Declining survival probability threatens the North Atlantic right whale.

The North Atlantic northern right whale (Eubalaena glacialis) is considered the most endangered large whale species. Its population has recovered only slowly since the cessation of commercial whaling and numbers about 300 individuals. We applied mark-recapture statistics to a catalog of photographically identified individuals to obtain the first statistically rigorous estimates of survival probability for this population. Crude survival decreased from about 0.99 per year in 1980 to about 0.94 in 1994. We combined this survival trend with a reported decrease in reproductive rate into a branching process model to compute population growth rate and extinction probability. Population growth rate declined from about 1. 053 in 1980 to about 0.976 in 1994. Under current conditions the population is doomed to extinction; an upper bound on the expected time to extinction is 191 years. The most effective way to improve the prospects of the population is to reduce mortality. The right whale is at risk from entanglement in fishing gear and from collisions with ships. Reducing this human-caused mortality is essential to the viability of this population.

Mass spectrometric contributions to the practice of phosphorylation site mapping through 2003: a literature review.

Reversible phosphorylation of proteins is among the most important post-translational modifications, and elucidation of sites of phosphorylation is essential to understanding the regulation of key cellular processes such as signal transduction. Unfortunately phosphorylation site mapping is as technically challenging as it is important. Limitations in the traditional method of Edman degradation of (32)P-labeled phosphoproteins have spurred the development of mass spectrometric methods for phosphopeptide identification and sequencing. To assess the practical contributions of the various technologies we conducted a literature search of publications using mass spectrometry to discover previously unknown phosphorylation sites. 1281 such phosphorylation sites were reported in 203 publications between 1992 and 2003. This review examines and catalogs those methods, identifies the trends that have emerged in the past decade, and presents representative examples from among these methods.

Documenting the prominence of forensic neuropsychology at national meetings and in relevant professional journals from 1990 to 2000.

Numerous authors have opined that forensic activities have become more prominent within clinical neuropsychology. To investigate the merits of these claims, the entire contents of Archives of Clinical Neuropsychology (ACN), Journal of Clinical and Experimental Neuropsychology (JCEN), and The Clinical Neuropsychologist (TCN) from 1990 through 2000 were reviewed and cataloged. These three journals were selected because they are the highest-ranking clinical subscription journals according to surveys of neuropsychology practitioners. Prior to rating journal content, various categories of interest were delineated and practice ratings were obtained until the two raters reached 92% agreement. Each of the raters read the journal contents and recorded content ratings for half of the journal issues under review. Results of the 8323 ratings demonstrated increases across time in the absolute numbers of articles related to forensic neuropsychology, although variable and different for each journal. Published articles that were partially or substantially forensic in nature in the three journals combined increased from 4% in 1990 to 14% in 2000. An annual peak in absolute number (n=32;16%) of forensic journal articles occurred in 1997. The most common topic of 139 articles published in ACN, JCEN, and TCN from 1990 to 2000 was malingering, which appeared in 86% of the general forensic articles. Forensic presentations at annual NAN meetings ranged from 3.9 to 11.3% (M=8%) of the convention programs, whereas within Division 40's programs at the American Psychological Association meeting, the average percentage ranged from 2.3 to 11.7% (M=6%). Results pertaining to each journal and to specific forensic topics are presented and implications of these and other results are discussed.

Organization and functioning of ISCEH, The International Society for Clinical and Experimental Hypnosis.

The first international society of hypnosis founded in this century, the International Society for Clinical and Experimental Hypnosis (ISCEH), was organized in 1958 as a direct result of societal conflicts between The Society for Clinical and Experimental Hypnosis (SCEH) and the American Society of Clinical Hypnosis (ASCH). Initially, it consisted of National Divisions built around key contributors in 30 different countries. This article describes the inception, organization, and development (including controversies and conflicts) of ISCEH up to 1973. At that time, through the cooperation of SCEH, ASCH, and other international groups, under the leadership of Dr. Ernest R Hilgard, ISCEH was reorganized, reconstituted, and renamed The International Society of Hypnosis (ISH). For the purpose of historical study, the society's voluminous correspondence, directories of officers and committees, awards, photographs, programs of congresses, election records, and other relevant documents have been cataloged and preserved in some 82 files in the Archives of the History of American Psychology at the University of Akron, Akron, Ohio.

A review of plant-derived and herbal approaches to the treatment of sexual dysfunctions.

Despite the increasing availability of effective conventional medical treatments, plant-derived and herbal remedies continue to provide a popular alternative for men and women seeking to improve their sex life. Nevertheless, the efficacy of most herbal agents in treating sexual problems remains uncertain. Therapists and consumers alike would benefit from an increased understanding of commonly used herbal agents on the market, their purported or supported effects, and their potential side effects. To this end, we cataloged the major prosexual herbal agents currently sold in several representative health food stores. We also specify the sexual problem purportedly ameliorated by each herbal agent. Finally, we evaluate eight herbal agents commonly promoted for the treatment of sexual problems. This evaluation includes a review of the research supporting the use, efficacy, dose, adverse effects, contraindications, and possible mechanism of action of each. We conclude by commenting on the quality of current research, pointing out gaps in our knowledge, and noting the need for rigorous research and product control to adequately address questions regarding the efficacy of these agents.

Spatial, sequence-order-independent structural comparison of alpha/beta proteins: evolutionary implications.

We present a unique sequence-order independent approach which allows examination of three dimensional structures, searching for spatially similar substructural motifs. If the amino acids composing the motifs are contiguous in the primary chain, that is, they follow each other in the sequence, a common ancestor and a divergent evolutionary process may be implied. On the other hand, if the three-dimensional substructural motif consists of amino acids whose positions in the sequences vary between the different proteins, a convergent evolution might have taken place. Starting from different, ancient sequences, mutations may have occurred that brought about formation and conservation of a truly structural motif. Such a motif might be particularly suitable for fulfilling a specific function. Clearly, in order to be able to carry out such a task one needs a technique which allows comparisons of protein structures absolutely independent of their amino acid sequence-order. Our novel, efficient, computer vision based technique treats atoms (residues) as unconnected points in space, using strictly the atomic (either all atoms or only the C alpha atoms) coordinates. The order of the residues is completely disregarded. Detection, cataloging and analysis of "real" three-dimensional, sequence-order independent motifs in the crystallographic database is expected to be an invaluable tool for protein folding. Here we demonstrate the power of the technique by applying it to alpha/beta proteins. Our studies indicate that for some of the proteins, the "classical" structural alignments (conserving the amino acid order) are the optimal ones. Nevertheless, for others, truly spatial (out of sequential-order) amino acid equivalencing results in a better geometrical match.

What's the use in getting mad? Anger and instrumentality in women's relationships.

In two studies, we examined women's anger expression and its instrumental function in relationships by addressing the following questions: What is the relationship between women's self-reports of instrumentality and their perceived styles of anger expression? In what ways and situations do women see their anger expression as instrumental or goal enhancing? In Study I, we expected that women's perceived styles of anger expression would be positively related to instrumentality, as measured with the Personal Attributes Questionnaire (PAQ; Spence, Helmreich, & Stapp, JSAS Catalog of Selected Documents in Psychology, 4, 43, 1974). Although our hypothesis was not supported, a positive relationship did emerge between assertiveness and instrumentality, as predicted. In Study II, we conducted three focus group discussions to elucidate women's experiences of anger and to provide clarification for the results of Study I. We identified group themes related to when women experienced their anger expression as instrumental as well as when women perceived themselves as noninstrumental in anger-arousing situations. In their narratives, women explained how they make decisions about expressing their anger based on relationship concerns.

Pesticides distribution in sediments of a tropical coastal lagoon adjacent to an irrigation district in northwest Mexico.

This work presents the content of organochlorine (OCs) pesticides in sediments of both, agricultural drains from the Irrigation District 076, El Carrizo, Sinaloa, and of the adjacent coastal lagoon ecosystem of Agiabampo-Bacorehuis-Jitzamuri, located in northwest Mexico. A questionnaire was applied to the farmers of this irrigation district, to evaluate the usage of pesticides and to determine if banned OCs are still used. Around 31 tons of active ingredients were applied in the autumn-winter cycle (1997-1998). There were eighteen different pesticides; of them 61% are insecticides, 28% herbicides, 6% fungicides and 6% other compounds. Around 73% of the pesticides applied are organophosphorus, carbamates and sulfur, OCs are still applied and represent 4% of the total. All the pesticides applied in the irrigation district are authorized in the Official Pesticide Catalog, nevertheless forbidden and restricted compounds were detected in the sediment samples. The presence of heptachlor, heptachlor epoxide, alpha-endosulfan, beta-endosulfan, endosulfan sulfate, alpha-HCH, beta-HCH, lindane, delta-HCH, aldrin, dieldrin p,p'-DDT, p,p'-DDE and p,p'-DDD was recorded in the sediment sample. The analysis of the sediments suggest that the principal pesticides input to the coastal ecosystem is mainly through three of the seven agricultural drains, that collect the excess irrigation water and runoff from the district surface. A gradient of pesticide residues in sediments exists from the agricultural drains (52 ng OCs g(-1) dry weight) to the Jitzamuri bay (32 ng OCs g(-1) dry weight).

Analysis of methamphetamine-induced changes in the expression of integrin family members in the cortex of wild-type and c-fos knockout mice.

Methamphetamine (METH) is an illicit drug that is also neurotoxic. Recent studies suggest that in addition to dopamine terminal degeneration in the striatum, METH causes apoptosis in cortical neurons. Earlier, we showed that c-fos knockout mice are more susceptible to the toxic effects of the drug. In order to identify possible pathways related to these differences, we have used cDNA array that provided us with a comprehensive catalog of METH affected genes. In the present study, we report on the effects of METH on the integrin family members that were shown to be involved in intracellular signaling cascades effecting cell survival. We found that, in comparison to wild type animals, c-fos knockout mice have lower baseline levels of the integrins in the cortex. Moreover, METH caused time-dependent decreases in their transcripts in both strains of mice. Quantitative RT-PCR confirmed the changes obtained in cDNA array. These findings are discussed in view of the possible role of integrins in METH-induced toxic effects on the cortical neurons.

Accessible microscopy workstation for students and scientists with mobility impairments.

An integrated accessible microscopy workstation was designed and developed to allow persons with mobility impairments to control all aspects of light microscopy with minimal human assistance. This system, named AccessScope, is capable of performing brightfield and fluorescence microscopy, image analysis, and tissue morphometry requisite for undergraduate science courses to graduate-level research. An accessible microscope is necessary for students and scientists with mobility impairments to be able to use a microscope independently to better understand microscopical imaging concepts and cell biology. This knowledge is not always apparent by simply viewing a catalog of histological images. The ability to operate a microscope independently eliminates the need to hire an assistant or rely on a classmate and permits one to take practical laboratory examinations by oneself. Independent microscope handling is also crucial for graduate students and scientists with disabilities to perform scientific research. By making a personal computer as the user interface for controlling AccessScope functions, different upper limb mobility impairments could be accommodated by using various computer input devices and assistive technology software. Participants with a range of upper limb mobility impairments evaluated the prototype microscopy workstation. They were able to control all microscopy functions including loading different slides without assistance.

Evaluation of the rate, location, and morphology of perforations in surgical gloves worn in urological operations.

Surgical perforation or intrinsic defects within the polymer membranes of surgical gloves are responsible for an increased exposure, and consequently infection risk, to HIV and HCV or other infectious diseases for the medical healthcare professional. A prospective field study was undertaken to evaluate the number and location of glove perforations, tested by the waterfill test. The morphology of these perforations was then inspected by stereo light and scanning electron microscopy. By comparing in-use defects to simulated defects created by medical instruments in unused gloves it was possible to categorize perforations caused by intrinsic defects or extrinsic mechanical stress. In six different types of surgical gloves collected from 720 urological operations the waterfill test revealed leakage rates of 6.6 percent to 12.3 percent. Further assessment of these gloves revealed intrinsic polymer material defect rates of 0.2 percent to 3.3 percent. The in-use acquired instrumental and mechanical perforation rate ranged from 4.6 percent to 12.15 percent. Perforations were mostly located at the index finger (left > right) or the thumbs. The morphology of the intrinsic defects was visualized as either incorporated membranous air bubbles or granular areas. The simulation of defects by surgical instruments in unused gloves resulted in the production of a catalog for comparison with defects seen in used gloves. This knowledge of the location and cause of these perforations highlights the need for a better avoidance strategy and/or establishment of a schedule for routine glove change by surgeons. In addition, the intrinsic polymer film quality of gloves can be improved by a lower incidence of air bubbles together with less granular sites.

Collection and preparation of molecular databases for virtual screening.

Drug discovery and development research is undergoing a paradigm shift from a linear and sequential nature of the various steps involved in the drug discovery process of the past to the more parallel approach of the present, due to a lack of sufficient correlation between activities estimated by in vitro and in vivo assays. This is attributed to the non-drug-likeness of the lead molecules, which has often been detected at advanced drug development stages. Thus a striking aspect of this paradigm shift has been early/parallel in silico prioritization of drug-like molecular databases (also database pre-processing), in addition to prioritizing compounds with high affinity and selectivity for a protein target. In view of this, a drug-like database useful for virtual screening has been created by prioritizing molecules from 36 catalog suppliers, using our recently derived binary QSAR based drug-likeness model as a filter. The performance of this model was assessed by a comparative evaluation with respect to commonly used filters implemented by the ZINC database. Since the model was derived considering all the limitations that have plagued the existing rules and models, it performs better than the existing filters and thus the molecules prioritized by this filter represent a better subset of drug-like compounds. The application of this model on exhaustive subsets of 4,972,123 molecules, many of which have passed the ZINC database filters for drug-likeness, led to a further prioritization of 2,920,551 drug-like molecules. This database may have a great potential for in silico virtual screening for discovering molecules, which may survive the later stages of the drug development research.

Sequence-based species delimitation for the DNA taxonomy of undescribed insects.

Cataloging the very large number of undescribed species of insects could be greatly accelerated by automated DNA based approaches, but procedures for large-scale species discovery from sequence data are currently lacking. Here, we use mitochondrial DNA variation to delimit species in a poorly known beetle radiation in the genus Rivacindela from arid Australia. Among 468 individuals sampled from 65 sites and multiple morphologically distinguishable types, sequence variation in three mtDNA genes (cytochrome oxidase subunit 1, cytochrome b, 16S ribosomal RNA) was strongly partitioned between 46 or 47 putative species identified with quantitative methods of species recognition based on fixed unique ("diagnostic") characters. The boundaries between groups were also recognizable from a striking increase in branching rate in clock-constrained calibrated trees. Models of stochastic lineage growth (Yule models) were combined with coalescence theory to develop a new likelihood method that determines the point of transition from species-level (speciation and extinction) to population-level (coalescence) evolutionary processes. Fitting the location of the switches from speciation to coalescent nodes on the ultrametric tree of Rivacindela produced a transition in branching rate occurring at 0.43 Mya, leading to an estimate of 48 putative species (confidence interval for the threshold ranging from 47 to 51 clusters within 2 logL units). Entities delimited in this way exhibited biological properties of traditionally defined species, showing coherence of geographic ranges, broad congruence with morphologically recognized species, and levels of sequence divergence typical for closely related species of insects. The finding of discontinuous evolutionary groupings that are readily apparent in patterns of sequence variation permits largely automated species delineation from DNA surveys of local communities as a scaffold for taxonomy in this poorly known insect group.

Assessing internal tobacco industry knowledge of the neurobiology of tobacco dependence.

The recent availability of internal tobacco industry documents provides a significant resource for evaluating industry understanding of the pharmacological, psychosocial, and behavioral mechanisms underlying tobacco dependence. In this study, we catalog the range of efforts undertaken by tobacco manufacturers seeking knowledge of these mechanisms. Some areas of industry research, such as cellular and molecular studies of nicotine and its effects, are widely available in the open literature. Of greater interest are internal research projects that have demonstrated direct influence on product development. These include studies of smoker psychology and behavior, evoked-response studies of tobacco-delivered nicotine, the effects of sensory perception, dose-related effects, and the development of nicotine analogs and synergists. Our findings suggest extensive industry knowledge of mechanisms that determine smoker perception and behavior, and application of this knowledge in product development, including control of sensory response, uptake of nicotine, and product effects. Independent research recently has begun to consider the contributions of tobacco product ingredients and design factors to the determination of risk, severity, and prevalence of addiction. However, the application of these findings to cessation and treatment efforts is still quite limited. We conclude that clinical research would greatly benefit from further examination of the decades of knowledge accumulated by tobacco manufacturers.

Disease status and use of ventilatory support by ALS patients. BDNF Study Group.

INTRODUCTION: Use of mechanical ventilation (MV), administered either invasively via tracheostomy, or more commonly non-invasively (CPAP, BiPAP), appears to be increasing in ALS. No prospective databases exist that describe the behavior of physicians and patients and the criteria for instituting MV in ALS. METHODS: 387 placebo patients in a Phase III trial of r-metHuBDNF were followed for 9 months. Although the use of MV was not the primary end-point of the trial, information was gathered regarding it by cataloging respiratory adverse events and tracking health resource utilization. RESULTS: 35 of 387 patients utilized MV during the trial. Twenty-eight (7%) patients received BiPAP. Seven (2%) were tracheotomized without first receiving BiPAP. Forced vital capacity (FVC): BiPAP patients had a mean ( SEM) FVC% of 71.8 +/- 2.8% and ALSFRS of 27.7 +/- 1.0 at baseline; non-BiPAP patients had a mean baseline FVC% of 88.7 +/- 1.0%, and an ALSFRS of 30.3 +/- 0.3. Symptom duration at entry was similar for both groups (2.1 +/- 0.4 years vs. 2.1 +/- 0.1 years). At the time of first use of BiPAP, average FVC% was 47.5 +/- 4.0% and ALSFRS score was 22.4 +/- 1.5. The range of FVC% at start of BiPAP was 15-87. The nine-month survival was 67.9% for BiPAP patients vs. 86% for non-BiPAP patients. The use of BiPAP varied tremendously among the 38 study sites, with some not employing it at all and others using it in as many as 40% of patients. CONCLUSIONS: Of the 9% of placebo patients who received MV, BiPAP patients were more rapidly progressing than non-BIPAP patients, and showed a greater eventual mortality rate. Patients began MV at a wide range of values of FVC%, and centers differed in their prescribing practices. Factors influencing BiPAP use are complex, and not strictly related to FVC%.

Tri (2-chloroisopropyl) phosphate--an unexpected organochlorine contaminant in some charcoal air-sampling sorbent tubes.

Air sampling in a government building was necessary in response to reports of a cancer cluster. SKC (Eighty Four, Pa.) charcoal coconut shell-based sorbent tubes (226-01 lot 120) were recommended for this procedure. A recently purchased supply was present at the University of British Columbia and consequently was used for this particular study. Analysis of the front charcoal section showed the presence of a flame retardant, tri (2-chloroisopropyl) phosphate, which was confirmed by gas liquid chromatography (GLC) and mass spectrometry analysis. In an effort to identify the source of this fire retardant in the building, it became apparent from the analysis done on unknown field blanks that tri (2-chloroisopropyl) phosphate was a contaminant of the sorbent tubes used. Analysis of additional blank tubes identified the foam separators as the most likely source of contamination. Levels of tri (2-chloroisopropyl) phosphate in the front charcoal section ranged from 1.3 to 5.9 micrograms. The foam separator contained between 11.4 and 16.5 micrograms, and the backup charcoal section contained between 14.5 and 24.0 micrograms of tri (2-chloroisopropyl) phosphate. In addition, another flame retardant, tri (1,3 dichloro-2-propyl) phosphate was also found. Because these contaminants have long column retention times in GLC, it may not be apparent that these contaminants are present and consequently are likely to have modified the sorbent characteristics of the activated charcoal. Another batch of sorbent tubes bearing the same catalog number and lot number was purchased from the supplier; no flame retardants were found in this batch.(ABSTRACT TRUNCATED AT 250 WORDS)
