differences between breathing gas and thorax tissues as well as the influences of soda-lime dust. Compared to compressed air diving the significantly increased oxygen partial pressures (p < 0.001) in closed circuit diving apparatuses may be an additional reason for the reversible airway obstructions.

[TGF-beta 1 in the experimentally induced inflammatory periodontal tissues in miniature swines].

TGF-beta 1 is a multifunctional molecule which has unique and potent effects on many target cells and tissues. TGF-beta 1 may promote inflammatory reaction by certain intercellular interaction. TGF-beta 1 at extremely low concentrations shows strong chemotatic activity for mononuclear phagocytes and stimulates bone resorption by enhancing production of PGE2. On the other hand, TGF-beta 1 plays a very important role in the regulation of extracellular matrix turnover presumably by modulating the action of other growth factors on matrix metalloproteinases (MMPs) and tissue inhibitor of metalloproteinases (TIMPs) expression. TGF-beta 1 was identified intra- and extracellularly in the inflamed gingival tissues and the distribution was associated with areas of inflammation. Sixteen miniature swines were used in this experimental gingivitis/periodontitis study. The ligatures were placed in situ for periods of 3, 5, 8 and 13 weeks and peroral innoculations of Porphyromonas gingivalis/Actinomyces viscosus into the ligatures were carried out only in the experimental group. ELISA was used to measure the levels of TGF-beta 1 in gingival tissues from the experimental and control groups. Recording of the clinical periodontal parameters was performed and the proportion of black-pigmented Bacteroides in the ligature (plaques) removed immediately prior to the biopsies was recorded. The results revealed that the concentration of TGF-beta 1 of the experimental group was higher and significantly different in statistics on the period of third week than that of the control group. The concentration of TGF-beta 1 was significantly different between the third week and the thirteenth week in the experimental group, and was negatively related to the time-length of ligatures. Furthermore, the concentration of TGF-beta 1 was negatively related to the changes of the calculus index and gingival index. These data indicated that the concentration of TGF-beta 1 of gingival tissue exhibited dynamic changes associated with the progression of experimental periodontal inflammation. The levels of TGF-beta 1 in gingival tissue may be valuable in detecting the inflammatory reaction of periodontal tissues.

Mandibular reconstruction in miniature pigs with prefabricated vascularized bone grafts using recombinant human osteogenic protein-1: a preliminary study.

A new technique of prefabrication of vascularized bone grafts in the latissimus dorsi muscle of miniature pigs, using recombinant human osteogenic protein-1 (rhOP-1) and xenogenic bone mineral as a carrier, is presented. The grafts were used to reconstruct a mandible using microsurgical anastomosis of the thoracodorsal vessels of the flap with a branch of the external carotid artery and the external jugular vein. The technique provided enough bone to reconstruct a mandible. The bone marrow stayed viable after transplantation, demonstrated by fluorescence microscopy and scintigraphy. Compared with identical defects of the contralateral side, which were treated with directly applied rhOP-1/bone mineral, a significantly better result was obtained with the prefabrication technique. The prefabrication technique may provide a means to treat irradiated patients with bone grafts produced by osteoinductive proteins and has a potential to become a clinical alternative to conventional vascularized bone grafts.

Hepatic haemodynamic changes following inhibition of endothelium-derived relaxing and hyperpolarising factors in anaesthetised miniature pigs.

The influence of endothelium-dependent vasodilatation in regulating the hepatic circulation has been investigated by intraportal infusion of inhibitors of either endothelium-derived relaxing factor (NG-nitro-L-arginine-methyl-ester [L-NAME]) or of endothelium-derived hyperpolarising factor (ATP-dependent K(+)-channel inhibitor, glybenclamide) in barbiturate anaesthetised miniature pigs. Intraportal infusion of acetylcholine (5.5 micrograms kg-1 min-1 over 2 min) produced a selective 3-fold increase in hepatic artery and coeliac trunk blood flow, while mesenteric, portal, systemic, and pulmonary vascular beds remained unchanged. Intraportal L-NAME or glybenclamide did not reduce the hepatic artery and coeliac trunk flows but increased systemic and mesenteric vascular resistances. The acetylcholine-induced hepatic artery vasodilatation was partially blocked by 59%, 76% and 66% by L-NAME, at 30, 100, and 300 mg/kg respectively. Glybenclamide pretreatment up to 3 mg/kg did not modify acetylcholine-induced vasodilatation of the hepatic artery and coeliac trunk. Furthermore, prior cyclooxygenase inhibition did not alter the hepatic vascular response to acetylcholine. These results suggest that, in contrast to what is observed in large vessels, the hepatic vascular tree may not be entirely regulated by nitric oxide under basal conditions, but nitric oxide is released readily upon stimulation with acetylcholine, a response that is largely but incompletely blocked by L-NAME pretreatment. Neither basal vascular tone nor acetylcholine-induced vasorelaxation are mediated by the opening of glybenclamide-sensitive K+ channels in the hepatic circulation in pigs.

[Formation of a gene pool of miniature Siberian swine "Minisibs" and their use in medico-genetic research].

The results of 35-year breeding of a new variety of laboratory animals for biological and medical studies, termed Minisibs (miniature Siberian swine), are summarized. Minisibs were obtained via hybridization between Swedish Landrace pigs, who have an epistatic white coat color, and black Vietnamese masked pigs of breed I, who have genes of dwarfism. Some genes of wild boars Sus scrofa scrofa and S. s. nigripes from Central Europe and Central Asia, respectively, were introduced to the Minisibs gene pool in order to strengthen the constitution, increase the general resistance, and improve the functions of the cardiac muscle and valves. The pigs were selected for a decrease in body weight and an increase in litter size over 15 generations. Immunogenetic and chromosomal polymorphisms were constantly monitored by Robertsonian translocations Rb 15/17 and Rb 16/17 introduced from boars. Long-term breeding produced a population of laboratory pigs with an average adult weight of approximately 60 kg, a litter size of seven to eight piglets, and quiet behavior. Most of animals used in selection were white, which is essential for radiobiological experiments and studies on skin histocompatibility. Introduction of chromosomal polymorphism to the minipig gene pool and long-term immunogenetic and cytogenetic monitoring made it possible to perform subchromosomal mapping of the loci of some blood-group systems associated with the loci controlling coat color, litter size, stress resistance and meat quality (halothane sensitivity), and histocompatibility. Minisibs have been successfully used in numerous biomedical studies and in therapeutic and surgical cardiology (for production of diagnostic sera controlling lipid metabolism and of biological prostheses of cardiac valves). The anatomical and physiological similarity of Minisibs to humans indicate that these animals may serve as donors of xenografts for orthotopic heart transplants to humans.

[Experimental study on supraeruption of the unopposed miniature pig molars].

Supraeruption of the molars in the absence of antagonist molars or without occlusal contacts was observed in a miniature pig. Tetracycline was used to label the new regenerated cementum and alveolar bone associated with the supraeruption. The results showed that the molar through root canal treatment migrated the same level as the molars with pulp, which means that the hydrodynamic and hydrostatic forces arising from the blood in pulp vessels look doubtful. Cementum and alveolar bone deposition was seen around the apical, cervical regions and under the furcation area. The thickness of the deposition layers was almost the same as the supraerupted volume from the model measurement.

[Genetic analysis of 35 microsatellite loci in 5 lineages of xishuangbanna miniature pig inbred line].

The polymorphism of 35 microsatellites in 5 lineages of Xishuangbanna small-ear miniature pig inbred line (XMI) was analysed. Nmuber of alleles of each lineage was counted, and rates of homozygote for 35 microsatellite loci in 5 lineages were calculated. According to gene frequencies of 35 microsatellites polymorphism information content (PIC) and mean heterozygosity were calculated for each lineage, and genetic distances between these lineage were estimated. The dendrograms were obtained based on genetic distances. The results suggest that rates of homozygote in these lineage are all high, and that is the highest in lineage 151. The results also suggest that polymorphism information content and mean heterozygosity in all the lineages are low. Composition of alleles of each lineage was quite different and the genetic relationship between lineages accorded with the process of inbred line. So it is suggested that the inbreeding degree of 5 lineages of XMI are all high, and the richness of genetic diversity is lower than general commercial pig breeds. It also shows each lineage has been different groups with individual genes.

[Anatomic, sialographic, histologic and ultrastructural studies of normal parotid gland of the miniature pig].

OBJECTIVE: To investigate the morphology of normal parotid gland of the miniature pig (minipig). METHODS: Anatomic, sialographic, histologic and ultrastructural studies of parotid glands were performed in 11 Chinese experimental minipigs. RESULTS: Sialograms showed a long main duct and a triangular shape gland. All branching ducts extended from the inferior-posterior of the main duct. No necessary glands were found. Typical serous acini were found microscopically. CONCLUSIONS: This study provides basic structural information of the parotid gland of the minipig. It is considered that minipig is a useful animal model in the investigation of parotid gland diseases.

[Pathological changes following retrograde infusion of methyl violet in the parotid gland of the miniature pig].

OBJECTIVE: To investigate the pathologic changes induced by infusion of 1% methyl violet in the parotid gland of the miniature pig (minipig). METHODS: Methyl violet (either 0.6 ml, 1.0 ml, or 4.0 ml) was injected into parotid glands of fifteen minipigs. The parotid glands were harvested for gross and histopathologic study after 1 week, 2 weeks, 1 month, 3 months and 6 months. RESULTS: The pathological changes induced by retrograde injection of methyl violet included necrosis or atrophy of acinar cells and fibrosis of the gland. The ductal system underwent atrophic changes, and thrombotic obliteration of main duct. CONCLUSION: Retrograde infusion of 1% methyl violet results in complete atrophy and fibrosis of the parotid gland in the minipig.

A new miniature catheter with side-holes for percutaneous transradial or transbrachial coronary angiography.

The percutaneous arm approach through the radial or brachial artery for diagnostic cardiac catheterization has advantages, such as a lower incidence of access-site complications and decreased patient discomfort, particularly when smaller diameter catheters are employed. However, the pressure produced by high-flow jets of contrast material exiting from an end-hole against the vascular wall can cause coronary dissection or myocardial blushing. To avoid this type of complication, we designed and developed a new miniature (4 French) catheter with two side-holes for coronary angiography. Under conditions similar to those of a clinical situation, we obtained the relationship between the pressure produced by the jets exiting from an end-hole and the force with which the tip of the catheter was pushed against the vascular wall. The presence of the side-holes substantially decreased the pressure at forces ranging between 0 and 15 gf, and at all injection rates (2.0 3.5 ml/second), but their location did not affect the pressure. In a preliminary clinical study, the new catheters with side-holes were feasible and effective for coronary angiography through the radial artery. Furthermore, the distribution of contrast material decreased the incidence of dislodgement of the catheter from the coronary ostia and enabled good visualization of the coronary ostia and proximal branches.

Aqueous infiltration into an implantable miniaturized telescope.

The implantable miniaturized telescope (IMT) is the first intraocular magnifying system proposed for optical correction in patients with age-related macular degeneration (ARMD). The optical component is embedded in a carrying device designed as an intraocular lens that is implanted after cataract surgery. In this study, we report findings on an IMT that was explanted because of aqueous infiltration into its optic and describe the configuration of this device and the surgical technique required for its implantation. The patient, a 75-year-old male with bilateral cataract and nonexudative ARMD, underwent phacoemulsification with implantation of an IMT in the right eye. The rigid device, weighing 46.1 mg in aqueous, has an overall diameter of 13.5 mm and requires implantation through a large limbal incision. It is fixated at the 6 to 12 o'clock meridian. Follow-up examination revealed the presence of numerous droplets inside the IMT optic. The device was explanted and sent to our center for evaluation. A large fissure was found on the carrying device. However, it was unlikely the site for aqueous infiltration. Microdefects at the level of the sealing of the optical cylinder appeared to provide the opening for the inflow of aqueous that formed droplets. Based on the findings of this report the manufacturer has modified the sealing technique to avoid this complication. Current clinical trials are now ongoing to assess the efficacy of this device in providing visual rehabilitation for ARMD patients.

[Therapeutic angiogenesis with the use of vascular endothelial growth factor 165 gene in the myocardium of miniature swine].

The purpose of the present study was to examine whether adenovirus-mediated vascular endothelial growth factor l65 (VEGFl65) can enhance collateral vessel formation of coronary artery and improve regional myocardial perfusion and function. Using a miniature swine model of chronic myocardial ischemia, the replication-deficient recombinant adenovirus vector containing complementary deoxyribonucleic acid (cDNA) for human VEGFl65 (Ad-VEGFl65) or for beta-galactosidase (Ad-Gal) was administered directly into the ischemic myocardium in the left circumflex (LCX) distribution. Myocardial perfusion and function were assessed by electrocardiogram-gated single photon emission computed tomography (SPECT) imaging and collateral vessel development of coronary artery was assessed by ex vivo coronary angiography (CAG). Four weeks after Ad-VEGF165 administration, SPECT imaging demonstrated a significant reduction in ischemic area (P<0.01) and ischemic severity (P<0.01), and a substantial improvement in left ventricular ejection fraction (P<0.01) and regional wall motion in the LCX distribution (P<0.05), as compared with that of control animals and that before administration of Ad-VEGFl65. Collateral vessel development with Rentrop Grading was also significantly greater in Ad-VEGF165 animals than in the Ad-Gal control animals (P<0.05). It's concluded that Ad-VEGFl65 can induce collateral vessel development in ischemic myocardium and result in significant improvement in myocardial perfusion and function.

Effect of nilvadipine on balloon catheterization-induced intimal thickening of the coronary artery in miniature pigs.

The effect of nilvadipine on balloon catheterization-induced intimal thickening of the coronary artery was examined in miniature pigs. A diffuse intimal thickening was observed in vehicle-treated controls 6 weeks after balloon catheterization. The histologic features of the intimal thickening resembled those of early atherosclerotic lesions and restenosis after percutaneous transluminal coronary angioplasty (PTCA). Nilvadipine given in a daily dose of 10 mg/kg (subcutaneously, s.c.) for 6 weeks significantly inhibited intimal thickening. The ratio of the cross-sectional area of the intima to the cross-sectional area of the media was significantly reduced by 62% in nilvadipine-treated animals. These results suggest that nilvadipine may prevent or exert beneficial effects on coronary arterial injuries such as atherosclerosis and restenosis after PTCA.

The potential use of fiber optics for detection in microchip separation and miniaturized flow-cell systems.

This article evaluates the potential of using fiber optics as a detection tool during electrophoretic separations or identification and quantification of analytes in miniaturized systems, including microchips. This paper covers reports in the literature from 1989 to June 2002. The articles have been chosen based on various aspects of the instrumentation including the microchip substrates, channel size and geometry, pathlength, fiber and detector types, light source, separation conditions, detection limits, and applications-all with the goal of comparing and contrasting optical detections. This review has been divided into three groups: fluorescence measurement-based studies, absorbance measurement-based studies, and innovative studies in chronological order.

Chronic catheterization of the inferior vena cava in Yucatan miniature swine.

Long-term venous access for leukapheresis, repeated blood sampling, and administration of drugs and fluids can be accomplished nonsurgically in Yucatan miniature swine. The catheter is placed under fluoroscopic guidance into the inferior vena cava using a needle and guidewire. This procedure has the advantage that it avoids a surgical incision, allows high flow rates, exists conveniently on the lower back, and can be replaced easily in the event of mechanical failure or thrombosis. Actuarial analysis of the duration of patency disclosed that of 41 catheters placed in 30 animals, the probability of function at 28, 42, and 54 days was 75%, 50%, and 25%, respectively. Eleven nonfunctioning catheters were replaced and nine of these continued to function until the completion of the experiment. No catheters were removed due to infection. Chronic catheterization of the inferior vena cava is a convenient method for long-term venous access in swine.

Characterization of a polymorphism of CD4 in miniature swine.

A polymorphism of CD4 in miniature swine has been identified by failure of cells from some animals to react with mAb 74-12-4. The phenotypic, molecular genetic, and functional characteristics of these animals have been defined. Cells from heterozygous animals bear approximately 50% the number of 74-12-4-reactive molecules on their surface as do cells from animals homozygous for the wild type. Animals of both phenotypes demonstrate similar flow cytometric profiles for CD8+ T cells. Northern blot analysis confirms the presence of mRNA for CD4 among PBL of animals failing to stain with 74-12-4. CD4 allelism is confirmed by Southern blot analysis, revealing RFLP. Function of the CD4 subset in vivo, as demonstrated by antibody production against a T cell-dependent Ag, is similar between animals of both phenotypes. Proliferative responses to PHA and alloantigen stimulation by a full haplotype mismatch or a class II mismatch alone are equivalent for animals of both phenotypes. These data suggest that the allelic form of CD4, designated CD4.2 in contrast to the wild-type CD4.1, is capable of performing normally as an accessory molecule in the generation of immune responses. Furthermore, antixenogeneic responses to C57B10.BR were equivalent, suggesting that both CD4 molecular types may be capable of interacting with xenogeneic class II molecules. Although the polymorphism includes differences in exons 3 and 4, regions thought to encode portions of the molecule interacting with MHC class II, these results imply that this naturally occurring CD4 polymorphism does not affect the interaction with class II molecules.
