Immobilisation of electroactive macrocyclic complexes within titania films.

The 4-carboxyphenyl-appended macrocyclic ligand trans-6,13-dimethyl-6-((4-carboxybenzyl)amino)-1,4,8,11-tetraazacyclotetradecane-6-amine (HL10) has been synthesised and complexed with Co(III). The mononuclear complexes [Co(HL10)(CN)]2+ and [CoL10(OH)]+ have been prepared and the crystal structures of their perchlorate salts are presented, where the ligand is bound in a pentadentate mode in each case while the 4-carboxybenzyl-substituted pendent amine remains free from the metal. The cyano-bridged dinuclear complex [CoL10-mu-NC-Fe(CN)5]2- was also prepared and chemisorbed on titania-coated ITO conducting glass. The adsorbed complex is electrochemically active and cyclic voltammetry of the modified ITO working electrode in both water and MeCN solution was undertaken with simultaneous optical spectroscopy. This experiment demonstrates that reversible electrochemical oxidation of the Fe(II) centre is coupled with rapid changes in the optical absorbance of the film.

Dimetallic complexes of a structurally versatile pyridazine-containing Schiff-base macrocyclic ligand with pendant pyridine arms.

The new [2 + 2] Schiff-base macrocyclic ligand L2, containing pyridazine head units and pyridine pendant arms, was synthesised as [Ba(II)2L2(ClO4)4(OH2)] 1 from the barium(II) ion templated condensation reaction of 3,6-diformylpyridazine and N1-(2-aminoethyl)-N1-(methylene-2-pyridyl)-ethane-1,2-diamine. Subsequent transmetallation reactions of 1 with copper(II), iron(II) and manganese(II) perchlorates led to the formation of [Cu(II)2L2](ClO4)4.2MeCN 2, [Fe(II)2L2(MeCN)2](ClO4)4 3 and two manganese complexes, 4 and 5, with the same formula, [Mn(II)2L2(MeCN)(OH2)](ClO4)4, but slightly different crystal structures, respectively. Single-crystal X-ray structural analyses reveal the variety of structures which can be supported by L2 in order to meet the coordination environment preferences of the incorporated metal ions. The barium(II) ions in 1 have an irregular ten-coordinate geometry whereas the copper(II) ions in 2 have a square pyramidal geometry and the iron(II) ions in 3 have an octahedral geometry, while in 4 and 5 every manganese(II) ion is seven-coordinate and the environment can be best described as distorted pentagonal bipyramidal. In 1, 4 and 5 the pyridazine moieties bridge the metal centres [Ba(1)...Ba(2) 4.9557(3)A 1; Mn(1)...Mn(2) 4.520(1)A 4; Mn(1)[dot dot dot]Mn(2) 4.3707(8)A 5] but this is not observed in the copper(II) and iron(II) complexes, 2 and 3, in which the metal ions are well separated [Cu(1)...Cu(2) 5.9378(6)A 2; Fe(1)...Fe(2) 5.7407(12)A 3]. In the cyclic voltammogram of [Cu2(II)L2](ClO4)4.2MeCN 2 in MeCN vs. Ag/AgCl two separate reversible one-electron transfer steps are observed [E(1/2)=0.04 V, DeltaE= 0.12 V and E(1/2)= 0.20 V, DeltaE=0.12 V; K(c)=510; in this system E(1/2)(Fc+/Fc)=0.42 V and DeltaE(Fc+/Fc)=0.08 V]. The other complexes cannot be reversibly reduced/oxidised.

Donor effect on supramolecular structures of silver(I) perchlorate complexes of macrocycles with O2S2X (X=S, O and NH) donor sets.

By virtue of simple one donor variations of O(2)S(2)X-donor macrocycles (L(1): X=S, L(2): X=O and L(3): X=NH) towards silver(I) perchlorates, four supramolecular complexes (1-3) with different topologies have been isolated; L(1) afforded solvent- and anion-free sandwich complex (1), otherwise L(2) and L(3) gave the dimeric (2a), 1-D polymeric (2b), and tetrameric bowl-type (3) complexes with solvent or anion coordination.

Basicity and coordination properties of a new phenanthroline-based bis-macrocyclic receptor.

The synthesis and characterisation of the new macrocyclic ligand 6-methyl-2,6,10-triaza-[11]-12,25-phenathrolinophane (L1), which contains a triamine aliphatic chain linking the 2,9 positions of 1,10-phenanthroline and of its derivative L2, composed by two L1 moieties connected by an ethylenic bridge, are reported. Their basicity and coordination properties toward Cu(II), Zn(II), Cd(II), Pb(II) and Hg(II) have been studied by means of potentiometric and spectroscopic (UV-Vis, fluorescence emission) measurements in aqueous solutions. L1 forms 1:1 metal complexes in aqueous solutions, while L2 can give both mono- and dinuclear complexes. In the mononuclear L2 complexes the metal is sandwiched between the two cyclic moieties. The metal complexes with L1 and L2 do not display fluorescence emission, due to the presence of amine groups not involved in metal coordination. These amine groups can quench the excited fluorophore through an electron transfer process. The ability of the Zn(II) complexes with L1 and L2 to cleave the phosphate ester bond in the presence has been investigated by using bis(p-nitrophenyl)phosphate (BNPP) as substrate. The dinuclear complex with L2 shows a remarkable hydrolytic activity, due to the simultaneous presence within this complex of two metals and two hydrophobic units. In fact, the two Zn(II) act cooperatively in substrate binding, probably through a bridging interaction of the phosphate ester; the interaction is further reinforced by pi-stacking pairing and hydrophobic interactions between the phenanthroline unit(s) and the p-nitrophenyl groups of BNPP.

Efficient stabilization of copper(III) in tetraaza pseudo-macrocyclic oxime-and-hydrazide ligands with adjustable cavity size.

Substitution of the amide donors in open-chain {2N(oxime), 2N(amide)} ligands by hydrazide donors gives new pseudo-macrocyclic copper complexes that show a significant decrease of the Cu(3+/2+) redox potentials in both mono- and polynuclear systems, thus demonstrating a pronounced capacity of such ligand systems to efficiently stabilize the trivalent copper.

Synthesis and characterisation of dimeric eight-coordinate lanthanide(III) complexes of a macrocyclic tribenzylphosphinate ligand.

The macrocyclic ligand 1,4,7,10-tetraazacyclododecane-1,4,7-triyl(methylenebenzyl-phosphinic acid) H3L3, has been prepared and its complexes with Eu, Gd and Tb(III) studied by NMR, relaxometry, luminescence and single crystal X-ray crystallography. In solution and in the crystal, the complexes have eight-coordinate metal centres with bridging phosphinate groups linking the two twisted square antiprismatic coordination polyhedra. A single stereoisomer crystallises from solution with an RRR and SSS configuration at the P centres in each sub-unit. The relaxivity of [GdL3]2 is low (1.9 mM-1 s-1, 298 K, 20 MHz), consistent with the absence of any proximate water molecules. The terbium dimer possesses a relatively long excited state lifetime (2.47 ms, 298 K).

Evaluation of the antiviral activity against Junin virus of macrocyclic trichothecenes produced by the hypocrealean epibiont of Baccharis coridifolia.

Four macrocyclic trichothecenes, roridin A, roridin E, verrucarin A and verrucarin J, produced by the hypocrealean epibiont of Baccharis coridifolia, were evaluated for their inhibitory activity against the arenavirus Junin (JUNV), the etiological agent of Argentine hemorrhagic fever. The trichothecenes achieved a dose-dependent inhibition of JUNV multiplication at concentrations not affecting cell viability. The 50 % inhibitory concentration (IC50) values determined by a virus yield inhibition assay were in the range 1.2 - 4.9 ng/ml. The most active compound was verrucarin J which reduced JUNV yield more than 2 log units and had a similar effect against the arenavirus Tacaribe. The trichothecenes lacked virucidal effects on JUNV virions. From time of addition and removal experiments, it can be concluded that verrucarin J inhibited a late stage in the replicative cycle of JUNV, after 5 h of adsorption.

New macrocyclic diterpenoids from Euphorbia esula.

The structures of two new macrocyclic jatrophane diterpenoid esters from the whole herb of Euphorbia esula, were established as 11,14-epoxy-3beta,5alpha,7beta,8alpha,9alpha,15beta-hexaacetoxy-12-oxo-13alphaH-jatropha-6(17)-ene (1) and 1alpha,3beta-diacetoxy-5alpha,7beta-dibenzoyloxy-9,14-dioxo-11beta,12alpha-epoxy-2alpha,8alpha,15beta-trihydroxy-13betaH-jatropha-6(17)-ene (2) by a combination of 1D- and 2D-NMR techniques as well as UV, IR and mass spectral data. Bioassay evaluation of all isolates against the human tumor cell lines (B16, KB, SMMC and BGC) indicated that ester 2 was cytotoxic to B16 with the IC50 value being 1.81 microg/ml. In addition, the irritant activity assay indicated that both diterpenoids were inactive (ID(24)50 > 100 microg/ear).

Isolation of a new 15-membered macrocyclic glycolipid lactone, Cuscutic Resinoside a from the seeds of Cuscuta chinensis: a stimulator of breast cancer cell proliferation.

While searching for new estrogenic compounds from the plant kingdom, we investigated an extract of the seeds of Cuscuta chinensis (Convolvulaceae) which showed potency for stimulating MCF-7 cell proliferation. A novel resin glycoside, cuscutic resinoside A ( 6) was isolated along with five known compounds from the extract. The structure was deduced from its spectral data as (11 S)-hydroxyhexadecanoic acid 11- O-alpha- L-(4- O-2 R,3 R-nilylrhamnopyranosyl)-(1-->2)- O-alpha- L-rhamnopyranosyl-(1,2-lactone) forming a unique 15-membered macrocyclic lactone. The compound significantly stimulated not only MCF-7 cell proliferation but also T47D human breast cancer cells at a concentration of 10 microM. Along with cuscutamine ( 1) and kaempferol ( 4), 6 was tested in the transient luciferase reporter assay and was found to have different luciferase inducing activity characteristics from the other compounds. These results suggest that 6 stimulated cancer cell proliferation by a different mechanism from 1 and 4.

Catalysis-based enantioselective total synthesis of the macrocyclic spermidine alkaloid isooncinotine.

A concise and efficient total synthesis of the spermidine alkaloid (-)-isooncinotine (1) incorporating a 22-membered lactam ring is outlined. The approach is largely catalysis-based, involving a selective iron-catalyzed alkyl-aryl cross-coupling reaction of a difunctionalized pyridine substrate, a heterogeneous asymmetric hydrogenation step to set the chiral center of the target, and a highly integrated ring-closing metathesis/hydrogenation sequence to forge the saturated macrocyclic edifice in a single operation.

Biomimetic macrocyclic receptors for carboxylate anion recognition based on C-linked peptidocalix[4]arenes.

Two neutral macrobicyclic anion receptors 4 and 6, containing a calix[4]arene in the cone conformation, two l-alanine units, and a 2,6-diacylpyridine or a phthaloyl bridge, are described. The x-ray crystal structure of the acetone complexes of the pyridine containing macrocycle 6 shows the four amide NH groups to be in close proximity to the chiral pocket delimited by the pyridine and one aromatic nucleus of the calix[4]arene. This conformation is also the most stable in acetone-d6 solution, as proven by one- and two-dimensional NMR spectral measurements. Electrospray ionization-MS and (1)H NMR experiments reveal that the two ligands strongly bind carboxylate anions in acetone solution. H-bonding interactions between the carboxylate anions and the amide NH groups, together with pi/pi stacking, are invoked to explain the efficiency and the selectivity of these anion receptors.

A macrocyclic zinc(II) fluorophore as a detector of apoptosis.

Our originally designed dansylamidoethylcyclen 4 as a biomimetic Zn(2+)-selective fluorophore has been demonstrated to be a good detector of the apoptosis (induced by an anticancer agent, etoposide, and H(2)O(2)) in cancer cells such as HeLa and HL60 cells. The macrocyclic Zn(2+) ligand 4 (mostly as a deprotonated form) is cell-permeable to show weak fluorescence (emission at 550 nm), which forms a strong fluorescent 1:1 Zn(2+) complex 5 (emission at 530 nm) when Zn(2+) is incorporated into the cells by a zinc(II) ionophore pyrithione. Thus formed, Zn(2+) complex 5 is cell-impermeable and remains intact over a few hours. When apoptosis in HeLa or HL60 cells is artificially induced, 4 selectively and strongly stains apoptotic cells only at early stages, which was verified by using the conventional apoptosis detection probe annexin V-Cy3. Detection of the apoptotic cells by 4 was perhaps due to significantly increased free Zn(2+) flux at early stages of apoptosis. Apoptotic detection by 4 has been compared with a presently available Zn(2+) fluorophore, Zinquin 1. We present that 4 has advantages in detection of apoptosis over annexin V-Cy3 and Zinquin 1.
